Hyaluronidase Inhibitory Activity of Extracts from Doenjang, Chungkookjang and Miso (된장, 청국장 및 미소추출물의 Hyaluronidase 저해활성)

  • The inhibitory effects of water and methanol extracts of Doenjang, Chungkookjang and Miso on bovine hyaluronidase were examined. The extracts were vacuum-dried and the resulting pellets were dissolved with 0.1 M acetate buffer with 1$\%$ DMSO to a final concentration of 50 mg/mL. The water extracts of Doenjang, Chungkookjang and Miso inhibited 62$\%$, 70$\% $, 56$\%$ of hyaluronidase activity, respectively, while the extract of crushed soybean inhibited 24$\%$ of the activity. Under the same condition, disodiumcromoglycate known as an anti-allergic drug inhibited 46$\%$ of hyaluronidase activity at the concentration of 0.35 mg/mL as a positive control. Also the methanol extracts of Doenjang, Chungkookjang and Miso inhibited 48$\%$, 52$\%$, 70$\%$ of hvaluronidase activity, respectively, while the extract of crushed soybean inhibited 46$\%$ of the activity. After heat treatment of the water extracts of Doenjang, Chungkookjang and Miso at 100"C for 10 min, hyaluronidase inhibitory effects of them were reduced approximately to half. However, hyaluronidase inhibitory effects of methanol extracts were maintained well even after heat treatment.

Parthenogenetic development of mouse eggs I. Parthenogenetic activation by ethanol and hyaluronidase treatments (생쥐 난자의 단위발생에 관한 연구 I. Ethanol 및 hyaluronidase처리에 의한 단위발생유기)

  • This experiment was carried out to find out the best condition for the parthenogenetic activation of mouse eggs by treating ethanol and hyaluronidase. For the parthenogenetic activation of eggs with ethanol, cumulus cell enclosed or denuded eggs were treated with 7% ethanol in D-PBS for 5, 7 or 9 minutes. For the activation of eggs with hyaluronidase, the eggs with cumulus masses were released into D-PBS with 100 unit hyaluronidase and treated for 10, 12 or 13 minutes. All of the treated eggs were incubated in BMOC-3 solution for 5 hours at $37^{\circ}C$ at an atmosphere of 5% $CO_2$ in air. The types of parthenogenetic eggs were morphologically classified into haploid, diploid, immediate cleavage eggs under an inverted microscope. The results obtained in this experiment were summarized as follows ; 1. High activation rate(99%) had been achieved by treating the eggs with 7% ethanol for 7 minutes. 2. With 100 IU hyaluronidase, high activation rate (94%) had been achieved by treating for 12 minutes. 3. The most frequent type of parthenogenetic eggs activated with ethanol or hyaluronidase was haploid (p<0.05). 4. The eggs collected from 18 to 22 hours post HCG injection showed higher activation rate than the eggs collected at 16 hours post HCG injection. 5. No significant difference (p>0.05) in activation rate was shown in strain of mouse and in presence of cumulus cells.

생약재 추출물의 hyaluronidase 저해 및 라디칼 소거활성 검색

  • For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined in vitro their hyaluronidase inhibitory effect and radical scavenging activity. Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix and Moutan Radicis Coritex showed more than 50% hyaluronidase inhibitory effects. Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed more than 90% in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical showed significantly activity.

  • Solvent extracts of 17 plants collected in Cheju Island were investigated for their biological properties related to cosmeceuticals such as tyrosinase and hyaluronidase inhibition and also radical scavenging effects. The chloroform fraction of Phytolacca esculenta root exhibited strong inhibition against tyrosinase activity. No fraction showed significant hyaluronidase inhibition. Some solvent extracts of plants such as Achyranthes japonica and Artemisia princeps showed considerable radical scavenging activities.

  • This study was carried out to evaluate glutathione S-transferase (GST) activity and hyaluronidase inhibitory effect of medicinal plants. The EtOH extracts of 20 species plants were tested. As the result, Acorus gramineus and Pueraria lobata exhibited GST activity. On the continuous experiment, the n-BuOH fraction of Acorus gramineus and the $H_2O$ fraction of Pueraria lobata showed the elevation of GST activity. On the experiment of hyaluronidase inhibitory effect, Acorus gramineus exhibited a potent inhibitory activity. These results suggest that the extract of Acorus gramineus can be applicable for the development of a new anti-inflammatory agent.

The Effect of Transforaminal Epidural Block with Hyaluronidase and Triamcinolone (Hyaluronidase를 사용한 경추간공 경막외 차단의 효과)

  • Background: Epidural steroid injections benefit certain patients with radicular pain, and often have only a limited duration. We compared the efficacy of hyaluronidase and triamcinolone and triamcinolone alone in patients with lumbar herniated disc disease treated with transforaminal epidural block. Methods: Forty patients who had undergone a transforaminal epidural injection were retrospectively reviewed. The T group received triamcinolone and local anesthetics; whereas, the HT group received hyaluronidase, triamcinolone and local anesthetics. We evaluated the improvement as being good, moderate, mild or no improvement, and in those where the improvement was good or moderate, also evaluated the duration of pain relief. Data were collected from the medical records of patients or via phone calls, which were analyzed using Student t- and chi-squared tests. A value of P < 0.05 was considered significant. Results: There were no significant differences in the degree of pain improvement or duration of pain relief between the two groups. Conclusions: A hyaluronidase and triamcinolone injection during transforaminal epidural block has on benefit with respect to the degree of pain improvement or its duration compared to a triamcinolone only injection.

