Characteristics of Volatile Flavor Compounds of Fuji Apples by Different Extraction Methods (추출방법에 따른 후지사과의 휘발성 향기성분 특성)

  • The characteristics of volatile flavor compounds of Fuji apples were compared by different extraction methods for information leading to a natural fragrance development. The volatile flavor components of Fuji apples were extracted using simultaneous steam distillation and extraction (SDE), solvent extraction (SE), and solid-phase microextraction (SPME) methods and then analyzed by gas chromatograph-mass spectrometer. A total of 66, 32 and 54 components were identified in SDE, SE and SPME extracts, respectively. (E,E)-$\alpha$-Farnesene, hexanol, butanol, 2-methyl butanol, hexyl hexanoate, hexyl 2-methyl butanoate, hexyl butanoate, and butyl hexanoate were the major flavor components in the extracts by different methods, but the composition of volatiles in the extracts were different. Alcohols and hydrocarbons were the major functional groups in SDE and SE extract whereas esters and hydrocarbons were the major functional groups in SPME extracts. SPME was the most suitable method for analysis of fresh volatiles from Fuji apples.

  • Vinyl chloride (VC) and vinylidene chloride (VDC), which may be present in poly(vinyl chloride) (PVC) and poly(vinylidene chloride) (PVDC) food packaging, were analyzed by using a headspace-gas chromatograph (GC)/mass spectrometer (MS) equipped with a PLOT Q column. 1-Chloropropane (1-CP) was used as an internal standard. The determinations of VC and VDC were carried out by monitoring the characteristic ions for each compound: m/z=61, 62, 64 and m/z=61, 96, 98, respectively. The method validated in this study can be suitable for the compliance test of EU regulation. Among 103 food packaging samples, VC and VDC were not detected, except in one PVDC food package. However, the detected level of VDC in the sample was below the EU regulatory specific limitation.

  • Analytical methods for the determination of the derivatives of the herbicide (${\pm}$)-2-(4-chloro-2-methylphenoxy)propionic acid (MCPP) by capillary column gas chromatography with mass spectrometer (GC-MS) and electron-capture detection (GC-ECD) were studied. A successful procedure was introduced for the ester preparation using $H_2SO_4$, as the catalyst and the alcohol 2,2,2-trichloroethanol (TCE) or 2,2,2-trifluoroethanol (TFE). The identificaiton and elucidation of MCPP by GC-MS spectrometry following the esterification with diazomethane, $BF_3$/methanol, $H_2SO_4$/methanol, TCE, TFE, or pentafluorobenzyl bromide (PFB) were carried out. A comparison of the response-sensitivities among those MCPP esters was made with GC-ECD. Although the methylation product of MCPP was confirmed by GC-MS, its low sensitivity to the ECD limited the detection of MCPP. TCE, TFE, and PFB derivatization methods resulted in a high rate of MCPP esterifications and very sensitive ECD molecular responses. Based on efficiency, convenience, worker safety, and least sample contamination, TFE esterificaiton was considered as the superior method for MCPP analysis to the other methods of derivatization. An accurate method is described for quantifying MCPP in soil leachates by GC-ECD at very low concentrations without the requirement of a complicated clean-up process. As a result, MCPP residues at concentrations of less than $0.1{\mu}g$ in 100ml soil leachate were detected.

Effects of Heat Treatment and Antioxidant Activity of Aroma on Garlic Harvested in Different Cultivation Areas (산지별 마늘의 향기 항산화활성과 열처리 효과)

