핑거루트(Boesenbergia pandura)와 생강(Zingiber oficinale Rosecoe)의 항산화 및 항균 활성 비교 (Comparison of Antioxidant and Antimicrobial Activities of Fingerroot (Boesenbergia pandura) and Ginger (Zingiber officinale Roscoe))

  • In this study, fingerroot (Boesenbergia pandurata) and ginger (Zingiber officinale Roscoe) were extracted with water, 70% and 100% ethanol, respectively. The contents of polyphenols, flavonoids and ascorbic acid, antioxidant and antimicrobial activities of these extracts were measured. With respect to the polyphenol compound content, both fingerroot and ginger extract showed the highest in 70% ethanol extract, and the fingerroot extract showed a generally higher content than the ginger extract. The content of flavonoids, fingerroot and ginger extracts showed values of 1,247.14~1,259.93 ㎍%, and there was no big difference between extracts. The content of ascorbic acid was the highest in 100% ethanol extract in both fingerroot and ginger. Fingerroot extract was slightly higher than ginger extract in all extracts. The DPPH radical scavenging activity of fingerroot and ginger extracts showed the highest antioxidant activity in 100% ethanol extract, and fingerroot showed a slightly higher activity than ginger. The ABTS radical scavenging activity was high in 70% and 100% ethanol extracts of both fingerroot and ginger. In the activity between fingerroot and ginger extracts, ginger was higher in water extract and fingerroot was slightly higher in ethanol extract. Fingerroot and ginger extracts showed antimicrobial activity against Staphylococcus aureus and Bacillus cereus.

  • Physiological activities of Korean-grown ginger (KG) and Chinese-grown ginger (CG) extracts were examined. Ginger was extracted with water, and 50 and 100% ethanol, and then nitrite-scavenging activity (NSA), superoxide dismutase (SOD)-like activity, and electron-donating ability (EDA) of extracts were investigated. NSA at pH 1.2 was the most effective in 50% ethanol extracts of both origins. SOD-like activities of water and 50% ethanol extracts of both samples were 8.66-35.95% lower than those of 1 and 0.1% L-ascorbate solutions. SOD-like activity of KG extracts was higher than that of CG extracts, and water extracts of samples were the highest. EDA of KG extract was higher (22.23-86.95%) than that of CG extract, while both sample extracts showed lower EDA than those of 1 and 0.1% L-ascorbate solutions.

과도한 운동 스트레스에 대한 숙성생강 추출물의 방어효과 (Protective Effects of Aged Ginger Extracts on Excessive Exercise-induced Stress)

  • 생강의 운동 스트레스에 대한 방어효과를 알아보기 위해 동결건조, 열풍건조, 숙성한 생강을 제조하여 추출물을 근육세포에 H2O2와 함께처리한 결과 생강을 숙성한 시료는 숙성하지 않은 시료에 비하여 우수한 근육세포 보호효과를 나타내었다. 또한 숙성생강 추출물을 마우스에 투여시 운동으로 인한 혈액내 LDH, lactate, GOT 증가를 억제시키는 경향을 나타내었다. 따라서 숙성생강을 섭취시 과도한 운동으로 인한 스트레스에 대한 방어효과를 기대할 수 있을 것으로 사료된다. 이러한 결과를 토대로, 향후 생강의 숙성 온도 및 시간에 따른 운동 스트레스 방어효과를 측정하여 가장 높은 활성을 나타내는 최적의 숙성 조건을 확립하는 연구를 진행할 예정이다.

Rosemary, Sage, Oregano와 Ginger의 메탄올과 헥산 추출물의 항산화 작용에 대한 카테킨과 아스코르브산의 상승 효과 (Synergistic Effects of Catechin or Ascorbic Acid on Antioxidative Activities of Hexane and Methanol Extracts from Rosemary, Sage, Oregano, and Ginger)

  • 역마이셀 미강유를 기질로 하여 rosemary, sage, oregano와 ginger의 메탄올과 헥산 추출물 500ppm에 카테킨 또는 아스코르브산 200 ppm을 각각 첨가하여 Rancimat으로 유도기간을 측정하여 항산화 상승효과를 조사하였다. Rosemary, sage, oregano와 ginger의 메탄을 추출물에 카테킨 첨가시 모두 상승효과를 나타내었으며, 아스코르브산을 첨가시 ginger를 제외하고는 항산화 상승효과를 보였다. 항산화 상승효과는 전반적으로 아스코르브산보다 카테킨이 높게 나타났다. 헥산 추출물의 경우, 카테킨 첨가시 oregano와 ginger에서 강한 상승효과를 보였으며, ascorbic acid 첨가시, rosemary와 sage에서 상승효과가 나타났다.

