Identification and Antioxidant Activity using Electron Spin Resonance Spectrometry of Antioxidant Producing Marine Actinomycetes Streptomyces sp. ACT-18 (항산화물질을 생산하는 해양방선균 Streptomyces Sp. ACT-18의 동정 및 Electron Spin Resonance Spectrometry를 이용한 항산화활성)

  • For the research of the natural marine antioxidant, an antioxidant-producing marine actinomycetes was isolated from sea water in Jeju coastal area. The strain was identified based on 16S rDNA sequencing, the morphology by a method of scanning electron microscopy, physiological and biochemical characteristics and cellular fatty acid analysis. The isolated strain ACT-18 was gram positive, aerobic, non-motile spores. Substrate mycelia are dark green and yellow gray aerial mycelia. The cell size of the strain was $0.5{\sim}1.0\;{\mu}m$. 16S rDNA sequence analysis showed that were Gram-positive bacteria grouped on Streptomyces sp. Results of cellular fatty acid analysis showed that major cellular fatty acids were $C_{15:0}$ anteiso (39.33%), $C_{16:1}$ cis 9 (11.96%), $C_{16:0}$ (13.08%) and $C_{17:0}$ anteiso (10.99%). The antioxidant activity of methanol extract from Streptomyce sp. ACT-18 was evaluated by measuring 1,1-diphenyl- 2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. DPPH radical scavenging activity of SBME (Streptomyces Broth Methanol Extract) A-18 was 46% at 0.1 mg/mL. Hydroxyl radical scavenging activity of SBME A-18 was 63% at 0.1 mg/mL. Alkyl radical scavenging activity of SBME A-18 was 39% at 0.1 mg/mL.

Antioxidative and Protective Activity of Polysaccharide Extract from Artemisia iwayomogi Kitamura Stems on UVB-Damaged Mouse Epidermis

  • Polysaccharide (PS) was fractionated from hot-water extract of Artemisia iwayomogi Kitamura stems. PS showed considerably higher hydroxyl radical scavenging activity than caffeic acid and glutathione. PS showed lower superoxide anion radical scavenging activity than hydroquinone and ascorbic acid. The scavenging activity of PS on the reactive oxygen species (ROS) induced by human neutrophils with zymosan was determined by the lucigenin-enhanced chemiluminescence assay. The scavenging effect of the PS on ROS as determined by the chemiluminescence assay was about 2-fold stronger than that of ascorbic acid at the same concentration. PS significantly decreased protein carbonyl and malonaldehyde contents in UVB irradiated skin homogenates, which was comparable to glutathione at the same concentration. This result suggested that PS derived from A. iwayomogi Kitamura stems may be a potent candidate as functional compound for the protection on UVB induced skin damage in cosmetics.

Growth and Antioxidant Activity on Lactic Acid Bacteria and Antimicrobial Activity on Fish Pathogenic Bacteria By Prunella vulgaris var. aleutica Fernald Extracts (꿀풀 추출물의 유산균에 대한 생육과 항산화 활성 및 어류 병원성 미생물에 대한 항균활성)

  • In this study was investigated the growth effect of Prunella vulgaris. aleutica Fernald(leaf and flower) extracts on various lactic acid bacterias, electron donating ability and hydroxyl radical scavenging activity. The total cell count of Enterococcus faecium KCCM 12118, Lactobacillus rhamnosus KCCM 32826, Lactobacillus plantarum KCCM 11542, Pediococcus pentosaceus KCCM 40464 in the absence at $37^{\circ}C$ after 48hr were $2.2{\times}10^9\;cfu/ml,\;2.1{\times}10^9\;cfu/ml,\;2.3{\times}10^9cfu/ml,\;2.2{\times}10^9\;cfu/ml$. On the other hand, the total cell count of E. faecium KCCM 12118, L. rhamnosus KCCM 32826, L. plantarum KCC 11542, P. pentosaceus KCCM 40464 in the presence of Prunella vulgaris. aleutica Fernald(leaf and flower) extracts(10%) at $37^{\circ}C$ after 48hr were $4.3{\times}10^9-4.5{\times}10^9\;cfu/ml,\;4.3{\times}10^9-4.5{\times}10^9\;cfu/ml,\;4.8{\times}10^9-4.9{\times}10^9\;cfu/ml,\;4.1{\times}10^9-4.1{\times}10^9\;cfu/ml$. The electron donating ability indicated to E. faecium KCCM 12118, L. rhamnosus KCCM 32826, L. plantarum KCCM 11542, P. pentosaceus KCCM 40464 added by 10% Prunella vulgaris. aleutica Fernald(leaf and flower) extracts, respectively. when 10% native plant extracts were added lactic acid bacterias, the electron donating ability is the highest. Hydroxyl radical scavenging activity of L. plantarum KCCM 11542, L. rhamnosus KCCM 32826, E. faecium KCCM 12118, P. pentosaceus KCCM 40464 showed higher than that of control.

