Effect of Basal Medium and Plant Growth Regulator on in vitro Plant Regeneration from Axillary Buds of Walnut New Cultiver "Sinlyeong"

  • The walnut (Juglans regia L.), a member of the Juglandaceae, is native to the mountain ranges of central Asia. This species of walnut is valued commercially for its nuts and in some areas for its timber. The seeds of walnut are recalcitrant and it has strong integument dormancy and their germination is irregular, making its natural propagation difficult. Low percentage of seed germination and long propagation cycle are the main problems of propagation. This study was conducted medium composition on in vitro plantlet regeneration from axillary buds of walnut. It has proved to be the most generally applicable and reliable method of in vitro propagation. Micropropagation culture that axillary buds are excised aseptically enables faster multiplication of plants. The axillary buds of walnut new cultivar "Sinlyeong" were cultured on two basal media which contained the different plant growth regulators depending on the respective shooting and rooting stage. After 12 weeks, the shoot generation rate was 85.3%, the shoot number and its length were 1.9/explant and 2.7 cm in the most favorable medium composition. The percentage of rooting was 25.4%. From these results, it was found the optimum basal medium and plant growth regulator for in vitro plant regeneration from axillary buds of walnut new cultivar "Sinlyeong". However, we have continued to search the other medium additives to enhance the rate of walnut root.

Propagation by In Vitro Zygotic Embryos Cultures of the Quercus myrsinifolia

  • Zygotic embryo culture was performed to propagate evergreen oak, Quercus myrsinifolia, which has recalcitrant seeds and is difficult to propagate by cuttings. Zygotic embryos appeared in WPM medium after 14 days, and after 56 days, they developed into complete plants with cotyledons and roots. The medium suitable for zygotic embryo culture was 1/4 WPM medium, showing a shoot growth of 2.43 cm and root growth of 8.7 cm after 8 weeks of culture. As a result of investigating the effect of GA3 on the growth of plants germinated from zygotic embryos through GA3 treatment, the best growth was shown in 0.5 mg/l GA3 treatment. The in vitro rooting and growth of IBA-treated zygotic embryo-derived plants were good in the 0.5 mg/l IBA treatment and rooting and shoot growth were not observed at higher concentrations. And the callus induction rate also increased as the concentration of IBA increased. Plants grown in vitro were transferred to a plastic pot containing artificial soil and acclimatized in a greenhouse for about 4 weeks, resulting in more than 90% survival. As a result of this study, the zygotic embryo culture method was confirmed to be effective for mass propagation of Q. myrsinifolia. The results of this study are expected to contribute significantly to the mass propagation of elite Q. myrsinifolia.

In vitro seed germination of Cymbidium aloifolium (L.) Sw., a potential medicinal Orchid from Eastern Ghats of Tamil Nadu, India

  • Cymbidium aloifolium (L). Sw. is an exquisite epiphytic orchid of the Kolli Hills (Eastern Ghats) of Tamil Nadu in Southern India. It is fast disappearing from its natural habitats due to deforestation and low germination rate in natural habitat. In the present study, an attempt was made to germinate the seeds from un-dehisced capsule of Cymbidium aloifolium (L). Sw under in vitro condition. The seed germination and protocorm development were recorded in three different well known media namely Knudson C (KC), Half strength Murashige & Skoog (1/2 MS) and Vacin & Went (VW) media. The highest seed germination of 90% was observed KC basal media after $30^{th}$ days whereas germination percentages were 40% and 30% on 1/2 MS and VW media respectively. The well-developed protocorm were transferred to KC media supplemented with 6-Benzyl Amino Purine (BAP) and Naphthalene acetic acid (NAA) where BAP (1.0 mg/l) and NAA (1.0 mg/l) together were found to be optimum for the highest shoot formation. About 90% of the shoots found to be well rooted after transfer to the KC medium differently supplemented with 1.5 mg/l Indole-3-acetic acid (IAA) and 1.0 mg/l Indole-3-butyric acid (IBA). Though rooting also took place in the two basic media but the duration was longer when compared with the hormone-supplemented media. The rooted plantlets were hardened and kept under greenhouse conditions which can be relocated in natural habitats.