  • The objectives of this study were to compare the aroma characteristics and antioxidant activity of raw and heated garlic ($130^{\circ}C$, 2 hr) from different cultivation areas (Danyang, Seosan, Uiseong Namhae, Namdo, Daeseo and China). The volatile compounds were extracted by simultaneous steam distillation extraction and identified with gas chromatography/mass spectrometer. The major volatile compounds of raw garlic were sulfur compounds such as diallyl disulfide, methyl-2-propenyl trisulfide, di-2-propenyl trisulfide etc. After heating, the major volatile compounds were allyl mercaptan, methyl pyrazine, 2,2-dimethyl-1,3-dithiane, 2-propenyl propyl disulfide, allyl methyl sulfide, allyl alcohol, and allyl sulfide etc. The DPPH radical scavenging activity (EDA, %) of volatile compounds from raw garlic and heated garlic was increased in a dose-dependent manner. The antioxidant activities (EDA, %) of volatile compounds from raw garlic cultivated in Danyang, Namhae and China were 20.07, 34.62, and 9.71% respectively. After heating, these values were increased to 79.90, 93.59, and 77.26% respectively. Results showed that heat treatment significantly enhanced the antioxidant activities (EDA, %) of the garlic.

  • In this study, we mainly focused on the PM (Particulate Matter) emission characteristics of a diesel engine. To analyze particle behavior in the tail-pipe, particle emission was measured on the engine-out (downstream of turbocharger), each upstream and downstream both of DOC (Diesel Oxidation Catalyst) and DPF (Diesel Particulate Filter). Moreover, particle emission contours on each sampling point were constructed. The reduction efficiency of particle number concentration and mass through the DOC and DPF was studied. Parameters such as EGR (Exhaust Gas Recirculation) and the main injection timing were varied in part load conditions and evaluated using the engine-out emissions. The DMS500 (Differential Mobility Spectrometer) was used as a particle measurement instrument that can measure particle concentrations from 5 nm to 1000 nm. Nano-particles of sizes less than 30 nm were reduced by oxidation or coagulated with solid particles in the tail-pipe and DOC. The DPF has a very high filtration efficiency over all operating conditions except during natural regeneration of DPF.

  • Essential oils from Fennel fruit(Foeniculum vulgare Mill), Olibanum resin(Boswellia carteii Birew) and Needl Juniperus stem(Juniperus rigida Sieb.) were extracted by a supercritical fluid extraction system(SFE) and biological activity of each essential oils were observed. SFE technique was applied for the isolation and purification of nonpolar biologically active essential oils from each samples. The quantitative analysis of essential oils was carried out by gas chromatography-mass spectrometer(GC/MS). About 60% of the growth of AGS and A549 cells were inhibited by adding 1.0g/l of the crude essential oils and below 40% was observed by the control. Cytotoxicity on human normal lung cell(HEL299) was scored as $15{\sim}18%$ for the crude essential oils and 12% for control, respectively. It meant that the essential oils were more effective than the control in anti-mutagenecity tested by CHO V79 cells. The effect of the essential oils on the growth of nerve cells, PC12 was observed as follows: The viable cell density was about two times higher than control.

Simultaneous Determination of Octylphenol, Nonylphenol and Bis(2-ethylhexyl)phthalate in Fish Samples Using Two Internal Standards (2가지 내부표준물질을 이용하여 어류시료 중 Octylphenol, Nonylphenol, Bis(2-ethylhexyl)phehalate의 동시정량)

  • A comprehensive analytical method of endocrine distruptors[i.e., nonylphenol(NP), octylphenol(OP), bis(2-ethylhexyl)phthalate(BEHP)] is fish samples was developed using two internal standards. This method employed closed culture tube extraction with dichloromethane and solvent exchange to iso-hexane and SPE(2g) aminopropyl column, followed by determination on gas chromatograph linked to mass spectrometer(GC/MS) operated in the single ion monitoring(SIM) mode. The recoveries of nonylphenol and octyphenol in the range of $0.2{\sim}20{\mu}g/g$ using 1-phenyl decanol as one internal standard were over 75%, and recovery of bis(2-ethylhexyl)phthalate in the range of $0.4{\sim}40{\mu}g/g$ using bis(2-ethylbutyl)phthalate(BEBP) as the other internal standards was showed over 102%. The present method was applied to fish samples from Korea and UK. The range of concentrations for nonylphnol(NP) and bis(2-ethylhexyl)phthalate in Korean fish were $0.02{\sim}0.06{\mu}g/g$ in 2 samples and $0.18{\sim}2.03{\mu}g/g$ in 9 samples respectively, but bis(2-ethylhexyl)phthalate(BEHP) in UK samples was found $2.99{\mu}g/g$ in just 1 sample. But octylphenol(OP) was not dected in any samples by this method. This two internal standard method provides a more precise analytical tool to investigate endocrine disruptors in a biological matrices of limited quantity.

