  • The major garlic component, Allicin [diallylthiosulfinate, or (R, S)-diallyldissulfid-S-oxide] is known for its medicinal effects, such as antihypertensive activity, microbicidal activity, and antitumor activity. Allicin and diallyldisulfide, which is a converted form of allicin, inhibited the cholesterol level in hepatocytes, in vivo and in vitro. The metabolism of allicin reportedly occurs in the microsomes of hepatocytes, predominantly with the contribution of cytochrome P-450. However, little is known about how allicin affects the genes involved in the activity of hepatocytes in vivo. In the present study, we used the short-term intravenous injection of allicin to examine the in vivo genetic profile of hepatocytes. Allicin up-regulate ten genes in the hepatocytes. For example, the interferon regulator 1 (IRF-I), the wingless-related MMTV (mouse mammary tumor virus) integration site 4 (wnt-4), and the fatty acid binding protein 1. However, allicin down-regulated three genes: namely, glutathione S-transferase mu6, a-2-HS glycoprotein, and the corticosteroid binding globulin of hepatocytes. The up-regulated wnt-4, IRF-1, and mannose binding lectin genes can enhance the growth factors, cytokines, transcription activators and repressors that are involved in the immune defense mechanism. These primary data, which were generated with the aid of the Atlas Plastic Mouse 5 K Microarray, help to explain the mechanism which enables allicin to act as a therapeutic agent, to enhance immunity, and to prevent cancer. The data suggest that these benefits of allicin are partly caused by the up-regulated or down-regulated gene profiles of hepatocytes. To evaluate the genetic profile in more detail, we need to use a more extensive mouse genome array.

  • The growth and health of the fetus and neonate are directly influenced by the nutritional and physiological status of sows. Sows are often under catabolic conditions due to restrict feeding program during pregnancy and low voluntary feed intake during lactation. The current restrict feeding program, which aims at controlling energy intake during gestation, results in an inadequate supply of dietary protein for fetal and mammary gland growth. Low voluntary feed intake during lactation also causes massive maternal tissue mobilization. Provision of amino acids and fatty acids with specific functions may enhance the performance of pregnant and lactating sows by modulating key metabolic pathways. These nutrients include arginine, branched-chain amino acids, glutamine, tryptophan, proline, conjugated linoleic acids, docosahexaenoic acid, and eicosapentaenoic acid, which can enhance conception rates, embryogenesis, blood flow, antioxidant activity, appetite, translation initiation for protein synthesis, immune cell proliferation, and intestinal development. The outcome is to improve sow reproductive performance as well as fetal and neonatal growth and health. Dietary supplementation with functional amino acids and fatty acids holds great promise in optimizing nutrition, health, and production performance of sows and piglets. • Research in low cost feeds with high nutritional value and immunogenicity is important to reduce production cost and increase yields in the shrimp industry. In this study, immunostimulants of bacterial origin (peptidoglycan and lipopolysaccharides) and egg white were incorporated in shrimp diets as feed additives to determine the growth, survival and tolerance of Penaeus monodon to white spot syndrome virus (WSSV). Although the results obtained were not statistically significant (p>0.05) among the treatments, shrimp fed with bacterial additives and egg white showed higher weight gain, specific growth rate and survival than those fed on commercial shrimp diet. Shrimp fed with artificial diet showed 100% mortality when challenged with WSSV. However, shrimp fed on peptidoglycan supplemented diet had higher survival than their counterpart, whereas shrimp fed on egg white supplemented diet had a higher specific growth rate and better tolerance when challenged with WSSV. Further studies are required to determine the effectiveness and optimization of bacterial strains and egg white as feed additives to increase production and enhance the shrimp immune response to diseases.

  • Successful anticancer strategies require a differential response between tumor and normal tissue (i.e., a therapeutic ratio). In fact, improving the effectiveness of a cancer therapeutic is of no clinical value in the absence of a significant increase in the differential response between tumor and normal tissue. Although radiation dose escalation with the use of intensity modulated radiation therapy has permitted the maximum tolerable dose for most locally advanced cancers, improvements in tumor control without damaging normal adjacent tissues are needed. As a means of increasing the therapeutic ratio, several new approaches are under development. Drugs targeting signal transduction pathways in cancer progression and more recently, immunotherapeutics targeting specific immune cell subsets have entered the clinic with promising early results. Radiobiological research is underway to address pressing questions as to the dose per fraction, irradiated tumor volume and time sequence of the drug administration. To exploit these exciting novel strategies, a better understanding is needed of the cellular and molecular pathways responsible for both cancer and normal tissue and organ response, including the role of radiation-induced accelerated senescence. This review will highlight the current understanding of promising biologically targeted therapies to enhance the radiation therapeutic ratio.

