고지방 식이로 유도된 비만 생쥐에서 창출약침의 항비만 효과 (Anti-obesity Effect of Rhizoma Atractylodis Herbal Acupuncture in High Fat Diet-induced Obese ICR Mouse)

  • Objectives : The aim of this study was to investigate the anti-obesity potential and mechanisms of action of Rhizoma Atractylodis(RA) herbal acupuncture in high fat diet- induced obese ICR mice. Methods : Sample solutions for herbal acupuncture were prepared from the Rhizoma Atractylodis water extract powder at concentration of 150mg/kg and 300mg/kg with distilled water. Five week-old ICR mice acclimatized to the laboratory environment for 1 week were allocated into four groups: regular diet group (RD), high fat diet group(HFD), groups fed HFD with 150mg/kg RA herbal acupuncture treatment (RAE 150) and with 300mg/kg RA herbal acupuncture treatment(RAE 300). Herbal acupuncture groups were injected with either 150mg/kg or 300mg/kg of Rhizoma Atractylodis(RA) subcutaneously onto both Sinsu($BL_{23}$) alternately on the same time everyday for 30days. Body weight, gross appearance of epididymal fat area, blood glucose, insulin, insulin resistance(HOMA-IR), non-esterified fatty acid, cholesterol, triglyceride, AST, ALT, histological analysis of white adipose tissue, gene expression responsible for adipocyte differentiation and AMPK activation were analyzed. Results : RA herbal acupuncture inhibited the development of weight gain, hyperglycemia, hyperinsulinemia, hyperlipidemia, increases of AST and ALT, and the enlargement of fat cell size induced by HFD. Also, RA herbal acupuncture inhibited the expression of PPAR-${\gamma}$, C/$EBP{\alpha}$, aP2, LPL, FAS, SCD-1 and enhanced the activation of AMP-activated protein kinase. Conclusions : The results of this study demonstrate that RA herbal acupuncture can exert the anti-obesity effect and it is partially mediated by activation of AMPK and inhibition of the gene expressions responsible for adipocyte differentiation. Further studies will be required to ascertain the nti-obesity effect and mechanisms of action of RA herbal acupuncture in animal models and human for aclinical application.

고지방식이 유도 비알콜성지방간 마우스 모델에서 육신국(六神麯)의 지방간(脂肪肝) 개선효과 (Massa Medicata Fermentata improves fatty liver in high fat diet-fed nonalcoholic fatty liver disease's mouse model)

  • Objectives : This study was undertaken to verify the effects of Massa Medicata Fermentata (MMF) on nonalcoholic fatty liver disease (NAFLD) using high fat diet-fed male mice. Methods : Fifty four male C57BL/6N mice (age matched) were used for all experiments. Nine standard chow diet-fed mice were used as normal group and forty five high fat diet-fed obese mice were randomly divided into 5 groups: control, atorvastatin-10mg/kg, MMF(1)-62.5mg/kg, MMF(2)-125mg/kg and MMF(3)-250mg/kg. After all groups were treated with several kinds of diets for 8 weeks, we measured body weight gain, adipose tissue weights, plasma lipid and glucose metabolism, visceral organ weights, histological analysis for liver on the mice. Results : MMF-treated mice had lower body weight gain compared with controls. Among MMF-treated mice, the effect was magnified in MMF(2). MMF(3)-treated mice had lower blood plasma total cholesterol (TC) and glucose level compared with controls. MMF decreased hepatic lipid accumulation, liver fibrosis and liver inflammation of mice compared with controls. The effects was maximized in MMF(2) and atorvastatin. Blood plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyltransferase (${\gamma}$-GT) concentrations tends to be decreased by MMF compared with controls. Blood plasma AST, ALT, ${\gamma}$-GT concentrations and organ weights were not changed by MMF, indicating that all three kinds of MMF do not show any hepatotoxicity. Conclusions : These results suggest that MMF improves NAFLD by reducing body weight gain, hepatic lipid accumulation, liver fibrosis, liver inflammation.

