Five Di-caffeoylquinic Acid Derivatives and a Coumarin from the Hot-water Extract of Artemisia annua (개똥쑥(Artemisia annua) 열탕추출물의 Di-caffeoylquinic Acid 및 Coumarin 성분)

  • Artemisia annua L. (Compositae) is an annual herb, which has been traditionally used as an antipyretic and hemostatic agent with the herbal medicine name of Cheong Ho (菁蒿) in Korea and China. In this study, five di-caffeoylquinic acid derivatives and a coumarin were determined from the hot-water extract of aerial parts of A. annua. The structures of isolates were elucidated to be 1,3-di-O-caffeoylqunic acid (1), 3,4-di-O-caffeoylquinic acid (2), 3,5-di-O-caffeoylqiunic acid (3), 1,5-di-O-caffeoylquinic acid (4) and 4,5-di-O-caffeoylquinic acid (5) and scopoletin (6). The presence of 1,5-di-O-caffeoylquinic acid (4) is firstly reported from A. annua in the current study.

Antioxidant and Anti-inflammatory Activities of Artemisia annua L. According to Extract Methods (개똥쑥 용매추출 방법에 따른 항산화 활성 및 항염증 효과 )

  • This study was to investigate the antioxidant and anti-inflammatory activity of leaf and stem of Artemisia annua L. extract by hot water and 70% ethanol. The total polyphenol content was significantly higher in the 70% ethanol extract of Artemisia annua L. than in the hot water extract, but there was no significant difference in the total flavonoid content between the hot water extract and the 70% ethanol extract. The DPPH radical and ABTS radical scavenging ability was the highest in the vitamin C treatment group at the concentration of 62.5~500 ㎍/mL, followed by the 70% ethanol extract and the hot water extract. As a result of measuring the NO production inhibitory effect at 125 ~ 500 ㎍/mL, the hot water extract showed strong NO production inhibition at the concentration of 500 ㎍/mL, and showed the inhibition of NO production with concentration-dependent pattern. In addition to inhibitory activity of NP production, the 70% ethanol extract also showed an inhibitory effect on inflammatory cytokines production. It is thought that it can be widely used in the treatment and improvement of inflammatory diseases because it shows antioxidant effects and significantly reduces the expression of inflammatory cytokines.

Inhibition of Free Radical-Induced Lipid Oxidation by the Extract from Submerged-Liquid Culture of Mushrooms in the Medium Containing Mulberry Tree Powders (뽕나무가루 첨가 배지에서 배양한 버섯균사체 배양물의 자유라디칼 유도 산화 억제)

  • Antioxidant activity of extracts from the submerged-liquid culture of mushrooms was measured using two systems : linoleic acid and mouse liver microsomes induced by various free radical sources. Mushrooms of Pleurotus ostreatus (Neutari), Phellinus linteus (Sanghwang), Paecilomyces japonicus (Dongchunghacho), Hericicum erinacium (Norugungdengyee) and Agaricus blazei (Shinryeong) in 1% mulberry tree powder-supplemented medium were incubated in a shaking incubator (200 rpm, $25^{\circ}C$) for 3 days. Hot water extracts of mycelial cultures were freeze-dried, followed by fractioning with hexane, chloroform, ethylacetate and butanol in the order. Antioxidant activity of each sample was examined in free radical-induced linoleic acid oxidation in phosphate-buffered saline (PBS ) solution by measuring the amount of malonaldehyde (MA), and mouse liver microsomal systems by measuring the amount of thiobarbituric acid reactive substances (TBARS). In linoleic acid oxidation system, hot water extracts from the cultures of Pleurotus ostreatus, Phellinus linteus, and Paecilomyces japonicus exhibited stronger antioxidant activity than aqueous or butanol fraction and the combined fraction of hexane, chloroform and ethylacetate, but the hot water extract from Pleurotus ostreatus culture was the strongest activity. The antioxidant activity of the hot water extract from Pleurotus ostreatus culture was stronger than any other fractions in mouse microsomal system. These results suggest that hot water extract of Pleurotus ostreatus culture, and the cultures of Phellinus linteus and Paecilomyces japonicus could be useful for functional materials to reduce the oxidation of lipids in food systems induced by free radicals.

The Effect of the Hot Water Extract from Taraxacum coreanum Nakai on Hepatocarcinogenesis Induced by N-nitrosodiethylamine in Rats (흰민들레 열수추출물이 N-nitrosodiethylamine로 유발된 흰쥐의 간암 발생에 미치는 효과)

  • The present study was carried out to investigate the hepatoprotective effect of hot water extract of Korean white dandelion (Taraxacum coreanum Nakai) on liver cancer induced in rats by a single dose of N-nitrosodiethylamine (NDEA). The animals were divided into three groups. The animals in the normal (Nor) group were fed basal diet. Control (Con) group were administered with NDEA (200 mg/kg, i.p.). Hot water extract of Korean white dandelion (KDDA, KDDB) group treated with hot water extracts of Korean white dandelion (30, 60 mg/kg/day) for 8 weeks after NDEA. Serum AST and ALT were measured, enzyme activity of liver were examined by spectrophotometer and histological alternation of liver were examined by light microscopy. KDDA and KDDB group were decreased the increase of serum AST, ALT level induced by NDEA. The activities of SOD and CAT were significantly increased in KDDA, KDDB then in the Con. In the histological observation, KDDA and KDDB group were structure of liver lobula and nuclear shape was relatively cleared. In addition, the cytoplasm staining was similar to those of the Nor group. vacuolization of cytoplasm was not observed. These results suggest that administration of hot water extract of Korean white dandelion suppress or retard liver cancer induced by NDEA in rats.