옻나무 종자의 기내 발아에 의한 무균묘 육성 (Development of Aspceptic Seedling by In Vitro Germination in Lacquer Tree Seed)

  • 옻나무 묘목대량증식법의 하나로 조직배양 방법을 이용하여 기내 유식물체를 다량으로 증식함으로써 옻나무 증식의 효율성을 높이기 위하여 기내 무균묘를 얻고자 실시하였다. 비중 1.0으로 선종한 결과 1차선종은 평균 50.7%의 선종율을 보였고, 98% sulfuric acid에 2시간 교반 후 2차로 선종한 결과 선종율은 20.8%였다. 발근율은 외종피를 함유한 종자(5.4%) 보다는 외종피를 제거하거나(18.3%) 내종피를 제거한 종자(32.4%)가 높았다. 전처리에 따른 종자의 발아율은 생장조절물질 처리에 의한 효과가 인정되지 않았으며, 각 처리별 유의수준도 인정되지 않았으나 sulfuric acid를 처리했을 경우 발아율이 3%로써 생장조절물질 처리보다는 약간 효과적인 것으로 나타났다. 외종피를 제거하면 발아율은 약간 높게 나타났는데, 생장조절물질을 첨가함으로써 약 4%정도 향상되었다. 특히 BA 1.0mg/L와 NAA 0.05mg/L의 혼용처리와 BA 1.0mg/L 단독처리가 10% 내외의 발아율을 보였다. 옻나무 종자는 배지에 치상한 후 약 10일 만에 정상적으로 발아하였고, 치상 후 약 3주만에 본엽이 출현한 정상적인 무균묘를 획득할 수 있었으나 발아율이 10%내외에 불과하여 매우 저조하였다.

야생 참당귀(Angelica gigas N.)의 기내발아 및 기관유도에 미치는 PGRs의 영향 (Effects of Plant Growth Regulators on In Vitro Germination and Organ Formation of Wild Angelica gigas N.)

  • 본 연구는 야생 참당귀의 무균묘 확보, 대량생산체계 확립 및 품종개발을 위해 기내발아와 기관유도에 미치는 PGRs의 영향을 알아보고자 실시하였다. 종자발아 실험은 전처리로 $GA_3$ 침지시간(0, 24h, 48h, 72h) 및 종자의 처리 방법(대조구, 기계적가상 처리, 표면 메탄올 가열 소독법)을 달리하여 실시하였다. 그 결과 $GA_3$에 침지시키지 않고 메탄올 가열법으로 처리한 종자를 0.5 mg/L $GA_3$가 첨가된 배지에서 가장 높은 발아율(15%)을 보였으나 저조하여 발아율 향상을 위해 초음파(frequency 80 KHz) 및 PGRs($GA_3$, BA, TDZ)를 처리하였다. 그 결과 초음파 처리 후 메탄올 가열법으로 처리 한 종자를 0.1 mg/L $GA_3$가 첨가된 배지에 치상하여 발아율(60%)을 증진 시킬 수 있었다. 기내 발아된 식물체로부터 기관유도에 미치는 PGRs(2,4-D, NAA, TDZ, BA)의 영향을 살펴본 결과 캘러스 유도는 줄기, 뿌리, hypocotyl을 1.0 mg/L NAA + 0.5 mg/L BA 처리구에 치상하였을 때 높은 유도율을 보였다. 신초와 뿌리유도는 각각 hypocotyl(PGRs 무첨가)과 뿌리(0.1 mg/L NAA 첨가구)에서 효율적으로 분화되었다. 또한 줄기(1.0 mg/L 2,4-D 처리구)와 hypocotyl(0.1 mg/L NAA 처리구) 조직에서 체세포배가 발생되어 MS 기본배지에 계대배양하여 정상적인 식물체로의 육성이 가능하였다. 본 연구를 통해 무균묘 확보 및 대량생산 체계를 확립하였으며 이를 통해 신품종 개발에 유용할 것으로 판단된다.

