가미옥천산의 ICR 마우스 경구 투여에 의한 급성 독성시험 연구 (Acute Toxicity of Kami-Okchun-San in ICR-Mice)

  • Kami-Okchun-San(OCS) is known as an effective herbal medicine on Type 2 diabetes. We peformed to investigate acute toxicity of OCS on ICR mice. ICR mice in acute toxicity experiment were administered orally with dosages of 3,200mg/kg (low dosage group), 4,000mg/kg (middle dosage group), 5,000mg/kg (high dosage group) per single time, respectively. Body weights, clinical signs, motalities and histopathological finding were observed daily for 14 days according to the Regulation of Korean Food and Drug Administration(1999. 12. 22). Single oral administration of OCS with different dosages, no animals died of the test drug. Autopsy of animal revealed no abnormal gross findings. Therefore, LD50 value of OCS for ICR mice was more than 5,000mg/kg on oral route. Normally increasing changes were observed in body weight, drinking water and food intake in every dosage group. Hematological parameters were also observed normally in all animals. No histopathological lesions were observed in both control and treated animals. Above data suggest that no toxic dose level of OCS in ICR mice is considered to be more than 5,000mg/kg. Therefore, it was concluded that OCS have no effect on acute toxicity and side effect in ICR mice.

Alternaria mycotoxins and its incidence in fruits and vegetables

  • Alternaria is a ubiquitous fungal genus, widely distributed in the environment and a range of different habitats. It includes both plant pathogenic and saprophytic species, which can affect crops in the field or cause post-harvest spoilage of plant fruits and kernels. Numerous Alternaria species cause damage to agricultural products including cereal grains, fruits and vegetables, and are responsible for severe economic losses worldwide. Most Alternaria species have the ability to produce a variety of secondary metabolites, which may play important roles in plant pathology as well as food quality and safety. Alternariol (AOH), alternariol monomethyl ether (AME), tenuazonic acid (TeA), tentoxin (TEN) and altenuene (ALT) are considered the main Alternaria compounds thought to pose a risk to human health. However, food-borne Alternaria species are able to produce many additional metabolites, whose toxicity has been tested incompletely or not tested at all. Both alternariols are mutagenic and their presence in cereal grain has been associated with high levels of human esophageal cancer in China. TeA exerts cytotoxic and phytotoxic properties, and is acutely toxic in different animal species, causing hemorrhages in several organs. The possible involvement of TA in the etiology of onyalai, a human hematological disorder occurring in Africa, has been suggested. Altertoxins (ALXs) have been found to be more potent mutagens and acutely toxic to mice than AOH and AME. Other metabolites, such as TEN, are reported to be phytotoxins, and their toxicity on animals has not been demonstrated up to now. Vegetable foods infected by Alternaria rot are obviously not suitable for consumption. Thus, whole fresh fruits are not believed to contribute significantly with Alternaria toxins to human exposure. However, processed vegetable products may introduce considerable amounts of these toxins to the human diet if decayed or moldy fruit is not removed before processing. The taxonomy of the genus is not well defined yet, which makes it difficult to establish an accurate relationship between the contaminant species and their associated mycotoxins. Great efforts have been made to organize taxa into subgeneric taxonomic levels, especially for the small-spored, food associated species, which are closely related and constitute the most relevant food pathogens from this genus. Several crops of agricultural value are susceptible to infection by different Alternaria species and can contribute to the entry of Alternaria mycotoxins in the food chain. The distribution of Alternaria species was studied in different commodities grown in Argentina. These food populations were characterized through a polyphasic approach, with special interest in their secondary metabolite profiles, to understand their full chemical potential. Alternaria species associated with tomato, bell pepper, blueberry, apples and wheat cultivated in Argentina showed a surprisingly high metabolomic and mycotoxigenic potential. The natural occurrence of Alternaria toxins in these foods was also investigated. The results here presented will provide background for discussion on regulations for Alternaria toxins in foods.