고지방식이 동물모델에서 크릴오일의 아디포카인 조절을 통한 항비만 효과 (Anti-Obesity Effect of Krill Oil by Regulation of Adipokines in High Fat Diet-Induced Mouse Model)

  • 본 연구는 고지방식이로 비만이 유도된 동물모델에서 크릴오일 섭취를 통한 비만 개선 효능을 확인하고자 하였다. 실험동물은 16주간 고지방식이를 섭취시켜 비만 동물모델을 유도하였으며, 실험 종료 4주 전 크릴오일을 100, 200, 500 mg/kg/day의 농도로 4주간 투여하였다. 실험동물의 체중 변화, 장기 무게 변화, 식이 효율과 혈청 내 아디포카인 인 렙틴과 아디포넥틴 농도를 측정하여, 농도별 크릴오일의 비만 개선 효과를 확인하였다. 고지방식이를 유도한 대조군은 정상식이를 유도한 군에 비해 유의적으로 체중이 증가하여 비만이 유도됨을 확인하였다. 반면 크릴오일 200, 500 mg/kg/day 농도로 투여한 군에서 비만을 유도한 대조군에 비해 유의적으로 체중을 감소시키는 효과를 확인하였다. 또한 고지방식이를 유도한 대조군은 혈청 렙틴 증가 및 아디포아인 감소로 비정상적인 아디포카인 조절능을 나타낸 반면 크릴오일을 500 mg/kg/day 농도로 투여한 군에서는 대조군에 비해 유의적으로 혈청 렙틴 감소 및 아디포넥틴 증가를 나타내어, 아디포카인 조절을 통한 비만 개선 효과를 나타내었다. 따라서 크릴오일의 투여는 아디포카인 분비 조절을 통해 체중을 감소시켜 비만 개선 효능이 있음을 알 수 있었으며, 체지방 개선용 기능성 소재로서의 활용 가능성이 있을 것이다.

  • Objectives : Although the anti-obesity effect of Schizandrae Fructus water extract has been demonstrated, its underlying mechanism is still unclear. Therefore, we aimed to evaluate the anti-obesity effect of Schizandrae Fructus water extract through the p-AMP-activated protein kinase (p-AMPK), sirtuin1 (Sirt1), and peroxisome proliferator-activated receptor-gamma coactivator 1α (PGC-1α) signaling in 60% high-fat diet (HFD)-induced obese mouse model. Methods : Male C57BL/6 mice were divided into four groups. The Normal group was fed a normal diet and the obese groups were fed 60% HFD. Except for the Control group, the GG group was supplemented with 0.5% Garcinia gummigutta and the SCW group was supplemented with 0.5% Schizandrae Fructus water extract. After 6 weeks, obesity-related biomarkers in serum were measured and the expressions of protein for lipid-related factors in liver tissue were analyzed by western blot. Results : Treatment with SCW significantly down-regulated body weight compared to the Control group. SCW down-regulated levels of triglyceride and total cholesterol in serum and significantly increased p-AMPK, Sirt1, and PGC-1α in liver tissue. In addition, the expressions of fatty acid oxidation-related proteins such as peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase 1A (CPT-1A), uncoupling protein 1 (UCP1), and uncoupling protein 3 (UCP3) were significantly up-regulated. However, fatty acid synthesis-related proteins including sterol regulatory element-binding protein-1 (SREBP-1), phospho-Acetyl-CoA Carboxylase (p-ACC), and fatty acid synthase (FAS) were significantly down-regulated. Conclusions : Taken together, SCW treatment showed anti-obesity effect by regulating both fatty acid oxidation-related and fatty acid synthesis-related proteins through AMPK/Sirt1/PGC-1α signaling in 60% HFD-induced obese mice.

고지방식이 비만마우스 모델에서 파키스탄산 및 중국산 마황으로 조성된 강지환(降脂丸)의 체중감량 효과 비교 (Comparison of Pakistani Ephedra Herba and Chinese Ephedra Herba Containing Gangjihwan in the Improvement Effects of Weight Loss in a High Fat Diet-Fed Obese Mice)

  • Objectives : This study investigated the improvement effects of Pakistani (DF-a) and Chinese Ephedra herba-containing Gangjihwan (DF-b) on obesity in a high fat diet-fed obese mouse model. Methods : Eight-week-old C57BL/6N mice were divided into four groups: a normal lean group given a standard diet, an obese control group given a high fat diet, and DF-a and DF-b groups given a high fat diet with DF-a (80 mg/kg), and DF-b (80 mg/kg), respectively. After 8 weeks of treatment, body weight gain, feeding efficiency ratio, blood lipid markers, fat weight and histology were examined. Results : 1. Body weight gain and fat mass were significantly decreased in DF-a and DF-b groups compared with control. The extent of decreases was eminent in DF-a group. 2. Feeding efficiency ratio and circulating leptin concentration were significantly decreased in DF-a and DF-b groups compared with control, whereas circulating adiponectin concentration was increased in DF-a and DF-b groups compared with control. 3. Consistent with their effects on body weight gain and fat mass, circulating concentrations of triglyceride, glucose and insulin were decreased in DF-a and DF-b groups compared with control. 4. The size of adipocytes were decreased by DF-a and DF-b compared with control, whereas the adipocyte number per unit area was increased by them, suggesting that DF-a and DF-b decreased the number of large adipocytes. 5. Consistent with their effects on body weight gain, liver fibrosis was reduced in DF-a and DF-b groups compared with control. Conclusions : In conclusion, these results suggest that DF-a and DF-b not only decrease feeding efficiency ratio, plasma leptin concentration, and blood anti-obesity biomarkers, but also reduce fat mass, contributing to the improvement of obesity. DF-a and DF-b also inhibit liver fibrosis. In addition, these effects were similar between Pakistani Ephedra herba and Chinese Ephedra herba-containing Gangjihwan.

