Study on the Regional Deposition of Smoke Particles in Human Respiratory Tract under the Variation of Fire and Breathing Conditions (화재 및 호흡조건 변화에 따른 연기입자의 인체 호흡기 내 영역별 침착량 분석)

  • Smoke generated in a fire consists of gaseous substances and particulate matter, such as unburned carbon that adsorbed the gases. Human injury caused by inhalation of gaseous substances present in smoke is mostly short-term, whereas damage caused by inhalation of particulate matter is relatively a long-term phenomenon depending on the state of the gas-phase adsorption. The amount and location of the deposited smoke particles are important factors in estimating the damage caused to humans, which are affected by the breathing conditions as well as particle conditions, such as the size and concentration affected by the combustion conditions. In this study, in order to understand the characteristics of the deposition of smoke particles in the respiratory tract related to the study of human smoke inhalation injury, the number and mass concentration of smoke particles deposited in different areas of the respiratory tract for different fuel types, combustion conditions and breathing conditions were calculated. In addition, the amount of mass deposition of smoke in the respiratory tract for a certain period of inhalation was compared with the atmospheric standard of fine dust.

Estrogen Mediates Ischemic Damage and the Migration of Human Umbilical Cord Blood Cells

  • Human umbilical cord blood cells(HUCBC) are rich in mesenchymal progenitor cells, endothelial cell precursors and hematopoietic cells. HUCBC have been used as a source of transplantable stem and progenitor cells. However, little is known about survival and development of HUCBC transplantation in the CNS. Estrogen has a neuroprotective potential against oxidative stress-induced cell death so has an effect on reducing infarct size of ischemic brain. We investigated the potential use of HUCBC as donor cells and tested whether estrogen mediates intravenously infused HUCBC enter and survive in ischemic brain. PKH26 labeled mononuclear fraction of HUCBC were injected into the tail vein of ischemic OVX rat brain with or without $17\beta$-estradiol valerate(EV). Under fluorescence microscopy, labeled cells were observed in the brain section. Significantly more cells were found in the ischemic brain than in the non-ischemic brain. HUCBC transplanted into ischemic brain could migrate and survive. Some of cells have shown neuronal like cells in hippocampus, striatum and cortex tissues. These result suggest that estrogen reduces ischemic damage and increases the migration of human umbilical cord blood cells. Safety countermeasures for the marine casualties of fishing vessels in Korea (우리나라의 어선 해양사고에 대한 안전대책)

  • Marine casualties of fishing vessels were analyzed to reduce the sacrifice of human life using data of the Korean Maritime Safety Tribunal from 1995 to 2004 in Korea. The occurred number of fishing vessel casualties were likely to be higher portion than non-fishing vessels, but the occurring ratio of fishing vessel casualties were marked 2.96 times lower than that of non-fishing vessel casualties. The occurring ratios of bigger fishing vessel casualties were higher than smaller ones. Most marine casualties were resulted from the human factors such as poor watchkeeping, negligent action for engine and etc. The trend of marine casualties showed that the machinery damage hold the first and collision accidents took the second, but on a point of cause of them, operating errors took first and poor handling or inspection of machinery held the second place. Because those two casualties took major portion, and very important problems for safety of fishing vessels, so we ought to try to reduce the factors before everything else. In addition, since collision, sinking and capsizing in marine casualties have led to death, missing and injury of lives, it is necessary for navigation operators to take more educations and training intended to reduce the marine casualties systematically and continuously.

Metronidazole Reduced Ammonia Toxicity in Human Hep G2 cell and Rat Hepatocytes (Hep G2 세포와 rat 간세포에서 Metronidazole에 의한 암모니아 독성 감소)