  • 이상의 결과에서 다음과 같은 생강 추출물의 항산화 효과를 요약할 수 있었다. 1. 항온 저장시에는 생강 추출물을 첨가한 모든 시료에서 항산화 효과를 나타내었으며 그 효과는 3%를 첨가시 가장 높았고 그 정도는 BHT와는 비슷한 효과를 보였으나 tocopherol에 비해서는 월등히 높은 항산화 효과를 나타내었다. 2. 가열 처리시에도 생강 추출물의 농도에 관계없이 비슷한 정도의 항산화 효과를 나타내었으며 특히 대두유의 경우에는 BHT, tocopherol보다 뚜렷하게 높은 항산화 효과를 보였다. 3. 건강편을 튀김유에 5 및 10%씩 넣고 가열한 후 튀김 횟수에 따른 튀김유의 산패 정도를 측정시 두 시료 모두에서 산패가 억제되었으며 그 중 5%의 건강편이 더 우수한 것으로 나타났다.

용매에 따른 카레 향신료 추출물의 항산화 효과 및 혼합효과 (Antioxidant and Mixture Effects of Curry Spices Extracts Obtained by Solvent Extraction)

  • 카레분의 단품향신료와 카레분(curry powder)을 메탄올, 에틸아세테이트, 헥산으로 추출하여 수율, 항산화 효과, 총페놀함량, 총토코페롤 함량을 측정하였다. 메탄올, 에틸아세테이트, 헥산의 추출 수율은 각각 $6.4{\sim}42.9$, $3.9{\sim}26.2$, $1.6{\sim}29.2\;%$로 극성이 높아질수록 수율이 증가하였다. 미강유의 각 용매별 추출물을 1,500ppm 첨가하여 Rancimat로 유도기간을 측정한 결과 메탄올 추출물은 rosemary, sage, ginger, turmeric, nutmeg 순으로 우수하였고, 에틸아세테이트 추출물은 rosemary, sage, oregano, thyme, ginger의 순이었으며 헥산 추출물은 rosemary, sage, oregano, ginger, thyme 순이었다. 1,500ppm을 미강유에 첨가하였을 때 BHT보다 유도기간이 길게 나타난 향신료는 3가지 용매 추출물에서 공통적으로 rosemary, sage, ginger, oregano, thyme이었으며 이들 향신료를 200, 500, 1,000, 1,500, 2,000 ppms 농도로 미강유에 첨가한 후 유도기간을 측정한 결과 rosemary와 sage는 헥산 추출물 2,000ppm에서, ginger는 메탄올 추출물 2,000ppm에서 우수한 효과를, oregano와 thyme은 에틸아세테이트 추출물이 1,500ppm과 2,000ppm에서 큰 차이 없이 우수한 효과를 나타내었다. Rosemary의 혼합에 따라 ginger와 thyme의 항산화 효과가 크게 증가함을 알 수 있었으나 상승효과라기 보다는 rosemary로 인한 유도기간의 연장 효과였다. 총페놀 함량은 메탄올 추출물은 $17,924{\sim}801\;mg%$, 에틸아세테이트 추출물은 $13,893{\sim}57\;mg%$, 헥산 추출물은 $6,864{\sim}12\;mg%$이었으며 총페놀함량과 항산화 효과가 일치하지는 않았다. 토코페롤의 경우 oregano가 3194.8mg%로 가장 높았으며, sage 1014.8 mg%, rosemary 571.1 mg%, thyme 187 mg%였고 ginger에서는 토코페롤이 검출되지 않았으며 토코페롤함량과 항산화 효과는 뚜렷한 상관관계가 발견되지 않았다.

생강 추출물 투여가 전구염증성 사이토카인 $IFN-{\gamma}$와 항염증성 사이토카인 IL-10 분비량에 미치는 영향 (The Effects of Zingiber officinale Roscoe Extracts on Mouse $IFN-{\gamma}$ and IL-10 Production)

  • Ginger(Zingiber officinale Roscoe) has been used as a raw material in many various traditional preparations since the ancient times. As a component of traditional health products, ginger is known to be effective as an appetite enhancer, and has anti-cold and anti-inflammatory activities. This study was performed to investigate the immunomodulative effects of Zingiber officinale Roscoe in mice, using ex vivo experiments. In order to elucidate ginger's immunomodulative effects of Ginger, water extracts were orally administered to mice, and isolated macrophages were used as the experimental model. To identify the ex vivo effects, six to seven week old Balb/c mice were fed a chow diet ad libitum and the ginger water extracts were administered orally every other day for two or four weeks at two different concentrations(50 and 500 mg/kg b.w.). The results show that IL-IO and $IFN-{\gamma}$ were detected in the 500 mg/kg b.w. supplemented group with LPS stimulation in all cases. Also, the $IFN-{\gamma}$ /IL-10 ratio ranged from 3~5 with mitogen stimulation such as Con A and LPS. In conclusion, this study suggests that ginger extracts may enhance the immune function by regulating the cytokine(IL-10 and $IFN-{\gamma}$) production capacity of activated macrophages in mice.

