Effects of Oral Administration of Phellinus linteus on the Productions of the Th1- and Th2-type Cytokines in Mice

  • Background: The mushroom Phellinus linteus (PL) has been shown to have the anti-tumor and immunostimulatory effects. We hypothesized that the hot water extract of PL (WEPL) exerts its significant immunostimulatory effect by inducing production of the Th1-derived cytokine interferon-${\gamma}$ (IFN-${\gamma}$) by T lymphocytes. Methods: T lymphocytes were isolated from the mice fed with 200 mg/kg of WEPL once a day for 4 weeks, and then stimulated with the mitogen concanavaline A (Con A). IFN-${\gamma}$ gene and intracellular protein expressions were analyzed by RT-PCR and flow cytometry, respectively. The production of IFN-${\gamma}$ was measured by enzyme-linked immunosorbent assay. Results: WEPL significantly enhanced the transcription of IFN-${\gamma}$ mRNA. The effect of WEPL on IFN-${\gamma}$ expression was further supported by a concomitant increase in the number of cells with intracellular IFN-${\gamma}$ protein as well as the secretion of IFN-${\gamma}$. However, WEPL did not modulate either gene expression or protein secretion of interleukin-4, a Th2-associated cytokine, by Con A-stimulated T lymphocytes. Conclusion: Our results demonstrate that one of the potentially beneficial anti-tumor and immunostimulatory effects of WEPL may be mediated through the enhancement of IFN-${\gamma}$ secretion by T lymphocytes.

재조합 대장균과 효모의 고정화 혼합세포계에 의한 ${\gamma}$-Glutamylcysteine 생산 (Production of ${\gamma}$-Glutamylcysteine by Immobilized Mixed Microbial System of Recombinant E. coli and Yeast)

  • ${\gamma}$-Glutamylcysteine 생산에 있어서 재조합 대장 균 HB101/pGH501만을 이용한 단일세포반응계가 재조합 대장균과 효모를 이용한 흔합서l포반응계보다 반응시간이 짧고 생산농도가 높은 것으로 나타났다. 그러나 생산경제성 측면에서 ATP 재생공정을 위하 여 훈합세포반응계를 사용하였다. 재조합 대장균과 효모를 이용한 혼합세포반응계에서 대장균과 효모의 비율은 1:4가 적합함을 보였고, ATP 재생공정에 사용되는 glucose는 O.5M의 농도에서 가장 효율적 으로 나타났다. 재조합 대장균과 효모를 alginate를 이용하여 고정화하여 반응계로 사용하였을 경우 반 응에 필요한 시간이 걸어지고 생산놓도도 감소되냐 반응계의 안정성은 10% 정도 증가됨을 알 수 있었다. 실험결과 alginate로 고정화된 흔합세포반응계 를 사용하여 ${\gamma}$-glutamylcysteine를 연속 생산할 수 있음을 확인하였다.

  • 본 연구는 펄스 반응기 및 고정층 연속반응기를 사용하여 ${\gamma}$-알루미나에 담지된 니켈촉매상에서 이산화탄소에 의한 프로판의 개질반응 특성을 고찰한 것이다 Ni/${\gamma}$-$A1_2O_3$촉매가 NiO/${\gamma}$-$A1_2O_3$촉매보다 $CO_2$의 해리능력과 프로판의 개질 능력이 우수한 것으로 나타났다. 프로판과 $CO_2$의 혼합물에 산소를 추가한 결과 프로판의 전환율이 증가하였고 표면의 탄소침적도 감소함을 알 수 있었다. Ni/${\gamma}$-$A1_2O_3$$Ga_2O_3$의 기계적 혼합 촉매는 Ni/${\gamma}$-$A1_2O_3$자체보다 촉매의 활성이 오래 유지되었고 두 상의 협동에 의한 상승효과(synergistic effect)가 관찰되었다.

중성자와 감마선 중복 조사가 자주달재비 분홍돌연변이율에 미치는 영향 (Effects of Combined Irradiation of Neutrons and ${\gamma}$-Rays on the Pink Mutation Frequencies in Tradescantia)

  • 자주달개비 식물체에 감마선과 중성자를 중복 조사하였을 패 나타나는 수술털(TSH) 분흥돌연변이율의 변화를 분석하였다. 감마선은 $0{\sim}2$ Gy의 범위에서 선량별 조사를 하였으며 중성자는 $^{252}Cf$ 선원으로부터 일정한 거리에 식물체를 배열하여 조사하였다. 감마선을 선량별로 조사한 후 중성자를 조사한 때의 TSH 분홍돌연변이 감마선량-반응의 증가계수는 6.17로 감마선량-반응식의 5.98에 비해 12%의 방사선효율 증가를 초래했으며 중성자를 먼저 조사한 다음 감마선을 조사한 경우는 증가계수가 7.48로서 25%의 방사선 생물효율 증대가 나타났다. 두 종류의 방사선 중복 조사시 조사방식의 선 후 조합에 따른 방사선생물효율 증대가능성은 멸균 또는 세포사멸 등 방사선을 긍정적으로 이용하는 분야에 있어 매우 중요한 의미가 있다.

