As in many other country, the use of organic matter in Korea has long history. Farmers understand the value of organic matter as the source of plant nutrient and soil improving agent in general. Since 50 years ago, the sources of organic matter in paddy soils were compost, rice and barly straw, green manure, animal waste, fish and beancake, etc.. Application of green manures such as vetch and chinese milk vetch showed no significant effect on the yield of brown rice in paddy soil. On the other hand, the effects of compost and rice straw showed more significant on the yield of brown rice in paddy soil. Application of rice straw in rice cultivation is commonly made at different times between harvest, early spring and several weeks before transplanting. Considering the suitable paddy soil for application of rice straw under well to moderately well drained soil, the yield was pronounced more than poorly drained soil. Based on laboratory and field experimants, application of rice straw promoted the decrease of oxidation-reduction potential in well to moderately well drained soil. This results to be enhanced the release of some mineral nutrients,. such as potassium, calcium, silicon, and increase of availability of soil phosphorus. In the field experiments, results obtained from nitrogen fraction on the immobilization-mineralization of the tracer nitrogen applied in paddy soil,the amount and index of organic nitrogen incoporated in soil was more pronounced in rice straw application than control. Rice straw and its transformation products incoporated in the soil, provided the inflow of energy necessary to maintain heterotrophic microbes activities. Rice straw and its transformation products, especially soluble carbohydrate, enhanced the population of free-living heterotrophic $N_2$ - fixing microbes. Moreover, rice straw and its transformation products in paddy soil, enhanced the activities of soil enzymes such as dehydrogenase and urease.

Plant growth promoting (PGP) hormones, which are produced in a small quantity by bacteria, affect in plant growth and development. PGPs play an important role on the crop productivity in agricultural field. In this study, a photosynthetic bacterial strain producing the PGP was isolated from paddy soil. Bacterial isolate was gram negative, rod-shaped and motility positive. From the 16s rRNA gene sequence analysis, the isolate was identified as Rhodobacter capsulatus PS-2. The mass cultivation of R. capsulatus PS-2 was optimized by considering of the carbon, nitrogen and inorganic salt sources. Optimal medium composition was determined as Na-succinate 4.5 g, yeast extract 5 g, $K_2HPO_4$ 1 g, $MgSO_4$ 5 g, per liter. From the result of 500 L fermentation for 2 days using the optimal medium, the viable cells were $8.7{\times}10^9cfu/mL$. R. capsulatus PS-2 strain produced the carotenoid and indole-3-acetic acid (IAA). The carotenoid extraction and quantitative analysis were performed by HCl-assisting method. Total carotenoid contents from R. capsulatus PS-2 culture broth were measured as $7.02{\pm}0.04$ and $6.93{\pm}0.05mg/L$ under photoheterotrophic and chemoheterotrophic conditions, respectively. To measure the productivity of IAA, colorimetric method was employed using Salkowski reagent at optical density 535 nm. The results showed that the highest content of IAA was $197.44{\pm}5.92mg/L$ in the optimal medium supplemented with 0.3% tryptophan.

Emitted CO2 is an attractive material for microbial electrochemical CO2 reduction. Microbial electrochemical CO2 reduction (i.e., microbial electrosynthesis, MES) using biocatalysts has advantages compared to conventional CO2 reduction using electrocatalysts. However, MES has several challenges, including electrode performance, biocatalysts, and reactor optimization. In this study, an MES system was investigated for optimizing reactor types, counter electrode materials, and CO2-converting microorganisms to achieve effective CO2 upcycling. In autotrophic cultivation (supplementation of CO2 and H2), CO2 consumption of Rhodobacter sphaeroides was observed to be four times higher than that with heterotrophic cultivation (supplementation of succinic acid). The bacterial growth in an MES reactor with a single-chambered shape was two times higher than that with a double chamber (H-type MES reactor). Moreover, a single-chambered MES reactor equipped with titanium mesh as the counter electrode (anode) showed markedly increased current density in the graphite felt as a working electrode (cathode) compared to that with a graphite felt counter electrode (anode). These results demonstrate that the optimized conditions of a single chamber and titanium mesh for the counter electrode have a positive effect on microbial electrochemical CO2 reduction.