산업장내(産業場內) 건강검진(建康檢診) 자료(資料)를 활용(活用)한 사상체질(四象體質)의 평가(評價) (The Evaluation of Sasang Constitution with Use of Health Examination Data in Workplace)

  • The subjects were 420 workers who received oriental and western health examination both in the workplaces for 1 month. This study was planned for the reflection of western health examination results through evaluation of Sasang constitution with use of data from western health examination, and for the presentation of basic data applicable to constitutional medicine with preventive medicine. Data that were collected in field research were analyzed into frequencies, One-Way ANOVA, Chi-square testing by the study purposes, and the research results were as follows: 1. The distribution of Sasang constitution in 420 subjects was Taeumin > Soumin > Soyangin, and the general characteristics of subject by Sasang constitution were no significant difference. 2. For the measured values for body by the subject of Sasang constitution, Taeumin had higher value for height, body weight, systolic and diastolic blood pressure than other 2 constitutions. Soumin had the lowest values in average for all items, and on the other hand, Soyangin had high pulse pressure compared to other constitutions. It was significant difference between constitutions. 3. For the hematological average, Taeumin had high average for FBS, Total-C, GPT, and Taeumin ,and Soyangin had high in Hb compared to Soumin. Soyangin had the highest average for ${\gamma}-GPT$, and there were significant differences between FBS, Total-C, Hb, GPT, ${\gamma}-GTP$, excepting for GOT. 4. For the result of health examination by the Sasang constitution of subject, Taeumin showed the highest percentage in the items of obesity, primary care, secondary suspicion, and showed significant difference by the constitution. For the distribution of the past disease history by the health examination result of Sasang constitution, Taeumin and Soumin had high prevalence rate for hypertension, liver disease, and Soyangin had high prevalence rate for liver disease, hypertension. For the requirement of lifestyle improvement to the subject, the highest requirements by the Sasang constitution were physical exercise for Soumin, regulation of body weight for Taeumin, and it showed significant difference. For the requirement of primary care by the constitution, the highest requiremetns were obesity control for Taeumin, blood pressure control for Soumin, Soyangin, and liver suspicion was the highest distribution for each constitution in the case of having decision for secondary suspicion.

Biomonitoring of Toxic Effects of Pesticides in Occupationally Exposed Individuals

  • Background: Workers in pesticide manufacturing industries are constantly exposed to pesticides. Genetic biomonitoring provides an early identification of potential cancer and genetic diseases in exposed populations. The objectives of this biomonitoring study were to assess DNA damage through comet assay in blood samples collected from industry workers and compare these results with those of classical analytical techniques used for complete blood count analysis. Methods: Samples from controls (n = 20) and exposed workers (n = 38) from an industrial area in Multan, Pakistan, were subjected to various tests. Malathion residues in blood samples were measured by gas chromatography. Results: The exposed workers who were employed in the pesticide manufacturing industry for a longer period (i.e., 13-25 years) had significantly higher DNA tail length ($7.04{\mu}m$) than the controls ($0.94{\mu}m$). Workers in the exposed group also had higher white blood cell and red blood cell counts, and lower levels of mean corpuscular hemoglobin (MCH), MCH concentration, and mean corpuscular volume in comparison with normal levels for these parameters. Malathion was not detected in the control group. However, in the exposed group, 72% of whole blood samples had malathion with a mean value of 0.14 mg/L (range 0.01-0.31 mg/L). Conclusion: We found a strong correlation ($R^2=0.91$) between DNA damage in terms of tail length and malathion concentration in blood. Intensive efforts and trainings are thus required to build awareness about safety practices and to change industrial workers' attitude to prevent harmful environmental and anthropogenic effects.

