An endolichenic fungus Xylaria grammica EL000614 produces grammicin, a potent nematicidal pyrone derivative that can serve as a new control option for root-knot nematodes. We optimized an Agrobacterium tumefaciens-mediated transformation (ATMT) protocol for X. grammica to support genetic studies. Transformants were successfully generated after co-cultivation of homogenized young mycelia of X. grammica with A. tumefaciens strain AGL-1 carrying a binary vector that contains the bacterial hygromycin B phosphotransferase (hph) gene and the eGFP gene in T-DNA. The resulting transformants were mitotically stable, and PCR analysis showed the integratin of both genes in the genome of transformants. Expression of eGFP was confirmed via fluorescence microscopy. Southern analysis showed that 131 (78.9%) out of 166 transformants contained a single T-DNA insertion. Crucial factors for producing predominantly single T-DNA transformants include 48 h of co-cultivation, pretreatment of A. tumefaciens cells with acetosyringone before co-cultivation, and using freshly prepared mycelia. The established ATMT protocol offers an efficient tool for random insertional mutagenesis and gene transfer in studying the biology and ecology of X. grammica.
Nucleotide metabolism in endothelium is variable between different species. Recent studies demonstrated that this variability could contribute coagulation dysfunction, even though organs of the alpha 1,3-galactosyltransferase gene knockout pig were transplanted into the primate. CD73 (ecto-5'-nucelotidase) is an enzyme at cell surface catalyzing the hydrolysis of adenosine triphosphate to adenosine, which plays role on a substance for anti-inflammatory and anti-coagulant. Thus, overexpression of CD73 in endothelial cells of the pig is considered as an approach to reduce coagulopathy. In this study, we constructed a human CD73 expression vector under control of porcine Icam2 promoter (pIcam2-hCD73), which is expressed specifically at endothelial cells, and of CMV promoter as a control (CMV-CD73). First, we transfected the CMV-CD73 vector into HEK293 cells, and then confirmed CD73 expression at cell surface by flow cytometry analysis. Next, we transfected the pIcma2-CD73 and CMV-CD73 vectors into primary porcine fibroblasts and endothelial cells. Consequence was that the pIcma2-CD73 vector was expressed only at the porcine endothelial cells, meaning that the pIcam2 promoter lead to endothelial cell-specific expression of CD73 in vitro. Finally, we nucleofected the pIcam2-hCD73 vector into passage 3 fibroblasts, and enforced hygromycin selection of 400mg/ml. We were able to obtain forty three colonies harboring pIcam2-CD73 to provide donor cells for transgenic cloned porcine production.
선발마커로 두 종류의 항생제 (hpt와 nptII)와 제초제(bar) 유전자를 사용하여 아그로박테리움을 이용한 수정된 들깨 형질전환방법을 개발하였다. 들깨 배축 절편을 pMOG6-Bar 운반체 혹은 pCK-Bar 운반체를 가진 아그로박테리움 EHA 105와 각각 3일간 공동배양 하였다. 1차로 형성된 신초는 하이그로마이신(15mg/L)이나 카나마이신(125 mg/L)을 사용하여 선발하였고 재분화 된 신초들은 확실한 형질전환체를 얻기 위해서 포스피노트리신(1.2 mg/L)에서 한번 더 선발하였다. 뿌리는 호르몬이 없는 MS배지에서 재분화 신초로 부터 유도하였으며 80개의 재분화개체를 획득하였다. 들깨 지놈으로 형질전환유전자의 삽입은 서던 블럿으로 확인하였고 유전자의 발현은 노던 블럿으로 분석하였다. To 식물체로부터 유도된 T1들깨 종자들은 후대로의 안정된 유전자 전이를 확인하기 위하여 0.3% 바스타 제초제 살포로 확인하였다.
Main strategy for a treatment of Parkinson's disease (PD), due to a progressive degeneration of dopaminergic neurons, is a pharmaceutical supplement of dopamine derivatives or ceil replacement therapy. Both of these protocols have pros and cons; former exhibiting a dramatic relief but causing a severe side effects on long-term prescription and latter also having a proven effectiveness but having availability and ethical problems Embryonic stem (ES) cells have several characteristics suitable for this purpose. To investigate a possibility of using ES cells as a carrier of therapeutic gene(s), human ES (hES, MB03) cells were transfected with cDNAs coding for tyrosine hydroxylase (TH) in pcDNA3.1 (+) and the transfectants were selected using neomycin (250 $\mu /ml$). Expression of TH being confirmed, two of the positive clone (MBTH2 & 8) were second transfected with GTP cyclohydrolase 1 (GTPCH 1) in pcDNA3.1 (+)-hyg followed by selection with hygromycin-B (150 $\mu /ml$) and RT-PCR confirmation. By immune-cytochemistry, these genetically modified but undifferentiated dual drug-resistant cells were found to express few of the neuronal markers, such as NF200, $\beta$-tubulin, and MAP2 as well as astroglial marker GFAP. This results suggest that over-production of BH4 by ectopically expressed GTPCH I may be involved in the induction of those markers. Transplantation of the cells into striatum of 6-OHDA- denervated PD animal model relieved symptomatic rotational behaviors of the animals. Immunohistochemical analyses showed the presence of human cells within the striatum of the recipients. These results suggest a possibility of using hES cells as a carrier of therapeutic gene(s).
