Effects of Zn-L-Selenomethionine on Carcass Composition, Meat Characteristics, Fatty Acid Composition, Glutathione Peroxidase Activity, and Ribonucleotide Content in Broiler Chickens

  • The effects of organic Zn-L-selenomethionine (Zn-L-SeMet) at 0.3 ppm on carcass composition, meat characteristics, fatty acid composition, glutathione peroxidase activity, and ribonucleotide content were compared against the commercial inorganic sodium selenite (Na-Se) and the combination of the two, in commercial broilers. A total of 540 one day-old chicks were assigned at random to 3 dietary treatments : i) commercial inorganic selenium as control or T1, ii) a 1:1 ratio of inorganic and organic selenium as T2, and iii) organic selenium as T3. Carcass composition, meat characteristics, cholesterol content, fatty acid composition, and ribonucleotide content were generally unaffected by treatments. However, discrepancy were significantly observed in glutathione peroxidase activity (GSH-Px) and water holding capacity, with organic selenium showing higher glutathione peroxidase activity (p<0.01) and lower shrinkage loss (p<0.05), respectively. These findings could be explained by the contribution of organic selenium in bioavailability of GSH-Px. However, having conducted in a commercial close house system with sufficient amount of nutritional supplementation, the present study demonstrated little or no effects of organic Zn-L-SeMet on meat characteristics, fatty acid composition, and ribonucleotide content (flavor characteristic) in broiler chickens.

Production and Characterization of Selenium Peptide from Saccharomyces Cerevisiae (효모를 이용한 selenium peptide 생산 및 특성 연구)

  • Selenium containing peptide was produced by culturing yeast with selenium, Selenium was broadly incorporated in the various size of proteins based on the GPC analysis of the total yeast protein. The ratio of selenium to protein increased with the concentration of added selenium in the culture medium. Antioxidant activity (glutathione peroxidase-like activity) was proportional to the concentration of selenium concentration in the peptide. Different size of proteins were obtained by hydrolyzing the total yeast protein by protease XIV. Average molecular weight of selenium peptide was analyzed by GPC. Glutathione peroxidase (GPx) activity of the selenium peptide increased as the size of peptide decreased. Sodium selenite had strong inhibition on the yeast growth than sodium selenate. The ratio of selenium to protein was higher with sodium selenate than with sodium selenite. These results showed the potentials of selenium peptide production by yeast cultivation.

Oxidative Stress in Vivax Malaria

  • Malaria is still a leading cause of morbidity and mortality. The increase in lipid peroxidation reported in malaria infection and antioxidant status may be a useful marker of oxidative stress during malaria infection. The aim of this study was to investigate the role of antioxidant enzymes against toxic reactive oxygen species in patients infected with Plasmodium vivax and healthy controls. Malondialdehyde levels, superoxide dismutase, and glutathione peroxidase activities were determined in 91 P. vivax patients and compared with 52 controls. Malondialdehyde levels, superoxide dismutase, and glutathione peroxidase activities were $8.07{\pm}2.29$ nM/ml, $2.69{\pm}0.33$ U/ml, and $49.6{\pm}3.2$ U/g Hb in the patient group and $2.72{\pm}0.50$ nM/ml, $3.71{\pm}0.47$ U/ml, and $62.3{\pm}4.3$ U/g Hb in the control group, respectively. Malondialdehyde levels were found statistically significant in patients with vivax malaria higher than in healthy controls (P<0.001). On the other hand, superoxide dismutase and glutathione peroxidase activities were found to be significantly lower in vivax malaria patients than in controls (P<0.05). There was an increase in oxidative stress in vivax malaria. The results suggested that antioxidant defense mechanisms may play an important role in the pathogenesis of P. vivax.