  • 본 시험은 sialic acid가 23%를 함유하도록 제조한 유청가수분해단백분말제제(whey protein of hydrolysis)의 기능성 식품원료로 개발을 위한 동물안전성을 평가에 연구목표를 두었다. 시험물질은 sialic acid 23%(v/v)와 원료인 GMP(glycomacropeptide)가수분해 단백질이 87%(v/v)로 구성되어 있었다(시험명: 23%-GNANA). 시험물질의 독성 유무는 한국식품의약안전청(KFDA, 2014)과 OECD(2008)의 의약품 등의 독성시험 기준에 따라 실시하였다. 평가방법으로서, 시험물질의 투여용량을 0, 1,250, 2,500 및 5,000 mg/kg/day하여 SPF Sprague-Dawley 계열 암수 랫드에 90일 동안 반복경구투여하였을 때 나타나는 독성 여부를 평가하였다. 평가항목으로서는 사망률, 일반증상관찰, 체중 변화, 사료섭취량 측정, 안검사, 요검사, 혈액학적 및 혈액생화학적 검사, 부검 시 장기의 중량 측정, 부검 시 육안적 및 조직병리학적 검사 등을 평가하였다. 결과로서, 시험물질로 인한 일반증상 및 사망동물은 발생하지 않았다. 또한, 안과학적 검사, 요검사 그리고 혈액학적 및 혈액이화학적 이상 여부와 부검 시 육안적 검사 및 조직병리학적 검사에서 대조군 대비 특이한 변화는 관찰되지 않았다(P<0.05). 이외의 평가 항목에서, 암컷은 사료섭취량과 증체율이 다소 감소하였고, 그리고 수컷에서는 요의 SG와 PRO 및 고환 무게가 다소 증가하는 경향을 보였지만, 시험물질-유래 경미한 변화(non-adverse effect)가 고용량군(5,000 mg/kg/day)만 확인되었다. Weight-based classification(독성 강도에 따른 분류)를 적용한 최종 독성평가 결과는 다음과 같다. 수컷의 경우, NOEL(No Observed Effect Level)은 5,000 mg/kg/day 그리고 암컷의 경우는 NOAEL(No Observed Adverse Effect Level)은 5,000 mg/kg/day로 최종 확인되었다. 따라서, 암수 모두에서 시험물질의 NOAEL은 투여최대용량인 5,000 mg/kg/day로 확인되었다. 결론적으로, 시험물질인 유청가수분해단백분말제제는 건강기능식품의 특성상 임상허용용량인 1,000 mg/kg/day의 5배 수준에서 안전함을 확인되었다.

HCB와 PCBs에 노출된 나일틸라피아, Oreochromis niloticus의 혈액성분의 변화 (Changes of Haematological Constituents in the Nile tilapia, Oreochromis niloticus Exposed to HCB and PCBs)

  • 유용 양식어류인 틸라피아를 대상으로 어체중당 1일 0.05, 0.25 및 0.50mg/kg의 HCB와 PCBs의 경 구투여에 의해 30일 동안 혈액성상 및 혈액화학적 변동을 조사하였다. HCB의 투여에 의한 틸라피아의 RBC 수는 20일 이후 0.50mg/kg, 30일째는 0.25mg/kg의 농도, hemoglobin 농도와 hematocrit 값은 20일 이후 0.50mg/kg 농도에서 유의한 감소를 나타냈다. PCBs의 투여에 의한 틸라피아의 RBC 수, hemoglobin 농도 및 hematocrit 값은 20일 이후 0.25mg/kg 이상의 농도에서 유의한 감소를 나타냈다. 혈청 무기성분인 calcium, magnesium 및 inorganic phosphorus 농도는 HCB를 투여한 모든 실험에서 뚜렷한 차이가 인정되지 않았다. 또한, PCBs를 투여한 모든 실험에서도 magnesium과 inorganic phosphorus 농도의 변동은 관찰되지 않았으나, calcium 농도는 20일 이후 0.50mg/kg 농도, 30일째는 0.25mg/kg 이상의 농도에서 유의한 감소를 나타냈다 HCB의 경구투여에 의한 혈청 total protein 농도는 30일째 0.50mg/kg 농도에서, PCBs의 경구투여에 의해서는 20일 이후 0.25mgfg 이상의 농도에서 유의하게 감소하였다. HCB 및 PCBs의 경구투여에 의한 glucose 농도는 20일 이후 0.25mg/kg 이상의 농도에서 유의한 증가를 나타냈다. HCB 및 PCBs의 경구 투여에 의한 GOT와 GPT 활성은 20일 이후의 0.50mg/kg에서 유의한 증가를 나타냈다. 결론적으로 HCB 및 PCBs의 경구투여에 의한 틸라피아의 혈액화학적 변동은 0.25 mg/kg 농도 이상에서 적어도 20일 후부터 영향을 받을 것으로 생각된다.

