Production of Algal Biomass and High-Value Compounds Mediated by Interaction of Microalgal Oocystis sp. KNUA044 and Bacterium Sphingomonas KNU100

  • There is growing interest in the production of microalgae-based, high-value by-products as an emerging green biotechnology. However, a cultivation platform for Oocystis sp. has yet to be established. We therefore examined the effects of bacterial culture additions on the growth and production of valuable compounds of the microalgal strain Oocystis sp. KNUA044, isolated from a locally adapted region in Korea. The strain grew only in the presence of a clear supernatant of Sphingomonas sp. KNU100 culture solution and generated 28.57 mg/l/d of biomass productivity. Protein content (43.9 wt%) was approximately two-fold higher than carbohydrate content (29.4 wt%) and lipid content (13.9 wt%). Oocystis sp. KNUA044 produced the monosaccharide fucose (33 ㎍/mg and 0.94 mg/l/d), reported here for the first time. Fatty acid profiling showed high accumulation (over 60%) of polyunsaturated fatty acids (PUFAs) compared to saturated (29.4%) and monounsaturated fatty acids (9.9%) under the same culture conditions. Of these PUFAs, the algal strain produced the highest concentration of linolenic acid (C18:3 ω3; 40.2%) in the omega-3 family and generated eicosapentaenoic acid (C20:5 ω3; 6.0%), also known as EPA. Based on these results, we suggest that the application of Sphingomonas sp. KNU100 for strain-dependent cultivation of Oocystis sp. KNUA044 holds future promise as a bioprocess capable of increasing algal biomass and high-value bioactive by-products, including fucose and PUFAs such as linolenic acid and EPA.

샤페론단백질동시발현기술을이용하여 Helicobacter pylori 유래의 fucosyltransferase의수용성생산 (Soluble Expression of the Fucosyltransferase Gene from Helicobacter pylori in Escherichia coli by Co-expression of Molecular Chaperones)

  • Fucosyltransferase는 퓨코실화된 올리고당을 생성하는데 필수적인 효소로서, GDP-β-L-fucose로 부터 fucose를 수용체로 전이시켜 알파 글리코사이드 결합을 형성하는 과정을 촉매한다. 하지만 Escherichia coli 에서 발현시켰을 때, 대부분의 경우 inclusion body를 형성하여 비활성으로 생성되었다. 따라서 본 연구에서는 Helicobacter pylori 26695에서 유래한 α1,2-fucosyltransferase (FucT2) 유전자의 수용성 발현을 위하여, E. coli에서 샤페론 단백질인 GroEL, GroES, DnaK, DnaJ, GrpE과 함께 동시발현시켰다. SDS-PAGE 분석결과, 5가지 샤페론 단백질과 함께 발현되었으며, 수용성 FucT2 단백질이 증가하였고 효소활성은 5배 증가하였다. 결론적으로, 샤페론 동시발현기술을 이용하여 대장균에서 FucT2 수용성 생산을 증가시킬 수 있었으며 본 수용성 효소는 퓨코실화된 올리고당을 효율적으로 생산하는데 이용될 수 있다.

영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 항암활성 다당류의 구조분석 (Structural Analysis of the Antitumor Active Exo-polysaccharide Produced by Submerged Cultivation of Ganoderma lucidum Mycelium)

  • 영지 균사체의 액체 배양에 의하여 생산된 세포의 다당시료 분획 중 우수한 항암활성을 나타내었던 산성분획인 BWS-DA-GI의 구조를 균사체 유래의 산성 다당 분획(MWS-DA-GI)과 비교하면서 조사하였다. BWS-DA-GI의 구성성분을 검토한 결과, 다량의 glucose, mannose 및 galactose와 미량의 xylose 및 fucose를 함유하였으며, glucose, galactose 및 mannose의 몰비는 2.5:2.1:2.5이었다. 반면, 균사체 유래의 산성 다당 분획인 MWS-DA-GI는 다량의 galactose, fucose, mannose 및 glucose와 미량의 xylose를 함유하였다. 또 BWS-DA-GI의 구조를 균사체 유래의 다당 분획인 MWS-DA-GI와 비교하면서 과요오드 산화, Smith분해, affinity chromatography 및 메칠화 분석으로 조사한 결과, 두 시료 모두 ${\beta}-1,3-glucan$이었고, BWS-DA-GI는 1,6분지를 보유하고 있는 반면, MWS-DA-GI는 1,4 및 1,6분지를 함께 보유하는 것으로 추정되었으며, 분자량은 각각 1,200,000 및 1,000,000 dalton이었다.

