염료 추출조건에 따른 소목의 색소성분 분리 거동 (Separation of Chromophoric Substance from Sappanwood under Different Extraction Conditions)

  • 본 연구는 출토직물의 염료 성분 판정을 위한 장기적인 프로젝트의 일환으로서 천연염료 식물 중 우리나라는 물론 동서양 각지에서 과거에 염색식물로 널리 사용되었던 소목에 대하여 주 색소성분인 브라질린을 가스크로마토그라피 질량분석법(GC-MS)으로 검출할 수 있는 최적의 염료 추출 및 분석 조건을 확립하는데 그 목적을 두었다. 소목추출액에 대한 GC-MS 분석 결과 증류수보다는 메탄올이, 메탄올보다는 아세톤 추출이 브라질린 추출효과가 높았다. 추출기기 간에는 $30^{\circ}C$로 진행된 waterbath shaker보다 상온에서 진행된 shaking plate가 브라질린 추출이 높았으며 두 기계 모두 1시간보다는 12시간 추출이 월등히 높은 추출량을 보였다. 증류수를 사용해 pH $3{\sim}9$로 조절하여 브라질린의 추출효과를 조사한 결과 pH 5 조건에서 브라질린 검출량이 가장 많았다. GC-MS 분석 조건은 컬럼내 온도를 초기온도 $50^{\circ}C로 하고 온도 증가율을 $23^{\circ}C/min$로 하여 $210^{\circ}C$까지 끌어올리고 다시 온도증가율을 $30^{\circ}C/min$로 하여 최종적으로 $305^{\circ}C로 가열한 후 14분 동안 holding하여 전체 분리시간을 24.12분으로 한 방법이 가장 효과적이었으며 MSD 스캔범위는 $75{\sim}400m/z$이 적합하였다.

  • Triazole 계의 대표적인 농약 중 하나인 amitrole (3-amino-1,2,4-triazole)은 널리 사용되고 있는 제초제로, 환경 중에 유입될 경우 지하수 및 지표수를 오염시킴으로써 궁극적으로 음용수의 오염까지 일으킬 수 있다. 이 물질은 물에 대한 용해도가 높은 반면, 유기 용매에 대한 용해도가 낮고 극성이 강하기 때문에 수질 시료로부터 추출하기 매우 어렵다. 본 연구에서는 수질 시료중의 amitrole을 GC/MS를 이용하여 ppb 수준 이하의 극미량까지 분석하기 위한 방법을 조사하였다. 10 mL의 수질 시료를 진공회전증발기로 완전히 증발시킨 후, isobutyl chloroformate (iso-BCF)를 사용하여 실온에서 15~20분간 반응시켜 유도체화 하였다. 그 결과, N-isobutoxycarbonyl amitrole derivative를 생성함으로써 GC/MS에서의 감도가 향상됨을 확인할 수 있었다. $1.0{\mu}g/L$, $10.0{\mu}g/L$$100.0{\mu}g/L$ 농도의 수질에서 본 분석방법의 회수율은 각각 98.0%, 94.9% 및 105.3%였고, RSD는 2.4%, 2.8% 및 1.5%였다. 10 mL의 수질 시료로부터 얻어진 검량선의 상관계수는 $0.1{\sim}100.0{\mu}g/L$의 농도 범위에서 0.997로 나타나 직선성이 우수하였으며, 검출한계는 $0.1{\mu}g/L$로 나타났다.

Identification of Metabolites of Phytosterols in Rat Feces Using GC/MS

  • $\beta$-Sitosterol, campesterol and stigmasterol have been known to the phytosterols the most frequently found in plants. Metabolism of phytosterols was investigated using rat feces and liver microsomes. Feces were collected after phytosterols (a well characterized mixture of $\beta$-sitosterol 40%, campesterol 30% and dihydrobrasicasterol) were administered orally (0.5 ${g/kg$) to rats. Metabolites of phytosterols were identified using GC/MS. Three peaks were eluted at 12.47, 12.65, 12.87 min and had characteristic molecular ions m/z 428, 430, 432, respectively. Three fecal metabolites were identified as androstadienedione, androstenedione, and androstanedione. No metabolites could be detected in the rat liver microsomal reaction mixture. The results suggest that the metabolites of phytosterols in rat feces are formed by oxidation at 3- position, saturation at 5- and 6- position, and 17- side chain cleavage in the rat large intestine.

인삼 Sesquiterpene의 새로운 분리방법 (New Methods for Isolation of Sesquiterpene from Panax ginseng)

  • New simple methods for the Isolation of sesquiterpenes from Panax ginseng were developed. First, volatile compounds were isolated by simultaneous distillation and extraction (SDE) with 30% methanol and $\alpha$-hexane instead of water and ethyl ether/pentane (1:1). Secondly, head space volatiles in U-shaped tube at 7$0^{\circ}C$ were passed through C18 Sep-Pak by nitrogen gas streaming and the adsorbed volatiles were fluted by $\alpha$-hexane. TLC analysis showed that the volatile concentrates consisted mainly of terpenes when colored by vanillin-sulfuric and. GC/MS data revealed that approximately 30 sesquiterpenes of molecular weight 204 occupied 81.1% or more of the volatile concentrates isolated by those two newly developed methods. Among these, alloaromadendrene, germacrene B, isocaryophyllene, $\alpha$-neoclovene, ${\gamma}$-muurolene, $\beta$-panaslnsene, and $\alpha$-humulene were identified as being major sesqulterpenes by authentic samples or literatme search Key words : Panax ginseng, volatile compound, sesquiterpene, isolation, new method, GC/MS.

