Immobilization of Penicillium citrinum by Entrapping Cells in Calcium Alginate for the Production of Neo-Fructooligosaccharides

  • This work describes neo-fructooligosaccharides (neo-FOSs) production using the immobilized mycelia of Penicillium citrinum. Some critical factors were evaluated to optimize maximal production of neo-FOS. Optimal alginate and cell concentrations were determined to be $1.96\%$ alginate and $7.17\%$ cell, respectively, by statistical analysis. The optimal concentration of $CaCl_{2}$, which is related to bead stability, was determined to be 2 M. It was possible to increase the neo-FOS production by adding 15 units of glucose oxidase to the batch reaction. By co-immobilizing cells and glucose oxidase, neoFOS productivity increased $123\%$ compared with the whole-cell immobilization process. Based on the results above, a co-immobilization technique was developed and it can be utilized for large-scale production.

  • The on-line monitoring technique for the concentrations fo glucose and starch by FIA(Flow Injection Analysis)system was studied. Glucose oxidase(GOD) and amyloglucosidase(AMG) were immboilized on VA-Fpoxy carrier and integrated into the FLA system. The pH, buffer flow rate and temperature were optimized and the effects of salts and metabolites dissolved in the sample on the activity of immobilized enzyme were investigated. GOD-FIA and AMG/GOD-FIA were applied for the on-line monitoring of the glucose and starch in a simulated bioprocess. The on-line measurements of glucose concentrations by GOD-FIA agreed with off-line data well and the AMG/GOD-FIA with single cartidge system took and advantage over the FIA system with two separated cartridges for the on-line monitoring of starch concentrations.

Highly Sensitive and Selective Glucose Sensor Realized by Conducting Polymer Modified Nanoporous PtZn Alloy Electrode

  • Platinum is a well known element which shows a significant electrocatalytic activity in many important applications. In glucose sensor, because of the poisoning effect of reaction intermediates and the low surface area, the electrocatalytic activity towards the glucose oxidation is low which cause the low sensitivity. So, we fabricate a nanoporous PtZn alloy electrode by deposition-dissolution method. It provides a high active surface and a large enzyme encapsulating space per unit area when it used for an enzymatic glucose sensor. Glucose oxidase was immobilized on the electrode surface by capping with PEDOT composite and PPDA. The composite and PPDA also can exclude the interference ion such as ascorbic acid and uric acid to improve the selectivity. The surface area was determined by cyclic voltametry method and the surface structure and the element were analyzed by Scanning Electron Microscope (SEM) and Energy Dispersive X-ray spectroscopy (EDX), respectively. The sensitivity is $13.5{\mu}A/mM\;cm^2$. It is a remarkable value with such simply prepared senor has high selectivity.

  • pH-ISFETs, the semiconductor pH sensors, were combined with immobilized enzyme membranes to prepare FET type urea and glucose sensors and its operational characteristics were investigated. Photolithography techniques were applied to immobilize enzymes on the $H^{+}$ sensing membrane of the pH-ISFET with photo-sensitive polymers, PVA-SbQ. Fabricated urea and glucose sensors could determine $0.5{\sim}50{\;}mg/dl$ urea concentrations and $10{\sim}1000{\;}mg/dl$ glucose concentrations, respectively.

Performance Evaluation of Biofuel cell using Benzoquinone Entrapped Polyethyleneimine-Carbon nanotube supporter Based Enzymatic Catalyst (벤조퀴논 포집 폴리에틸렌이민-탄소나노튜브 지지체 기반 효소촉매의 바이오연료전지로서의 성능평가)

  • In this study, we synthesized biocatalyst consisting of glucose oxidase (GOx), polyethyleneimine (PEI) and carbon nanotube (CNT) with addition of p-benzoquinone (BQ) that was considered anodic catalysts of enzymatic biofuel cell (EBC). For doing this, PEI/CNT supporter was bonded with BQ by physical entrapping method stemmed from electrostatic attractive force ([BQ/PEI]/CNT). In turn, GOx moiety was further immobilized on the [BQ/PEI]/CNT to form GOx/[BQ/PEI]/CNT catalyst. This catalyst has a special advantage in that the BQ that has been usually dissolved into electrolyte was immobilized on supporter. According to the electrochemical analysis, maximum current density of the GOx/[BQ/PEI]/CNT catalyst was 1.9 fold better than that of the catalyst that did not entrap BQ with the value of $34.16{\mu}A/cm^2$, verifying that catalytic activity of the catalyst was enhanced by adoption of BQ. Also, when it was used as anodic catalyst of the EBC, its maximum power density was 1.2 fold better than that of EBC using the catalyst that did not entrap BQ with the value of $0.91mW/cm^2$. Based on such results, it turned out that the GOx/[BQ/PEI]/CNT catalyst was promising and viable as anodic catalyst of EBC.

