The Comparison of Antioxidative Activities of Sprouts Extract (발아 채소 추출물의 항산화 활성 비교)

  • This study was carried to develop the new material of functional food which antioxidant of natural substances of sprouts. We compared antioxdiative activity and antioxidant substances exist in sprouts. Antioxidaive activities of sprouts were measured by total polyphenolic acid contents, electron donating activity(EDA), SOD-liked activity and hydroxyl radical scavenging ability. Total polyphenolic acid content was very higher in Rape sprouts extract(Rap) than other sprout extracts. Also Rape extract was showed the most excellent antioxdiative activity in SOD liked activity at 86.94%. The EDA was ordered Rape sprouts extract(Rap)>Tatsoi sprouts extract(Tat)>Broccoli sprouts extract(Bro)>Alfalfa sprout extract(Alf). Hydroxy radical scavenging ability was the most effective in Rape sprouts extract(Rap). Therefore we could be certain that Rape sprouts extract(Rap) was the most effective in antioxidative activity from sprouts.

Antioxidant Activity of Fractions from 70% Methanolic Extract of Sonchus oleraceus L.

  • The potential antioxidant activities of different fractions from a 70% methanolic (MeOH) extract of Sonchus oleraceus were assayed in vitro. All of the fractions exception of n-hexane showed a strong antioxidant activity, especially the ethyl acetate (EtOAc) fraction, which showed the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity ($IC_{50}=19.25{\mu}g/mL$). The results of hydroxyl radical scavenging activity and a reducing power assay showed concentration dependence, the EtOAc fraction demonstrating a better result than the other fractions at the same concentration in the studies. Additionally, the fractions' total phenolic (TP) contents was measured, phenolic compounds such as tannic acid, p-coumatric acid, quercetin, epicathchin, and kaempferol being detected by high performance liquid chromatography (HPLC). Meanwhile, a regression analysis revealed a moderate-to-high correlation coefficient between the antiradical activity and the TP contents, suggesting that fractions obtained from the 70% MeOH extract of S. oleraceus are of potential use as sources of antioxidant material.

  • Aristolochia bracteolata is being used in traditional medicine for a variety of ailments. Three successive and two crude extracts of its leaves were subjected for in vitro antioxidant activity using seven different methods. The successive ethyl acetate and crude methanol extracts have shown potent antioxidant activity in ABTS method with $IC_{50}$ values of $17.08\;{\pm}\;0.44\;and\;28.12\;{\pm}\;2.87\;{\mu}g/ml$, respectively. The crude methanol and successive chloroform extracts have shown moderate antioxidant activity in scavenging of hydroxyl radical by p-NDA method. All the extracts have shown moderate to low antioxidant activity by other methods.

Catalpol Content and Antioxidant Activities in Various Cultivars of Rehmannia glutinosa (품종별 지황의 Catalpol 함량 및 항산화능)

  • This study was to investigate catalpol content and antioxidant activities in five cultivars (Kokang, Suwon 10, Suwon 11, Jiwhang 1 and Korea) of Rehmannia Radix. Catalpol content was analysed by HPLC using $C_{18}$ column ($150{\times}4.6\;mm$, $5{\mu}m$), 1% acetonitrile and a UV detector. The standard curve was found to be a linear regression of y=0.0483x-0.0841 ($R^2$=0.999). The catalpol content ranged from 85.2 mg/g to 144.9 mg/g, of which the Korea was the highest among cultivars. The total phenol content in Rehmannia Radix was 0.358~0.459 mg/g, of which the Korea was the highest among cultivars. The antioxidant activities determined by DPPH and hydroxyl radical scavenging activities were the highest in Korea with $IC_{50}$ of 205.8 mg/g for DPPH radical scavenging activity and $IC_{50}$ of 38.8 mg/g for hydroxyl radical scavenging activity. Based on these results, the Korea among Rehmannia Radix cultivars was suggested to be a potent medicinal plant material for antioxidant activities.