경동맥 내중막 두께에 따른 죽상경화반의 위험요인과의 상관관계 (Correlation between Carotid Intima-media Thickness and Risk Factors for Atherosclerosis)

  • 본 연구는 건강검진을 목적으로 내원하여 경동맥초음파를 실시한 113명을 대상으로 하였다. 경동맥의 내중막 두께는 심혈관 질환 및 뇌혈관 질환과 유의한 상관관계를 보이는 것으로 알려져 있다. 임상에서 선별 검사로 많이 이용되고 있는 경동맥 초음파를 이용하여 경동맥 내중막 두께와 체질량지수, 허리둘레, 혈청지질수치, 공복 혈당, 당화혈색소, 혈압등이 갖는 연관 관계에 대하여 알아보고자 하였다. 경동맥초음파상 IMT두께가 0.8 mm 이상을 비정상으로 간주하고 죽상경화증(atherosclerosis)의 유무를 평가하였으며, 혈청검사를 통하여 지질학적 수치 및 공복혈당수치, 당화혈색소를 분류하여 상관관계를 알아보았다. 그 결과 허리둘레(p=.022), 저밀도 콜레스테롤(p=.004), 공복혈당수치(p=.019), 당화혈색소 수치(p=.002)가 죽상경화증(atherosclerosis을 일으키는 예측인자로 분석되었다. ROC 곡선 분석에서 허리둘레에서 민감도 87.80%(95% CI : 73.8-95.9), 특이도 41.67%(95% CI : 30.2-53.9), 저밀도 콜레스테롤에서 민감도 78.05%(95% CI : 62.4-89.4), 특이도 50.00%(95% CI : 38.0-62.0 공복혈당에서 민감도 73.11%(95% CI : 57.1-85.8), 특이도 61.11 (95% CI : 48.9-72.4)), 당화혈색소에서 민감도 82.93%(67.9-91.8), 특이도 43.06%(31.4-55.3)를 나타내었다. 로지스틱 회귀분석에서 죽상경화증(atherosclerosis) 발생위험성은 허리둘레(WC)>76 cm에서 0.248배, 저밀도 콜레스테롤(LDL-C)${\geq}124mg/dL$에서 3.475배, 당화혈색소(HbA1C)>5.4%에서 0.618배로 나타났다. 향후 심뇌혈관질환의 효과적인 일차예방 역할을 위해 다수의 대상자를 대상으로 혈액검사수치를 고려하여 혈관의 추적관찰이 용이한 경동맥초음파를 통한 전향적연구가 필요할 것으로 사료 된다.