  • Lipophilic ammonia is toxic gas and can easily diffuse across cell membranes. Excess ammonia is implicated in the pathogenesis of several metabolic disorders including hepatic encephalopathy and may result in the death. The purpose of this study was to clarify the inhibition effect of metronidazole on liver cell damage due to ammonia in human Hep G2 cell and rat hepatocytes. The effects of metronidazole were studied in ammonium chloride treated human Hep G2 cell (75 mM) and rat hepatocyte (100 mM) following $0.1{\mu}M$ metronidazole treatment. In MTZ+AC group, cell viabilities increased prominently and LDH activities decreased over 25% than AC group. Furthermore, ammonia level according to ammonium chloride treatment reduced over 30% and lipid peroxidation as an index of cell membrane damage decreased more than twice. By comparison with control, catalase activity showed more than 30% reduction in AC group while less than 10% reduction in MTZ+AC group, respectively. In addition, MTZ+AC group showed the similar cell structure as control in cell morphology study by using light microscope, and represented fluorescent intensity decrement compared with AC group in fluorescent microscopic study with avidin-TRITC fluorescent dye. And cleaved PARP expression due to ammonia reduced twofold or more in MTZ+AC group. As the results suggest, metronidazole may protect the liver cell by inhibiting cell damages due to ammonia and be used for an effective antagonist of ammonia in hyperammonemia.

Effect of Arresting MCF-7 Human Breast Carcinoma Cell at G2/M Phase of Trichosanthes Kirilowii (천화분이 MCF-7 유방암 세포주의 G2/M 세포주기 억제에 미치는 영향)

  • The purpose of this study is to investigate the anti-proliferative mechanism by Trichosanthes kirilowii (TCK) in MCF-7 human breast carcinoma cell. In this study, we used human breast cancer cell line, Michigan cancer foundation-7 cells (MCF-7 cells). They were co-incubated with 30~200 ${\mu}g$/ml TCK for 48 hours, and cell viability was measured by Water-soluble tetrazolium salt-1 (WST-1) assay. After MCF-7 cells were exposed to 60 ${\mu}g$/ml of TCK for 0, 3, 6, 12, 24, 48 hours, We performed flow analysis cytometry sorting(FACS) and western blot analysis. We investigated the effect of dose-dependent cell growth inhibition by TCK, which could be proved by WST-1 assay. Also, flow cytometry analysis showed that TCK increased percentage of subG1 phase and G2/M phase cell cycle. In addition, TCK induced apoptosis through the expression of caspase-9, -3 and poly(ADP-ribose) polymerase(PARP) activation. Moreover, we showed that ATM-dependent G2/M phase arrest by DNA damage and phosphorylation of chk2, cdc25C, cdc2(Tyr15). Taken together, these results suggest that by G2/M phase arrest through DNA damage and inducing of apoptosis through intrinsic pathway, TCK may have potential tumor suppressor in breast cancer.

A Strategic Study on National Disaster Medical System (국가재난의료체계에 대한 정책적 고찰)

  • Due to major disasters Korea has been damaged, and they caused lots of casualties: for last ten years natural disasters caused 1288 deaths including missing people; human disasters including industrial disasters brought as many as 4,512.148 casual ties (126,372 deaths with 4,385,400 injuries); and they cost 44.1 trillion property damage. However, even though major disasters have brought about tremendous human loss and property damage, Koreas National Disaster Medical System to rescue casualties is insufficient, and it has not been activated. Fortunately, through major disaster management process, the National Disaster Management System has been developed, increasing its own efficiency, and resulting in to organize an Office of Firefighting and Prevention of Disasters under the central government. Considering the value of human lives, the disaster medical part, in the U.S.A. as well as in Korea, must have an independent organization in the government, not as one sector of the government department. It will have its own organizational structure, such as disaster planning, operation, and logistics, and interact with central and local government or between local government agencies. So each agency will cooperate and supply resources interchangeably. Also, with the system of disaster management and restoration, the disaster medical system must be advanced in keeping step. Its role must be extended due to the possibility of biological terror or SARS around the world, resulting in severe casualties. Korea has the Emergency Medical Service System based on the regulation of emergency medical care, yet it is a part of the National Disaster Management System. It must be managed independently apart from it. As we see the emergency medical technicians playing as the backbone in disaster medical care in the US, we should have legal foundations for Koreas emergency medical technicians, emergency medical providers, to participate in rescue operation actively. At the same time, we need to have a national register system to classify disaster medical resources, and a total plan to place resources according to the impact of disaster, and how to organize teams. We also need to draw up a scheme to activate civil disaster medical resources, as integrating public and private or voluntary organizations.