Protective Enzymes of Paraquat-Resistant Conyza bonariensis (Paraquat 저항성 망초의 protective 효소)

  • The resistance of Cornyza bonariensis to herbicide paraquat was investigated by evaluating the activities of three enzymes concerning in scavenging paraquat-generated toxic oxygen species such as superoxide radical and hydrogen peroxide in resistant and susceptible biotypes. Conyza bonariensis inhabited in cultivated area was more tolerant to paraquat than that of uncultivated area. This is the first report that a biotype of Cornyza bonariensis has appeared in an area with repeated paraquat treatments of Korea. Superoxide dismutase activity of resistant biotype was 20% higher as 150 than that of susceptible biotype. Ascorbate peroxidase activity of resistant biotype was 44% higher than that of susceptible biotype. Glutathione reductase activity of resistant biotype was 64% higher than that of susceptible biotype. It can be concluded from above results that the resistance of Conyza bonariensis to paraquat depends partially on the toxic oxygen species-scavenging efficiency of protective multienzymatic system which is composed of three enzymes, superoxide dismutase, ascorbate peroxidase, and glutathione reductase.

Studies on the Relationship of Lipid Peroxidation and Drug Metabolizing Enzyme in Regenerating Rat Liver (재생중인 흰쥐간의 Lipid Peroxidation과 약물대사효소의 상관관계에 관한 연구)

  • The activities of aminopyrine demethylase which is marker enzyme of the microsomal drug-metabolizing system, NADPH-cytochrome a reductase and glutathione peroxidase were measured during the course of liver regeneration after about seventy percent hepatectomy in Wistar rats. In addition, the extent of lipid peroxidation and contents of cytochrome P-450 were also measured. Partial hepatectomy produced a significant depression in aminopyrine demethylase, to reach a minium about 24 hours after operation, but this activity was increased to normal value during regeneration. On the other hand, in sham-operated animals, this showed no change. All the activities of NADPH-chrome P-450 contents of liver microsomes were rapidly decreased at the early stage of regeneration. These values returned to normal after 7 days. By contrast, the activity of glutathione peroxidase was nearly unchanged. According to these results, at the early stage of regeneration, the decrease of cytochrome P-450 and NADPH-cytochrome c reductase activity lead to decrease of lipid peroxidation and drug metabolizing enzyme activity. But these phenomena were not detected after 7 days of regeneration.

Effects of Pine Needle on Lipid Composition and TBARS in Rat Fed High Cholesterol (솔잎분말이 고 콜레스테롤 급여 흰쥐의 체지질구성과 TBARS량에 미치는 영향)

  • Effects of pine needle on lipid composition and TBARS(thiobarbituric acid reactive substances) were investigated in rat fed high cholesterol. Plasma total cholesterol showed a tendency to decrease in pine needle group(P<0.05), however plasma HDL-cholesterol and liver total cholesterol showed no significantly different(P>0.05). Contents of plasma TBARS ranged from 23.15 nmol/ml to 35.38 nmol/ml and pine needle feeding resulted in a decrease in plasma TBARS(P<0.05). Contents of TBARS in liver showed a tendency to increase in the cholesterol group, however these values were lowered by pine needle feeding(P<0.05). Activity of glutathione peroxidase in liver alues showed no significantly different among treatment groups (P>0.05).

  • Physiological cellular activities responses to cadmium (Cd) exposure in green algae with several reductases activities and viability of the cell were examined. The cell division of green algae, Selenastrum capricornutum treated with 5ppm was significantly decreased than that of normal algae. The mean cell number of normal algal culture was as twice much as than that of algae at 6 days after Cd treatment. The cellular viability of algae was analysed by flow-cytometry with fluorescent dye after esterase reaction on cell membrane. The 85.35% of cellular viability of normal culture was decreased to 34.35% when algae was treated with 5 ppm of Cd at 6 days after treatment. It was considered that those method of flow-cytometry is useful tool for toxicity test on micro-organisms in the respect of identifying cellular viability. Also, the activities of both glutathione peroxidase (GPX) and ascorbate peroxidase (APX), which are indirectly react against oxidative stress through reduction of glutathione by Cd were significantly increased with 25%. It is considered that both GPX and APX are involved in the metabolic pathway of Cd -detoxification with similar portion in Selenasturm capricornutum.