Identification of Coffee Fragrances Using Needle Trap Device-Gas Chromatograph/Mass Spectrometry (NTD-GC/MS)

  • A fast and simple sampling and sample preparation device, (NTD) has been developed and applied to sample and analyze volatile components from ground coffee beans. Coffee fragrances and other volatile organic compounds (VOCs) were sampled by the NTD and then analyzed by gas chromatograph-mass spectrometry (GC/MS). Divinylbenzene (DVB) particles (80/100 mesh size) were the sorbent bed of the NTD. More than 150 volatile components were first identified based on the database of the mass library and then finally 30 fragrances including caffeine were further confirmed by comparing experimental retention indices (i.e. Kovat index) with literature retention indices. Total sampling time was 10 minutes and no extra solvent extraction and/or reconstitution step need. Straight n-alkanes (C6-C20) were used as retention index probes for the calculation of experimental retention indices. In addition, this report suggests that an empty needle can be an alternative platform for analyzing polymers by pyrolysis-GC/MS.

  • Headspace hanging drop liquid phase micro-extraction (HS-HD-LPME) is studied as a novel solvent-based sample pretreatment method for floral volatile aroma compounds. This paper reports on application of the HSHD- LPME combined with GC-MS for the analysis of linalool component emitted from evening primrose flowers. The effect of several variables on the method performance was investigated. Additionally, the separation of enantiomers on a cyclodextrin capillary column was performed to identify chirality of (−)-linalool component. Since the unsurpassed volume of a few micro-liters of solvent is used, there is minimal waste or exposure to toxic organic solvents. This method enables to combine extraction, enrichment, clean-up, and sample introduction into a single step prior to the chromatographic process.

차가버섯 용매별 분획물의 항산화 활성과 Tyrosinase 저해 활성 (Antioxidative Activity and Tyrosinase Inhibition Effect of Fractions from the Extract of Inonotus obliquus)

  • This study was conducted to investigate the antioxidative activities and tyrosinase inhibition effects of fractions from the distilled water extract of Inonotus obliquus. Moreover, GC-MS based analysis with trimethylsilyl (TMS) derivatization was carried out for active compound in the extract of I. obliquus. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate fraction was 0.393 mg/ml as a result of the most effective than other fractions. Meanwhile, aqueous fraction showed higher effect in tyrosinase inhibitoty activity. In GC-MS based analysis with TMS derivatization, 7 compounds including syringic acid, vanillic acid and protocatechuic acid were observed in ethyl acetate fraction, and oxalic acid is the main compound in aqueous fraction. As a result, it was confirmed that oxalic acid in aqueous fraction from the distilled water extract of I. obliquus was a compound showing tyrosinase inhibition effect.

전자코 시스템을 이용한 우유의 품질에 따른 휘발성 향기성분 분석 (Analysis of Volatile Flavor Compounds in Milk Using Electronic Nose System)

  • Volatile flavor compounds from milk were analyzed and identified by using the analysis methods of headspace solid phase microextraction gas chromatography/mass spectrometry (HSPME-GC/MS) and electronic nose (E-Nose) system. About 30 volatile compounds were identified by HSPME-GC/MS for the fresh and off-flavor milk samples. Also, the correlation between rancidity and ageing days of milk was obtained by the aid of principal component analysis algorithm. It shows that the E-Nose system can identify the various types of milk flavor. These results imply that the analysis method based on the E-nose system can apply to the quality control of milk flavor and the rancidity.

택란(澤蘭)의 정유성분(精油成分)에 관한 연구(硏究) (Studies on Essential Oils of Lycopus lucidus Turcz)

  • The essential oils of Lycopus lucidus have been studied. The essential oil obtained from the dried herba by steam distillation followed by fractionation by column chromatography was analysed by gas chromatography-mass spectrometry(GC-MS). The compounds identified by GC-MS were carvacrol, 2,5-dimethoxy-p-cymene, trans-caryophyllene, spathurenol and $trans-{\beta}-farnesene$. Two compounds, thymol and caryophyllene were isolated by silica gel column chromatography and analyzed by TLC, IR, Mass and NMR. And the weak diuretic effects of essential oil and water extract from the dried drugs were observed in rats.

Catalytic Pyrolysis of Waste Polyethylene Terephthalate over Waste Concrete

  • The feasibility of waste concrete as a catalyst for the effective pyrolysis of polyethylene terephthalate (PET) was examined using thermogravimetric (TG) and pyrolyzer-gas chromatography/mass spectrometry (Py-GC/MS) analyses. TG analysis results indicated that the maximum decomposition temperature of PET is not altered by the use of waste concrete, showing similar values (407 ℃ and 408 ℃ at 5 ℃/min). Meanwhile, the volatile product distribution data obtained from the Py-GC/MS analysis revealed that the use of waste concrete promoted the deoxygenation reaction via converting the oxygen containing products such as benzoic acids, benzoates, and terephthalates to valuable deoxygenated aromatic hydrocarbons including benzene, toluene, ethylbenzene, and styrene. This suggests that the waste concrete can be used as a potential catalyst for the production of valuable aromatic hydrocarbons from PET pyrolysis.
