Influence Analysis on the Number of Ruptured SG u-tubes During mSGTR in CANDU-6 Plants (중수로 증기발생기 다중 전열관 파단사고시 파단 전열관 수에 대한 영향 분석)

  • An influence analysis on multiple steam generator tube rupture (mSGTR) followed by an unmitigated station blackout is performed to compare the plant responses according to the number of ruptured u-tubes under the assumption of a total of 10 ruptured u-tubes. In all calculation cases, the transient behaviour of major thermal-hydraulic parameters, such as the discharge flow rate through the ruptured u-tubes, reactor header pressure, and void fraction in the fuel channels is found to be overall similar to that of the base case having a single SG with 10 u-tubes ruptured. Additionally, as the conditions of low-flow coolant with high void fraction in the broken loop continued, causing the degradation of decay heat removal, the peak cladding temperature (PCT) would be expected to exceed the limit criteria for ensuring nuclear fuel integrity. However, despite the same total number of ruptured u-tubes, because of the different connection configuration between the SG and pressurizer, a difference is foud in time between the pressurizer low-level signal and reactor header low-pressure signal, affecting the time to trip the reactor and to reach the PCT limit. The present study is expected to provide the technical basis for the accident management strategy for mSGTR transient conditions of CANDU-6 plants.

  • Cellobiose dehydrogenase(CDH) as a hemoflavoenzyme is secreted out of cell in the cellulose degradation. As CDH strongly bound to amorphous cellulose, it helps cellulose hydrolysis by cellulase. CDH may have an important role of saccharification process for bioethanol production. In this study, Phanerochaete chrysosporium ATCC 32629 was selected for the production of CDH among other strains tested. The optimal temperature and pH of CDH produced by P. chrysosporium ATCC 32629 were ${55^{\circ}C}$ and 4, respectively. To improve the activity of CDH, the mutation of P. chrysosporium was performed using proton beam that has high energy level partially. As a result, P. chrysosporium mutant with the high activity was selected at 1.2 kGy in a range of 99.9% lethal rate. The CDH and $\beta$-glucosidase activities of mutant were 1.4 fold and 20 fold higher than those of wild strain. Therefore, P. chrysosporium mutant with the high activities of CDH and $\beta$-glucosidase was obtained from mutation by proton beam irradiation.

Effect of Temperature on Formation of Polymer in Oxidation of Methyl Linoleate (Methyl Linoleate 산화중 중합체 형성에 미치는 온도의 영향)

  • To investigate the effect of heating conditions on the polymerization of methyl linoleate, the esters were heated at $60^{\circ}C,\;90^{\circ}C,\;120^{\circ}C$ and $150^{\circ}C$, respectively, with sparging oxygen for different periods of time. On the basis of the peroxide curve obtained at each of the four temperatures, four heating times were chosen for the analysis of the polymers and total oxidation products. Significant linear relationships were found between polymer contents and total oxidation product contents. The contents of polymers and their linkage types were analyzed by High Performance Size Exclusion Chromatography. The polymers formed at four temperatures were qualitatively identified as dimers. The dimers with peroxide linkages were detected from methyl linoleate oxidized at $60^{\circ}C\;and\;90^{\circ}C$ but they were not detected from methyl linoleate oxidized at $120^{\circ}C\;and\;150^{\circ}C$. Therefore, all dimers formed at $120^{\circ}C\;and\;150^{\circ}C$ seemed to be the ones with ether linkage or carbon to carbon linkage. The degradation rate of the dimers with peroxide linkages at $90^{\circ}C$ was faster than at $60^{\circ}C$.

The Analysis of the Breakdown Voltage according to the Change of JTE Structures and Design Parameters of 4H-SiC Devices (4H-SiC 소자의 JTE 구조 및 설계 조건 변화에 따른 항복전압 분석)

  • Silicon Carbide(SiC) has large advantage in high temperature and high voltage applications because of its high thermal conductivity and large band gap energy. When using SiC to design power semiconductor devices, edge termination techniques have to be adjusted for its maximum breakdown voltage characteristics. Many edge termination techniques have been proposed, and the most appropriate technique for SiC device is Junction Termination Extension(JTE). In this paper, the change of breakdown voltage efficiency ratio according to the change of doping concentration and passivation oxide charge of each JTE techniques is demonstrated. As a result, the maximum breakdown voltage ratio of Single Zone JTE(SZ-JTE), Double Zone JTE(DZ-JTE), Multiple Floating Zone JTE(MFZ-JTE), and Space Modulated JTE(SM-JTE) is 98.24%, 99.02%, 98.98%, 99.22% each. MFZ-JTE has the smallest and SZ-JTE has the largest sensitivity of breakdown voltage ratios according to the change of JTE doping concentration. Additionally the degradation of breakdown voltage due to the passivation oxide charge is analyzed, and the sensitivity is largest in SZ-JTE and smallest in MFZ-JTE, too. In this paper, DZ-JTE and SM-JTE is the best efficiency JTE techniques than MFZ-JTE which needs large doping concentration in short JTE width.