Induction of DNA Breakage by the Hot-water Extracts of Fructus Chaenomelis (Chaenomeles sinensis Koehne)

  • The possible mechanism of the DNA strand breaking activity of the hot-water extract of Fructus Chaenomelis (dried fruit of Chaenomeles sinensis) in a closed circular duplex replica form DNA (RFI DNA) was studied through agarose gel electrophresis under various conditions. Induction of DNA strand scission by the hot-water extract of C. sinensis occurred in dose and time-dependent manners. $Cu^{2+}$ was indispensable for the induction of DNA strand breakage. Exogeneous chelating agents inhibited the DNA breaking activity, conforming the catalytic action of $Cu^{2+}$ on generation of free radicals responsible for oxidative damage. Antioxidant enzymes and some radical scavengers were used to investigate the major radical species triggering the DNA strand scission, demonstrating that a highest inhibitory activity was found in the presence of catalase, while less in the presence of tiron (a scavenger for superoxide radical), 2-aminoethyl-isothiuroniumbromide-HBr, cysteamine (scavengers for hydroxyl radical), and 1,4-diazabicyclo [2,2,2] octane (a scavenger for singlet oxygen) in decreasing order. The findings implied that oxygen radical species generated in presence of transition divalent cation during the oxidation of some compounds contained in the hot-water extract of C. sinensis is mainly responsible for inducing genotoxicity.

Nitric Oxide Production and Acetylcholinesterase Inhibitory of Activity Various Extracts from Codonopsis lanceolata by Steaming Times (증숙 더덕 용매별 추출물의 Nitric Oxide 생성 저해 효과 및 Acetylcholinesterase 저해활성)

  • Steaming is a method that has traditionally been used for medicinal plant extraction. This study investigated nitrite oxide production, ferrous ion chelating activity, α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities of ethanol, acetone and hot-water extracts of Codonopsis lanceolata prepared by steaming seven times. MTT assay showed that each extract was non-toxic up to a concentration of 700 ㎍/mL confirming that there was no cytotoxicity in all extracts. The α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities exhibited by the hot-water extract obtained from steaming seven times were higher (83.1%) than the other extracts. Higher production of nitrite oxide and better ferrous chelating activity was recorded with hot-water extract compared to ethanol and acetone extracts. These results indicated that more steaming of Codonopsis lanceolata extracts would be required to validate the possibility of developing antioxidants. Also, further study is needed to determine if the components present in the tested extracts might be useful in the prevention of Alzheimer's disease. These results showed that hot-water extracts may be useful for their antioxidant and the production inhibitory activity of nitrite oxide. It will be helpful in the investigation of the constituent analysis of the steam-processed product of Codonopsis lanceolata.

Study on the Improvement Effect of Antibacterial and Allergic Contact Dermatitis on Torreyae Semen Extracts (비자의 항균 및 알레르기성 접촉 피부염 개선 작용 연구)

  • The purpose of this study was to investigate the efficacy of Torreyae semen extract against antimicrobial and allergic contact dermatitis to explore its utility as a herbal resource. To this end, the antibacterial effects of Torreyae Semend extracted with hot water and ethanol (30%, 60%, 90%) were investigated using Staphylococcus aureus, Staphylococcus epidermidis, pseudomonas aeruginosa, and Propionibacterium acnes. And observed the anti-atopic dermatitis efficacy with a mouse model that allergic contact dermatitis induced by DNCB. Experimental results showed that the antibacterial activity against S. epidermidis and P. aeruginosa was significantly higher in the hot water extract of Torreyae Semen. We observed that the extracts of 30% ethanol and hot water of Torreyae Semen inhibited growth in a concentration-dependent manner in P. acnes & S. epidermidis. We observed that the administration of hot water extracts and 30% ethanol extract of Torreyae Semen significantly reduce the thickness of ear & back and gross skin condition compared to the control group in the mice with allergic contact dermatitis. As a result of H & E histological staining, it was observed that the Torreyae Semen extraction group had markedly reduced epidermis and skin thickness, granules, and keratinization compared to the control group. The groups administrated with Torreyae Semen hot water extracts had decreased TNF-α and IL-10 contents compared with the control group. As a result, Torreyae Semen may be used as a useful herbal resource because it has a certain level of antibacterial and anti-atopic effects.