Radical Scavenging Effect and Neuroprotective Activity from Oxidative Stress of Engelhardtia chrysolepis Leaf (Engelhardtia chrysolepis의 라디칼 소거능 및 신경세포의 산화 스트레스 보호효과)

  • In this study, the radical scavenging activity and protective effect of ethanol extract from leaf of Engelhardtia chrysolepis HANCE (ECE) against oxidative stress were investigated under in vitro and cellular system. ECE showed strong radical scavenging activities in 1,1-diphenyl-2-picrylhydrazyl, hydroxyl(${\cdot}OH$) and nitric oxide(NO) radical as a concentration-dependent manner. Particularly, strong scavenging activity against the ${\cdot}OH$ and NO radical were observed with the $IC_{50}$ value of $1.30{\mu}g/ml$ and $12.61{\mu}g/ml$, respectively. Furthermore, the cellular oxidative stress was induced by amyloid beta($A{\beta}_{25-35}$) in C6 glial cells. The treatment of $A{\beta}_{25-35}$ to C6 glial cells generated high levels of reactive oxygen species(ROS) and declined cell viability. However, production of ROS was decreased by the treatment of ECE. In addition, the cell viability was significantly increased at each concentration(10, 25, $50{\mu}g/ml$) as dose-dependent manner. The Alzheimer's disease-related protein expressions in $A{\beta}_{25-35}$-treated C6 glial cells were analyzed. The ECE treatment inhibited expression of amyloid precursor protein(APP), C-terminal fragment-${\beta}(CTF-{\beta})$, ${\beta}$-site APP cleaving enzyme(BACE), phosphorylated tau(p-tau) proteins in C6 glial cells induced by $A{\beta}_{25-35}$. The present study indicated that ECE has strong radical scavenging activity and neuroprotective effect through attenuating oxidative stress.

Purification and Identification of Antioxidant Peptides from Enzymatic Hydrolysate of Spirulina platensis

  • The aim of this study was to isolate antioxidant peptides from an enzymatic hydrolysate of Spirulina platensis. A novel antioxidant peptide was obtained by ultrafiltration, gel filtration chromatography, and reverse-phase high-performance liquid chromatography, with the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay used to measure the antioxidant activity, and the sequence was determined to be Pro-Asn-Asn (343.15 Da) by electrospray ionization tandem mass spectrometry. This peptide was synthesized to confirm its antioxidant properties, and it exhibited 81.44 ± 0.43% DPPH scavenging activity at 100 μg/ml, which was similar to that of glutathione (82.63 ± 0.56%). Furthermore, the superoxide anion and hydroxyl free-radical scavenging activities and the SOD activity of the peptide were 47.84 ± 0.49%, 54.01 ± 0.82%, and 12.55 ± 0.75%, respectively, at 10 mg/ml. These results indicate that S. platensis is a good source of antioxidant peptides, and that its hydrolysate may have important applications in the pharmaceutical and food industries.

Anti-oxidant activities of ethanol extract and fractions from defatted Camellia japonica L. seeds (동백 유박 에탄올추출물 및 분획물의 항산화 활성)

  • The aim of this study was to investigate in vitro antioxidant activities of defatted Camellia japonica L. seeds (DCJS). The DCJS were extracted using ethanol and then fractionated with butanol (BuOH), ethyl acetate (EtOAc), chloroform, and hexane. To evaluate antioxidant activity of extract and fractions from DCJS, we investigated free radical scavenging activities such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+), hydroxyl radical (OH), and superoxide anion (O2-) radicals. The five extract and fractions of DCJS dose-dependently increased DPPH, ABTS+ and O2- radical scavenging activities. The BuOH fraction of DCJS showed the highest free radical scavenging activities among other extract and fractions. The contents of total polyphenol and flavonoid in BuOH fraction of DCJS were 23.26 mg GAE/g and 32.39 mg QE/g, respectively. The polyphenol and flavonoids contents of BuOH fraction has highest than other extract and fractions. In addition, BuOH and EtOAc fraction of DCJS contained 102.37 and 165.05 ㎍/g of camelliaside B, respectively. Therefore, DCJS has higher antioxidant activity and may be useful as a natural antioxidant material.