Evaluation of Microcracks in Thermal Damaged Concrete Using Nonlinear Ultrasonic Modulation Technique (비선형 초음파 변조 기법을 이용한 열손상 콘크리트의 미세균열 평가)

  • This paper concentrates on the evaluation of microcracks in thermal damaged concrete on the basis of the nonlinear ultrasonic modulation technique. Since concrete structure exposed to high temperature accompanies the development of microcracks due to the physical and chemical changes from temperature and exposed time, the adoption of nonlinear approach is required. Instead of using the conventional ultrasonic nondestructive methods which have the limitation in evaluating excessive microcracks, accordingly, a nonlinear ultrasonic modulation method which shows better sensitivity in quantifying microcracks is introduced. Upon the analysis for the modulation of ultrasonic wave and low frequency impact to measure the nonlinearity parameter, which can be used as an indicator of thermal damage, the verification processes for the introduced technique are followed: SEM investigation and permeable pore space test are performed to characterize thermally induced microcracks in concrete, and ultrasonic pulse velocity tests are performed to confirm the outstanding sensitivity of nonlinear ultrasonic modulation technique. In advance, compressive strength of thermal damaged concrete is measured to represent the effect of microcracks on performance degradation. Correlation studies between experimental data and measured data show that nonlinear ultrasonic modulation technique can effectively be used to quantify thermally induced microcracks, and to estimate the compressive strength of thermally damaged concrete.

  • Deterioration of fish muscle is known to occur more quickly in the dark fleshed fish than in the white fleshed fish, causing by their high intestinal proteolytic activity. Muscle degradation which suffer post-mortem autoproteolysis is affected by trypsin with its unique activation function towards other enzymes. To compare physicochemical and enzymatic properties for the trypsins of the dark fleshed fish, trypsins from the viscera of anchovy (Engraulis japonica), and the pyloric caeca of mackerel (Scomber japonicus), yellowfin tuna (Thunnus albacores) and albacore (Thunnus alalunga) were purified through ammonium sulfate fractionation, benzamidine-Sepharose 6B, DEAE-Sephadex A-50, and Sephadex G-75 chromatography Two trypsins from mackerel (designated mackerel trypsin A and mackerel trypsin B), and one each from anchovy, yellowfin tuna and albacore were isolated as electrophoretical homogeneity, The purities of anchovy trypsin, mackerel trypsin A and B, yellowfin tuna trypsin, and albacore trypsin increased to 78.1, 4.8, 9.3, 120, and 160-fold, respectively, compared to crude enzyme solutions. Molecular weights of the trypsins from the dark fleshed fish estimated by SDS-polyacrylamide electrophoresis were ranged from 22kDa to 26kDa. The trypsins contained higher amount of glycine, serine and aspartic acid, and less amount of tryptophan, methionine, lysine and tyrosine. Optimal conditions for amidotici reactions of the enzymes were pH 8.0 and 45$^{\circ}C$ for anchovy trypsin, pH 8.0 and 5$0^{\circ}C$ for mackerel trypsin A and B, pH 9.0 and 55$^{\circ}C$ for yellowfin tuna trypsin, and pH 9.0 and 5$0^{\circ}C$ for albacore trypsin. It was supposed that the habitat temperature of the dark fleshed fish is slightly connected with the optimal reaction temperature of the trypsins of the fish.

Characteristics of The Wastewater Treatment Processes for The Removal of Dyes in Aqueous Solution(1) - Chemical Precipitation or Biological Treatment - (수용액 중의 염료 제거를 위한 폐수처리공정의 특성(1) -화학적 응집 및 생물학적 처리-)

  • This study was conducted to remove the dyes in dye wastewater by the chemical precipitation or biological treatment which are one of the main pollutants in dye wastewater. In order to remove the disperse dyes effectively in aqueous solution by chemical precipitation process, coagulation and flocculation tests were carried out using several coagulants on various reaction conditions. It was found that the Ferrous sulfate was the most effective coagulant for the removal of disperse dye(DB79), and we could get the best result for the removal of disperse dye(DB56) in the aspects of TOC removal efficiency and sludge yield. When the Ferrous sulfate dosage was 800mg/l, the sludge settling velocity was very fast$(SV_{30}=4\%)$, and the color was effectively removed in the disperse dye(DB79) solution. Although the color removal was ineffective when the Alum was used as a coagulant, the sludge yield decreased in comparison with the Ferrous sulfate or the Ferric sulfate being used in the disperse dye(DB56) solution. In order to decolorize disperse dye(DR17) by using biological treatment process, a strain which has potential ability to degrade disperse dyes was isolated from natural system. The optimal culture conditions of temperature and pH were found to be $40^{\circ}C\;and\;8.5\~9$, respectively. When yeast extract was mixed with polypeptone at the mixing ratio of 1:1 as a nitrogen source, decolorization efficiency was highest$(93\%)$ among the nitrogen sources. The strain screened was excellent to adjust to pH, and it seems to have ability to control pH needed to growth. The optimal culture conditions in concentration of $MgSO_{4.}\cdot7H_2O\;and\;KH_2PO_4$ were $0.1\%(w/v)\;and\;0.2\%(w/v)$, respectively. Strains degrading and decolorizing reactive dyes, RB198 and RR141 which were isolated from water system, are named RBK1 and RRK. And the cell growth characteristics of RBK1 and RRK were investigated. The optimal culture conditions of temperature and pH were found to be 30t' and 7.0, respectively. Optimum nitrogen source was peptone, and it was found that decolorization efficiencies by strains RBK1 and RRK, were $85\%\;and\;62\%$, respectively, with introduction of 4,000mg/l of peptone. In the case of RBK1, color removal efficiencies were very high below 400mg/l. Decolorization efficiency was over $90\%$ at 20hours of culture time. The Color degradation ability of RRK was lower than that of RBK1.

