Geosmin and 2-methylisoborneol (2-MIB), responsible for earthy or musty smell, are a major concern for safe drinking water supplies. This study investigated the effects of environmental factors on odorous compound production and cell growth in cyanobacterial strains. Anabaena sp. FACHB-1384, a 2-MIB producer, was sensitive to low temperature (<$20^{\circ}C$). However, geosmin producers, Anabaena sp. Chusori and Anabaena sp. NIER, were sensitive to high light intensity (>$100{\mu}mol/m^2/sec$), but not to low temperature. Geosmin concentrations increased under higher nitrate concentrations, being linearly proportional to cell density. A P-limited chemostat showed that P-stress decreased the geosmin productivity and extracellular geosmin amount per cell in Anabaena sp. NIER. However, only 2-MIB productivity was reduced in Planktothrix sp. FACHB-1374 under P-limitation. The extracellular 2-MIB amount per cell remained constant at all dilution rates. In conclusion, high light intensity and P-stress can contribute to the lower incidence of geosmin, whereas 2-MIB reduction could be attainable at a lower temperature.
Geosmin is volatile metabolites produced by a range of filamentous cyanobacteria which causes taste and odor problems in drinking water. Molecular ecological methods which target biosynthetic genes (geoA) are widely adopted to detect geosmin-producing cyanobacteria. The aim of this study was to investigate the potential production capability of 8 strains isolated from the Nakdong River. Ultimately, a suggestion for a genetical monitoring tool for the identification of geosmin producers in domestic waters was to be made. Geosmin was detected using solid phase microextraction gas chromatography mass spectrometry (SPME GC-MS) in two strains of Dolichospermum plactonicum (DGUC006, DGUC012) that were cultured for 28 day. The highest concentrations during the experiment period was $17,535ngL^{-1}$ and $14,311ngL^{-1}$ respectively. Additionally, geoA genes were amplified using two primers (geo78F/971R and geo78F/982R) from strains shown to produce geosmin, while amplification products were not detected in any of non-producing strains. PCR product (766 bp) was slightly shorter than the expected size for geosmin producers. According to the BLAST analysis, amplified genes were at nucleotide level with Anabaena ucrainica (HQ404996, HQ404997), Dolichospermum planctonicum (KM13400) and Dolichospermum ucrainicum (MF996872) between 99 ~ 100 %. Both strains were thus confirmed as potential geosmin-producing species. We concluded that the molecular method of analysis was a useful tool for monitoring potential cyanobacterial producers of geosmin.
활성탄 재질별 geosmin과 2-MIB의 최대 흡착량은 석탄계 재질의 활성탄이 가장 우수한 것으로 나타났고, 다음으로 야자계, 목탄계 활성탄 순으로 나타났으며, geosmin과 2-MIB에 대한 석탄계 활성탄의 최대 흡착량(X/M)은 신탄의 경우 야자계와 목탄계 활성탄에 비해 각각 $1.2\sim1.9$배 및 $2.1\sim2.6$배 정도 높은 것으로 조사되었다. 또한, 3.1년 사용탄의 경우는 석탄계와 목탄계 재질의 활성탄에서 높게 나타났으며, 5.9년 사용탄의 경우는 목탄계 재질의 활성탄이 석탄계 재질의 활성탄보다도 높게 나타났다. 활성탄에서의 흡착용량을 나타내는 k값의 경우 활성탄 재질별, 사용연수별 geosmin과 2-MIB에 대해 전체적으로 geosmin이 크게 나타나고 있어 활성탄 흡착공정에서 2-MIB 보다 제거가 용이한 것으로 조사되었다. 활성탄 사용율(CUR)은 석탄계 재질의 활성탄이 geosmin과 2-MIB에 대해 1.72 g/day 및 1.44 g/day, 야자계나 목탄계 활성탄의 경우는 각각 1.72와 2.05 g/day 및 2.12와 1.90 g/day의 활성탄을 사용하여야만 제어가 가능한 것으로 조사되었으며, 또한, 3.1년과 5.9년 사용탄의 경우는 목탄계 재질의 활성탄이 geosmin과 2-MIB에 대해 각각 3.13과 4.57 g/day 및 2.87과 4.14 g/day로 나타나 다른 재질의 활성탄들에 비해 적은 양으로도 geosmin과 2-MIB를 제어할 수 있는 것으로 나타났다. 석탄계와 야자계 재질의 신탄, 3.1년 및 5.9년 사용탄들에 대해 geosmin과 2-MIB의 최대 흡착량(ng/g)과 비표면적$(m^2/g)$ 및 총 세공용적$(cm^3/g)$에 대한 상관성 조사결과, 최대 흡착량은 비표면적 보다는 총 세공용적이 높은 상관성을 가지는 것으로 나타났으며, 최대 흡착량과 총 세공용적의 상관식은 geosmin의 경우는 $y=264,459\times-79,047(R^2=0.95)$, 2-MIB는 $y=319,650\times-101,762(R^2=0.93)$으로 나타났다.
