Two bacterial strains, Bacillus licheniformis YB-1413 and YB-1414, producing extracellular α-galactosidase, were obtained from homemade Doenjang. On the basis of their biochemical properties, 16S rRNA sequences and random amplified polymorphic DNA patterns by polymerase chain reaction, they were found to be somewhat different from one another. α-Galactosidase productivities of the two isolates were increased by wheat bran, but drastically decreased by melibiose, raffinose and sucrose which were used as carbon sources. The enzyme productivities were increased by yeast extract as a nitrogen source with maximum levels of 1.87 U/ml for YB-1413 and 1.69 U/ml for YB-1414, respectively. The enzymes of both isolates exhibited maximum activity for hydrolysis of para-nitrophenyl-α-D-galactopyranoside (pNP-αGal) under reaction conditions of pH 6.0 and 45℃. Their hydrolyzing activities for pNP-αGal were drastically decreased by the addition of low concentrations of ribose and galactose. They were capable of hydrolyzing completely α-1,6 linked galactosyl residue in melibiose, raffinose and stachyose, which are known to be anti-nutritional factors in products of soybean and legume. In relation to the latter, the isolates YB-1413 and YB-1414 have potential applicability in improving soybean-fermented foods and the nutritional value of soybean feed.
This study will examine the current condition of outdoor play in child care centers and the overall perception of teachers on outdoor play in regards to its related programs and children's reaction to them. Through examining such issues, this study will provide the basic data that can be applied in planning appropriate outdoor play activities, programs and environments. For the study, we surveyed teachers and directors of child care centers using a structured questionnaire. The objects of the survey were 22 child care centers in Daejeon and Seoul, who agreed to conduct the survey. The survey was conducted from August 1 to September 30 2006; 220 copies were distributed, and 188 copies were collected. The results of the study can be summarized as follows: 1) The number of outdoor plays per week were on average 1~2 in most cases, although teachers believed the play should be done everyday. Outdoor play time was mostly 21~30 minutes, while teachers thought 31~40 minutes as appropriate. Children's reaction scored the highest when they played outdoors for 30 minutes every day with their teachers involved in the play. As a result, we have concluded that the appropriate outdoor play should be conducted everyday for 30~40 minutes. 2) Outdoor play was mostly conducted as part of the overall educational plan rather than a separate plan. Since outdoor play affects children's development as much as indoor play, there is a need for development of a specific and separate plan for various outdoor play programs. 3) The outdoor play program that was most often conducted was equipment play and sand play. This indicated the monolithic nature of the outdoor program. Given the fact that children enjoy outdoor plays a lot (M=4.71), various programs including adventure play, sensitive play, and carpenter play should be developed. 4) In outdoor play, the teacher's role included supervising and interacting with children. According to the survey, teachers had a good understanding of the importance on safety and supervising issues in outdoor plays. However they seemed to overlook the importance of having better programs and improving teacher's participation to the play. Despite the importance of outdoor play to children's development, the result shows that teachers have tendencies to limit and obstruct outdoor plays by emphasizing supervising and safety issues instead. 5) Children's safety should not be the only consideration for outdoor plays; teachers ought to better understand that children's development can be maximized by the play. Furthermore, there should be emphasis placed on the program development and teacher's participation; teacher's participation should actively encourage the interactions between a child and a teacher, as well as among children.

The use of pile reinforcement is considered as one of the most promising techniques for improving load carrying capacity of piles in offshore area. In this study, to consider the horizontal and uplift bearing capacity of submerged breakwater bearing pile, exclusive analysis on load-transfer behaviour of pile was conducted. First of all, check the reinforcing effect from the three-dimensional finite element method, and estimate load transfer curve (ground reaction force). Based on these results, the reinforcing effect was quantified by estimating the coefficients of horizontal and uplift reinforcement of reinforced piles. Load transfer function with consideration of the reinforcing effect was proposed from estimated coefficients. A comparison of the analysis using the proposed load transfer function with three-dimensional finite element analysis has resulted that the proposed load transfer function is displaying good accuracy of predicting behavior of the load transfer between the pile and soil reinforcement. Interpretation of the submerged structure by applying a load transfer function considering the reinforcing effect, has shown that the reinforced pile's shear, bending moment and displacement are less than that of non-reinforced piles, while the subgrade reaction modulus arises greater. Thus, it is expected to be relatively cost effective in terms of design.