Construction and Characterization of Vitreoscilla Hemoglobin (VHb) with Enhanced Peroxidase Activity for Efficient Degradation of Textile Dye

  • Pollution resulting from the discharge of textile dyes into water systems has become a major global concern. Because peroxidases are known for their ability to decolorize and detoxify textile dyes, the peroxidase activity of Vitreoscilla hemoglobin (VHb) has recently been studied. It is found that VHb and variants of this enzyme show great promise for enzymatic decolorization of dyes and may play a role in achieving their successful removal from industrial wastewater. The level of VHb peroxidase activity correlates with two amino acid residues present within the conserved distal pocket, at positions 53 and 54. In this work, sitedirected mutagenesis of these residues was performed and resulted in improved VHb peroxidase activity. The double mutant, Q53H/P54C, shows the highest dye decolorization and removal efficiency, with 70% removal efficiency within 5 min. UV spectral studies of Q53H/P54C reveals a more compact structure and an altered porphyrin environment (λSoret = 413 nm) relative to that of wild-type VHb (λSoret = 406), and differential scanning calorimetry data indicate that the VHb variant protein structure is more stable. In addition, circular dichroism spectroscopic studies indicate that this variant's increased protein structural stability is due to an increase in helical structure, as deduced from the melting temperature, which is higher than 90℃. Therefore, the VHb variant Q53H/P54C shows promise as an excellent peroxidase, with excellent dye decolorization activity and a more stable structure than wild-type VHb under high-temperature conditions.

Biodegradation of Biphenyl by Sphingbium yanoikuyae BK-10 (Biphenyl의 Sphingobium yanoikuyae BK-10에 의한 분해 특성)

  • Bacterium capable of using biphenyl as a sole source of carbon and energy were isolated from soil, and based on the results of 16S rDNA sequence, strain BK10 identified as a Sphingobium yanoiktiyae. The optimum cultural conditions were as follows; $NH_4NO_3$ 1g, $K_2HPO_4$ 1g, $MgSO_4{\cdot}7H_2O$ 0.5g, $CaCO_3$ 0.2 g per 1 liter of distilled water. The Sphingobium yanoikuyae BK10 strain was completely utilized biphenyl in mineral salt media containing biphenyl at concentration 500 $\mu$g/ml of biphenyl as a sole carbon and energy source within 48 hours. Optimumal pH and temperature for biphenyl degradation and cell growth of strains were 6.0$\sim$8.0 and 20$\sim$50$^{\circ}C$, respectively. Especially, at 30$^{\circ}C$, cell-growth were higher than other temperature. Cell grown on biphenyl has been shown to have a higher removal rate for biphenyl than grown on sucrose. This study shows that Sphingobium yanoikuyae BK10 strain had a high biodegradation capability of biphenyl and can be simulate a candidate compounds the bioremediation of PCBs (Polychlorinated biphenyl) contaminant soil and water.

Purification and Characterization of Endo-polygalacturonase Produced by Plant Pathogenic fungus, Botrytis cinerea (식물 병원진균 Botrytis cinerea가 생산하는 Endo-polygalacturonase의 순수정제와 특성)

  • Botrytis cinerea T91-1 has shown to produce at least four different polygalacturonases in a liquid medium containing citrus pectin as a carbon source. One of the enzymes, its molecular weight was estimated as 37 kDa by denatured polyacrylamide gel electrophoresis, was purified by a series of procedures including acetone precipitation, ion exchange, heparin affinity, and reverse phase column chromatographies. By viscometric analysis, the enzyme was revealed as an endo-polygalacturonase. The enzyme activity was inhibited by divalent cations such as $Ca^{2+}$, $Co^{2+}$, and $Cu^{2+}$. Km and Vmax for polygalacturonic acid hydrolysis were 0.33 mg/ml and 28.6 nM/min, respectively. The optimum temperature for enzymatic activity was $55^{\circ}C$ and the enzyme showed optimal pH values between 4.0 and 4.5. The enzyme was stable up to 12 hours in the range of pH 4 to 7 and at the temperature below $30^{\circ}C$. Amino acid sequence from N-terminal up to 6 amino acids determined by Edman degradation showed little homology with polygalacturonases from fungi and plants.