Variation in Fucoidan Contents and Monosaccharide Compositions of Korean Undaria pinnatifida (Harvey) Suringar (Phaeophyta)

  • Three different forms of Undaria pinnatifida, the southern form (U. pinnatifida f. typica), the northern form (U. pinnatifida f. distans), and Samcheok form (recently cultivated strain), were examined for the contents and compositions of fucoidans. Fucoidans were extracted from the dried edible portions of three forms of U. pinnatifida in low pH condition, mainly by ethanol precipitation and CaCl2 treatment. It was shown that Samcheok form contains 1.8 and 3.5 times more fucoidans than the northern and the southern forms, respectively. The monosaccharide compositions of individual fucoidans were also varied. The fucoidans from the southern and the northern forms were shown to be composed of mainly fucose and galactose with the molar percentage ratios of 83.5%:16.5% and 87.4%:12.6%, respectively, indicating that these are F-type fucoidans. The fucoidan from Samcheok form, however, consisted of fucose (62.7%), galactose (32.9%), and small amount of glucose (4.4%). The results of this study showed that both amount and monosaccharide compositions of fucoidans are variable depending on U. pinnatifida forms.

미역(Undaria pinnatifida) 부위별로 추출한 다당의 이화학적 특성 (Chemical Composition and Rheological Properties of Polysaccharides Isolated from Different Parts of Brown Seaweed Undaria pinnatifida)

  • The chemical and rheological properties of fucoidan and alginate prepared from different parts of Undaria pinnatifida (sporophyll, frond, stipe) were investigated. The algal materials were extracted with HCl (pH 2.0, 3 h at 70℃) to prepare fucoidan, and the remaining solid was continuously re-extracted with Na2CO3 (pH 10.0, 70℃, 3 h) to prepare alginic acid. The fucoidan and alginic acid contents in the sporophyll, frond, and stipe were 11.14%, 3.84%, and 1.73% and 22.04%, 37.14%, and 31.74%, respectively. The content of fucoidan and alginate depends on the part extracted. The fucoidan extracted from the sporophyll mainly consists of fucose and galactose, but the fucoidan extracted from frond and stipe contains mannose in addition to fucose and galactose. Fourier-transform infrared spectroscopy analysis of fucoidan and alginate suggests the presence of sulfate groups (1261 and 840 cm-1) and carboxyl groups (1626 and 1419 cm-1), respectively. Alginate solutions (5%) had a low viscosity of 10.84-31.63 mPa·s. The activation energies of fucoidan and sodium alginate were 14.45-18.38 kJ/mol and 18.61-22.06 kJ/mol, respectively. The D-mannuronic acid/L-guluronic acid (M/G) ratios of alginate showed a relatively high (frond, 3.72; stipe, 2.88; and sporophyll, 1.80).

상황버섯(Phellinus linteus) 균사체로부터 항보체 활성 다당류의 정제 및 특성 (Purification and Characterization of Anti-complementary Polysaccharide from Phellinus linteus Mycelia)

  • 식용버섯으로부터 항보체 활성물질을 탐색하여 새로운 소재화 연구의 일환으로 상황버섯(Phellinus linteus) 균사체로부터 항보체 활성 물질을 분리, 정제를 하였으며 화학적 특성을 검토하였다. 정제는 상황버섯의 열수추출한 후, 열수추출물(PL-0)를 한외여과하여 분자량별로 5개의 획분을 얻었다. 이 중, 300kDa 이상의 획분(PL-5)을 DEAE-Sepharose FF 이온교환 크로마토그래피, Sepharose CL-4B 겔여과 크로마토그래피를 실시하여 PL-5-IIIa 획분을 얻었다. PL-5-IIIa 획분을 HPLC를 행한 결과 단일 피크로 나타났으며 정제다당 PL-5-IIIa의 분자량은 800,000Da으로 측정되었다. 정제다당 PL-5IIIa는 64.2%의 당과 29.6%의 단백질로 구성되었으며 구성당은 galactose가 43.1%, mannose가 40.6%, fucose가 15.8%로 나타났다.

Structural and Physiological Characteristics of Rhamnogalacturonan II from Fruit Wines

  • To characterize the polysaccharides which exist as soluble forms in fruit wines, crude polysaccharides were isolated from red, white, raspberry, wild grape, and pear wine, respectively. Among them, the crude polysaccharide (RW-0) in red wine showed the highest yield and considerable amounts of thiobarbituric acid (TBA)-positive materials. The pectic polysaccharide RW-2 was purified to homogeneity from RW-0 by subsequent size-exclusion chromatography using Sephadex G-75 and its structure was characterized. RW-2 consisted of 14 different monosaccharides which included rarely observed sugars in general polysaccharides, such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose (Api), 3-C-carboxy-5-deoxy-L-xylose (aceric acid, AceA), 3-deoxy-D-manno-2-octulosonic acid (Kdo), and 3-deoxy-D-lyxo-2-heptulosaric acid (Dha). Methylation analysis indicated that RW-2 comprised at least 20 different glycosyl linkages such as 3,4-linked fucose, 2,3,4-linked rhamnose, 3'-linked apiose, and 2,3,3'-linked apiose, being characteristic in rhamnogalacturonan II (RG-II). High performance size-exclusion chromatography indicated that RW-2 mainly comprised RG-II of higher molecular weight (12,000), and that the changes of molecular weight to apparent 7,000 under less than pH 2.0 were observed. These analyses indicated that the higher molecular weight polysaccharide in RW-2 was mainly present as a RG-II dimer.