본 연구는 antisense 유전자를 이용하여 웅성불임 식물체를 유기하는 웅성불임 유기 유전자의 발현을 억제함으로써 임성을 회복하기 위하여 실시하였다. 약특이 promoter(GBAN215-6 promoter)와 antisense 방향의 diphtheria toxin(DTx-A) 유전자로 제작된 pKDA215b로 담배(cv. petit Havana SR1)를 형질전환시키고 형질전환이 확인된 76개의 $R_0$ 세대를 자가수분하여 $R_1$ 세대를 획득하였다. $R_1$ 세대의 유전 분석후 antisene 유전자가 복수로 존재하는 5개의 $R_1$ 계통 (21505, 21507, 21511, 21522, 21525)을 선발하고 동형접합체를 획득하기 위해 자가수분을 하였다. 임성회복을 유도하고 회복친을 선발하기 위하여 antisense 유전자를 가진 $R_2$ 계통과 웅성불임 식물체를 교배하였다. 그 결과 꽃가루가 완전이 회복된 개체, 부분적으로 회복된 개체, 회복되지 않은 개체 등 3종류의 식물체를 획득하였으며 이들 식물체의 화분 이외의 표현형은 정상식물체와 같았고 그 기작 구명을 위해 선발하고 계속 연구가 진행되고 있다.
FMDV는 동물에서 구제역을 일으키는 병원체이며, VP1은 이 바이러스의 주요 capsid단백질이므로 구제역의 진단과 단백질 백신의 개발에 가장 많이 사용되는 재료 중 하나이다. 본 연구는 FMDV taiwan O형과 베트남에서 분리된 FMDV의 VP1 sequence를 기반으로 식물에서 VP1 유전자의 발현을 위하여 633 bp의 VP1유전자로 재편집하였으며, 이를 long-nucleotide를 사용한 multiple fragment extension 방법을 사용하여 인공적인 DNA 단편을 합성하였다. 또한 새로운 식물 형질전환 벡터로 pBI121 과 pCAMBIA1390의 장점을 수용하여, hygromycin 저항성과 CaMV 35S promoter를 포함하는 pCAMBIA II를 제작하였다. 제작된 벡터와 VP1 유전자 및 GFP유전자를 사용하여 담배를 형질 전환시켰고, 각각의 형질전환식물체내에서 전체길이의 target gene(VPl)의 성공적인 삽입을 확인하였다. 각 유전자의 발현은 RT-PCR과 Real-Time PCR의 결과로 측정하였으며, VP1 유전자의 전사가 담배 내에서 이루어졌음과 고효율의 전사체를 만드는 형질전환체 VP1-4를 선별하였다.
Thrombospondins (TSP-1, TSP-2) are secretory extracellular glycoproteins that are involved in a variety of physiological processes such as tumor cell adhesion, invasion, and metastasis. The present study was undertaken to elucidate the involvement of thrombospondins in the adhesion of osteoblast-like cells using the TSP-1 or TSP-2 antisense MG63 and MC3T3-E1 cell lines. For downregulation of TSPs expression, we prepared antisense constructs for TSP-1 and TSP-2 using the pREP4 an episomal mammalian expression vector, which be able to produce the specific antisense oligonucleotides around chromosome. MG63 and MC3T3-E1 osteoblast-like cells were transfected with the antisense constructs and nonliposomal Fugene 6, and then selected under hygromycin B (50 ${\mu}g/ml$) treatment for 2 weeks. Western blot analysis revealed that expression of the TSP proteins was downregulated in the antisense cell lines. The cell adhesion assay showed that adhesive properties of TSP-1 and TSP-2 antisense MG63 cells on the polystyrene culture plate were reduced to 17% and 21% of the control cells, respectively, and those of the TSP-1 and TSP-2 antisense MC3T3-E1 cells also decreased to 19% and 27% of control, respectively. Adhesion of TSP-1 and TSP-2 antisense MC3T3-E1 cells on Type I collagen-coated culture plate decreased to 27% and 76%, respectively. These results indicate that TSP-1 and TSP-2 proteins may have an important role in adhesion of osteoblast-like cells to extracellular matrix.