황칠나무의 경정배양에 의한 기내번식 (In vitro Propagation using Shoot Tip Culture in Gold Tree[Dendropanax morbifera $L_{EV}]$.)

  • 황칠나무의 선발된 우량개체군의 집단재배시 우량해체간의 교잡에 의해서 형성된 종자를 증식하기 위한 기내배양체계를 확립하기 위해서 기내 발아, 증식, 발근에 적합한 식물생장조절제를 구명하기 위하여 본 실험을 실시한 결과는 다음과 같다. 황칠나무 종자의 무균발아에 적합한 배지로는 MS(Murasinge Skook)기본배지가 발아율이 높고 유식물체(plantlet)의 생장이 양호하여 알맞는 배지로 판단 된다. 기내에서 신초의 증식과 생육에 효과적인 식물생장조절제는 사이토키닌(BAP 0.1~l.0 mg/l)과 오옥신(NAA 0.5~1.0 mg/l)의 혼용처리가 효과적이었다. 또한 기내의 발근은 오옥신의 첨가에서 쉽게 유도되었으며, 그 중에서 NAA1.0 mg/l첨가에서 가장 양호하였다.

Development of Zygotic Embryos and Seedlings is Affected by Radiation Spectral Compositions from Light Emitting Diode (LED) System in Chestnut (Castanea crenata S. et Z.)

  • Among the environmental conditions employed in micropropagation, light quality plays an important role in growth, specially morphogenesis and photosynthesis. The effect of radiation quality (350-740 nm) on the development and growth of zygotic embryos and in vitro plantlets of open-pollinated chestnut (Castanea crenata S. et Z.) were studied. Two types of explants were exposed for 4 weeks to cool white (W, as control), monochromatic red (R, peak emission 650 nm), monochromatic blue (B, peak emission 440 nm), red+blue (R+B, 1:1), or red+far-red (R+Fr, 1:1, far-red peak emission 720 nm) radiation from a light-emitting-diode (LED) system. While the zygotic embryos showed positive photoblastic behavior, their germination was inhibited by blue radiation. Hypocotyl elongation and root development were promoted by red radiation. The emergence of primary leaf and its expansion were faster under blue than under red radiation. In the plantlets, red and red+far-red radiation significantly increased the formation and growth of the root, whereas blue light reduced rooting. Therefore, radiation quality appears to influence some steps in the development of zygotic embryos and plantlets in the chestnut.

Mass Propagation of Sundew, Drosera rotundifolia L. through Shoot Culture

  • In order to establish in vitro propagation method of sundew, Drosera rotundifolia L., the effects of MS medium concentration, cytokinin type and concentration, pH, and auxin type and concentration on shoot proliferation and root formation were investigated using shoots at 3 month after seed germination. The highest shoot production was obtained with the half strength of MS ($\frac{1}{2}$ MS) medium than with any other strength of MS medium tested. Addition of kinetin or BA in $\frac{1}{2}$ MS medium was strongly suppressed shoot proliferation. The suppression of shoot proliferation was more effective in BA-supplemented $\frac{1}{2}$ MS medium than kinetin-supplemented. The optimum pH of the media for shoot proliferation was pH 5.7-6.7. Shoots were subcultured in $\frac{1}{2}$ MS medium supplemented with 0.5mg/L 2,4-D for rooting every 8 weeks. All subcultured shoots produced extensive root systems after 5 to 6 week culture. Plantlets after root development were planted in plastic pots filled with moss. The survival rate of plantlets was almost 100%. On subculturing every 8 weeks, hundreds of the plants were propagated from a single plant within a year.