Methods : Eight-week-old C57BL/6N mice were divided into four groups: a normal lean group given a standard diet, an obese control group given a high fat diet, and DF and DF+GSH groups given a high fat diet with DF (40 mg/kg), and DF+GSH (80 mg/kg), respectively. After 8 weeks of treatment, body weight gain, feeding efficiency ratio, blood lipid markers, fat weight and histology were examined. Results : 1. Body weight gain and fat mass were significantly decreased in DF and DF+GSH groups compared with control. The extent of decreases was eminent in DF+GSH group. 2. Feeding efficiency ratio and circulating concentration of leptin were decreased in DF and DF+GSH groups compared with control. These decreases were significant in DF+GSH group. 3. Consistent with their effects on body weight gain and fat mass, circulating concentrations of triglyceride, glucose and insulin were decreased in DF and DF+GSH groups compared with control. 4. Conclusions : In conclusion, these results suggest that DF and DF+GSH groups decrease feeding efficiency ratio, plasma leptin concentration, blood anti-obesity biomarkers and fat mass, improves body weight gain contributing to the inhibition of liver fibrosis. In addition, these effects were more effective in DF+GSH combination group than in DF-only group.

Monascus pilosus로 발효시킨 후발효 녹차가 고지방 식이 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향 (Dietary Effects of Post-fermented Green Tea by Monascus pilosus on the Body Weight, Serum Lipid Profiles and the Activities of Hepatic Antioxidative Enzymes in Mouse Fed a High Fat Diet)

  • Monascus pilosus로 발효시킨 후 발효 녹차의 첨가식이가 고지방식이 ICR 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향을 조사하였다. 정상대조군(NC), 고지방식이대조군(HC), 비발효녹차 분말을 2% 첨가한 고지방식이군(CHA) 및 M. pilosus로 발효시킨 후발효녹차 분말을 2% 첨가한 고지방식이군(mCHA)으로 구분하여 7주간 사육하였다. 체중은 CHA군과 mCHA군간의 유의적인 차이가 없으나 HC군에 비하여 각각 24.15 및 29.66%가 감소되었으며 식이섭취량에는 차이가 없었다. 체중 당 간 및 신장의 중량은 CHA군 및 mCHA군이 HC군에 비해 증가하였다. 체중 당 부고환주위지방의 무게는 mCHA군에서 유의하게 감소하였다. 혈청 TG와 의총콜레스테롤 함량은 mCHA군에서 NC군 수준으로 감소하였다. CHA군과 mCHA군의 HDL-cholesterol 함량은 HC군에 비하여 96.55 및 119.32%가 높았다. CHA군과 mCHA군의 CHA군 및 mCHA군의 LDL-cholesterol 함량은 각각 0.67 및 0.41을 나타내었다. 간 조직의 GSH 함량은 HC군에 비하여 CHA군이 11.24%, mCHA군은 23.68%가 높았다. LPO 함량은 CHA군과 mCHA군이 HC군에 비하여 43.70 및 45.38%가 낮았다. CHA군과 mCHA군의 xanthine oxidoreductase D type 효소활성은 HC군에 비하여 각각 18.52 및 33.33%가 감소하였으며 mCHA군은 NC군 수준으로 높았다. XOD O type 효소활성은 CHA군과 mCHA군이 HC군에 비하여 각각 16.67 및 33.33%가 높았다. superoxide dismutase 활성은 CHA군과 mCHA군이 HC군에 비하여 유의차는 없었다. mCHA군의 glutathione S-transferase 활성은 HC군에 비하여 유의적으로 높았으며 특히, mCHA군의 glutathione peroxidase 활성은 HC군의 2.55배, CHA군의 1.5배로 높았으며 NC군과 대등하였다. CHA군과 mCHA군의 ALT 활성 HC군에 비하여 현저하게 낮았다. 간 조직의 현미경 관찰결과, CHA군과 mCHA군에서는 간 세포 내 축적된 지방의 크기가 작은 포말형으로 변화되고, 이로 인해 간 세포판과 동양혈관 형성이 간 소엽단위로 뚜렷하게 구분되었다. 이상의 실험결과로 보아 M. pilosus로 발효시킨 후발효 녹차는 고지방식이에 의해 유발되는 비만과 고지혈증 및 지방간을 예방 혹은 경감시켜줄 수 있는 유효한 가능성을 시사하였다.