The Combined Effect of Gamma Knife Irradiation and p53 Gene Transfection in Human Malignant Glioma Cell Lines

  • Objective: The purpose of this study is to elucidate in vitro responses to combined gamma knife irradiation and p53 gene transfection on human malignant glioma cell lines. Methods: Two malignant human glioma cell lines, U87MG (p53-wild type) and U373MG (p53-mutant) were transfected with an adenoviral vector containing p53 (MOI of 50) before and after applying 20Gy of gamma irradiation. Various assessments were performed, including, cell viability by MTT assay; apoptosis by annexin assay; and cell cycle by flow cytometry, for the seven groups: mock, p53 only, gamma knife (GK) only, GK after LacZ, LacZ after GK, GK after p53, p53 after GK. Results: Cell survival decreased especially, in the subgroup transfected with p53 after gamma irradiation. Apoptosis tended to increase in p53 transfected U373 MG after gamma irradiation (apoptotic rate, 38.9%). The G2-M phase cell cycle arrest markedly increased by transfecting with p53, 48 hours after gamma knife irradiation in U373 MG (G2-M phase, 90.8%). Conclusion: These results suggest that the in vitro effects of combined gamma knife irradiation and p53 gene transfection is an augmentation of apoptosis and G2-M phase cell cycle arrest, which are more exaggerated in U373 MG with p53 transfection after gamma knife irradiation.

  • Let ${\Delta}$ be a simplicial complex, $I_{\Delta}$ its Stanley-Reisner ideal and $K[{\Delta}]$ its Stanley-Reisner ring over a field K. Assume that ${\Gamma}(R)$ denotes the zero-divisor graph of a commutative ring R. Here, first we present a condition on two reduced Noetherian rings R and R', equivalent to ${\Gamma}(R){\cong}{\Gamma}(R{^{\prime}})$. In particular, we show that ${\Gamma}(K[{\Delta}]){\cong}{\Gamma}(K^{\prime}[{\Delta}^{\prime}])$ if and only if ${\mid}Ass(I_{\Delta}){\mid}={\mid}Ass(I_{{{\Delta}^{\prime}}}){\mid}$ and either ${\mid}K{\mid}$, ${\mid}K^{\prime}{\mid}{\leq}{\aleph}_0$ or ${\mid}K{\mid}={\mid}K^{\prime}{\mid}$. This shows that ${\Gamma}(K[{\Delta}])$ contains little information about $K[{\Delta}]$. Then, we define the squarefree zero-divisor graph of $K[{\Delta}]$, denoted by ${\Gamma}_{sf}(K[{\Delta}])$, and prove that ${\Gamma}_{sf}(K[{\Delta}){\cong}{\Gamma}_{sf}(K[{\Delta}^{\prime}])$ if and only if $K[{\Delta}]{\cong}K[{\Delta}^{\prime}]$. Moreover, we show how to find dim $K[{\Delta}]$ and ${\mid}Ass(K[{\Delta}]){\mid}$ from ${\Gamma}_{sf}(K[{\Delta}])$.

혈부축어탕이 대동맥 평골근 세포에서 NO 생성에 미치는 영향 (Effects of Hyeolbuchukeo-tang (Xiefuzhuyutang) on NO Production in Aortic Vascular Smooth Muscle Cells)

  • Objective : This study was designed to investigate the effect of Hyeolbuchukeo-tang (HCT) on NO production and the molecular mechanism of NO production modulated by HCT in the primary VSMC (vascular smooth muscle cells). Method : Primary VSMC was established from aorta and cultured VSMC used in this study. NO production of VSMC was assayed by Griess reagent and the expression of iNOS gene was assayed by Western, RT-PCR. Result : $TNF-{\gamma}$ induced NO production, but $IFN-{\gamma}$ or HCT alone did not induce NO production in cultured VSMC. However, $IFN-{\gamma}$ or HCT potentiated NO production in $TNF-{\gamma}-treated$ VSMC in a time- and dose-dependent manner. $TNF-{\gamma}$ induced the iNOS gene expression corresponding to NO production in $TNF-{\gamma}-treated$ VSMC. HCT potentiated NO production in $TNF-{\gamma}-treated$ VSMC by about 20%, but HCT did not increase the level of iNOS mRNA in $TNF-{\gamma}-treated$ VSMC. HCT slightly increased the level of iNOS protein in $TNF-{\gamma}-treated$ VSMC. Calcium ionophore A23187 decreased NO production in $TNF-{\gamma}-treated$ VSMC, but HCT attenuated the effect of A23187. Conclusion : As NO is deeply involved in the development of arteriosclerosis and dilation of blood vessels, drugs or chemicals modulating NO production in VSMC could be used for preventing and treating arteriosclerosis. Considering the effect of HCT on the modulation of NO production in VSMC, MCT has a potential capacity for preventing and treating diseases of the circulation system including arteriosclerosis.

