A Cell-Based Assay System for Monitoring NF-$\kappa$B Activity in Human Epidermal Keratinocytes: A Screening Tool of the Antioxidants and Anti-inflammatories for Dermatological Purpose

  • A cell-based assay system for monitoring NF-$textsc{k}$B activity was developed to determine the influence of activated NF-$textsc{k}$B in human HaCaT cells. The pNF-$textsc{k}$B-SEAP-NPT plasmid that permits expression of the secreted alkaline phosphatase (SEAP) reported gene in response to the NF-$textsc{k}$B activity and contains neomycin phosphotransferase (NPT) gene for the geneticin resistance in host cells was constructed and transfected into human keratinocyte cell line HaCaT. Human HaCaT transfectant cells secreted the SEAP enzyme into the culture medium in a time-dependent manner until 72h. NF-$textsc{k}$B activities were measured in the SEAP reporter gene assay using a fluorescent detection method. The treatment of HaCaT cell transfectants with known antioxidants [e.g., N-acetyl-L-cysteine and vitamin C] showed inhibition of NF-$textsc{k}$B activity in a time-and concentration-dependent manner. The phorbol 12-myristate 13-acetate (PMA) known as a stimulator of NF-$textsc{k}$B expression demonstrated that it increased NF-$textsc{k}$B activity in a time- and concentration-dependent manner. This assay system could be used to determine the quantitative measurement of NF-$textsc{k}$B activity in the human skin and allow the screening of anti-inflammatory agents from various synthetic chemicals and natural products for dermatological purpose. 조협 추출물이 HaCaT cells의 항염증과 SEB-1 cells의 피지분비 억제에 미치는 영향 (The Effects on Anti-inflammatory Action in HaCaT Cells and Inhibiting Sebum Secretion in SEB-1 Cells by Gleditsiae Fructus Extract)

  • Objectives The purpose of this study is to investigate the effects of Gleditsiae Fructus 70% EtOH extract (JS_E) on anti-inflammatory action in HaCaT cells (A spontaneously immortalized human keratinocyte cell line) and inhibiting triglyceride genesis in SEB-1 cells (Immortalized human sebocyte). Methods The anti-inflammatory effect of JS_E was analyzed by enzyme-linked immunosorbent assays (ELISA) which measured levels of IP-10, RANTES and MDC in HaCaT cells. Also the effect on secretion of sebum of JS_E was analyzed by TG-S kit which measured the quantity of triglyceride in SEB-1 cells. Results JS_E inhibited IP-10, RANTES and MDC expression in a dose dependent manner. IP-10 expression was inhibited significantly in comparison to TNF-${\alpha}$ and IFN-${\gamma}$ recombination (TI) control group at concentration of JS_E $200{\mu}g/ml$ and RANTES and MDC expressions were inhibited significantly at concentration of JS_E 100, $200{\mu}g/ml$. JS_E also inhibited triglyceride secretion of SEB-1 cells significantly in comparison to the control group in a dose dependent manner. Conclusions This study shows that JS_E has the effects of anti-inflammatory action and inhibiting sebum secretion. According to these results, JS_E can be used for treating skin diseases such as acne and dermatitis caused by inflammation and excessive secretion of sebum by controlling the activity of the HaCaT and SEB-1 cells.

황기의 자외선에 의한 세포 손상을 막는 보호 효과 (The Protective Effects of Astragali Radix Against UV Induced Cellular Damage in Human Keratinocytes)

  • The root of Astragalus membranaceus Bunge (Leguminosae) has been used in the Korean oriental medicine for strengthening the vital energy. UV irradiation has been suggested as a major cause of photo aging in skin. In order to investigate protective effects against UV induced cellular damage, Astragali Radix was extracted with 70% ethanol and dissolved in DMSO. The protective effect was detected by MTT assay, LDH assay, and Comet assay in immortalized human keratinocyte cell line, HaCaT cell system after UV irradiation. Astragli Radix 70% EtOH extract reduced UV induced cellular damage in cell survival, membrane integrity and DNA damage.

