막걸리에서 분리한 다당의 면역자극 활성에 미치는 효과 (Immunomodulatory Activity of Crude Polysaccharides from Makgeolli)

  • 본 연구에서는 막걸리로부터 다당을 분리하여 자연면역계를 구성하고 있는 보체계 및 macrophage에 대한 면역자극활성을 측정하였다. 열수추출한 뒤 ethanol 침전을 통하여 조다당을 추출한 후 특성을 검토한 결과, 막걸리 조 다당체(RWW)의 neutral sugar 함량은 87.3%, uronic acid 함량은 2.5%, protein 함량은 10.2%이었다. RWW는 인체 초기 면역반응에 있어 중요한 역할을 담당하는 보체계에 대한 활성을 나타냈으며, 마우스 RAW 264.7 macrophage에서 세포증식 및 면역유도 cytokine 분비활성을 나타내었다. RWW를 1, 10, $100{\mu}g/mL$ 농도로 macrophage에 처리하여 면역자극을 유도한 결과 모든 처리 농도에서 IL-6, IL-12, TNF-${\alpha}$의 분비가 증가되었다. 이상의 결과를 종합해 볼 때, 막걸리 유래 다당체는 자연면역계를 구성하고 있는 보체계와 macrophage의 활성인자로 작용할 수 있음이 확인되었고, RWW를 통해 cytokine 분비를 유도, 조절함으로써 체내 면역기능을 증강시킬 가능성이 있을 것으로 사료되었다.

  • Pinus densiflora belongs to the Pinaceae family which has been widely used for health promoting purposes as folk medicine or as a food. Various curative effects of different parts of the pine have been reported including as a remedy for carcinoma. We examined the effects of pine needle water extracts (PNE) on macrophage function using peritoneal macrophage, pre-osteoclast bone macrophage (Raw 264.7 cell) and brain macrophage (C6 microglia). When peritoneal macrophages were treated with various concentrations of PNE ($1{\sim}100{\mu}g/mL$) for 24 hours, phagocytic activity was significantly increased, whereas it had no effect on tumoricidal activity and NO production. However, the treatment of Raw 264.7 with PNE resulted in the enhancement of NO production at high concentration ($100{\mu}g/mL$). Furthermore, the treatment of C6 with PNE increased the production of NO in a concentration-dependent manner, whereas PNE suppressed NO production in $LPS/IFN-{\gamma}-stimulated$ microglia. These results suggest that PNE has differential immunomodulatory effects on macrophages.

  • Streptococcal diseases are known as serious problems in fresh water and marine fish culture industry worldwide. Recently, the importance of the infection of Streptococcus parauberis has been increased among the streptococcicosis since severe outbreaks in cultured olive flounder (Paralichthys olivaceus) were recorded in Korea. The aims of the present study were to investigate immunomodulatory differences between S. parauberis and S. iniae. In this study, S. parauberis isolated from diseased olive flounder was investigated on the characteristics of morphological and immune responses. Immune response of the fish to the pathogen was characterized by bactericidal activity in the serum, phagocytic activity and reactive oxygen intermediaters (ROIs) production of olive flounder. The capsules of both bacteria, S. parauberis and S. iniae seem to be associated with the ability to resist killing activity of normal sera. Also, capsulated strains could survive in the phagocytes and induce to lower ROIs production.

Prosuction of Cytokine and NO by RAW 264.7 Macrophages and PBMC In Vitro Incubation with Flavonoids

  • Flavonoids, a group of low molecular weight phenylbenzopyrones, have various pharmacological properties including antioxidant activity, anticancer, and immunomodulatory effects. In the present study, lipopolysaccharide (LPS) and phorbol 12-myristate 13-acetate/phytohemagglutinin (PMA/PHA) were used as stimulants for RAW 264.7 macrophages and human peripheral blood mononuclear cell (hPBMC), and tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-2 productions were measured. In addition, flavonoids were examined for their effects on LPS-induced NO production in RAW 264.7 macrophages. The results showed that all compounds were not strongly cytotoxic at the tested concentrations on hPBMC and RAW 264.7 macrophages. On immunomodulatory properties, catechin, epigallocatechin (EGC), naringenin, and fisetin repressed NO production and TNF-${\alpha}$ secretion. Furthermore, catechin, epigallocatechin gallate (EGCG), epicatechin (EC), luteolin, chrysin, quercetin, and galangin increased IL-2 secretion while EGC, apigenin, and fisetin inhibited the secretion. These results indicated that flavonoids have the capacity to modulate the immune response and have a potential anti-inflammatory activity. There was no obvious structure-activity relationship regard to the chemical composition of the flavonoids and their cell biological effects.

Modulatory Activity of CpG Oligonucleotides from Bifidobacterium longum on Immune Cells

  • The purpose of this study was to characterize and investigate the immune activity of CpG oligodeoxynucleotides (ODNs) from Bifidobacterium longum. Bacterial CpG motifs have attracted considerable interests because of their immunomodulatory activities. Genomic DNA from B. longum was prepared and amplified for 4 different 180-188-mer double-stranded ODNs (BLODN1-BLODN4). When immune cells (RAW 264.7 murine macrophages and JAWS II dendritic cells) with these ODNs were treated, BLODN4 induced the highest immune activity. To assess the effectiveness of the CpG sequences within BLODN4, single-stranded 40-mer ODNs containing CpG sequences (sBLODN4-1, sBLODN4-2) were synthesized. sBLODN4-1 induced higher level of cytokines such as interleukin (IL)-12p40 and tumor necrosis factor (TNF)-$\alpha$ by macrophage and IL-6 and TNF-$\alpha$ by dendritic cells than did sBLODN4-2. The results suggest that CpG ODNs-enriched components of B. longum might be useful as an immunomodulatory functional food ingredient.

