This study was performed to investigate improving immune activities of natural water-soluble sulforaphane extracted from Brassica oleracea var. italica by nano encapsulation process. The nanoparticles of the sulforaphane extracted with ultrasonification process at $60^{\circ}C$ promoted human B and T cell growth, about $7{\sim}35%$ compared to the control. The secretion of IL-6 and TNF-${\alpha}$ from T cells were also enhanced as $2.6{\times}10^{-4}pg/cell$ and $2.1{\times}10^{-4} pg/cell$, respectively, by the adding nano samples. NK cell activation was improved about 8%, compare to the control in adding cultured medium of T cell added nano samples. It was also found that sulforaphane extracted from B. oleracea var. italica had highly inhibitory activity on hyaluronidase as $IC_{50}$ about $200\;{\mu}g/m{\ell}$. It can be concluded that natural water-soluble sulforaphane samples by nano-encapsulation, each size is 200 nm, extracted from B. oleracea var. italica has high immune activities through higher efficiency of bio-activation than conventional extracts.
The aim of this study was to investigate the immune activity of Ailantias altissima as an active ingredient on the immune enhancement by mixing ethanol extract with lactic acid bacteria (LAB). The activity of $NF-{\kappa}B/AP1$ transcription factor increased by $NF-{\kappa}B$ activity when mixed with LAB samples rather than with the extract alone. Nitric oxide (NO) production was similar in ethanol extract alone group and LPS treatment group. Mixing Ailantias altissima extract and lactic acid bacteria led to low NO production. The cytokine productivity of $TNF-{\alpha}$ significantly increased in Ailantias altissima extract when treated with LPS, and increased even more when mixed with lactic acid bacteria. The $IL-1{\beta}$ cytokine production was high when the Ailantias altissima extract were treated alone, but no $IL-1{\beta}$ cytokine was produced in the mixtures with isolates. The combination of the ethanol extract of the Ailantias altissima and the lactic acid bacteria was found to be effective in the immune function. Consequently, the ingredient to combine Ailantias altissima extract and lactic acid bacteria can be effectively used for development of the health functional food on the prevention and treatment of hypoimmunities.

In this study, the effects of the immune stimulator Euglena gracilis (Euglena) in cyclophosphamide (CCP)-induced immunocompromised mice were assessed. The key component β-1,3-glucan (paramylon) constitutes 50% of E. gracilis. Mice were orally administered Euglena powder (250 and 500 mg/kg body weight (B.W.)) or β-glucan powder (250 mg/kg B.W.) for 19 days. In a preliminary immunology experiment, ICR mice were intraperitoneally injected with 80 mg of CCP/kg B.W. during the final 3 consecutive days. In the main experiment, BALB/c mice were treated with CCP for the final 5 days. To evaluate the enhancing effects of Euglena on the immune system, mouse B.W., the spleen index, natural killer (NK) cell activity and mRNA expression in splenocytes lungs and livers were determined. To detect cytokine and receptor expression, splenocytes were treated with 5 ㎍/ml concanavalin A or 1 ㎍/ml lipopolysaccharide. The B.W. and spleen index were significantly increased and NK cell activity was slightly enhanced in all the experimental groups compared to the CCP-only group. In splenocytes, the gene expression levels of tumor necrosis factor-α, interferon-γ, interleukin (IL)-10, IL-6, and IL-12 receptor were increased in the E. gracilis and β-glucan groups compared to the CCP-only group, but there was no significant difference. Treatment with 500 mg of Euglena/kg B.W. significantly upregulated dectin-1 mRNA expression in the lung and liver compared to the CCP-only group. These results suggest that Euglena may enhance the immune system by strengthening innate immunity through immunosuppression.