A psychrotolerant cyanobacterium, Nodularia spumigena KNUA005, was isolated from a cyanobacterial bloom sample collected near Dasan Station in Ny-${\AA}lesund$, Svalbard Islands during the Arctic summer season. To generate an axenic culture, the isolate was subjected to three purification steps: centrifugation, antibiotic treatment and streaking. The broad antibacterial spectrum of imipenem killed a wide range of heterotrophic bacteria, while the cyanobacterium was capable of enduring both antibiotics, the remaining contaminants that survived after treatment with imipenem were eliminated by the application of an aminoglycoside antibiotic, kanamycin. Physical separation by centrifugation and streaking techniques also aided axenic culture production. According to the cold-tolerance test, this mat-forming cyanobacterium was able to proliferate at low temperatures ranging between 15 and $20^{\circ}C$ which indicates the presence of cold-tolerance related genes in N. spumigena KNUA005. This suggests the possibility of incorporating cold-resistance genes into indigenous cyanobacterial strains for the consistent production of algae-based biofuel during the low-temperature seasons. Therefore, it is needed to determine the cold-tolerance mechanisms in the Arctic cyanobacterium in the next research stage.
Quantification of the airborne microorganisms (bacteria and fungi) at a swine wastewater treatment plant was performed. Microbial samples were collected at three different phases of the treatment process over a 1-yr period. Cultivation methods based on the viable counts of mesophilic heterotrophic bacteria and fungi were performed. The concentrations of airborne bacteria ranged up to about $5{\times}10^3$ colony-forming unit (CFU)/$m^3$, and those of airborne fungi ranged up to about $9{\times}10^2CFU/m^3$. The primary treatment (e.g., screen, grit removal, and primary sedimentation) was found to be the major source of airborne microorganisms at the site studied, and higher levels of airborne bacteria and fungi were observed in summer. High levels of the respirable bioaerosol (0.65 to $4.7{\mu}m$ in size) were detected in the aeration phase. Among the environmental factors studied, temperature was strongly associated with fungal aerosol generation (with a Spearman correlation coefficient of 0.90 and p-value <0.01). Occupational biorisks are discussed based on the observed field data.
This study evaluated microbial risk that could develop within soil microbial communities after amended with organic fertilizers from stabilized swine manure waste. For this purpose, we assessed the occurrences and competitiveness of antibiotic resistance and pathogenicity in soil microbial communities that were amended with swine manure wastes stabilized by a traditional lagoon fermentation process and an autothermal thermophilic aerobic digestion process, respectively. According to laboratory cultivation detection analysis, soil applications of the stabilized organic fertilizers resulted in increases in absolute abundances of antibiotic resistant bacteria and of two tested pathogenic bacteria indicators. The increase in occurrences might be due to the overall growth of microbial communities by the supplement of nutrients from the fertilizers. Meanwhile, the soil applications were found to reduce competitiveness for various types of antibiotic resistant bacteria in the soil microbial communities, as indicated by the decrease in relative abundances (of total viable heterotrophic bacteria). However, competitiveness of pathogens in response to the fertilization was pathogens-specific, since the relative abundance of Staphylococcus was decreased by the soil applications, while the relative abundance of Salmonella was increased. Further testes revealed that no MAR (multiple antibiotic resistance) occurrence was detected among cultivated pathogen colonies. These findings suggest that microbial risk in the soil amended with the fertilizers may not be critical to public health. However, because of the increased occurrences of antibiotic resistance and pathogenicity resulted from the overall microbial growth by the nutrient supply from the fertilizers, potential microbial risk could not be completely ruled out in the organic-fertilized soil samples.
Objectives: This study was performed in order to evaluate the generation characteristics of airborne bacteria and fungi while operating a household humidifier, in consideration of user habits. Methods: Microbial samples were collected in a closed chamber with a total volume of 2.76 $m^3$, in which a humidifier was operated according to experimental strategies. A cultivation method based on the viable counts of mesophilic heterotrophic bacteria and fungi was performed. Experimental strategies were divided into three classes: the type of water in the water reservoir (tap water, cooled boiled water); the frequency of filling the reservoir (refill every day, no refill); and the sterilization method (sterilization function mode, humidifier disinfectants). Results: Significant increases in the concentration of airborne bacteria were observed while the humidifier was in operation. The concentration had increased to 2,407 $CFU/m^3$ by 120 hours when tap water filled the reservoir without any application of sterilization, while for cooled boiled water, it was merely 393 $CFU/m^3$ at a similar time point. Usages of disinfectant in the water tank were more effective in decreasing bioaerosol generation compared to sterilization function mode operation. Generation characteristics of airborne fungi were similar to those of bacteria, but the levels were not significant in all experiments. Calculated exposure factor can be used as an indicator to compare biorisk exposure. Conclusion: This study identified the potential for bioaerosol generation in indoor environments while operating a household humidifier. User practices were critical in the generation of bioaerosol, or more specifically, airborne bacteria. Proper usage of a humidifier ensures that any biorisks resulting from generated bioaerosol can be prevented.