The following results are from the test of semi-gel electrolyte to store energy efficiently and use advanced VRLA batteries by photovoltaic and wind power generation. Semi-Gel electrolyte with Silica 5% became Gel after 1 and half hour. It shows it is the most suitable time that the electrolyte can be absorbed into the separator and active material of plate to be gel. The test also says that semi-gel electrolyte shows the much better performance for low-rate discharge and the liquid electrolyte is good for high-rate discharge because the reaction rate of gel electrolyte is slower than liquid one for high-rate discharge performance. The test with DOD10% and DOD100% says that 5% silica electrolyte shows much better performance for life efficiency than liquid one. Because semi-gel electrolyte increase the efficiency of gas recombination at the chemical reaction of VRLA battery and it makes minimizing the reduction of electrolyte. Using the 5% silica electrolyte in order to improve the stroage efficiency and life performance for photovoltic and wind power generation, it causes improving by 4.8% for DOD100% and 20% for DOD10%.

The purpose of this study was to optimize experimental conditions (time ($X_1$) (ranging of 26.36 - 93.64 min), concentration of sulfuric acid ($X_2$) (ranging of 0-2.5%) and temperature ($X_3$) (ranging of $136.4-203.6^{\circ}C$) for an organosolv pretreatment process to extract lignin from waste wood. The resulting quadratic model equation using RSM (response surface methodology) represented y (lignin yield) = $-79.89+0.91X_1+9.8X_2-2.54{\times}10^{-3}X_1{^2}-2.11X_2{^2}$. The $R^2$ (coefficient of determination) value of 0.8531 for a model indicates this model has statistically significant predictors at the 10% levels. The predictive results optimized by quadratic model produced a lignin yield of 12.46 g/100 g of dry wood under conditions of $178.2^{\circ}C$ and 2.32% $H_2SO_4$. The lignin yield was more affected by the acid catalyst concentrations than the reaction temperature, but the reaction time was not an influential factor for improving lignin extraction from waste wood in this organosolv pretreatment. According to ANOVA (analysis of variance), the significance probability (p-value) of model was smaller than 0.001 and simulation of obtained model equations showed a good reproducibility based on actual organosolv tests under optimal conditions.

Korea Food and Drug Administration (KFDA) has officially announced 2,4-dinitrophenylhydrazine (DNPH) derivatization - high performance liquid chromatography (HPLC) methods for analysis of formaldehyde. This study was conducted to develop a convenient derivatization method for cosmetics by improving complex pre-treatment procedures included in KFDA method. To simplify pre-treatment procedures of KFDA method, reaction conditions including pH, time and temperature were optimized. This pre-treatment method does not require complicate pre-treatment steps of KFDA method such as pH adjustment of test solution with acetate buffer (pH 5.0), solvent-solvent partitioning with dichloromethane and concentrating procedure with vacuum evaporator. Formaldehyde-dinitrophenylhydrazone (formaldehyde-DNP) product produced by derivatization reaction was separated and quantified with a reversed-phase HPLC, which was slightly modified with KFDA method. The linearity test showed good results with 0.9999 of correlation coefficient ($r^2$) in the range of 2 ~ 40 ppm of standard solutions. In this method, limit of detection (LOD) and limit of quantitation (LOQ) values for formaldehyde were 0.2 ppm and 0.5 ppm, respectively. In addition, recovery test demonstrated that the method was also accurate and reproducible. Therefore, the proposed method can be applicable to rapid analysis of formaldehyde in cosmetics.
The objective of this study is to identify the kinematic variables of giant swing backward to handstand as well as individual variations of each athlete performing this skill, which in turn will provide the basis for developing suitable training methods and for improving athlete's performance in actual games. For this end, 3 male athletes, members of the national team, who are in ${\Box}{\Box}H{\Box}{\Box}$ University, have been randomly chosen and their giant swing backward to handstand performance was recorded using two digital cameras and analyzed in 3 dimensional graphics. This study came to the following conclusion. 1. Proper time allocation for giant swing backward to handstand are: Phase 1 should provide enough time to attain energy for swing track of a grand round movement. The phase 3 is to throw the body up high in the air and stay in the air as long as possible to smoothen up the transition to the next stage and the phase 4 should be kept short with the moment arm coefficient of the body reduced. 2. As for appropriate changes of locations of body center, the phase 1 should be comprised of horizontal, perpendicular, compositional to make up a big rotational radius. Up to the Phase 3 the changes of displacements of vertical locations should be a good scale and athlete's body should go up high quickly to increase the perpendicular climbing power 3. When it comes to the speed changes of body center, the vertical and horizontal speed should be spurred by the reaction of the body in Phase 2 and Phase 3. In the Phase 4, fast vertical speed throws the body center up high to ensure enough time for in-the-air movement. 4. The changes of angles of body center are: in Phase 2, shoulder joint is stretching and coxa should be curved up to utilize the body reaction. In the Phase 4, shoulder joint and coxa should be stretched out to get the body center as high as possible in the air for stable landing. 5. The speeds of changes in joints angles are: in the Phase 2 should have the speed of angles of shoulder joints increase to get the body up in the air as quickly as possible. The Phase 3 should have the speed of angles in shoulder joint slow down, while putting the angles of a knee joint up to speed as quickly as possible to ensure enough time for in-the-air movement.
Numerical analysis was done to evaluate the fluid distribution inside of the mixing quencher to increase the reaction efficiency of the aqueous hydrogen peroxide solution in the scrubbing column which is used for simultaneous desulfurization and denitrification. Effective injection of the aqueous hydrogen peroxide ($H_2O_2$) solution in the mixing quencher has major effects for improving the reaction efficiency in the scrubbing column by enhancing the mixing of the aqueous $H_2O_2$ solution with the exhaust gas. The current study is to optimize the array of nozzles and the spray angles of the aqueous $H_2O_2$ solution in the mixing quencher by using the computational method. Main concerns of the analysis are how to enhance the uniformity of the $H_2O_2$ concentration distribution in the internal flow. Numerical analysis was done to check the distribution of the internal flow in the mixing quencher in terms of RMS values of the $H_2O_2$ concentration at the end of quencher. The concentration distribution of $H_2O_2$ at the end of is evaluated with respect to the different array of the nozzle pipes and the nozzle tip angles, and we also analyzed the turbulence formation and fluid mixing in the zone. The effect of the spray angle was evaluated with respect to the mixing efficiency in different flow directions. The optimized mixing quencher had the nozzle array at location of 0.3 m from the inlet duct surface and the spray angle is $15^{\circ}$ with the co-current flow. The RMS value of the $H_2O_2$ concentration at the end of the mixing quencher was 12.4%.