Cells Transformed by PLC-Gamma 1 Overexpression are Highly Sensitive to Clostridium difficile Toxin A-Induced Apoptosis and Mitotic Inhibition

  • Phospholipase C-${\gamma}l$ (PLC-${\gamma}l$) expression is associated with cellular transformation. Notably, PLC-${\gamma}$ is up-regulated in colorectal cancer tissue and breast carcinoma. Because exotoxins released by Clostridium botulinum have been shown to induce apoptosis and promote growth arrest in various cancer cell lines, we examined here the potential of Clostridium difficile toxin A to selectively induce apoptosis in cells transformed by PLC-${\gamma}l$ overexpression. We found that PLC-${\gamma}l$-transformed cells, but not vector-transformed (control) cells, were highly sensitive to C. difficile toxin A-induced apoptosis and mitotic inhibition. Moreover, expression of the proapoptotic Bcl2 family member, Bim, and activation of caspase-3 were significantly up-regulated by toxin A in PLC-${\gamma}l$-transformed cells. Toxin A-induced cell rounding and paxillin dephosphorylation were also significantly higher in PLC-${\gamma}l$-transformed cells than in control cells. These findings suggest that C. difficile toxin A may have potential as an anticancer agent against colorectal cancers and breast carcinomas in which PLC-${\gamma}l$ is highly up-regulated.

High-Molecular-Weight Poly-Gamma-Glutamate Protects Against Hypertriglyceridemic Effects of a High-Fructose Diet in Rat

  • We studied the effects of 2 different dosages of high-molecular-weight poly-${\gamma}$-glutamic acid (hm ${\gamma}$-PGA) derived from Bacillus subtilis chungkookjang on lipid metabolism in a high-fructose diet-induced hypertriglyceridemic animal model. For 4 weeks, rats were fed either AIN-93 diet (normal control, NC; n = 10) or modified AIN-93 diet in which cornstarch was substituted with 63% fructose (n = 30) to induce hypertriglyceridemia. After 4 weeks, the hypertriglyceridemic rats were treated with daily oral doses of 0 mg (hypertriglyceridemic control, HC), 2.5 mg (hypertriglyceridemic, low hm ${\gamma}$-PGA, HL), or 5 $mg{\cdot}kg{\cdot}bw^{-1}{\cdot}d^{-1}$ (hypertriglyceridemic, high hm ${\gamma}$-PGA, HH) hm ${\gamma}$-PGA for 4 weeks. The HL and HH groups exhibited significantly lower levels of serum triglyceride, total cholesterol, LDL cholesterol, and free fatty acids than the HC group. The administration of hm ${\gamma}$-PGA reduced serum ALT and AST levels. The activities of lipogenic enzymes such as hepatic malic enzyme and glucose-6-phosphate dehydrogenase as well as glucose-6-phosphate dehydrogenase mRNA expression were significantly decreased by hm ${\gamma}$-PGA administration (p < 0.05). These results indicate that hm ${\gamma}$-PGA has an anti-hypertriglyceridemic effect in high-fructose diet-induced hypertriglyceridemic rats.

Effects of Gamma Irradiation on Nutrient Composition, Anti-nutritional Factors, In vitro Digestibility and Ruminal Degradation of Whole Cotton Seed

  • Whole cotton seed (WCS) has become one of the major feed ingredients in TMR for dairy cattle in Korea, and WCS for feed use is mostly imported from abroad. Since this genetically modified oil seed is usually fed to the animal in raw state, its germination ability, if last long, often causes concerns about ecological disturbances. In the process of looking for effective conditions to remove germination ability of WCS this study had the objectives to evaluate the nutritional effects of gamma irradiation at doses of 8, 10 and 12 kGy on changes in nutrient contents, anti-nutritional factors, in vitro digestibility and ruminal degradability. No significant differences were found in proximate analysis of nutrients between raw WCS and gamma irradiated one. Glycine and threonine contents significantly increased when the WCS was exposed to gamma ray as compared to untreated WCS (p<0.05). As for fatty acid composition, no significant differences were observed with the irradiation treatment. Free gossypol in WCS was decreased (p<0.05) by gamma irradiation treatment. Of the 3 different levels of gamma irradiation, a dose of 12 kGy was found to be the most effective in reducing free gossypol concentration. Results obtained from in situ experiment indicated that gamma irradiation at a dose of 10 kGy significantly (p<0.05) lowered rumen degradability of both dry matter and crude protein as compared with raw WCS. However, there were no significant differences in rapidly degradable and potentially degradable fractions of crude protein due to 10 kGy gamma irradiation. Overall, this study show that gamma irradiation at a dose of 10 kGy is the optimum condition for removing germination ability of WCS, and could improve nutritive value for the ruminant with respect to the decrease in both ruminal protein degradability and gossypol content of WCS.