청도반시 추출물에 의한 라디칼소거 활성과 면역조절 활성의 상호 비교 (Comparison of Radical Scavenging and Immunomodulatory Activities Exhibited by an Aqueous Extract of Diospyros kaki Thunb. Fruit (Persimmon))

  • 청도반시 추출물로부터 라디칼 소거활성, T-bet promoter 활성, IL-4 발현 활성의 상관관계를 분석하고자 7월부터 10월까지 감을 시기별로 수확하여 분자염증 활성 여부를 확인하였다. 감의 무게는 시기가 경과할수록 무거워지면서 항산화 활성에 있어서는 감이 미숙과일수록 항산화활성이 우수하였다. 4가지의 용매로 추출한 감의 항산화 활성도 유사한 양상을 보였다. T-bet promoter 활성은 추숙이 될 수록 억제되는 양상을 보였는데, 이에 반하여 IL-4의 발현 정도는 숙성함에 따라 증가하는 것을 알 수 있었다. 이로 미루어 보아 수확시기에 따라 이러한 분자염증에 관련한 일련의 활성의 억제 효과는 감이 미숙과일수록 활성이 상승하였다.

국내산 백화사설초 전초 및 뿌리 메타놀층의 면역조절 효과 (Immunomodulatory Effect of Methanol Extracts of Korean Oldenlandiae Herba and Radix)

  • For the evaluation of immunomodulatory effect of Korean Oldenlandiae Herb (OH) and Radix (OR), our experiment was performed with methanol extracts of Korean Oldenlandiae Herba and Radix. After administration of methanol extracts of Korean OH and OR for 7 days, Balb/C mice were immunized with sheep red blood cells. Four days later, splenic leukocytes were isolated and immunological experiments were performed. Rosette forming cells and plaque forming cells were significantly increased in Korean OH and OR treated mice compared with PBS treated control. Korean OH and OR also enhanced T and B lymphocytes, macrophage and natural killer cells by flowcytometric analysis. LPS-induced TNF-α and IL-6 levels were increased by OH and OR compared with untreated control. These results suggest that Korean OH and OR have immunomodulatory activity through regulation of cell-mediated immune and humoral immune response.

  • Background: A red seaweed, Callophyllis japonica has been traditionally eaten in the oriental area. In a recent study, it has been demonstrated that the ethanol extract of C. japonica have antioxidant activity. However, there are few studies about the effects of C. japonica on the function of immune cells. We investigated the immunomodulatory effects of C. japonica on the function of dendritic cells, the potent antigen-presenting cells. Methods: Bone marrow-derived dendritic cells (DCs) were used and the viability was measured by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay and trypan blue exclusion test. Cytokine and nitric oxide (NO) levels were determined by using ELISA and Griess reagent, respectively. The expression levels of DC surface markers were measured by flow cytometric analysis. Results: C. japonica ethanol extract did not significantly affect the DCs viability and the IL-12 production from DCs, irrespective of the presence of lipopolysaccharide (LPS). In addition, it did not significantly change the expression of DC surface markers. However, C. japonica ethanol extract significantly inhibited the LPS-induced NO production and also increased the proliferation of allogeneic lymphocytes activated by DCs. Conclusion: Our data suggests that C. japonica ethanol extract enhances the proliferation of allogeneic lymphocytes activated by DCs which is associated with inhibition of NO production from DCs induced by LPS.

영지버섯 생장점 단백다당체 GLB-A, GLB-B의 항암효과 및 면역 활성에 관한 유세포 분석학적 연구 (Flow Cytometrical Analysis of the Antitumor and Immunomodulatory Activities of GLB-A and GLB-B, the Protein-polysaccharide Fractions of the Growing Tips of Ganoderma Lucidum)

  • In the previous study we described the antitumor effect of GLB, a protein-polysaccharide fraction separated from the growing tips of Ganoderma lucidum, against sarcoma 18 0 solid tumor in ICR mice. In this study, we separated an acidic protein-polysaccharide fraction, GLB-A, and a basic protein-polyaccharide fraction, GLB-B, from GLB by differential precipitation, and elucidated their antitumor and immunomodulatory activities. When ip injected at the dose of 50mg/kg/day into the ICR mice, GLB-A and GLB-B inhibited the growth of ip implantated sarcoma 180 cells by 32.4% and 21.0%, respectively. Of these, GLB-A increased the % lymphoblast in the spleen of the tumor-bearing and the normal mice by 20.9% and 123.0%, and the CD4/CD8 ratio by 73.3% and 22.4%, respectively. GLB-A also increased the expression of CD25 (IL-2 receptor alpha ch0ain) in normal mice by 82.0%. These results strongly suggest that GLB-A is a promising candidate for antitumor immunomodulatory medicine.

  • Evodiae Fructus(EF) has been used as Traditional medicine for the treatment of headache, abdominal pain, hemorrhage, and menorrhae in many Asian countries. The present study was conducted to investigate the immunomodulatory effect on cyclophosphamide(CY)-induced immunesuppression of water extracted EF(EFE). In the mouse spleen cell proliferation assay, EFE enhanced mitogenic activity and restored the CY-induced cell suppression. In the nitric oxide(NO) assay, EFE inhibited NO production and iNOS protein levels in LPS-stimulated RAW 264.7 cells. In the GC-MS analysis, many ingredients of EFE were detected by solvents. These results indicated that EFE can modulate immune response through immune cell proliferation, the regulation of NO production and the inhibition of CY-induced immunotoxicity.
