IMMUNOMODULATING ACTIVITIES OF BRAZILIN AND HEMATOXYLIN IN NORMAL YOUNG MICE

  • Experiments were undertaken to evaluate the effects of brazilin and hematoxylin on immune functions in normal young mice. Brazilin and hematoxylin decreased the circulating leucocyte counts. Brazilin and hematoxylin decreased the cirulating leucocyte counts. Both compounds did not produce a significant change of immunoorgan weights in comparison with those in the control group. Brazilin decreased significantly IgM plaque forming cells. Brazilin and hematoxylin decreased significantly Arthus reaction at 3 hours-post challenge with respect to aggregated bovine serum albumin.

  • Mushrooms are ubiquitous in nature and have high nutritional attributes. They have demonstrated diverse biological effects and therefore have been used in treatments of various diseases, including cancer, diabetes, bacterial and viral infections, and ulcer. In particular, polysaccharides, including ${\beta}$-glucan, are considered as the major constituents responsible for the biological activity of mushrooms. Although an overwhelming number of reports have been published on the importance of polysaccharides as immunomodulating agents, not all of the healing properties found in these mushrooms could be fully accounted for. Recently, many research groups have begun investigations on biologically active small-molecular weight compounds in wild mushrooms. In this mini-review, both structural diversity and biological activities of novel bioactive substances from Korean native mushrooms are described.

Antitumor Effects and Immunomodulating Activities of Phellinus linteus Extract in a CT-26 Cell-Injected Colon Cancer Mouse Model

  • The antitumor effects of Phellinus linteus extract (Keumsa Linteusan) were investigated in a CT-26 cell-injected colon cancer mouse model. When administered orally (250${\sim}$1,000 mg/kg body weight), Keumsa Linteusan significantly inhibited the growth of solid colon cancer. The highest dose was highly effective, reducing tumor formation by 26% compared with the control group. The anticomplementary activity of Keumsa Linteusan increased in a dose-dependent manner. Lysosomal enzyme activity of macrophages was increased by 2-fold (100 ${\mu}$/ml) compared with the control group. Keumsa Linteusan can be regarded as a potent enhancer of the innate immune response, and can be considered as a very promising candidate for antitumor action.

원유에서 분리한 Lactobacillus fermentum 450의 생리적 특성과 면역활성 (Physiological Characteristics and Immunomodulation Activity of Lactobacillus fermentum 450 isolated from Raw Milk)

  • 본 연구는 원유로부터 면역증진 젖산균을 분리 및 동정하고, 이 균주의 생리적 특성을 규명하여 상업적으로의 이용가능성을 검토하고자 실시하였다. 이를 위해 Modified MRS 분별배지를 사용하여 노란색 집락을 형성하는 균주를 대상으로 면역활성이 높은 균주를 선발한 결과, D205 균주가 최종 선발되었다. D205 균주는 $IL-1{\alpha}$, $TNF-{\alpha}$, NO 값이 각각 2,500pg 이상/mL, 2,000 pg 이상/mL, $11.55{\pm}2.95{\mu}M$로 나타났으며, 동정결과 Lactobacillus fermentum으로 판명되었고, Lactobacillus fermentum 450으로 명명하였다. Lactobacillus fermentum 450의 최적 생장 온도는 $40^{\circ}C$이었으며, 답즙산과 산성의 pH에서 모두 우수한 생존력을나타내었다. 효소활성은 leucine arylamidase와 acid phosphatase가 비교적 높게 나타났다. 항생제 내성 실험 결과, kanamycin, neomycin, polymyxin B에 내성이 있는 반면 novobiocin에 감수성을 나타냈으며, Salmonella Typhimurium에 대해 63.86%의 억제 효과를 지니고 있으나, Escherichia coli와 Staphylococcus aureus에 대해서는 항균력이 거의 없는 것으로 나타났다.