고구마樹脂중 잘라핀의 糖構成에 관한 硏究 (Sugar Constituents of Jalapin from Sweet Potato Tubers)

  • 고구마중 잘라핀의 糖構成을 究明하기 위하여 고구마塊根에서 정제한 잘라핀을 脫아실化한 후 가수분해한 결과 L-rhamnose, D-fucose, D-glucose가 1:1:1의 分子比 및 上記順序의 酸安定度를 가짐을 알았다. 脫아실化한 잘라핀 1몰은 2몰을 過沃素酸에 의하여 산화되었으나 D-glucose는 산화되지 아니하였다. 따라서 고구마중 脫아실化한 잘라핀의 가능한 化學構造를 다음과 같이 제안하였다. L-$Rha_f$-(1${\to}$4)-D-$Fuc_p$-(1${\to}$3)-D-$Glu_p$-(1${\to}$11)-jalapinolic acid.

Molecular Simulations and Conformational Studies of Fucoseα1-3)Gal(β1-X)GlcNAc where X=3, 4, or 6 Oligosaccharides

  • Energy minimization and conformational studies of molecular ions generated by ESI (electrospray ionization) tandem mass spectrometry (MS/MS) can be used for the discrimination of stereoisomeric permethylated and sodium cationized trisaccharides. Sets of fucose-containing trisaccharides having different internal and terminal linkages have been synthesized to analyze the reducing terminal linkage positions using BT and IT fusion approaches. A detailed investigation has been undertaken on the conformational behaviors of four trisaccharide fragments from human milk and blood group determinants of Type 1 and Type 2, namely Fuc($\alpha$1- 3)Gal($\beta$1-3)GalNAc and Fuc($\alpha$1-3)Gal($\beta$1-X)GlcNAc where X = 3, 4 and 6 using molecular modeling methods. Three dimensional rigid and adiabatic phi-psi-energy maps (Surfer program) describing the energy as a function of rotation around corresponding glycosidic linkages were calculated by SYBYL molecular modeling and MM4 force field programs conjunction with cleavage energies of ESI MS/MS for the side group orientations. This approach predicted conformational behaviors exhibited by isomer saccharides for future applications on biologically active glycoconjugates and to exploit a faster method of synthesizing a series of structural isomeric oligosaccharides.

삼림토양(森林土壤)의 층위별(層位別) 가수분해물중(加水分解物中) 단당류(單糖類)의 함량(含量) (Contents of Monosaccharides in the Hydrolysates of Some Forest Soil Horizons)

  • 강원도(江原道)에 위치(位置)한 점봉산(點鳳山)과 오태산(五台山)의 삼림토양(森林土壤)에서 침엽수림(針葉樹林)과 활엽수림(闊葉樹林)으로 나누어 층위별(層位別)로 채취(採取)하여 단당류(單糖類)의 함량(含量)을 분석(分析)하여 조사(調査)한 결과(結果)는 다음과 같다. 1. 단당류(單糖類)의 함량(含量)은 유기층(有機層)에 가장 많았고 심층(深層)으로 내려갈수록 감소(減少)되었는데 이는 유기물함량(有機物含量)과 직접적인 관계가 있었다. 2. Hexose (galactose, glucose, mannose)의 함량(含量)이 절대적으로 많았으며 그 다음으로는 Pentose(arabinose, ribose, xylose)의 순서이었고 deoxyhexoe(furose, rhamnese)의 함량(含量)이 가장 적었다. 單糖類의 함량(含量)은 아래와 같다. glucose>mannose>galactose>arabinose>xylose>rahmnose>fucose>ribose. 3. 침엽수림토양(針葉樹林土壤)의 유기층(有機層)에는 극히 적은 량(量)의 ribose만이 함유(含有)되어 있었으며 활엽수림토양(闊葉樹林土壤)에서는 ribose가 검출되지 않았다. 4. 전토양(全土壤) 유기물(有機物)에 대한 단당류(單糖類)의 비율(比率)은 토양(土壤)의 깊이에 따라 감소(減少)했는데 이는 부식화(腐植化)의 효과(效果)라고 생각된다. 표면유기층(表面有機層)의 단당류(單糖類) 함량(含量)은 유기탄소(有機炭素)에 대해 27~50%, 유기물함량(有機物含量)으로 환산(換算)하면 15.7~29%에 해당하며 이 중에서 hexose는 유기탄소(有機炭素)에 대해 20~30%, 유기물(有機物)에 대해서는 12~22 %의 비율(比率)로 함유(含有)되어 있었다. 5. 각각(各各)의 단당류(單糖類)가 차지하는 비율(比率)은 토양단면(土壤斷面)을 통해 종합해 볼 때 어떤 규칙적인 양상을 보이지 않았다. 이는 토양미생물(土壤微生物)의 활동(活動)이 일정치 않았기 때문이 아닌가 사료된다.