Transcription Profiles of Human Cells in Response to Sodium Arsenite Exposure

  • Arsenic exposure is associated with several human diseases, including cancers, atherosclerosis, hypertension, and cerebrovascular diseases. In cultured cells, arsenite, an inorganic arsenic com-pound, was demonstrated to interfere with many physiological functions, such as enhancement of oxidative stress, delay of cell cycle progression, and induction of structural and numerical changes of chromosomes. The objective of this study is to investigate the effects of arsenic exposure on gene expression profiles by colorimetric cDNA microarray technique. HFW (normal human diploid skin fibroblasts), CL3 (human lung adenocarcinoma cell line), and HaCaT (immortalized human keratinocyte cell line) were treated with 5 $\mu\textrm{M}$ or 10 $\mu\textrm{M}$ sodium arsenite for 6 or 16 h, respectively. By a dual-color detection system, the expression profile of arsenite-treated cultures was compared to that of control cultures. Several genes expressed differentially were identified on the microarray membranes. For example, MDM2, SWI/SNF, ubiquitin specific protease 4, MAP3K11, RecQ protein-like 5, and Ribosomal protein Ll0a were consistently induced in all three cell types by arsenite, whereas prohibitin, cyclin D1, nucleolar protein 1, PCNA, Nm23, and immediate early protein (ETR101) were apparently inhibited. The present results suggest that arsenite insults altered the expression of several genes participating in cellular responses to DNA damage, stress, transcription, and cell cycle arrest.

진세노사이드 Rb1과 Rg1에 의한 HaCaT 피부각질세포의 전사체 분석 (Transcriptome Analysis of Human HaCaT Keratinicytes by Ginsenosides Rb1 and Rg1)

  • 인삼(Panax ginseng C. A. Meyer)의 주요 생리활성물질인 진세노사이드(ginsenoside) Rb1과 Rg1의 효능검증 및 작용점을 규명하고자 HaCaT 피부각질세포에서 유전체 분석(gene expression profiles)을 실시하였다. 진세노사이드 Rb1과 Rg1 각각의 처리 농도 및 시간에 따른 HaCaT 세포에 대한 세포독성은 나타나지 않았으며, $10{\mu}g/mL$의 진세노사이드 Rb1과 Rg1 각각을 6 및 24 시간 처리하여 유전체 분석 결과, 진세노사이드 Rb1과 Rg1의 24 시간 처리군에서 항노화 및 피부탄력 관련 유전자인 fibroblast growth factor (FGF2)의 활성이 증가된 것으로 나타났다. 또한 진세노사이드 Rb1의 24 시간 처리군에서는 항산화 작용점에 있는 일련의 유전자군, FANCD2, FGF2, LEPR, FAS 등의 활성을 확인하였다. 향후 확인된 항노화 및 피부탄력 관련 주요인자들의 작용 및 상관관계를 구체적으로 확인하고, 특히 진세노사이드 Rb1의 신호전달을 완성하고자 한다.

  • 본 연구에서는 증폭시킨 홍삼으로부터 분리한 ginsenoside Rh2(Rh2)와 compound K(CK)의 융복합적 항염증 및 항암효과를 연구하여 홍삼 내 유용한 기능성분에 대한 기초 자료를 제공하고자 한다. 이에 Hep3B에서의 세포독성과 IL-6 유도 STAT3 루시퍼라아제 활성, B16F10과 Hacat 세포의 생존 농도를 측정하였고 Apoptosis와 관련된 분자의 발현양상을 확인하기 위해 FAC (fluorescence activated cell sorting) 분석을 수행하였다. 실험결과 Rh2, CK mixture가 10 ug/ml일 때 Hep3B 세포에서 세포독성이 없고 IL-6 감소율이 102%로 항염증 효과가 있는 것으로 나타났다. 또한 Rh2, CK mixture 50 uM에서 melanoma 세포인 B16F10과 human keratinocyte인 Hacat에서 독성을 보여 사멸하는 것을 관찰하였다. FACS 분석 결과 annexin V가 발현되지 않고 흑색종 세포와 keratinocyte가 탈착되면서 사멸되는 것을 확인하였다. 이러한 현상을 통하여 사멸되는 메카니즘이 anoikis 방식의 세포사멸로 인한 것으로 추정할 수 있으며 그것에 대한 명확한 세포사 신호 체계 규명을 위하여 향후 세포부착 단백질의 변화에 대한 연구가 필요할 것으로 판단된다.

