Toxicological Studies on the Essential Oil of Eugenia caryophyllata Buds

  • The essential oil (EC-oil) obtained from the buds of Eugenia caryophyllata (Myrtaceae) was examined for its free radical-scavenging activity, cytotoxicity, and in vivo toxicity. To find the xenobiotic properties of EC-oil, serum thiobarbituric acid reactive substances (TBARS) level and hepatic drug-metabolizing enzyme activities were measured. It was found that EC-oil displayed xenobiotic properties like bromobenzene. The cytotoxicities of eugenol and of the EC-oil were greatly attenuated by the sulfhydryl-containing N-acetyl-L-cysteine (NAC), suggesting that eugenol was susceptible to nucleophilic sulfhydryl. In addition, eugenol also showed potent free radical-scavenging activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Moreover, methyleugenol considerably exhibited less cytotoxicity and less potent free radical-scavenging activity than eugenol, and the cell viability of the methyleugenol was more increased with NAC treatment than the eugenol. These results indicate that the phenolic OH in eugenol may play a crucial role in both cytotoxicity and free radical-scavenging activity. The fashion on oxidative stress and hepatic drug-metabolizing enzyme activities of eugenol resembled those of bromobenznene.

Schisandra Chinensis Baillon regulates the gene expression of phase II antioxidant/detoxifying enzymes in hepatic damage induced rats

  • BACKGROUND/OBJECTIVES: This study investigated the antioxidant activities and hepatoprotective effects of Schisandra chinensis Baillon extract (SCE) against tert-butyl hydroperoxide (t-BHP)-induced oxidative hepatic damage in rats. MATERIALS/METHODS: Sprague-Dawley (SD) rats were pretreated with SCE (300, 600, and 1,200 mg/kg BW) or saline once daily for 14 consecutive days. On day 14, each animal, except those belonging to the normal control group, were injected with t-BHP (0.8 mmol/kg BW/i.p.), and all of the rats were sacrificed 16 h after t-BHP injection. RESULTS: Although no significant differences in AST and ALT levels were observed among the TC and SCE groups, the high-dose SCE group showed a decreasing tendency compared to the TC group. However, erythrocyte SOD activity showed a significant increase in the low-dose SCE group compared with the TC group. On the other hand, no significant differences in hepatic total glutathione (GSH) level, glutathione reductase (GR), and glutathione peroxidase (GSH-Px) activities were observed among the TC and SCE groups. Hepatic histopathological evaluation revealed that pretreatment with SCE resulted in reduced t-BHP-induced incidence of lesions, such as neutrophil infiltration, swelling of liver cells, and necrosis. In particular, treatment with a high dose of SCE resulted in induction of phase II antioxidant/detoxifying enzyme expression, such as glutathione S-transferase (GST) and glutamate-cysteine ligase catalytic subunit (GCLC). CONCLUSIONS: Based on these results, we conclude that SCE exerts protective effects against t-BHP induced oxidative hepatic damage through the reduction of neutrophil infiltration, swelling of liver cells, and necrosis. In addition, SCE regulates the gene expression of phase II antioxidant/detoxifying enzymes independent of hepatic antioxidant enzyme activity.

  • This study was performed to determine the changes of serum enzyme activities in rats with hepatic injury induced by the administration of AAPH. Minor behavioral change, brittleness of skin hair and decreased water and fled intake were observed in rats administered intraperitoneally with AAPH. Serum AST and ALT activities pre-treatment were $65{\pm} 13.8 and 32{\pm}$ 12.6 IU/L, respectively and increased sharply from 2 hours of administration and reached $1248{\pm} 77.6 and 946{\pm}$ 45.6 IU/L, respectively at 48 hours of administration. Serum ALP and $\gamma -GTP activities pretreatment were 221 {\pm} 75.6 and 2.2{\pm}$ 0.35 IU/L respectively and increased sharply from 8 hours of administration and reached $767{\pm} 44.9 IU/L and 8.0{\pm} 1.23 IU/L,$ respectively at 48 hours of administration.