Cytotoxicity and DNA Damage in Human Lung Cells Treated with Lanthanum Oxide and Neodymium Oxide (산화란타늄, 산화네오디뮴이 세포독성 및 DNA손상에 미치는 영향)

  • Objectives: The present study investigated cytotoxicity and DNA damage in human lung cells in vitro. Methods: Neodymium oxide and lanthanum oxide were dispersed by ultrasonic treatments. The assay was performed with MRC-5 (Human male fetus lung cell). Cytotoxicity and comet assay of lanthanum oxide and neodymium oxide were measured after 24 and 48 hours incubation. Results: After 24 hours of exposure to rare earth metals, the cytotoxicities of lanthanum oxide in more than $1{\mu}M$ concentration groups were significantly increased when compared to the control group, but the cytotoxicities of neodymiun oxide in more than $100{\mu}M$ concentration groups were statistically increased. After 48 hours exposure, cytotoxicities of both materials were statistically increased in $100,000{\mu}M$ concentration groups. Olive tail moments of the lanthanum oxide treated group were significantly increased when compared to the control group. Conclusions: The cytotoxicity of lanthanum oxide was higher than that of neodymium oxide. The DNA of MRC-5 cells treated with lanthanum oxide for 48 hours were significantly damaged.

Life Risk Assessment of Landslide Disaster Using Spatial Prediction Model (공간 예측 모델을 이용한 산사태 재해의 인명 위험평가)

  • The spatial mapping of risk is very useful data in planning for disaster preparedness. This research presents a methodology for making the landslide life risk map in the Boeun area which had considerable landslide damage following heavy rain in August, 1998. We have developed a three-stage procedure in spatial data analysis not only to estimate the probability of the occurrence of the natural hazardous events but also to evaluate the uncertainty of the estimators of that probability. The three-stage procedure consists of: (i)construction of a hazard prediction map of "future" hazardous events; (ii) validation of prediction results and estimation of the probability of occurrence for each predicted hazard level; and (iii) generation of risk maps with the introduction of human life factors representing assumed or established vulnerability levels by combining the prediction map in the first stage and the estimated probabilities in the second stage with human life data. The significance of the landslide susceptibility map was evaluated by computing a prediction rate curve. It is used that the Bayesian prediction model and the case study results (the landslide susceptibility map and prediction rate curve) can be prepared for prevention of future landslide life risk map. Data from the Bayesian model-based landslide susceptibility map and prediction ratio curves were used together with human rife data to draft future landslide life risk maps. Results reveal that individual pixels had low risks, but the total risk death toll was estimated at 3.14 people. In particular, the dangerous areas involving an estimated 1/100 people were shown to have the highest risk among all research-target areas. Three people were killed in this area when landslides occurred in 1998. Thus, this risk map can deliver factual damage situation prediction to policy decision-makers, and subsequently can be used as useful data in preventing disasters. In particular, drafting of maps on landslide risk in various steps will enable one to forecast the occurrence of disasters.

Alterations in the Neuro-Mechanical Properties of Human Ankle Dorsiflexor after Maximum Eccentric Exercise (최대 신장성 수축운동에 따른 인체 하지 족배굴곡근의 신경-기계학적 특성 변화)

  • The purpose of this study was to investigate changes in mechanical properties of human tibialis anterior following eccentric exercise. Healthy subjects (n=12) performed 120 maximum eccentric contraction of ankle dorsiflexor. Before and 1- and 24- hour after the eccentric exercise, ankle dorsiflexion moment-angle relationships were obtained. Along with significant decrease in maximum isometric muscle strength, the shift of the optimum ankle joint angle toward the longer muscle length direction was observed, independent of the ranges of motion of the eccentric exercise. The results of this study demonstrated that eccentric exercise-induced micro muscle damage(Morgan & Allen, 1999) does rut seem to be a sole mechanism of eccentric contraction-induced muscle damage, suggesting further investigation for the better understandings of this phenomenon.