Mitochondria-Targeted Vitamin E Protects Skin from UVB-Irradiation

  • Mitochondria-targeted vitamin E (MVE) is designed to accumulate within mitochondria and is applied to decrease mitochondrial oxidative damage. However, the protective effects of MVE in skin cells have not been identified. We investigated the protective effect of MVE against UVB in dermal fibroblasts and immortalized human keratinocyte cell line (HaCaT). In addition, we studied the wound-healing effect of MVE in animal models. We found that MVE increased the proliferation and survival of fibroblasts at low concentration (i.e., nM ranges). In addition, MVE increased collagen production and downregulated matrix metalloproteinase1. MVE also increased the proliferation and survival of HaCaT cells. UVB increased reactive oxygen species (ROS) production in fibroblasts and HaCaT cells, while MVE decreased ROS production at low concentration. In an animal experiment, MVE accelerated wound healing from laser-induced skin damage. These results collectively suggest that low dose MVE protects skin from UVB irradiation. Therefore, MVE can be developed as a cosmetic raw material.

Methylanthranilate, a Food Fragrance Attenuates Skin Pigmentation through Downregulation of Melanogenic Enzymes by cAMP Suppression

  • Methyl anthranilate (MA) is a botanical fragrance used in food flavoring with unexplored potential in anti-pigment cosmetics. MA dose-dependently reduced melanin content without affecting cell viability, inhibited dendrite elongation and melanosome transfer in the co-culture system of human melanoma cells (MNT-1) and human keratinocyte cell line (HaCaT), and downregulated melanogenic genes, including tyrosinase, tyrosinase-related protein 1 and 2 (TRP-1, TRP-2). Additionally, MA decreased cyclic adenosine monophosphate (cAMP) production and exhibited a significant anti-pigmentary effect in MelanodermTM. These results suggest that MA is a promising anti-pigmentary agent for replacing or complementing existing anti-pigmentary cosmetics.

연잎, 연꽃, 연꽃 수술 추출물이 UVB 자외선 조사에 의한 각질형성세포의 보호 및 피부 노화 방지에 미치는 영향 (The Effects of Nelumbo nucifera on Utraviolet-B Irradiated human Keratinocytes)

  • Objectives : The purpose of this study was to investigate the anti-aging effects on cultured human keratinocytes with Nelumbo nucifera extracts. Methods : Each parts of leaves, flowers and stamen were extracted with water or 70% ethanol. These extracts were tested for cell viability on HaCaT cells (human keratinocyte line) by MTT assay. We investigated the effects of Ultraviolet-B (UVB) irradiation on cytotoxicity and lipid peroxidation in cultured skin keratinocytes. Results : The ethanol extract of Nelumbo nucifera flowers showed maximun cell viability as 111.39% in 30 ug/ml concentration. The water extracts of stamen, flowers, leaves showed cell viability as 107.12, 101.65, 101.46%, respectively. HaCaT keratinocytes were survived 63.06% at $20mJ/cm^2$ UVB irradiation. The cell membrane lipid peroxidation was measured by accumulation malondialdehyde (MDA). The levels of MDA were decreased by the ethanol extract of Nelumbo nucifera flowers and the water extracts of stamen. Conclusions : These finding suggest that the ethanol extract of Nelumbo nucifera flowers prevent anti-aging effects on cultured human keratinocytes during UVB irradiation.

  • Ceramium boydenii belongs to euphorbia humitusa of red algae, and is distributed along the coast of Jeju island. This study was conducted to examine the anti-inflammatory effect and action mechanism of C. boydenii in the human keratinocyte cell line HaCaT. The 80% EtOH extract of C. boydenii inhibits the production of MDC (macrophage derived chemokine), an inflammatory chemokine, induced by IFN-${\gamma}$ and TNF-${\alpha}$ in a concentration dependent manner. It also suppressed the phosphorylation and nuclear translocation of STAT1, a key transcription factor in IFN-${\gamma}$ and TNF-${\alpha}$ signaling pathway, at the effective concentrations. These results suggest that C. boydenii demonstrates the anti-inflammatory activity via the suppression of STAT1 activation and has the significant value as an anti-inflammatory source.