  • A study on blood pressure and hematocrit values of 1,559 people in 19 Korean rural areas was carried out in 1974 and the results were analyzed statistically. Obtained as follows: 1. The blood pressure according to sex and age groups (from the twenties to the seventies) was as follows: 1) The blood pressure of male by age group il) In the twenties, M (mean) was 125. 85/74, 15mm/Hg, a (standard deviation) was 15.9/10.2, and ill (standard error) was 1.55/0.99. (2) In the thirties, ${\delta}$ was 123.93/77.19 mm/Hg, a was 14.4/10.8, and m was 1.24/0.93. (3) In the forties, M was 128.44/81.15 mm/Hg, a was 23.9/14.7, and m was 2.16/1.33. (4) In the fifties, M was 128.48/181.24 mm/Hg, a was 24.7/13.9, and m was 2.05/1.16. (5) In the sixties, M was 135.80/81.70 mm/Hg, a was 27.4/18.8, and ${\delta}$ was 2.74/1.88. (6) In the seventies, M was 146.84/83. 16mm/Hg, ${\delta}$ was 24.5/10.0, and m was 5.62/2.30. 2) The blood pressure of female by age group (1) In the twenties, M was 117.89/73.33 mm/Hg, ${\delta}$ was 15.7/12.1, and m was 1.42/1.09. The range of distolic blood pressure in male was 50~160 mm/Hg and in female was 40~140 mm/Hg. 2. The hematocrit value according to sex and age groups was as follows: 1) The hematocrit values of male by age group (1) In the twenties, M was 42.72%, ${\delta}$ was 3.05, and m was 0.30. (2) In the thirties, M was 41.77%, ${\delta}$ was 3.29, and m was 0.28. (3) In the forties, M was 41.39, ${\delta}$ was 3.86, and m was 0.35. (4) In the fifties, M was 40.12%, ${\delta}$ was 3.65, and m was 0.30. (5) In the sixties, M was 39.88%, ${\delta}$ was 3.81. and m was 0.38. (6) In the seventies, M was 38.47%, ${\delta}$ was 2.27, and m was 0.52. 2) The hematocrit values of female by age group (1) In the twenties, M was 35.40%, ${\delta}$ was 3.37, and m was 0.30. (2) In the thirties, M was 35.50%, ${\delta}$ was 3. 35, and m was 0.22. (3) In the forties, M was 35.75%, ${\delta}$ was 3. 18, and m was 0.22. (4) In the fifties, M was 35.84%, ${\delta}$ was 3.30, and m was 0.22. (5) In the sixties, M was 35.70%, ${\delta}$ was 3.35, and m was 0.30. (6) In the seventies, M was 35.08%, ${\delta}$ was 3.08, and m was 0.60. The range of hematocrit values in male was 23~50% and in female was 18~50% (un associated with age groups). 3. In comparison with the blood pressure and the value of hematocrit of study groups showed that the blood pressure raised higher but the value of hematocrit got lowered on the contrary as the groups are getting older. 4. Total number of patients with hypertension was 165(10.6%) which were consisted with 71 male (11.3%) and 94 female (10.1%). But only two cases of the male patient and one case of the female patient were associated with protein uria. 5. The incidence of anemia by hematocrit values was as follows: 1) The incidence of male anemia patients based on$\leqq$41% ($\leqq$39%). (1) In the twenties, incidence was 43.90% (16.98%). (2) In the thirties, 41.48% (25.93%). (3) In the forties, 42.62% (25.41%). (4) In the fifties, 62.76% (40.69%). (5) In the sixties, 70% (38%). (6) In the seventies, 84.21% (73.68%). 2) The incidence of female anemia patients bailed on $\leqq$35% ($\leqq$34%). (1) In the twenties, incidence was 43.90% (37.39%). (2) In the thirties, 48.21 % (33.93%). (3) In the forties, 43.58% (32.57%). (4) In the fifties, 45.91 % (34.09%). (5) In the sixties, 47.11% (37.19%). (6) In the seventies, 50% (46.15%) 6. The incidence rate of anemia patients with hypertension was highest in the age group 51~60 in male and 31~40 in female.

양식어류의 선별과정중 수심감소와 어류의 수조이동에 따른 스트레스 반응 (Stress Responses of Cultured Fishes Elicited by Water Level Reduction in Rearing Tank and Fish Transference during Selection Process)