Effects of Aminotriazole on Lung Toxicity of Paraquat Intoxicated Mice (Paraquat중독에 의한 폐독성에 미치는 Aminotriazole의 영향)

  • Background: Paraquat, a widely used herbicide, is extremely toxic, causing multiple organ failure in humans. Paraquat especially leads to irreversible progressive pulmonary fibrosis, which is related to oxygen free radicals. However, its biochemical mechanism is not clear. Natural mechanisms that prevent damage from oxygen free radicals include changes in glutathione level, G6PDH, superoxide dismutase(SOD), catalase, and glutathione peroxidase. The authors think catalase is closely related to paraquat toxicity in the lungs Method: The effects of 3-amino-1,2,4-triazole(aminotriazole), a catalase inhibitor, on mice administered with paraquat were investigated. We studied the effects of aminotriazole on the survival of mice administered with paraquat, by comparing life spans between the group to which paraquat had been administered and the group to which a combination of paraquat and aminotriazole had been administered. We measured glutathion level, glucose 6-phosphate dehydrogenase(G6PDH), superoxide dismutase(SOD), catalase, and glutathione peroxidase(GPx) in the lung tissue of 4 groups of mice: the control group, group A(aminotriazole injected), group B(paraquat administered), group C(paraquat and aminotriazole administered). Results: The mortality of mice administered with paraquat which were treated with aminotriazole was significantly increased compared with those of mice not treated with aminotriazole. Glutathione level in group B was decreased by 20%, a significant decrease compared with the control group. However, this level was not changed by the administration of aminotriazole(group C). The activity of G6PDH in all groups was not significantly changed compared with the control group. The activities of SOD, catalase, and glutathione peroxidase(GPx) in the lung tissue were significantly decreased by paraquat administration(group B); catalase showed the largest decrease. Catalase and GPX were significantly decreased by aminotriazole treatment in mice administered with paraquat but change in SOD activity was not significant(group C). Conclusion: Decrease in catalase activity by paraquat suggests that paraquat toxicity in the lungs is closely related to catalase activity. Paraquat toxicity in mice is enhanced by aminotriazole administration, and its result is related to the decrease of catalase activity rather than glutathione level in the lungs. Production of hydroxyl radicals, the most reactive oxygen metabolite, is accelerated due to increased hydrogen peroxide by catalase inhibition and the lung damage probably results from nonspecific tissue injury of hydroxyl radicals.

Improvement of In Vitro Development of Bovine Embryos in a Medium Containing Selenium

  • The objective of this study was to investigate the effect of selenium (Se) on in vitro development of bovine embryos. In CR1BSA, FBS-free medium, the bovine embryos could not proceed past the developmental block more efficiently to morula stage than in chemically undefined media. Addition of glutathione precursor, cysteine, with $\beta$-mercaptoethanol did not improve the development in chemically defined medium and neither did glutathione alone. Exogenous selenium improved the embryonic development to the morula and blastocyst stages at 6 days post-insemination (dpi) significantly (67.1% vs 57.5%, p<0.05), and blastocyst stage at 8 dpi (30.1% vs 20.5%, p>0.05). These improvements might be induced by elevated glutathione peroxidase activity due to addition of Se, and a possible mechanism of selenium to elevate the activity of glutathione peroxidase is discussed.

Protective Effect of an Aged Garlic-bamboo Salt Mixture on the Rat with the Alcohol-salicylate Induced Gastropathy (마늘-죽염 제제가 위장 장애 유발 흰쥐의 항산화 효소활성에 미치는 영향)

  • Garlic has been known to be effective against the gastrointestinal diseases which can be induced by production of oxygen-derived free radical. It has been shown that bamboo salt is effective on the treatment and prevention of various gastrointestinal disorders. Bamboo salt is a processed salt invented by a Korean, ll-Hoon Kim. It has been reported that garlic and bamboo-salt are useful to the treatment of gastric disorders in Korea. To clarify the protective mechanism of the garlic-bamboo salt mixture, the gas-tropathy was induced in rats with alcohol-salicylate and the activities of the free radical scavenging enzymes were examined. In this study, we found that the garlic-bamboo salt mixture reduced the severity of hemorrhagic lesion in gastric mucosa in the rats. In addition, the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR) were significnatly increased and the total content of GSH was recovered. From those results, we concluded that the protective effect of the garlic-bamboo salt mixture on gastropathy in rats is its ability to recover the level of GSH and to increase the activities of the free radical scavenging enzymes (SOD, GPx, GR).