Development of Extracts of Lycii folium Having High Antioxidant Activity (항산화 활성이 높은 구기엽 추출물 제조)

  • The aim of this study was to analyze the antioxidant properties of Lycii folium extracts prepared from different solvents. Lycii folium were extracted with water, 80% ethanol (80% EtOH), 80% methanol (80% MeOH) and 100% methanol (100% MeOH) in water bath at $40^{\circ}C$. The antioxidant activity of the extracts was evaluated using DPPH, hydroxyl and hydrogen radical scavenging activities, and SOD-liked activity. Total phenolic acid contents were 1.085 mg/mL in 100% MeOH, 1.382 mg/mL in 80% EtOH, 1.420 mg/mL in 80% MeOH and 1.084 mg/mL in water. DPPH radical scavenging activity of the extracts were 65.60% in 80% EtOH, 56.80% in 80% MeOH, 83.85% in 100% MeOH and 54.65% in water. Hydroxyl radical scavenging activities were 66.65% in 100% MeOH, 73.13% in 80% ethanol, 73.58% in 80% MeOH and 70.73% in water. Hydrogen radical scavenging activity of the extracts prepared from Lycii folium were 11.70% in 100% MeOH, 33.73% in 80% EtOH, 35.40% in 80% M eOH and 23.86% in water. SOD-liked activity of the extracts prepared from Lycii folium was 71.58% in 100% MeOH, 74.29% in 80% EtOH, 88.46% in 80% MeOH and 67.47% in water. Our result showed that Lycii folium extracts prepared from 80% methanol were found to be promising biomaterials with antioxidant effects.

Studies on Bioactive Substances and Antioxidant Activities of Marine Algae from Jeju Island (제주도 연안 서식 해조류에 대한 주요 활성성분 및 항산화효능 연구)

  • The objective of this study was to investigate the useful components and potential antioxidant activities of 30 marine algae, collected from Jeju Island in Korea. The contents of bioacitve substance of extracts from marine algae, such total polyphenol and carbohydrate, were determined. The extract of Distromium decumbens had the highest amount of total polyphenol content (21.27%), and that of Gracilaria incurvata Okamura had the highest amount of total carbohydrate content (10.18%). The antioxidant activities of extracts obtained from algae were tested through the evaluation of DPPH radical and hydroxyl radical scavenging activity. The extracts of Distromium decumbens, Sargassum hemiphyllum (Turner) C.Agardh, Sargassum serratifolium (C. Agardh) C. Agardh and Acrosorium yendoi Yamada were found to have more than 80% DPPH radical scavenging activity and that of Dictyota okamurae (Dawson) $H{\ddot{o}}tning$, Schnetter, et Prud'homme van Reine, Myagropsis myagroides (Martens ex Turner) Fensholt, Sargassum serratifolium (C. Agardh) C. Agardh and Cladophora wrightiana Harvey showed more than 50% hydroxyl radical scavenging activity. These results suggest that algae collected from Jeju Island would be good raw materials for antioxidant.

Antimicrobial Activities and Free Radical Scavenging Effect of Korean Folk Plants (민속식물의 항균활성 및 산화적 스트레스 개선 효과)

  • We investigated the antioxidative and antimicrobial activities of the methanol extracts from Korean folk plants (MKs) in Chungcheong Province. Among 30 MKs, 16 plants at $100{\mu}g/ml$ showed over 90% scavenging activity of 1,1-diphenyl-2-picrylhydrazy (DPPH) and 30 plants exerted the hydroxyl radical scavenging effect over 55%. Fourteen plants at the concentration $50{\mu}g/30{\mu}l$ showed strong microbial inhibition activity against Escherichia coli and Staphylococcus aureus, with clear zone greater than 11 mm in disc assays. Furthermore, the protective effect against anti-inflammatory system using RAW 264.7 macrophage cell was also studied. The treatment of LPS & INF-${\gamma}$ to RAW 264.7 cell induced nitric oxide (NO), however inhibit the formation of NO less than 50% of 5 plants. The present result indicates that the 30 species of MKs exerts protective effect of oxidative stress, antimicrobial activities and anti-inflammatory. In particular, Rhus javanica and Cornus controversa showed stronger effect on not only radical scavenging activity and inhibits growth of S. aureus but also highest protective effects from inflammation.