Hepatoprotective Effects of Gardenia jasminoides Ellis Extract in Nonalcoholic Fatty Liver Disease Induced by a High Fat Diet in C57BL/6 Mice

  • This study was carried out to investigate the potential effects of Gardenia jasminoides (GJ) extracts, on hepatic steatosis and lipid metabolism in mice fed with high-fat diet (HFD). GJ extracts (100 mg/kg, ${\times}10$ weeks) fed mice showed reduced body weight, adipose tissue weight, reduced aminotransferase level in plasma and hepatic lipid (triglyceride, total cholesterol) content. These effects were accompanied by decreased expression of lipogenic genes, sterol regulatory element binding protein-1c (SREBP-1c), liver X receptor (LXR), fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), cluster of differentiation 36 (CD36), lipoprotein lipase (LPL) and decreased lipogenic enzyme FAS and HMG-CoAR enzyme activities while elevating carnitine palmitoyltrasferase-1 (CPT) activity. Based on these results, we speculated that the inhibitory effect on hepatic steatosis of GJ extract containing geniposide is the result of suppression of lipid synthesis in mice fed with HFD, suggesting that GJ extract may be beneficial in preventing hepatic steatosis.

Effect of High Dietary Carbohydrate on the Growth Performance, Blood Chemistry, Hepatic Enzyme Activities and Growth Hormone Gene Expression of Wuchang Bream (Megalobrama amblycephala) at Two Temperatures

  • The effects of high carbohydrate diet on growth, serum physiological response, and hepatic heat shock protein 70 expression in Wuchang bream were determined at $25^{\circ}C$ and $30^{\circ}C$. At each temperature, the fish fed the control diet (31% CHO) had significantly higher weight gain, specific growth rate, protein efficiency ratio and hepatic glucose-6-phosphatase activities, lower feed conversion ratio and hepatosomatic index (HSI), whole crude lipid, serum glucose, hepatic glucokinase (GK) activity than those fed the high-carbohydrate diet (47% CHO) (p<0.05). The fish reared at $25^{\circ}C$ had significantly higher whole body crude protein and ash, serum cholesterol and triglyceride, hepatic G-6-Pase activity, lower glycogen content and relative levels of hepatic growth hormone (GH) gene expression than those reared at $30^{\circ}C$ (p<0.05). Significant interaction between temperature and diet was found for HSI, condition factor, hepatic GK activity and the relative levels of hepatic GH gene expression (p<0.05).

울금(Curcuma longa L.) 섭취가 당뇨성 흰쥐의 크레아티닌 및 간 기능 효소 활성에 미치는 영향 (4Effects of Turmeric (Curcuma longa L.) Supplementation on Creatinine and Hepatic Functional Enzyme Activities in Streptozotocin-induced Diabetic Rats)

  • Streptozotocin (STZ, 45 mg/kg body weight)으로 유도된 Sprague Dawley (SD)계 수컷 흰쥐에 5% 울금을 섭취시켰을 때, 혈청 크레아티닌(creatinine) 및 blood urea nitrogen (BUN)의 농도와 alkaline phosphatase (ALP), lactate dehydrogenase (LDH), aminotransferase (AST, ALT), 카탈라아제(catalase), 아밀라아제(amylase) 및 리파아제(lipase) 활성에 미치는 영향을 알아보기 위해 본 실험을 수행한 결과, 기본식이를 급여시킨 대조군(control)인 정상군은 BD군, 기본식이 실험군에 5% 울금 급여군(BT군), 당뇨 유발 실험군(BS군)인 질환 모델 대조군(control-diabetic)과 질환 실험군(diabetic, BS군)에 5% 울금을 급여시킨 실험군(ST군)으로 나누었다. 흰쥐의 혈청 크레아티닌 및 BUN 농도와 ALP, AST 및 ALT 활성은 유사한 경향으로 5% 울금의 급여가 감소시킨 것으로 확인되었다. SD계 수컷 흰쥐의 혈청 LDH 활성은 BS군에서 유의적인 차이를 나타내며 높은 활성을 보였으나(p<0.05), 5% 울금의 급여가 감소 효과를 나타내었다(p<0.05). 카탈라아제 활성은 BS군에서 유의적으로 낮은 활성을 나타내었고, ST군에서 활성을 증가시켰다(p<0.05). 혈청 아밀라아제 및 리파아제의 활성은 5% 울금의 급여가 감소시킨 것으로 확인되었다. 본 연구 결과, SD계 수컷 흰쥐를 통한 in vivo 실험에서 5% 울금의 섭취가 당뇨, 내인성 항산화 효소 및 간 기능 개선에 효과가 있는 것으로 나타났다.