  • Purpose: To promote condom use for young men, there is necessary to meet their varying needs for condom. The purpose of this study was to examine what predicts condom use intention according to experience of sexual intercourse among Korean male college students. The conceptual framework for the study was the theory of planned behavior and the health belief model. Methods: A cross-sectional descriptive design was used with self-report questionnaires, which were completed by 399 subjects. Logistic regression analysis was done to examine predictors of condom use intentions according to experience of sexual intercourse. Results: The susceptibility to sexual transmitted diseases/ Acquired Immune Deficiency Syndrome (STDs/AIDS) and pregnancy and benefits from condom use were significant predictors of condom use intentions in the non-experienced group. The significant predictors of condom use intention for the experienced group were barriers to condom use and condom use self-efficacy. Conclusion: These findings suggest that sex education program should be developed to overcome barriers to condom use, enhance condom use self-efficacy, and maximize the benefits from condom use as well as increase the perceived susceptibility to STDs/AIDS and pregnancy. To enhance condom use self- efficacy, practicing and increasing ability in interactive negotiation to use condom should be included.

  • In order to investigate the effects of Kyegyoksan on antitumor effects after Sarcoma 180 cells transplantation into the peritoneal cavity or left groin in mice, and immune depression in mice induced by methotrexate, the extracts of its herbal medicines were orally administered for 14 or 21 days. Experimental studies were performed for measureance of $IC_{50}$ in MTT assay, mean survival days, tumor and body weights for antitumor effects, delayed type hypersensitivity, hemagglutinine titer, hemolysin titer, rosette forming cells, interleukin-2 productivity, lymphocyte transformation, natural killer cell activity and phagocytic activity for immune responses in the immune depressed ICR mice, and SGOT, SGPT, BUN and creatinine for liver and kidney protective function in SO-rats. The results were obtained as follows: 1. From the results of MTT assay, the Kyegyoksan exstracts for SUN-1 and SUN-C4 were inhibited cell viability. 2. Mean survival time in Kyegyoksan-treated group was slightly increased with no effectiveness, as compared with the control group. 3. Tumor weight in Kyegyoksan-treated group was depressed with the statistical significance, as compared with the control group(p<0.01). 4. Body weight in Kyegyoksan-treated group was incresed with the statistical significance, as compared with the control group(p<0.05). 5. Delayed type hypersensitivity in Kyegyoksan-treated group was slightly incresed with no effctiveness, as compared with the control group. 6. Hemagglutinin titer in Kyegyoksan-treated group was incresed with the statistical significance(p<0.05), but hemolysin titer was slightly incresed with no effectiveness, as compared with the control group. 7. Rosette forming cells in Kyegyoksan-treated group was incresed with the statistical significance, as compared with the control group(p<0.001). 8. Interleukin-2 productivity in Kyegyoksan-treated group was incresed with the statistical significance, as compared with the control group(p<0.001). 9. Lymphocyte transfomation in Kyegyoksan-treated group was incresed with the statistical significance, as compared with the control group(p<0.01). 10. Natural killer cell activity in Kyegyoksan-treated group at E/T ratio 100 : 1 was incresed with the statistical significance(p<0.01), but at E/T ratio 50 : 1 and 10 : 1 was slightly incresed with no effectiveness, as compared with the control group. 11. Phagocytic activity in Kyegyoksan-treated group was slightly incresed with no effectiveness, as compared with the control group. 12. The levels of serum glutamic oxalacetic transaminase, serum glutamic pyruvic transaminase, blood urea nitrogen and serum creatinine in Kyegyoksan-treated group were not effective change, as compared with the control group. According to the above results, it could be suggested that Kyegyoksan have prominent antitumor effects, enhance both cellular and humoral immunity, and have no injury to liver and kidney functions.

  • We report non-specific immune responses and its disease resistance against Edwardsiella tarda by alga mixture (HE; Hizikia:Ecklonia) in olive flounder for the first time. Five isonitrogenous (44% crude protein) and isocaloric (17.1 MJ $kg^{-1}$) diets were formulated to have 0%, 2%, 4%, 6% and 8% of the alga mixture. One of five experimental diets was fed triplicate groups of fish (30 fish/group) to apparent satiation in a flow through system. After a two week feeding, blood was sampled at 3, 6, 12, 24 h after the last feeding for a kinetic measurement of nitroblue tetrazolium (NBT) activity and healthy fish with similar sizes in each tank were selected and injected with 1 mL of E. tarda suspension ($1.0\times10^7$ CFU/mL) to evaluate the disease resistance of the fish. Dietary supplementation of alga mixtures resulted in significantly higher non-specific immune responses compared with the fish fed the control diet. The cumulative mortality was significantly lower in the fish groups fed alga mixture containing diets than control group in the challenge test with E. tarda. Therefore, the results in this study indicate that dietary supplementation of Hizikia and Ecklonia mixtures enhance the non-specific immune responses and a disease resistance of olive flounder.