FPS (farnesyl diphosphate synthase) plays an essential role in organ development in plants. However, FPS has not previously been identified as a key regulatory enzyme in triterpene biosynthesis. In order to investigate the effect of FPS on ginsenosides biosynthesis, we over-expressed FPS of Centella asiatica (CaFPS) in Panax giseng adventitious roots. PCR analysis showed the integrations of the CaFPS and hygromycin phosphotransferase genes and we ultimately selected three lines. The result of Southern blot analysis demonstrated the introduction of the CaFPS gene into genome of ginseng. In addition, the results of RT-PCR analysis revealed that CaFPS gene overexpression induced an accumulation of its transcription in the ginseng adventitious roots. To determine whether or not the overexpression of the CaFPS gene contributes to the downstream gene expression associated with triterpene biosynthesis, the level of mRNAs was analyzed by real-time PCR. The result showed that no differences were detected in any expression of all genes. To determine quantitatively the content of ginsenosides in transgenic ginseng adventitious roots, HPLC analysis was conducted. The content of total 7 ginsenosides was increased to 1.8, 1.4, and 1.7 times than that of the controls, respectively. This indicated that the overexpression of CaFPS in ginseng adventitious roots causes an increase in ginsenoside content, although down stream genes of FPS gene were suppressed by CaFPS overexpression.
본 연구실에서 개발된 particle inflow gun (PIG)은 조작이 간편하고, 사용비용도 저렴하며, 식물 세포 내의 유전자 도입효율이 높은 특징을 갖고 있다. PIG 장비를 이용하여 벼 캘러스 내로의 유전자 도입 조건을 검토하기 위해서 사용된 vector는 pIG121Hm으로서 T-DNA 내부에 intron GUS ($\beta$-glucuronidase)와 hygromycin 및 kanamycin 저항성 유전자를 포함하고 있다. 또한 벼 캘러스 내에 물리적으로 DNA를 도입할 때에 DNA 도입 효율과 관계가 높은 요인들을 GUS의 발현빈도를 통하여 조사하였다. 그 결과 gold particle에 DNA를 부착하는 과정에 사용되는 spermidine과 calcium chloride의 경우 무첨가구에 비해 16 mM의 spermidine과 1.5 M의 calcium chloride 첨가구에서 GUS 발현율이 각각 2배, 3배 증가하였다. 그리고 1회 분사되는 gold particles양이 2 mg의 경우 가장 높은 GUS 발현율을 보여주었으며, 또한 PIG장비의 분사거리와 헬륨의 압력은 벼의 배양세포의 경우 12cm의 분사거리에서 3.5 bar (50 psi)의 헬륨압력으로 분사하였을 때 GUS 발현율이 가장 높았다. 이상의 결과에서 PIG 장비를 이용한 유전자 도입은 본 연구에서 검토한 최적의 조건을 이용하였을 경우 기존에 많이 사용되고 있는 Biolistic Gun (Bio-Rad 사)과 거의 비슷한 유전자 도입효율을 보여 주었다. 특히 PIG 장비의 경우 조작이 매우 간편하고, 분사에 사용되는 일회용 부품이 필요하지 않기 때문에 대량의 반복실험을 필요로 하는 연구에서 손쉽게 사용되리라 기대된다.
Plant secondary metabolites have always been a focus of study due to their important roles in human medicine and nutrition. We transferred the isoflavone synthase (IFS) gene into soybean [Glycine max (L.) Merr.] using the Agrobacterium-mediated transformation method in an attempt to produce transformed soybean plants which produced increased levels of the secondary metabolite, isoflavone. Although the trial to produce transgenic plant failed due to unestablished hygromycin selection, transformed callus cell lines were obtained. The induction rate and degree of callus were similar among the three cultivars tested, but light illumination positively influenced the frequency of callus formation, resulting in a callus induction rate of 74% for Kwangan, 67% for Sojin, and 73% for Duyou. Following seven to eight subcultures on selection media, the isoflavone content of the transformed callus lines were analyzed by high-performance liquid chromatography. The total amount of isoflavone in the transformed callus cell lines was three- to sixfold higher than that in control callus or seeds. Given the many positive effects of isoflavone on human health, it may be possible to adapt our transformed callus lines for industrialization through an alternative cell culture system to produce high concentrations of isoflavones.