Water Extracts of Paecilomyces tenuipes Inhibit Cathepsin S-induced Adipocyte Differentiation in 3T3-L1 Cells

  • Cathepsin S is a cysteine protease that affects extracellular matrix remodeling. Recently, several studies have reported that cathepsin S is involved in obesity. Both mouse and human adipose cells produce this enzyme in the early phase of adipocyte differentiation, where it degrades fibronectin. Cathepsin S gene expression is elevated in the adipose tissue of obese mice as compared to that of lean mice. Paecilomyces tenuipes water extracts (PTW) are shown to have an inhibitory effect on cathepsin S activity. In this study, Z-Val-Val-Arg-MCA was used as a cathepsin S-specific substrate in order to examine inhibitory effect of PTW. Supplementing 3T3-L1 cell media with PTW clearly reduced lipid droplet accumulation and cathepsin S-induced adipogenesis. Furthermore, PTW decreased weight gain, subcutaneous adipose tissue growth, the level of serum triglyceride, and total cholesterol in mice fed a high-fat diet. These data suggest that PTW work against adipose cathepsin S and presumably contribute to anti-obese activities.

고지방식이급여 마우스에서 잠상산물의 항당뇨 효능 (Anti-Diabetic Effect of Sericultural Product in High Fat Diet-Fed Mice)

  • 본 연구는 고지방식이로 유도된 제2형 당뇨 동물모델에서 잠상산물의 항당뇨 및 지질개선 효과를 규명하기 위하여 C57BL/6N 마우스에 14주간 잠상산물 분말(ML, SK, MF, CT)을 급여한 후 제2형 당뇨의 임상적 지표 및 인슐린 신호 전달체계에 미치는 영향을 분석하였다. HFD군보다 잠상산물을 급여한 군들의 체중증가량이 감소하는 경향이었으며, 혈장 포도당 및 인슐린 농도는 HFD군보다 ML분말을 급여한 군에서 특히 감소하였다. 혈장 T-C 농도는 HFD군에 비하여 ML과 SK 분말 급여 군에서 유의적으로 감소하였으며, 혈장 HDL-C 농도 및 HTR은 HFD군보다 ML군과 SK군에서 현저히 증가하였다. 간 조직 지질 농도는 HFD군보다 잠상산물을 급여한 네 군 모두에서 TG와 T-C 농도의 유의적인 감소가 나타났다. 이상의 결과 잠상산물 중 ML분말이 혈장 포도당, 인슐린, T-C 및 TG 농도를 가장 효과적으로 감소시켰으며, 혈장 HDL-C 농도 및 HTR은 증가시켰다. 또한, 내장지방 조직의 AKT, S6K, 그리고 AMPK의 활성도 증가시킨 것으로 보아 고지방식이에 의한 인슐린 저항성 및 지질 대사를 개선한 것으로 생각된다. 향후 ML의 인슐린 저항성 개선 효과에 대해 더 깊이 있는 기전연구가 수행된다면 제2형 당뇨환자를 위한 건강기능성 식품으로 개발될 수 있을 것으로 생각한다.

Obesity Exacerbates Coxsackievirus Infection via Lipid-Induced Mitochondrial Reactive Oxygen Species Generation

  • Coxsackievirus B3 (CVB3) infection causes acute pancreatitis and myocarditis. However, its pathophysiological mechanism is unclear. Here, we investigated how lipid metabolism is associated with exacerbation of CVB3 pathology using high-fat diet (HFD)-induced obese mice. Mice were intraperitoneally inoculated with 1×106 pfu/mouse of CVB3 after being fed a control or HFD to induce obesity. Mice were treated with mitoquinone (MitoQ) to reduce the level of mitochondrial ROS (mtROS). In obese mice, lipotoxicity of white adipose tissue-induced inflammation caused increased replication of CVB3 and mortality. The coxsackievirus adenovirus receptor increased under obese conditions, facilitating CVB3 replication in vitro. However, lipid-treated cells with receptor-specific inhibitors did not reduce CVB3 replication. In addition, lipid treatment increased mitochondria-derived vesicle formation and the number of multivesicular bodies. Alternatively, we found that inhibition of lipid-induced mtROS decreased viral replication. Notably, HFD-fed mice were more susceptible to CVB3-induced mortality in association with increased levels of CVB3 replication in adipose tissue, which was ameliorated by administration of the mtROS inhibitor, MitoQ. These results suggest that mtROS inhibitors can be used as potential treatments for CVB3 infection.