In this study we evaluated the immune-enhancing effects of β-glucan, the main component of Euglena gracilis (Euglena), and Euglena on inflammatory factor expression in RAW264.7 macrophages and ICR mice with cyclophosphamide-induced immunosuppression. Macrophages were treated with β-glucan or Euglena for 48 h. The β-glucan and Euglena groups exhibited higher levels of inducible nitric oxide synthase, nitric oxide, and tumor necrosis factor (TNF)-α than the control (vehicle alone) group. Animals were fed saline and β-glucan (400 mg/kg body weight (B.W.)) or Euglena (400 or 800 mg/kg B.W.) for 19 days, and on days 17-19, cyclophosphamide (CCP, 80 mg/kg B.W.) was administered to induce immunosuppression in the ICR mouse model. CCP reduced the body weight, spleen index, and cytokine expression of the mice. To measure cytokine and receptor expression, splenocytes were treated with concanavalin A (ConA) or lipopolysaccharide (LPS) as a mitogen for 24 h. In vivo, ConA stimulation significantly upregulated the expression of interferon (IFN)-γ, interleukin (IL)-10, IL-12 receptor β1, IL-1β, and IL-2 in splenocytes from the β-glucan- or Euglena-treated groups compared with those in the splenocytes from the CCP-treated group; LPS stimulation increased the levels of the cytokines TNF-α, IL-1β, and IL-6 in splenocytes from the β-glucan- or Euglena- treated groups compared with those from the CCP-treated group, but most of these differences were not significant. These results demonstrate the effect of Euglena in ameliorating macrophages and immunosuppression in CCP-treated mice. Thus, Euglena has the potential to enhance macrophage- and splenocyte- mediated immune-stimulating responses.

The objective of the present study was to determine the possible immune-enhancing effects of a substance extracted from a submerged culture of Lentinus edodes with rice bran (SLRB). According to the results obtained by measuring the in vitro macrophage activity of the exo-biopolymer from SLRB, it appears to exhibit activity similar to that of LPS, and this activity seems to occur in a dose-dependent manner. According to the results obtained by measuring splenocyte proliferation, the exo-biopolymer appears to induce an increase in proliferation of approximately 1.4-fold compared to the control group. We measured the proliferation of bone marrow cells in order to evaluate gut immunity and, according to our results, proliferation was increased to 109% that of the control group, and was similar to that associated with LPS. In order to characterize the enhancement of immunological activity in vivo, we orally administered the exo-biopolymer (25, 50, 250 mg/kg bw) to C3H/He mice, and then measured the macrophage activity, determining that the activity was higher than that of the controls at concentrations of 50 and 250 mg/kg. Therefore, the exo-biopolymer from SLRB can be considered to be a useful a BRM agent, as it clearly allows some protection against immunological diseases.
Background: It is necessary for human beings to uptake vitamin C through diet or supplements. It is also well-known that vitamin C plays an important role in the prevention of scurvy, enhancement of collagen synthesis and anti-tumor immune response. In addition, there are several recent reports regarding the effective role of vitamin C on the regulation of allergic responses, such as atopic dermatitis and asthma. However, the effective therapeutic and preventive measures using vitamin C are not established yet, since vitamin C is seriously unstable in aqueous solution. Therefore, we have investigated the best way to maintain the stability of vitamin C. Methods: After we making a mixture of polyphenol (0.001, 0.01, 0.1%) and vitamin C (1 mM), the mixtures were placed at room temperature both with/without light protection. And then the concentration of ascorbic acid was measured with HPLC. To analyze the in vivo effect of vitamin C on the regulation of skin allergic reaction, polyphenol (0.1%)-vitamin C (1 mM) mixture was applied to the skin and the production of histamine from mast cell was analyzed by Evans blue dye staining. Results: We have found that the polyphenol has preventive power of oxidation of vitamin C. In addition, the production of histamine was suppressed by the polyphenol (0.1%)-vitamin C (1 mM) mixture. Conclusion: We have reached the conclusion that our study suggests the research guideline for the therapy of atopic dermatitis through vitamin C.
Acute respiratory virus infectious diseases are a growing health problem, particularly among children and the elderly. Much effort has been made to develop probiotics that prevent influenza virus infections by enhancing innate immunity in the respiratory tract until vaccines are available. Lactobacillus plantarum GB-LP2, isolated from a traditional Korean fermented vegetable, has exhibited preventive effects on influenza virus infection in mice. To identify the molecular basis of this strain, we conducted a whole-genome assembly study. The single circular DNA chromosome of 3,284,304 bp was completely assembled and 3,250 protein-encoding genes were predicted. Evolutionarily accelerated genes related to the phenotypic trait of anti-infective activities for influenza virus were identified. These genes encode three integral membrane proteins, a teichoic acid export ATP-binding protein and a glucosamine - fructose-6-phosphate aminotransferase involved in host innate immunity, the nonspecific DNA-binding protein Dps, which protects bacteria from oxidative damage, and the response regulator of the three-component quorum-sensing regulatory system, which is related to the capacity of adhesion to the surface of the respiratory tract and competition with pathogens. This is the first study to identify the genetic backgrounds of the antiviral activity in L. plantarum strains. These findings provide insight into the anti-infective activities of L. plantarum and the development of preventive probiotics.