Lactobacillus plantarum HY7712 Protects Against the Impairment of NK-Cell Activity Caused by Whole-Body ${\gamma}$-Irradiation in Mice

  • While searching for lactic acid bacteria that can restore aging-impaired immune responses, we isolated the Toll-like receptor (TLR) 2/NF-${\kappa}B$-activating strain Lactobacillus plantarum HY7712 from kimchi and investigated its immunomodulating effect in whole-body ${\gamma}$-irradiated mice. Exposure to HY7712 strongly activated NF-${\kappa}B$ signaling in RAW264.7 cells, but inhibited lipopolysaccharide-stimulated NF-${\kappa}B$ activation. Moreover, HY7712 protected against the downregulation of interferon (IFN)-${\gamma}$ and upregulation of interleukin (IL)-13 caused by ${\gamma}$-irradiation in mice. In mice, ${\gamma}$-irradiation impaired NK-cell activity against YAC-1 tumor cells, but following HY7712 exposure, the activity of NK cells was restored to 91.5% of the level measured in control mice (p < 0.05). These findings suggest that HY7712 activates the TLR2/NF-${\kappa}B$ signaling pathway and protects against the impairment of NK-cell activity caused by ${\gamma}$-irradiation or aging.

Antitumor Activity of the Korean Mistletoe Lectin is Attributed to Activation of Macrophages and NK Cells

  • Inhibitory effect of the lectins (KML-C) isolated from Korean mistletoe (KM; Viscum album coloratum) on tumor metastases produced by murine tumor cells (B16-BL6 melanoma, colon 26M3.1 carcinoma and L5178Y-ML25 lymphoma cells) was investigated in syngeneic mice. An intravenous (i.v.) administration of KML-C (20-50 ng/mouse) 2 days before tumor inoculation significantly inhibited lung metastases of both B16-BL6 and colon 26-M3.1 cells. The prophylactic effect of 50 ng/mouse of KML-C on lung metastasis was almost the same with that of 100 $\mu$ g/mouse of KM. Treatment with KML-C 1 day after tumor inoculation induced a significant inhibition of not only the experimental lung metastasis induced by B16-BL6 and colon 26M3.1 cells but also the liver and spleen metastasis of L5178Y-ML25 cells. Furthermore, multiple administration of KML-C given at 3 day-intervals after tumor inoculation led to a significant reduction of lung metastasis and suppression of the growth of B16-BL6 melanoma cells in a spontaneous metastasis model. In an assay for natural killer (NK) cell activity. i.v. administration of KML-C (50 ng/mouse) significantly augmented NK cytotoxicity against Yac-1 tumor cells 2 days after KML-C treatment. In addition, treatment with KML-C (50 ng/mouse) induced tumoricidal activity of peritoneal macrophages against B16-BL6 and 3LL cells. These results suggest that KML-C has an immunomodulating activity to enhance the host defense system against tumors, and that its prophylactic and therapeutic effect on tumor metastasis is associated with the activation of NK cells and macrophages.

편평세포암 동물 모델에서 고려인삼잎-다당체(MB40)의 항암치료효과 (The Anti-tumor Effect of Polysaccharide from the Leaves of Panax Ginseng C.A. Meyer(MB40) in a Murine Squamous Cell Carcinoma Model)

  • Background and Objectives : Panax ginseng C.A. Meyer is a medical plant that has been widely utilized as a tonic and nutritional agent since ancient times in Korea. Ginseng has anti-metastatic property of cancer and immunomodulating activity. The novel acidic polysaccharide compound(MB40) was isolated from the leaves of Panax ginseng C.A. Meyer. To determine immunomodulating activities of MB40, we evaluate anti-cancer and anti-metastatic effects of MB40 in tumor bearing immune competent mice. Material and Methods : C3H mice were divided into three equal groups(Cisplatin treatment group, MB40 treat-ment group, Cisplatin and MB40 treatment group) and were transplanted SCC(Squamous Cell Carcinoma) cells(2${\times}$106) to the lateral side of abdomen. From day 4 after transplantation, MB40 was administrated at dose of 10mg/kg, respectively, every other day by intratumoral injection. Cisplatin was systemically administrated at doses of 1mg/kg, respectively, every week by intraperitoneal injection. Results : 5 days after administration, tumors can be palpated in every mice group. After 13 days of administration, the mice group to which MB40 were administrated exhibited reduction in tumor size respectively, compared to cisplatin group. Overall status of mice such as body weight and activity were superior in MB40 group than cisplatin group. Conclusion : The result of this study indicates MB40 may have significant therapeutic effect and decreases complications induced by systemic chemotheraphy. MB40 may be developed as a novel and potent immunotropics to improve the cell immune system and anti-cancer drug for the treatment of cancer patients in head and neck squamous cell carcinoma.