The purpose of this study was to inhibit biofouling on a selectively permeable membrane (SPM) and increase biomass productivity in marine photobioreactors (PBRs) for microalgal cultivation by chemical treatment. Surfaces of a SPM, composed of polyethylene terephthalate (PET), was sulfonated to decrease hydrophobicity through attaching negatively charged sulfonic groups. Reaction time of sulfonation was varied from 0 min to 60 min. As the reaction time increased, the water contact angle value of SPM surface was decreased from $75.5^{\circ}$ to $44.5^{\circ}$, indicating decrease of surface hydrophobicity. Furthermore, the water permeability of sulfonated SPM was increased from $5.42mL/m^2/s$ to $10.58mL/m^2/s$, which reflects higher nutrients transfer rates through the membranes, due to decreased hydrophobicity. When cultivating Tetraselmis sp. using 100-mL floating PBRs with sulfonated SPMs, biomass productivity was improved by 34% compared with the control group (non-reacted SPMs). In addition, scanning electron microscopic observation of SPMs used for cultivation clearly revealed lower degree of cell attachment on the sulfonated SPMs. These results suggest that sulfornation of a PET SPM could improve microalgal biomass productivity by increasing nutrients transfer rates and inhibiting biofouling by algal cells.

Soil stabilization can make the soils becoming more stable by using an admixture to the soil. Lime stabilization enhances the engineering properties of soil, which includes reducing soil plasticity, increasing optimum moisture content, decreasing maximum dry density and improving soil compaction. Silica fume is utilized as a pozzolanic material in the application of soil stabilization. Silica fume was once considered non-environmental friendly. In this paper, the materials required are kaolin grade S300, lime and silica fume. The focus of the study is on the determination of the physical properties of the soils tested and the consolidation of kaolin mixed with 6% silica fume and different percentages (3%, 5%, 7% and 9%) of lime. Consolidation test is carried out on the kaolin and the mixtures of soil-lime-silica fume to investigate the effect of lime stabilization with silica fume additives on the consolidation of the mixtures. Based on the results obtained, all soil samples are indicated as soils with medium plasticity. For mixtures with 0% to 9% of lime with 6% SF, the decrease in the maximum dry density is about 15.9% and the increase in the optimum moisture content is about 23.5%. Decreases in the coefficient of permeability of the mixtures occur if compared to the coefficient of permeability of kaolin soft clay itself reduce the compression index (Cc) more than L-SF soil mix due to pozzolanic reaction between lime and silica fume and the optimum percent of lime-silica fume was found to be (5%+6%) mix. The average coefficient of volume compressibility decreases with increasing the stabilizer content due to pozzolanic reaction happening within the soil which results in changes in the soil matrix. Lime content +6% silica fume mix can reduce the coefficient of consolidation from at 3%L+6%SF, thereafter there is an increase from 9%L+6%SF mix. The optimal percentage of lime silica fume combination is attained at 5.0% lime and 6.0% silica fume in order to improve the shear strength of kaolin soft clay. Microstructural development took place in the stabilized soil due to increase in lime content of tertiary clay stabilized with 7% lime and 4% silica fume together.