Whitening and Antioxidative Effect of Rice Bran Fermented by Bacillus subtilis (발효된 미강의 항산화 효과 및 미백 효과)

  • In this study, we investigated rice bran fermented by Bacillus subtilis (RFB) at $40^{\circ}C$ for 36 h to develop a new natural antioxidant and whitening agent for new natural cosmetics. RFB showed reactive oxygen species (ROS) scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide radical scavenging activities and hydroxyl radical scavenging activities at a concentration of $100 \;{\mu}L$/mL. DPPH, superoxide radical scavenging activities and hydroxyl radical scavenging activities were higher in the RFB than in the RB. Inhibitory activity on auto-oxidation of linoleic acid is also highest at a concentration of $100 \;{\mu}L$/mL. RFB showed the higher inhibitory activity than RB in auto-oxidation. RFB reduced intracellular tyrosinase activity about 80 % at a concentration of $100 \;{\mu}L$/mL. Therefore, we suggest that RFB could be used as a useful antioxidant and whitening agent.

Schisandra Chinensis Inhibits Oxidative DNA Damage and Lipid Peroxidation Via Antioxidant Activity

  • Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties

Change of Antioxidant Activities in Carrots (Daucus carota var. sativa) with Enzyme Treatment (효소처리 가공이 당근(Daucus carota var. sativa)의 항산화 활성 변화에 미치는 영향)

  • The purpose of this research is to minimize the loss of nutrients in carrots (Daucus carota var. sativa). A protopectinase was used to enzymatically macerated and separate cells without damage. The enzyme modification group's collection rate was 81% (residue rate 19%), while the grinding process group's collection rate was 56% (residue rate 44%)-an over 20% of collection rate difference. Thus we predicted a big difference in transference number after the process and wastage. In comparing ingredient changes in the enzyme modification group versus the grinding process group, the content of ${\beta}$-carotene (the carrot's main ingredient) showed a change in protection factor (PF) ($2.2{\pm}0.2$ PF, $1.4{\pm}0.4$ PF, respectively), total polyphenol content ($89{\pm}3.42{\mu}g/g$, $64{\pm}4.16{\mu}g/g$, respectively), and total flavonoid content ($68{\pm}2.73{\mu}g/g$, $41{\pm}3.26{\mu}g/g$, respectively). Thus we confirmed that nutrient destruction, due to cell membrane preservation, occurred less often in the enzyme modification process than the mechanical grinding process group. We also measured DPPH radical scavenging activity, hydroxyl radical scavenging activity, and nitrite scavenging activity. DPPH radical scavenging activity was $87{\pm}0.29%$ and $74{\pm}1.56%$ in the enzymatic modification group compared to the mechanical grinding process group, respectively. Hydroxyl radical scavenging activity was $44{\pm}0.49%$ and $32{\pm}0.48%$ in the enzymatic modification group compared to the mechanical grinding process group, respectively. Nitrite scavenging activity was $59{\pm}0.53%$ and $46{\pm}0.62%$ in the enzymatic modification group compared to the mechanical grinding process group, respectively. Our results show that cell membrane preservation, via the protopectinase enzyme process, decreases the loss of nutrients and still preserves inherent antioxidants.

Analysis of the Antioxidant Properties of 2,2-diphenyl-1 Picrylhydroazyl, Hydroxyl Radicals, and Nitric Oxide in Alaska Pollock Roe, with or without Natural Fermented Seasoned (알래스카 명란의 DPPH, OH, NO의 항산화 특성 분석)

  • Alaska pollock (Gadus chalcogrammus) is a marine fish species belonging to the family Gadidae. In this study, whether the Alaska Polloack Roe have antioxidant properties, 2,2-diphenyl-1 picrylhydroazyl (DPPH), hydroxyl radical (OH) reducing activity, and nitric oxide radical (NO) scavenging activity were evaluated in distilled water extract (DWE) and ethanol extract (ETE) of raw Alaska pollock roe, Gochujang Pollock roe, and fermented seasoned Pollock roe. The DPPH scavenging activity of the DWE with Gochujang Pollock roe was 71.9% at a concentration of 1.0 mg/ml and that of the ETE was 73.7% at the same concentration. The DPPH scavenging activity of the DWE with fermented seasoned Pollock roe was 78.0% at 1.0 mg/ml, whereas that of ETE was 78.4% at the same concentration. The $IC_{50}$ values of the DWE and ETE of raw Pollock roe for DPPH were $11.65{\mu}g/ml$ and $11.47{\mu}g/ml$, respectively. The OH scavenging activities of raw Pollock roe, Gochujang Pollock roe, and fermented seasoned Pollock roe ethanolic extracts at a concentration of 1.0 mg/ml were 70.9%, 79.0, and 80.6%, respectively. The $IC_{50}$ values of the DWE and EWE of raw Pollock roe for NO were $11.45{\mu}g/ml$ and $11.41{\mu}g/ml$, respectively. The DPPH, OH, and NO scavenging abilities in DWEs and ETEs of Gochujang and fermented seasoned Pollock roe were higher than those of instant (no Gochujang or season treatment) treatment Pollock roe. Both the Gochujang and fermented seasoned Pollock roes have natural radical scavenging ability and may be useful potential antioxidant food supplements.