A Dipstick-Type Electrochemical Immunosensor for The Detection of The Organophosphorus Insecticide Fenthion

  • A dipstick-type immunochemical biosensor for the detection of the organophosphorus insecticide fenthion was developed using a screen-printed electrode system as an amperometric transducer with polyclonal antibodies against fenthion as a bioreceptor. The assay of the biosensor involved competition between the pesticide in the sample and pesticide-glucose oxidase conjugate for binding to the antibody immobilized on the membrane. This was followed by measurement of the activity of the bound enzyme by the supply of the enzyme substrate (glucose) and amperometric determination of the enzyme reaction product ($H_2O_2$). The activity of the bound enzyme was inversely proportional to the concentration of pesticide. The optimized sensor system showed a linear response against the logarithm of the pesticide concentration ranging from $10^{-2}$ to $10^3\;{\mu}g/L$.

  • Screen printed carbon electrodes (SPEs) modified with co-immobilized osmium-based redox polymers can be used to apply multi-detecting biosensors. In this study, we report our initial studies of multi-detecting biosensor concepts using two osmium-based redox polymers for horseradish peroxidase-mediated reduction of ${H_2}{O_2}$ coupled to glucose oxidase-mediated oxidation of glucose. We target to synthesize two osmium redox polymers of potentials use, a chloride-containing redox polymer ($E^{O'}$ + 0.520 vs. Ag/AgCl) and a methoxy-containing redox polymer $E^{O'}$ + 0.150 vs. Ag/AgCl). The former show good catalytic electrical signals with horseradish peroxidase and the latter's redox polymer is to be an effective redox mediator of glucose oxidation by glucose oxidase.

  • Graphene has a large surface area to volume ratio and good mechanical and electrical property and biocompatibility. This study described the electrochemical deposition and reduction of graphene oxide on the surface of indium tin oxide (ITO) glass slide and electrochemical characterization of graphen-modified ITO. Cyclic voltammetry was used for the deposition and reduction of graphene oxide. The surface of graphen-coated ITO was characterized using scanning electron microscopy and energy dispesive X-ray spectroscopy. The electrodes were evaluated by performing cyclic voltammetry and electrochemical impedance spectroscopy. The number of cycles and scan rate greatly influenced on the coverage and the degree of reduction of graphene oxide, thus affecting the electrochemical properties of electrodes. Modification of ITO with graphene generated higher current with lower charge transfer resistance at the electrode-electrolyte interface. Glucose oxidase was immobilized on the graphene-modified ITO and has been found to successfully generate electrons by oxidizing glucose.

  • The multi-wall carbon nano-tube composite mixed with carbon paste electrode presented more sensitive and selective amperometric signals in the oxidation of glucose than general screen-printed carbon electrodes(SPCEs). Redox mediators to transport electrodes from enzyme to electrodes are very important part in the biosensor. A novel osmium redox complex was synthesized by the coordinating pyridine group containing primary amines which were electrochemically immobilized onto the MWCNT-SPCEs surface. Electrochemical studies of osmium complexes were investigated by cyclic voltammetry, chronoamperometry. The surface coverage of osmium complexes on the modified carbon nano-tube electrodes were significantly increased at 100 time (${\tau}_0=2.0\;{\times}\;10^{-9}\;mole/cm^2$) compared to that of the unmodified carbon electrodes. It's practical application of the glucose biosensor demonstrated that it shows good linear response to the glucose concentration in the range of 0-10 mM.

Enhanced Performance of Immunoassays with Affinity-Purified Analyte-Enzyme Conjugates as Signal Generators

  • In a competitive enzyme immunoassay, the performance was tested with different analyte-enzyme conjugates (signal generators) in their binding constants to antibody. Analyte (progesterone)-enzyme (glucose oxidase; GO) conjugates were chemically synthesized and purified by using a gel column with an immobilized antibody to progesterone. In an elution range from the column, four peaks were detected by measuring total enzyme activities. Results from further analysis indicated that the first peak contained mainly unreacted GO while the next three peaks conjugated GO with progesterone. These three conjugate preparations were compared in dose-response curves along with the unpurified mixture. The purified conjugates showed higher detection capabilities than did the mixture. Especially, the preparation in the second peak next to the free GO peak improved the detection limit five times. This performance was comparable to that of a progesterone-horseradish peroxidase conjugate that has been identified to have one progesterone ligand.