  • Recently many investigators have initiated searches for immunomodulating substances from natural food sources. Ginger (Zingiber officinale Roscoe) has been used as a raw material in many traditional preparations since the ancient time. This study was performed to investigate the immunomodulative effects of Zingiber officinale Roscoe in mice, using ex vivo experiments. In order to elucidate the immunomodulative effects of Ginger, water extracts of the plant were orally administrated into mice, and isolated splenocytes and macrophages were used as experimental model. In order to identify its ex vivo effect six to seven week old Balb/c mice were fed ad libitum on a chow diet and water extracts of ginger were orally administrated every other day for two weeks at two different concentrations (50 and 500 mg/kg b.w.). After preparing the single cell suspension, the proliferation of splenocytes was determined by MTT assay. The result of ex vivo study showed that the highest proliferation of splenocytes and macrophage activatation was seen in the mice orally administrated at the concentration of 500 mg/kg b. w. of ginger water extracts. In conclusion, this study suggests that ginger extracts nay enhance the immune function by regulating the splenocyte proliferation and cytokine prodution capacity by activated macrophages in mice.

생강(生薑) 추출물의 위장관 기능개선 및 체온저하 억제효과에 대한 실험적 연구 (Effect on Promoting Gastrointestinal Function and Inhibiting of Decreasing Body Temperature of Ginger Extracts(Zingiber Officinale))

  • This study was performed to investigate the effect of promoting gastrointestinal function and inhibiting of decreasing body temperature of ginger extract(Zingiber officinale) in rats. In order to elucidate the gastrointestinal function and inhibiting effect of body temperature of native ginger and improved ginger, water extracts of ginger were orally administrated into rats. The results are as follows: The gastrointestinal transit time was significantly decreased in native ginger(7.66hrs) and improved ginger(7.72hrs) extract administrated groups compare to control group(8.44hrs). The mean red faecal weight was increased in native ginger(30.6%) and improved ginger(31.1%) extract administrated groups compare to control group(24.9%) for 24hrs. Inhibiting effect of decreasing body temperature induced by serotonin was increased in native ginger($1.116^{\circ}C$) and improved ginger($1.416^{\circ}C$) extract administrated groups compare to positive control group($0.384^{\circ}C$) during 40 minutes. Gastrin and CGRP immunoreactive density was more strongly expressed in native ginger and improved ginger extract administrated groups compare to control group. Serotonin immunoreactive density was more weakly expressed in native ginger and improved ginger extract administrated groups compare to control group. These results suggest that ginger extracts may enhance physiological activity such as gastrointestinal motility, protection of mucosa and gastric acid secretion in gastrointestinal tracts, and inhibits decreasing body temperature

생강 추출물 투여가 마우스 면역세포 활성에 미치는 영향 (Effect of Zingiber Officinale Roscoe Extracts on Mice Immune Cell Activation)

  • Ginger (Zingiber officinale Roscoe) has been used as a raw material in many traditional preparations since the ancient time. As a component of traditional health products, Ginger is known to be effective as appetite enhancer, anticold and anti-inflammation. This study was performed to investigate the immunomodulative effects of Ginger in mouse, using in vitro and ex vivo experiments. In vitro experiment, the mice splenocytes proliferation and three kinds of cytokines (IL-1 $\beta$, IL-6, and TNF-$\alpha$) prodution by peritoneal macrophages cultured with ethanol and water extracts of Ginger were used to indicate the immunomodulative effect. In order to elucidate the immunomodulative effects of Ginger ex vivo, water extract of Ginger was orally administrated into mice, and isolated splencytes and macrophages were used as experimental model. Ex vivo experiment, six to seven week old mice were fed ad libitum on a chow diet, and water extract of finger was orally administrated every other day for four weeks at two different concentractions (50 and 500 mg/kg B.W./day). In vitro study, the splenocytes proliferation was increased when water extract was supplemented in the range of 50-500 $\mu$l/ml concentration. In case of cytokines production, IL-1 $\beta$, IL-6 and TNF-$\alpha$ released by activated peritoneal macrophages were augmented by the supplementation of water extract of the Ginger. Ex vivo experiment, the highest proliferation of splenocytes and production of cytokines by activated peritoneal macrophages were seen in the mice orally administrated at the concentration of 500 mg/kg B.W./day. In conclusion, this study suggests that Ginger extracts may enhance the immune function by regulating the splenocytes proliferation and enhancing the cytokine prodution capacity by activated macrophages in mice.