Performance Evaluation of Hazardous Substances using Measurement Vehicle of Field Mode through Emergency Response of Chemical Incidents

  • BACKGROUND: Chemical accidents have increased owing to chemical usage, human error and technical failures during the last decades. Many countries have organized supervisory authorities in charge of enforcing related rules and regulations to prevent chemical accidents. A very important part in chemical accidents has been coping with comprehensive first aid tool. Therefore, the present research has provided information with the initial applications concern to the rapid analysis of hazardous material using instruments in vehicle of field mode after chemical accidents. METHODS AND RESULTS: Mobile measurement vehicle was manufactured to obtain information regarding field assessments of chemical accidents. This vehicle was equipped with four instruments including gas chromatography with mass spectrometry (GC/MS), Fourier Transform Infrared Spectroscopy (FT-IR), Ion Chromatography (IC), and UV/Vis spectrometer (UV) to analyses of accident preparedness substances, volatile compounds, and organic gases. Moreover, this work was the first examined the evaluation of applicability for analysis instruments using 20 chemicals in various accident preparedness substances (GC/MS; 6 chemicals, FT-IR; 2 chemicals, IC; 11 chemicals, and UV; 1 chemical) and their calibration curves were obtained with high linearity ( r 2 > 0.991). Our results were observed the advantage of the high chromatographic peak capacity, fast analysis, and good sensitivity as well as resolution. CONCLUSION: When chemical accidents are occurred, the posted measurement vehicle may be utilized as tool an effective for qualitative and quantitative information in the scene of an accident owing to the rapid analysis of hazardous material.

  • The objective of this study was to compare the aroma characteristics and antioxidant activity of Chrysanthemum indicum Linne (CIL), C. morifolium Ramat (CMR) and C. zawadskii var Latilobum (CZL). The volatile compounds were extracted by simultaneous steam distillation extraction and identified with gas chromatography/mass spectrometer. The major volatile compounds of Chrysanthemum sp. were camphene, 1,8-cineole, benzene, pinocarvone, bicyclo-2,2,1-heptan-2-ol, ${\beta}$-caryophyllene, 3-cyclohexen-1-ol, ${\gamma}$-curcumene, zingiberene and ${\beta}$-bisabolene. The DPPH radical scavenging activity (EDA, %) of volatile compounds in CIL, CMR and CZL were 30.57, 46.36, and 51.72%/g sample, respectively. The ascorbic acid equivalent antioxidant capacity (AEAC) of volatile compounds were 34.99, 35.31, and 38.48 mg AEAC/g, respectively.

40Ar-39Ar Age Determination for the Quaternary Basaltic Rocks in Jeongok Area (전곡 지역 제4기 현무암질 암석의 40Ar-39Ar 연대 측정)

  • Quaternary basaltic rocks occur as volcanic plateau and/or river cliff in the watershed area of Hantan River in Jeongok, central Korea. We measured $^{40}Ar-^{39}Ar$ ages for the basaltic rocks from Jeongok area using the multi-collector noble gas mass spectrometer and laser heating device introduced for the first time in Korea. The basaltic rocks from the river cliff in Eundae-ri area show the systematic change in $^{40}Ar-^{39}Ar$ ages from $0.54{\pm}0.07Ma$ through $0.48{\pm}0.01Ma$ to $0.12{\pm}0.01Ma$ toward the top. The other sample from Jeongok-ri area yields the age of $0.43{\pm}0.04Ma$. This results suggest that there might be a episodic volcanic eruption between 0.12-0.54 Ma in Jeongok area.