당화된 레스베라트롤의 대식세포 RAW 264.7세포의 생존능력과 레스베라트롤의 면역제어 활성을 증가 (Glucosylation of Resveratrol Improves its Immunomodulating Activity and the Viability of Murine Macrophage RAW 264.7 Cells)

  • 레스베라트롤의 면역제어 성질과 대식세포의 생존능력과 관련하여 당화된 레스베라트롤의 효과를 확인하기 위해 대식세포 RAW 264.7에서 연구하였다. 인비토로에서 대식세포에서 총 4개의 레스베라트롤 및 당화된 유도체 (E)-resveratrol, (E)-resveratrol 3-O-${\beta}$-${\small{D}}$-glucoside (R-3-G), 및 (E)-resveratrol 4'-O-${\beta}$-${\small{D}}$-glucoside (R-4'-G)를 여러 가지 농도로 처리한 후 일산화질소 (NO)와 인터루킨 6 (IL-6) 발현을 연구하였다. 앞서 언급한 물질로 처리한 후 인비토로에서 RAW 264.7 세포의 생존능력도 연구하였다. 대식세포 생존능력 평가분석 결과를 보면, 두 개의 레스베라트롤 모노글루코사이드인 R-3-G와 R-4'-G은 (E)-resveratrol와 비교하여 A549 and HepG2 세포에서 50-80% 감소된 독성을 보여준다. 당이 없는 레스베라트롤과 비교하면, 당화된 레스베라트 유도체는 긍정적으로 전사적으로 IL-6 및 iNOS 발현이 높아지는 방향으로 NO 및 대식세포에서 IL-6의 생산이 조절된다. 레스베라트롤의 당의 역할은 RAW 264.7 세포의 생존능력과 레스베라트롤의 면역제어 활성을 증가시켜 주는 것으로 보여주고 있다.

Effect of chloroform extract of traditional Dicranopteris linearis leaves against paracetamol- and CCl4-induced liver toxicity in rats

  • The present study aimed to determine the hepatoprotective activity of the chloroform extract of D. linearis leaves (CEDL) using the paracetamol (PCM)- and carbon tetrachloride ($CCl_4$)-induced liver injury models in rats. The rats received $dH_2O$ (negative control), 200 mg/kg of silymarin (positive control) or CEDL (50, 250 and 500 mg/kg) orally once daily for 7 days and then were subjected to the hepatotoxic induction on the $7^{th}$ day. The samples (i.e. blood and liver) were collected and underwent biochemical and microscopical analysis, respectively. From the data obtained, both inducers caused significant (p < 0.05) increase in the levels of AST and ALT when compared to the control group, which were significantly (p < 0.05) reduced by CEDL in a generally dose-dependent manner. These biochemical findings were supported by the histopathological analysis and histological scoring. In conclusion, CEDL possesses potential hepatoprotective activity, which could be associated with its flavonoid and tannin contents with the mechanisms of hepatoprotection linked to either its antioxidant or anti-inflammtory/immunomodulating activities. Further in-depth studies are required to identify the responsible bioactive compound.

Probiotic Potential of Enterococcus faecium Isolated from Chicken Cecum with Immunomodulating Activity and Promoting Longevity in Caenorhabditis elegans

  • Probiotics, including Enterococcus faecium, confer a health benefit on the host. An Enterococcus strain was isolated from healthy chicken cecum, identified as E. faecium by 16S rDNA gene sequence analysis, and designated as E. faecium L11. To evaluate the potential of E. faecium L11 as a probiotic, the gastrointestinal tolerance, immunomodulatory activity, and lifespan extension properties of the strain were assayed. E. faecium L11 showed >66% and >62% survival in artificial gastric juice (0.3% pepsin, pH 2.5) and simulated small intestinal juice (0.5% bile salt and 0.1% pancreatin), respectively. Heat-killed E. faecium L11 significantly (p < 0.05) increased immune cell proliferation compared with controls, and stimulated the production of cytokines (IL-6 and $TNF-{\alpha}$) by activated macrophages obtained from ICR mice. In addition, E. faecium L11 showed a protective effect against Salmonella Typhimurium infection in Caenorhabditis elegans. In addition, feeding E. faecium L11 significantly (p < 0.05) extended the lifespan of C. elegans compared with the control. Furthermore, genes related to aging and host defense were upregulated in E. faecium L11-fed worms. In conclusion, E. faecium L11, which prolongs the lifespan of C. elegans, may be a potent probiotic supplement for livestock.