  • 양식현장에서 선별작업시 처리되는 사육수조의 수심감소와 어류이송에 따른 넙치 (대, 소)와 큰민어의 스트레스 반응을 알아보기 위하여 스트레스 지표로 알려져 있는 혈액학적 요인, 코티졸, 글루코스, 젖산 및 삼투질 농도를 조사하였다. 수심감소에서 넙치대의 Ht는 실험전 $14.6\%$였던 것이 10시간째까지 계속 높아져 $23.5\%$를 보였으나, 실험종료시 (46시간째) $14.4\%$로 회복되었다. 넙치대의 코티졸 농도는 실험개시시에 1.9ng/mL였던 것이 22시간째에 13.7 ng/mL로 높아졌고, 46시간째에는 4.0ng/mL로서 개시시의 수준으로 회복되었다. 큰민어는 4시간째 282.3ng/mL, 22시간째 350.5ng/mL로 높아졌으나, 실험종료시에는 16.0ng/mL로 낮아졌다. 큰민어의 글루코스는 실험개시시 57.0mg/dL로부터 22시간째에는 138.0mg/dL조 유의하게 높아졌다 젖산은 모든 어종에서 유의차가 인정되지 않았다. 어류이송에 따른 넙치대의 RBC는 실험개시시의 $1.9\times10^6 cell/{\mu}L$로부터 실험종료시의 $4.2\times10^6 cell/{\mu}L$로 높아졌다. 큰민어의 Hb는 실험개시시의 9.8g/dL로부터 실험종료시에 20.4g/dL로 개시시 보다 유의하게 높았다. 넙치대의 코티졸 농도는 실험개시시의 1.2ng/mL로부터 이동 1시간째에 95.3ng/mL로 높아졌고, 큰민어도 실험개시시의 5.5ng/mL로부터 1시간째에 175.5ng/mL로 높아졌다. 큰민어의 글루코스는 1, 3시간째 각각 132.5 mg/dL, 129.5 mg/dL로 실험개시시 46.5 mg/dL 보다 높은 수준을 보였다.

  • 연구배경: 속립성 결핵환자에서 말초혈액의 변화는 골수생검상 결핵성 병변을 보인 군에서 더 많은 것으로 알려져 있고, 폐결핵보다 속립성 결핵에서 세포성 면역의 변화가 심한 것으로 연구되어지고 있다. 방법: 대상은 1990년에서 1994년 사이에 원주의과 대학부속 원주기독병원에 내원하여 속립성 결핵으로 진단받고 골수생검을 시행받은 환자 40예 였다. 이들을 골수생검상 결핵성 병변을 보인 양성군과 보이지 않은 음성군으로 나누어 임상상, 말초 혈액의 혈청 ADA, 수용성 인터루킨 2 수용체, 임파구 아형 분류를 시행하여 비교하였다. 결과: 1) 대상 환자의 평균연령은 39세로, 남녀는 23:17예 이었다. 2) 동반된 폐외 결핵으로는 결핵성 뇌막염이 9예, 결핵성 관절염이 6예, 결핵성 흉막염이 2예 있었다. 3) 골수검사상 60%(24/40)가 결핵성 병변을 보였다. 4) 말초혈액검사상 빈혈은 60%(24/40)가 있었으며 골수생검 양성군에서 11예 음성군에서 13예였고, 백혈구감소증은 12%(5/40)로 양성군에서 4예 음성군에서 1예였고, 혈소판 감소증은 10%(4/40)로 양성군이 3예 였다. 5) 혈청 ADA는 평균 83 U/L로 양성군에서 90 U/L, 음성군에서 70.6 U/L 이었다(p=0.23). 6) 혈청 가용성 interleukin 2 수용체 활성도는 평균 4,643 pmol/L 였으며 양성군에서 $6,840{\pm}7,446\;pmol/L$(n=10), 음성군 $1897{\pm}1663\;pmol/L$(n=8)으로 양성군에서 더 높았다(p=0.06). 7) 혈청내 T 임파구 아형분류에서 총 T 임파구는 평균 64%으로 양성군에서 $62{\pm}19%$(n=18), 음성군에서 $73{\pm}10%$ 였고(n=7)(p=0.2), $T_4/T_8$ ratio는 평균 $1.16{\pm}0.5$으로 양성군에서 $1.14{\pm}0.5$, 음성군에서 $1.18{\pm}0.5$였다(p=0.8). 8) 일부 환자(9예)에서 BAL의 T 임파구 아형분류을 시행하였으며 $T_4/T_8$ ratio는 $1.97{\pm}1.2$으로, 말초혈액소견과 비교하여 더 증가되어 있었다. 결론: 이상의 결과로 속립성 결핵의 골수생검 양성률은 60% 였으며, 골수생검상 결핵성 병변을 보인 군에서 말초혈액내 변화와 세포성 면역의 변화가 더 심한 경향을 보였다.