We conducted a screening and checked the cultivation methods of lactic acid bacteria, which have anti-atopic dermatitis functions, by determining the lactic acid bacteria's immune enhancement by FACS, and antimicrobial activity against Staphylococcus aureus. The increase of Tcell CD4+/CD25+/foxp3+ was bigger in Lactobacillus plantarum than Lactococcus lactis subsp. lactis (Lc. lactis) and the antimicrobacterial activity against S. aureus was the opposite. The antimicrobial activity of Lb. plantarum culture with medium containing Lc. lactis culture broth was not enhanced, but the antimicrobial activity of Lc. lactis cultured in a medium containing Lb. plantarum culture broth was enhanced. As the optimal method caltivation of Lc. lactis in a medium containing 10% of heat-killed Lb. plantarum culture broth was chosen. By this method, the antibacterial activity of the pure Lc. lactis culture increased sharply at the end of the log phase, while a restraint effect on the growth of S. aureus increased 1.29 times.
Plasma protein which has been known as one of nonspecific immunostimulators was added to feedstuff to examine its effect on the enhancement of cellular immune response in porcine immune system. A total of 40 piglets, 20 male and 20 female each, were fed for 30 days with or without plasma protein. The peripheral blood were collected and analyzed for the investigation of leukocyte subpopulations and their activities by using a panel of monoclonal antibodies specific to porcine leukocyte differentiation antigens and flow cytometry. The results obtained as follows. 1. Total weight gain, daily feed intake and feed conversion rate for 10 days were significantly improved to 56%, 20% and 22% in the piglets fed plasma protein, respectively. 2. A significant increase in N (null or non T/non B) cells was also noticed. Leukocyte proportion from piglets fed plasma protein was 20.2-24.7%, otherwise that from piglets fed without plasma protein was 12.3-13.4%, respectively. 3. A significant increase in the proportion of B cells and cells expressing poCD1 was not found in piglets fed plasma protein. 4. Reaction with monoclonal antibodies specific to adhesion molecules, poCD11a, poCD11b, poCD44 and poCD45A and poCD45B, has shown that leukocyte subpopulation from piglets fed plasma protein did not significantly higher than that from piglets fed without plasma protein. 5. Total proportion of granulocytes and monocytes was about 50% in both group and the proportion after treated with Hypaque/Ficoll was 2.7% and 5.8% in each group, respectively.

Crude polysaccharide (CP) was fractionated from the submerged culture (containing both mycelia and culture broth, SC) with Phellinus linteus (PL) in mushroom complete medium (MCM) supplemented with ginseng extract ($65^{\circ}$Bx, GE) to enhance the immune activity. PL-GE-15-CP from SC cultivated in MCM supplemented with GE-15% (v/v, a ratio of MCM volume to GE) showed significantly higher macrophage stimulation (1.45 fold of the saline control at $100{\mu}g$/mL) than PL-GE-5 and 10-CP with GE-5 and 10%, or PL-CP from SC without GE. The potent intestinal immune system modulating activity through Peyer's patch was also obtained by PL-GE-15-CP (1.46 fold). When PL-GE-15-CP further fractionated on DEAE-Sepharose CL-6B (Cl- form), PL-GE-15-CP-II was the significantly higher than others from PL-GE-15-CP or PL-CP on macrophage stimulation, interleukin (IL)-12 production and intestinal immune system modulation (1.54, 3.96 and 1.56 fold, respectively). PL-GE-15-CP-II also had higher anti-metastatic activity against colon 26-M3.1 carcinoma cell (57.3% inhibition of tumor control, $200{\mu}g$/mouse) rather than PL-CP-II. This active fraction (PL-GE-15-CP-II) mainly contained neutral sugar (82.45%) and uronic acid (12.99%), and component sugar analysis showed that PL-GE-15-CP-II consisted mainly of uronic acid, Ara, Man, Gal and Glc (molar ratio of 0.52:0.97:0.63:1.00:0.54). Furthermore, the activity of GE culture was higher compared with culture without GE, indicating that GE helped to enhance the immune activity of P. linteus; also, it is assumed that the polysaccharide plays an important role in immune enhancement.