Effects of Hot Aqueous and Ethanol Extract from $Lonicera$ $japonica$ $Flos$ on NO and $PGE_2$ in Macrophage (금은화 추출액이 RAW 264.7 Macrophage에서의 NO와 $PGE_2$ 생성에 미치는 영향)

  • Objectives : The objective of this study is to study the effects of hot aqueous extract and ethanol extract from $Lonicera$ $japonica$ $Flos$ on nitric oxide(NO) and prostaglandin $E_2(PGE_2)$ production in macrophage. Methods : $Lonicera$ $japonica$ $Flos$ was extracted in two ways. One was extracted with distilled water(2L) for 4 h and the other one was extracted with 70% ethanol (2L) for 4h. The RAW 264.7 macrophage was subclutured. In order to evaluate cytotoxicity, MTT assay was performed. The concentrations of NO were measured by Griess assay. The concentrations of $PGE_2$ were measured by enzyme immunoassay. Results : 25, $125{\mu}g/m{\ell}$ hot aqueous extract from $Lonicera$ $japonica$ $Flos$ inhibited NO production in LPS-stimulated RAW 264.7 macrophages significantly. 25, 125, $625{\mu}g/m{\ell}$ ethanol extract from $Lonicera$ $japonica$ $Flos$ inhibited NO production in LPS-stimulated RAW 264.7 macrophages significantly. 150, $200{\mu}g/m{\ell}$ hot aqueous extract and ethanol extract from $Lonicera$ $japonica$ $Flos$ inhibited $PGE_2$production in LPS-stimulated RAW 264.7 macrophages significantly. Conclusions : This study suggests that hot aqueous extract and ethanol extract from $Lonicera$ $japonica$ $Flos$ suppress NO and $PGE_2$ production. So hot aqueous extract and ethanol extract from $Lonicera$ $japonica$ $Flos$ may have an anti-inflammation effect.

Analysis of Physiological Activity and Cytotoxicity of Residue Extracts after Pepper Harvest (고추 수확 후 잔재물 추출물의 생리활성과 세포독성 분석)

  • The purpose of this study was to investigate the value of residue discarded after harvesting a species of pepper (Capsicum annuum L.). This study analyzed the physiological activity and cytotoxicity of a fermented extract and a hot water extract gathered from the residues of the Cheongyang pepper and the Nokkwang pepper. The total nitrogen, phosphoric acid, potassium, calcium, and magnesium contents of the fermented extract of the Nokkwang pepper were approximately twice as high as those of the Cheongyang pepper, while the hot water extracts had similar levels of these components across both peppers. Among the microelements, only boric acid, iron, and silicon components were detected, whereas zinc, manganese, molybdenum, and copper components were not detected in either extract. The total polyphenol and flavonoid contents of the fermented extracts were more than two times higher than those of the hot water extracts for both peppers. The DPPH radical scavenging ability (indicating antioxidant activity) of the fermented extract was higher than that of the hot water extract, while the ABTS radical scavenging ability (indicating antioxidant activity) of the hot water extract was higher than that of the fermented extract. A cytotoxicity test was conducted on the extracts using an MTT assay. Both extracts exhibited weak cytotoxicity at all concentrations for both peppers. These results suggest that the extract of residue discarded after harvesting these peppers could be suitable for functional feed or agricultural materials.

Antioxidant activity of extracts from soybean and small black bean (대두와 쥐눈이콩 추출물의 항산화 활성에 관한 연구)

  • This study was carries out to analyzed the antioxidant activities of extracts extrated from soybean and small black bean for the development of functional materials. Yields of hot-water and ethanol extracts were higher in the soybean than small black bea, and yields of hot-water extracts were higher than ethanol extracts. Total phenol contents were 26.22 mg/g in the ethanol extracts from small black bean, and were higher than those in the soybean extracts. Total flavonoid contents were 30.52 mg/g of hot-water extracts from small black bean. The EDA values were increased by the increase of extract concentrations and were high in the hot-water and ethanol extracts from bean and small black bean The EDA values was high in the extracts of small black bean from low concentrations. The SOD-like activity of hot-water and ethanol extracts from small black bean was higher than those of soybean extracts. The SOD-like activity was 73%, 62% of the hot-water and ethanol extracts from small black bean in 10 ml/mL extract concentration, respectably. The nitrite scavenging ability was high extracts from soybean was 47.60% in pH 1.2, Those of hot-water extracts were higher than in ethanol extracts in 10 ml/mL extract concentration. The xanthine oxidase inhibitory activities were increased by the increase of extract concentrations and were high in ethanol extracts from small black bean of low concentration. The xanthine oxidase inhibitory activities was hot-water extracts from soybean were higher than 5 ml/mL extract concentration than those of other extracts. The inhibitory activities of tyrosinase were increased by increase of extract concentrations and were high each other 42.05%, 45.67% of hot-water and ethanol extracts from small black bean in 10 ml/mL extract concentration. Reducing power was increased by increase of extract concentrations in extracts of small black bean, and were high more than double in hot-water and ethanol from small black bean than extracts of soybean.