Antioxidant Activities and Inhibitory Effect on Oxidative DNA Damage of Nelumbinis Semen Extracts (연자육 추출물의 항산화 및 산화적 DNA 손상억제 활성)

  • Objective : This study was conducted to investigate the antioxidant activity and inhibitory effect on oxidative DNA damage of Nelumbinis Semen Extracts Methods : Nelumbins semen were extracted with hot-water and ethylacetate (EtOAC). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay were performed for antioxidative effect and ${\phi}X$-174 RF I DNA cleavage assay and intracellular DNA damage assay were used for inhibitory effect on intracellular DNA damage. Results : In DPPH, Hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 96.22%, 53.53%, 64.72%, while those of hot-water extracts were 20.86%, 10.72%, 29.74% at $200{\mu}g/m{\ell}$, respectively. In ${\phi}X$-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 76% and 6% at $200{\mu}g/m{\ell}$, respectively. Conclusion : These results indicated that the seed extracts of Nelumbo nucifera can be used as a natural antioxidants, which effectively inhibits the oxidative DNA damage.

Antioxidative effects of Kimchi under different fermentation stage on radical-induced oxidative stress

  • BACKGROUND/OBJECTIVES: Kimchi is a traditional Korean fermented vegetable containing several ingredients. We investigated the protective activity of methanol extract of kimchi under different fermentation stages against oxidative damage. MATERIALS/METHODS: Fresh kimchi (Fresh), optimally ripened kimchi (OptR), and over ripened kimchi (OvR) were fermented until the pH reached pH 5.6, pH 4.3, and pH 3.8, respectively. The radical scavenging activity and protective activity from oxidative stress of kimchi during fermentation were investigated under in vitro and cellular systems using LLC-$PK_1$ cells. RESULTS: Kimchi exhibited strong radical scavenging activities against 1,1-diphenyl-2-picrylhydrazyl, nitric oxide, superoxide anion, and hydroxyl radical. In addition, the free radical generators led to loss of cell viability and elevated lipid peroxidation, while treatment with kimchi resulted in significantly increased cell viability and decreased lipid peroxidation. Furthermore, the protective effect against oxidative stress was related to regulation of cyclooxygenase-2, inducible nitric oxide synthase, nuclear factor-${\kappa}B$ p65, and $I{\kappa}B$ expression. In particular, OvR showed the strongest protective effect from cellular oxidative stress among other kimchi. CONCLUSION: The current study indicated that kimchi, particularly OptR and OvR, played a protective role against free radical-induced oxidative stress. These findings suggest that kimchi is a promising functional food with an antioxidative effect and fermentation of kimchi led to elevation of antioxidative activity.

Reaction of ferritin with hydrogen peroxide induces lipid peroxidation

  • Lipid peroxidation is known to be an important factor in the pathologies of many diseases associated with oxidative stress. We assessed the lipid peroxidation induced by the reaction of ferritin with $H_2O_2$. When linoleic acid micelles or phosphatidyl choline liposomes were incubated with ferritin and $H_2O_2$, lipid peroxidation increased in the presence of ferritin and $H_2O_2$ in a concentration-dependent manner. The hydroxyl radical scavengers, azide and thiourea, prevented lipid peroxidation induced by the ferritin/$H_2O_2$ system. The iron specific chelator desferoxamine also prevented ferritin/$H_2O_2$ systemmediated lipid peroxidation. These results demonstrate the possible role of iron in ferritin/$H_2O_2$ system-mediated lipid peroxidation. Carnosine is involved in many cellular defense processes, including free radical detoxification. In this study, carnosine, homocarnosine, and anserine were shown to significantly prevent ferritin/$H_2O_2$ system-mediated lipid peroxidation and also inhibited the free radical-generation activity of ferritin. These results indicated that carnosine and related compounds may prevent ferritin/$H_2O_2$ system-mediated lipid peroxidation via free radical scavenging.