Optimization of Bovine Testicular PH-20 hyaluronidase Production in Pichia pastoris (소의 히아론산 분해효소(PH-20)의 Pichia pastoris에서의 생산 최적화)

  • Bovine testicular hyaluronidase PH-20 was cloned into pPIC9 vector and expressed in Pichia pastoris. Recombinant PH-20 was 75 kDa MW and 7460 units/L activity. Extracellular hyaluronidase activity was two times higher than that of intracellular activity. Non-buffered medium and $30^{\circ}C$ cultivation was favorable for PH-20 production. 1M sorbitol as an osmotic pressure and 0.3% methanol inducer increased cell growth and enzyme activity. 0.4 M arginine augmentation decreased the proteolytic degradation of recombinant hyaluronidase.

Control of spermatozoa penetration and polyspermy by cumulus cells in porcine oocytes matured in culture

  • The functional role of the cumulus cells on sperm penetration and polyspermy during in vitro fertilization was examined. The penetration rate was significantly higher(p<0.01) in oocytes with(61%) than without(25%) cumulus cells. No significant differences, however, was observed in polyspermy. When the hyaluronidase was supplemented to the fertilization medium with different concentrations, penetration rates in oocytes with cumulus cells were higher than oocytes without cumulus cells at 0(61 vs 34% ; p<0.05), 0.01(56 vs 35% ; p<0.05), 0.1(66 vs 30% ; p<0.05) and 1.0 mg/$m\ell$(39 vs 27%). On the other hand, the polyspermy rates were lower oocytes without than with cumulus cells, and had a tendency to decrease with high concentrations of hyaluronidase. In another experiment, the penetration and polyspermy rates had a tendency to increase as time of sperm-oocytes culture was prolonged. At 16 and 20hrs after insemination, the penetration rates were significantly higher(p<0.05) in oocytes with(48 and 62% for 16 and 20hrs) than without(25 and 31% for 16 and 20hrs) cumulus cells in medium with hyaluronidase. However, the polyspermy rates were significantly(p<0.05) lower in oocytes without(3 and 16%) than with(37 and 48%) cumulus cells at 16 and 20hrs after insemination. In cumulus-free oocytes inseminated in medium with or without hyaluronidase at different concentrations of cumulus cells, the penetration rates were significantly(p<0.05) higher in medium with than without hyaluronidase at different concentrations of cumulus cells. The proportions of polyspermy were lower in medium without than with hyaluronidase at 0 (10 vs 0%), 10$^2$(25 vs 0%), 10$^4$(24 vs 14%) and 10$\^$6/(29 vs 10% ; p<0.05) cumulus cells/ml. These results suggest the advantage of culture in medium with cumulus cells and denuded oocytes to inhibit polyspermy with no decrease in the penetration rates during the fertilization in vitro in the porcine.

  • Hyaluronidases enhance therapeutic drug transport by breaking down the hyaluronan barrier to lymphatic and capillary vessels, facilitating their tissue absorption. Commercially available hyaluronidases are bovine in origin; however, they pose risks such as bovine spongiform encephalopathy. The present study aimed to develop a novel, highly active hyaluronidase and assess its function. Therefore, in order to find the most efficient active hyaluronidase, we produced several shortened hyaluronidases with partial removal of the N- or C-terminal regions. Moreover, we created an enzyme that connected six histidines onto the end of the hyaluronidase C-terminus. This simplified subsequent purification using $Ni^{2+}$ affinity chromatography, making it feasible to industrialize this highly active recombinant hyaluronidase which exhibited catalytic activity equal to that of the commercial enzyme. Therefore, this simple and effective isolation method could increase the availability of recombinant hyaluronidase for research and clinical purposes.

Anti-skin aging activities of ethanol extract from Echinodorus cordifolius L. in human keratinocytes (물수선화 에탄올 추출물의 피부 노화 억제 효과)

  • Echinodorus cordifolius (L.) is an aquatic plant in the family Alismataceae. The anti-skin aging activity of E. cordifolius (L.) has not been yet reported. Therefore, the objective of the present study was to prepare 70% ethanol extract (ECEE) from E. cordifolius (L.) and investigate their antioxidant and anti-hyaluronidase activities for confirm the potential of anti-skin aging. ECEE showed good activities of DPPH, hydrogen peroxide scavenging, and hyaluronidase inhibition, with EC50 and IC50 values of 31.4, 300, and 450 ㎍/mL, respectively. ECEE also significantly improved cell viability and inhibited intracellular reactive oxygen species dose-dependently against 1 mM hydrogen peroxide-induced oxidative stress in immortalized human keratinocytes (HaCaT cells). Furthermore, ECEE upregulated hyaluronic acid (HA)-synthesizing enzyme hyaluronan synthase 2 (HAS2) expression level, but downregulated expression level of HA-degrading enzyme hyaluronidase 2, resulting in increased HA production in HaCaT cells. Taken together, these results suggest that ECEE shows antioxidant and anti-hyaluronidase potential and could be a functional cosmetic ingredients for anti-skin aging.