Purple non-sulfur (PNS) bacterium $Rhodobacter$$sphaeroides$ KD131 was studied with the aim of achieving maximum hydrogen production using various carbon and nitrogen sources at different pH conditions. Cells grew well and produced hydrogen using $(NH_4){_2}SO_4$ or glutamate as a nitrogen source in combination with a carbon substrate, succinate or malate. During 48h of photo-heterotrophic fermentation under 110$W/m^2$ illumination using a halogen lamp at $30^{\circ}C$, 67% of 30mM succinate added was degraded and the hydrogen yield was estimated as 3.29mol $H^2$/mol-succinate. However, less than 30% of formate was consumed and hydrogen was not produced due to a lack of genes coding for the formate-hydrogen lyase complex of strain KD131. Initial cell concentrations of more than 0.6g dry cell weight/L-culture broth were not favorable for hydrogen evolution by cell aggregation, thus leading to substrate and light unavailability. In a modified Sistrom's medium containing 30mM succinate with a carbon to nitrogen ratio of 12.85 (w/w), glutamate produced 1.40-fold more hydrogen compared to ammonium sulfate during the first 48h. However, ammonium sulfate was 1.78-fold more effective for extended cultivation of 96h. An initial pH range from 6.0 to 9.0 influenced cell growth and hydrogen production, and maintenance of pH 7.5 during photofermentation led to the increased hydrogen yield.
Molecular and cultivation techniques were used to characterize the bacterial communities of biobead reactor biofilms in a sewage treatment plant to which an Aerated Up-Flow Biobead process was applied. With this biobead process, the monthly average values of various chemical parameters in the effluent were generally kept under the regulation limits of the effluent quality of the sewage treatment plant during the operation period. Most probable number (MPN) analysis revealed that the population of denitrifying bacteria was abundant in the biobead #1 reactor, denitrifying and nitrifying bacteria coexisted in the biobead #2 reactor, and nitrifying bacteria prevailed over denitrifying bacteria in the biobead #3 reactor. The results of the MPN test suggested that the biobead #2 reactor was a transition zone leading to acclimated nitrifying biofilms in the biobead #3 reactor. Phylogenetic analysis of 16S rDNA sequences cloned from biofilms showed that the biobead #1 reactor, which received a high organic loading rate, had much diverse microorganisms, whereas the biobead #2 and #3 reactors were dominated by the members of Proteobacteria. DGGE analysis with the ammonia monooxygenase (amoA) gene supported the observation from the MPN test that the biofilms of September were fully developed and specialized for nitrification in the biobead reactor #3. All of the DNA sequences of the amoA DGGE bands were very similar to the sequence of the amoA gene of Nitrosomonas species, the presence of which is typical in the biological aerated filters. The results of this study showed that organic and inorganic nutrients were efficiently removed by both denitrifying microbial populations in the anaerobic tank and heterotrophic and nitrifying bacterial biofilms well-formed in the three functional biobead reactors in the Aerated Up-Flow Biobead process.

This research investigated the characteristics of antibiotic resistance of bacteria in microbial communities from municipal wastewater treatment plants (MWTPs), and monitored seasonal changes of antibiotic resistant bacteria (ARB) from MWTPs and Han river. When antibiotics were amended to either R2A agar (R2A) for general heterotrophs or MacConeky sorbitol agar (MSA) for coliform bacteria, all the MWTP samples exhibited multiple antibiotic resistance on the antibiotic-amended solid media. The antibiotic resistance appearing frequencies of ampicillin and sulfathiazole, respectively, were higher than reported data for other countries. The antibiotic resistance appearances differed depending upon the concentrations of primary substrate and nutrients and the types of cultivation media. The following 16S rRNA gene phylogenetic analysis showed that the identified multiple-antibiotic resistant microbes on R2A plates were more likely to be known human-pathogenic bacteria than the background heterotrophic bacteria were, suggesting a high risk of antibiotic resistance appearance to public health. In addition, according to our investigation of seasonal changes of ARB from urban MWTP and river samples, the frequency of ARB appearances was shown to correlate positively with temperature. This indicates a possibility that global warming result in increase in microbial risk to public health.