본 연구에서는 생물활성탄 공정에서 geosmin과 MIB의 제거시 생물분해와 흡착기작의 역할을 BAC와 biofilter를 이용하여 평가하였다. 정상상태에 도달한 BAC 공정에서 부착 박테리아들의 활성이 저하된 저수온기($9^{\circ}C$)에도 EBCT 30분의 조건에서는 geosmin과 MIB가 완전 제거되었다. 유입수의 수온이 $26^{\circ}C$일 때가 $9^{\circ}C$일 때보다 상층부분에서 부착 박테리아의 생체량과 활성도가 높았고, 여층의 하부로 갈수록 부착 박테리아의 생체량과 활성도의 감소율이 높게 나타났다. 이는 수온이 높을 때 상층부분에서 BOM 제거율이 높아 하층에 공급할 수 있는 BOM 양이 감소하기 때문이다. 부착 박테리아의 활성이 억제된 BAC 공정의 geosmin과 MIB의 제거율은 활성화된 BAC 공정에서의 제거율과 비교하여 큰 차이를 나타내지 않아 BAC 공정에서 geosmin과 MIB의 제거에 흡착기작의 기여도가 높게 나타났다. BAC 공정에서 geosmin과 MIB의 제거시 부착 박테리아에 의한 생물분해 기작과 활성탄 표면이 가지는 흡착 기작이 경쟁적으로 작용하였다.
This study was carried out to compare ozonation with photocatalysis degradation for removal Geosmin of algae byproduct. The change of pH was decresed from 7.02 to 2.8 after contact time 480 minute for ozonation. In case of UV-germicidal lamp, pH was very quickly increased from 7.02 to 7.5, but Halogen lamp did very slowly change pH. Geosmin degradation ratio was as following, UV-germicidal lamp/TiO2(100mg/L) O3>UV-germicidal lamp/TiO2(50mg/L)>UV-germicidal lamp(10W)>halogen lamp(50W). Instead of TiO2 suspension solution, Geosmin degradation ratio was very low using hollow bead and pellet as coated TiO2. As a result of identifing byproducts, ozonation generated three species of aldehyde such as 3-Heptanone and three species of alcohol such as Heptanal, but photocatalysis formed 1, 14-Tetradecanediol infinitesimally.
수중 이 취미를 유발시키는 대표적인 미량오염물질인 2-methylisoborneal (MIB)과 geosmin(지오스민)의 배제율을 소수성 polyethersulfone (PES) 나노분리막(분획분자량 : 400 Da)을 적용하여 다양한 용액조성에서 관찰하였다. 실험결과 적용된 모든 조건에서 지오스민이 2-MIB보다 다소 높은 배제율을 나타내었다. 용액의 pH 효과를 관찰한 결과 pH가 증가할수록 2-MIB와 지오스민 양쪽 모두 배제율이 증가하는 경향을 나타내었다. 한편, 수중 자연유기물질의 존재는 두 미량유기물질의 배제율을 크게 증가시켰으며 이와 같은 현상은 높은 pH일수록 더욱 뚜렷하게 나타났다. 소수성 분리막을 친수성 $TiO_2$ 입자로 표면코팅 시킨 후 배제율을 관찰한 결과 분리막의 표면을 친수화한 후 소수성인 2-MIB와 지오스민의 배제율은 증가하는 경향을 나타내었다. 따라서 소수성 상호작용은 미량유기물질 나노여과 배제율의 중요한 기작 중 하나임을 확인할 수 있었다.
SPME (Soild phase microextraction) has been used in the analysis of many volatile organic compounds, such as geosmin and 2-methylisoborneol (2-MIB), trihalomethanes (THMs) in drinking water. SPME fiber is characterized by high adsorption capacity (DVB/CAR/PDMS, DVB/PDMS etc.). Although the highly active adsorption capacities of the SPME fiber are often to the chemical functional group, surface properties play a significant role in determining the surface adsorption capacities. The objectives of this study were to evaluate effect of residual chlorine on analysis of geosmin and 2-MIB. Image taken by SEM before preloaded with chlorine, the surface and porous media was almost perfect spherical shape and no clogging of pores. However, after preloaded with chlorine the surface was aggregated and pore was blocked. The recovery rate of geosmin and 2-MIB coexisting with chlorine was reduced by 35 to 62%. The recovery rate with preloaded with chlorine was reduced by 25 to 43%. The lower concentration of geosmin and 2-MIB and the higher concentration of chlorine existed in water, the lower the recovery rate was.