한국전통차 재료의 열수추출물이 납투여 흰주의 간조직 중 유해 활성산소 생성과 제거효소 활성에 미치는 영향 (Effect of Korean Traditional Tea Materials Water Extract on Hepatic Oxygen Free Radical Generating and Scavenging Enzyme Activities in Lead Administered Rats)

  • 한국전통차로 알려진 녹차, 감잎, 홍화 및 두충 열수추출물이 납투여된 흰쥐의 유해 활성산소 대사에 미치는 영향을 구명하기 위하여 체중 kg당 25 mg의 납을 매주 1회 경구 투여 하였다. 녹차, 감잎, 홍화 및 두충 열수추출물은 매일 일정시간에 체중 kg당 1.26 g 수준이 되도록 4주간 경구 투여 하여 사육한 결과 간조직 중 과산화지질 함량은 납 단독투여군이 정상군에 비하여 유의적으로 증가되었으며 한국전통차 급여시 과산화지질 생성을 현저하게 억제하는 것으로 나타났다. 간조직 중의 CYP 함량과 AD 활성은 납 투여시 유의적으로 감소되었으나 각각의 열수추출물 급여시 납투여로 억제된 활성이 회복됨으로써 한국전통차 급여가 CYP 함량과 산화적 디메틸화율을 증가시키는 것으로 관찰되었다. 또한 납투여로 증가된 간조직의 XO와 SOD의 활성은 녹차, 감잎, 홍화 및 두충 열수추출물 급여시 활성이 유의적으로 억제되었다. MAO 활성은 납 투여시 정상군에 비하여 유의적이지는 않으나 증가되었으며 녹차와 감잎 열수추출물 급여로 활성 증가가 현저하게 억제되었다. 간조직 중 CAT 활성은 납투여시 약 3.3배 증가되었는데 감잎군 37%, 녹차군 34%, 두충군 30%, 홍화군 28%, 순으로 억제되는 것을 관찰할 수 있었다. 또한 GSH-Px와 GST 활성 및 글루타티온 함량은 정상군에 비하여 납 단독투여시 유의적인 감소를 보였으나 한국전통차 급여로 회복되었다. GR 활성은 실험군간에 변화가 관찰되지 않았으나 G6PD 활성은 납투여로 증가된 활성이 실험식이 급여시 유의적으로 감소되었다.

  • To evaluate the antioxidative effect of maengjong-juk (Phyllostachys pubescens) extract coated rice in vivo system, maengjong-juk extract coated rice diets were fed to C57BL/6 mice for 16 weeks. Plasma total antioxidative capacity, hepatic lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content were measured. Plasma total antioxidative capacity was elevated significantly in maengjong-juk extract diets supplemented group in a dose dependant manner. Hepatic TBARS contents were significantly decreased in maengjong-juk extract diets supplemented group compared to high cholesterol group. Maengjong-juk extract coated rice diets suppressed the protein oxidation significantly in liver. Activities of hepatic antioxidative enzymes such as total SOD, CuㆍZn-SOD, Mn-SOD, GSH-Px and catalase activities of maengjong-juk extract coated rice diets were significantly higher than those of high cholesterol diet. Total hepatic glutathione content was significantly increased by maengjong-juk extract coated rice diets administration. According to this study, numerous antioxidative materials and phytochemicals containing in maengjong-juk extracts appear to protect antioxidative systems in C57BL/6 mice fed bamboo extract coated rice diet. Effects of Intravenous Administration of Taurocholate on Hepatic Catechol-O-Methyltransferase Activity in Common Bile Duct Ligated Rats

  • Possible mechanism of decreased catechol-O-methyltransferase (COMT) activity in cholestatic rat liver was studied. Hepatic and serum COMT activities were determined from the experimental rats with common bile duct ligation (CBDL). The Michaelis-Menten constants in this hepatic enzyme were also measured. The activities of cytosolic, mitochondrial and mircosomal COMT as well as their Vmax values were found to be decreased significantly in CBDL plus taurocholic acid (TCA) injected group than in the control group, such as CBDL alone groups. However, their Km values in the experimental groups did not vary. Serum COMT activity increased slightly in the CBDL plus TCA injected group than in the control group. The above results suggest that TCA represses biosynthesis of the COMT in the liver. The elevated activity of the serum COMT is believed to be caused by the increment of membrane permeability of hepatocytes upon TCA mediated liver cell necrosis.

Antioxidant Activities of Decursinol Angelate and Decursin from Angelica gigas Roots

  • The anti-oxidant activities of decursinol angelate (1) and decursin (2) isolated from Angelica gigas were investigated. These two coumarins exhibited decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4-intoxication$ in rats. They also showed increase in anti-oxidant enzyme such as hepatic cytosolic superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH-px) in $CCl_4-intoxicated$ rats. These results suggest that decursinol angelate (1) and decursin (2) from A. gigas possess not only the anti-oxidant, but also the hepatoprotective activities in rats.