Geosmin and 2-methylisoborneol (2-MIB) are two of the most common taint compounds that adversely affect the quality of aquacultural animals. In the present study, 94% of geosmin and 97% of 2-MIB in suspended growth reactors producing bioflocs (SGRs) with aquaculture waste were removed after inoculation with Bacillus subtilis, significantly higher than that of control SGRs (70% of geosmin and 86.4% of 2-MIB). The lowest concentrations of geosmin and 2-MIB achieved in the effluent of the SGRs were 2.43 ± 0.42 ng/l and 2.23 ± 0.15 ng/l, respectively. The crude protein content of the bioflocs produced in the SGRs was 35 ± 4%. The NH4+-N and NO2--N concentrations in the effluent of the reactors were 1.13 ± 0.21 mg/l and 0.42 ± 0.04 mg/l, respectively. These results suggest that inoculated with Bacillus subtilis, SGRs have a better performance to reuse the nitrogen in fish waste and to remove geosmin and 2-MIB from the culture water efficiently.
3가지 재질의 생물활성탄 부착 박테리아들을 부리 동정한 결과 총 9종류의 부착 박테리아가 동정되었다. Pseudomonas 속이 차지하는 비율이 평균 56.5%로 나타나 가장 높은 우점비율을 나타내었고, 다음으로 Pasteurella속 18.9%, Chryseomonas 속 4.0%, Agrobacterium속 3.5%, Aeromonas속 2.0% 순으로 검출되었다. 순수 분리된 9종의 박테리아들의 성장곡선을 조사한 결과 24~96시간 내에 최대의 생체량을 나타내어 geosmin을 유기탄소원으로 활용하는 능력이 뛰어난 것으로 조사되었다. $4^{\circ}C$와 $25^{\circ}C$의 운전조건에서 geosmin에 대한 생분해능을 조사한 결과 Pseudomonas vesicularis, Pseudomonas fluorescens, Agrobacterium radiobacter 및 Stenotrophomonas maltophilia 등이 뛰어난 생분해율을 나타낸 반면 Chryseomonas luteola, Spingomonas paucimobilis, Spirillum spp. 등은 비교적 낮은 geosmin 생분해능을 나타내었다. Geosmin의 생분해능은 수온이 $4^{\circ}C$일 경우 생분해율 속도상수가 $0.00006{\sim}0.00020\;hr^{-1}$의 범위에서 $25^{\circ}C$에서는 $0.0043{\sim}0.0046\;hr^{-1}$의 범위로 나타나 수온 상승에 따라 큰 폭으로 증가하였으며, 또한 투입된 geosmin의 농도가 10~10,000 ng/L로 증가할수록 생분해율 속도상수도 $0.0003{\sim}0.0882\;hr{-1}$로 증가하였다.
본 연구는 최근 북한강 수계에서 번성하고 있는 Anabaena strain의 16S rDNA 염기서열을 이용한 종 수준의 동정과 geosmin 합성 유전자의 탐침을 통해 이취미 물질의 잠재 생산능력을 분석하였다. 현장(경기도 양평군 조암면 삼봉리 수역)에서 분리 배양한 Anabaena는 직선형과 나선형 두 가지의 형태적 변이를 보였다. 이들은 세포의 크기와 사상체에서 형태적 차이를 나타냈으며, A. circinalis 및 A. crassa와 유사한 형태적 특징들을 보여주었다. 그러나 16S rDNA 계통수 및 유연관계를 분석한 결과, 직선형과 나선형 모두 동일한 A. circinalis 종으로 확인되었다(98%~100%의 유전적 유사도). 또한 직선형과 나선형 strain 모두에서 geosmin을 합성하는 유전자 구간이 발견되어, 북한강 수계에 존재하는 Anabaena circinalis는 종의 형태적 변이에 관계없이 geosmin을 생산할 수 있음을 보여주었다. 본 연구의 결과는 유전자 수준에서 A. circinalis의 geosmin 생산에 대한 직접적인 증거를 제공하는 국내 최초의 보고로서 북한강 수계에서 geosmin 증가의 원인종 확인 및 관리에 중요한 자료를 제공한다.
