Extraction yield and nutraceuticals of mushroom Pholiota species (비늘버섯류의 추출수율 및 영양 약리학적 특성)

  • This study was performed to obtain basal data for development of new functional food by using mushroom Pholiota spp.. Among several kinds of extracts, water extracts of Pholiota adiposa ASI 24027 showed the highest solid yield of 58.7%. Crude protein of Pholiota adiposa ASI 24015 and ${\beta}$-glucan contents of Pholiota adiposa ASI 24005 were 25.0% and 0.66%, respectively. K and Mg were also contained plentifully in the fruiting body of Pholiota adiposa ASI 24004 (3,649 mg and 138 mg/100g dried fruiting body, respectively). Amino acid and total fatty acid contents were similar between almost all of Pholiota spp..

Artificial Cultivation and Antioxidant Activity of Korean Native Mushroom Pycnoporus coccineus (한국산 간버섯의 인공재배 및 항산화 활성 평가)

  • The mycelial growth of P. coccineus strain was good in PDA and YMA, but mycelial growth was low in MEA. Light irradiation during the incubation period affected the pigment formation and density of mycelia. Mushroom of P. coccineus strain was able to produce fruiting bodies in both bottle and bag cultivation, and oak sawdust was found to be the most suitable substrate for spawn culture and cultivation. In artificial cultivation using sawdust medium, fruiting body was grown to the extent that visual observation was possible from the 15th day, and it formed about 5 days fast in the treatment group with low relative humidity. From 40 to 45 days of mushroom development, mature fruiting bodies could be harvested, and the lower relative humidity of the growing room favored mushroom development and growth. Antioxidant activity of fruiting bodies harvested from artificial cultivation showed that ABTS radical scavenging activity of bottle-cultivated and wild fruit bodies were shown at 505㎍/㎖ and 515㎍/㎖, respectively. However, fruiting bodies harvested in bag cultivation were high at 910㎍/㎖. As a result of DPPH radical scavenging activity, all extracts were found to be inactive, exhibiting IC50 value of more than 2,000㎍/㎖ concentration. The ethyl acetate extract of mushrooms obtained from bottle cultivation showed the highest activity with 1,550㎍/㎖ IC50 value. Methanol extract of wild fruit bodies had the highest ABTS radical scavenging activity at the same concentration (10mg/㎖).

  • Some of men have been suffered from the insufficient secretion of testosterone causing by physical factors, social and psychological factors. Testosterone is an essential steroid hormone controlling male reproductive function. Alternative medicines in plants, fungi, and insects have been studied to enhance sexuality. $Cordyceps$ species including $Cordyceps$ $sinensis$ (CS) and $C.$ $militaris$(CM) has been used as for the enhancement of sexual functionfor hundreds of years in Far East Asian. In the present study, we determined the effect of fruiting bodies of $C.$ $militaris$ which cultured on bee drone medium (CMD) and brown rice medium (CMB) on testosterone concentration in Sprague-Dawley rats. Eighteen rats per group were housed to regular diet or diet supplemented with CMB and CDD, respectively for 4 weeks. Serum was collected from 6 rats per group. Results showed that changes of the body weight, food and water intake of the rats were not observed in this study. However, both CMB and CDD increased the serum testosterone concentration in rats. Furthermore, CMD significantly stimulated testosterone production (p <0.05) compared to the control. Hence, it suggests that $C.$ $militaris$ fruiting bodymight be developed as a complementary medicine to improve sexual hormones.

Characterization of a new commercial strain 'Guseol' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 품종 '구슬'의 육성 및 그 특성)

  • To develop new variety of oyster mushroom, 63 intra-specific hybrids between the strain Suhan and #Nongi201 were developed using hyphal anastomosis technique in 2004. The Po2008-275 hybrid between the dikaryon strain 04-154(Suhan x #Nongi201) and the monokaryon strain derived from ASI2487 were developed using hyphal anastomosis in 2008. The Po2008-275 was shown the best cultural characteristics, selected to be a new variety and named as 'Guseol'. The new commercial strain, 'Guseol' had dark grey pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Guseol' were of an excellent quality in that not only the stipe was thick and long but also the pileus was small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $10{\sim}16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body was about 3 to 5 days depending on the temperatures. The shape of fruiting body was thin funnel shape. Fruiting body production per box($43{\times}43{\times}12cm$) was about $1545{\pm}400.9g$ which was almost 137% quantity compared to that of parental strain 04-154. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Guseol' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the mycelial growth of this mushroom were much faster in mushroom complete medium. Similar results were observed with other variety '#Chunchu2'. Analysis of the genetic characteristics of the new commercial strain 'Guseol' showed a major DNA profile as that of the parental 04-154 when primer URP 1, primer URP 2 and primer URP 5 were used, but different to '#Chunchu2' that was used as a control. This new variety of the dark grey oyster mushroom had smart and high quality image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.

Characterization of a new commercial strain 'Goni' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 백색느타리 품종 '고니'의 육성 및 그 특성)

  • To develop new white variety of Oyster mushroom, all white varieties which have been collected and kept in the lab were revived and screened their cultural characteristics. 84 intra-specific Oyster mushroom hybrids between the white-colored mutants Suhan and Wonhyeong were developed using hyphal anastomosis technique in 2007. The Po2007-63 ($2842-7{\times}0205-7$) was shown the best cultural characteristics, selected to be a new variety and named as 'Goni'. The new commercial strain, 'Goni' has white pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Goni' are of an excellent quality in that not only the stipe is thick and long but also the pileus is small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25-30^{\circ}C$ and $10-16^{\circ}C$, respectively. When two different media including potato dextrose medium and mushroom complete medium were compared, the growth of mushroom were much faster in mushroom complete medium at $20-25^{\circ}C$, but not at $25^{\circ}C$. Similar results were observed with other variety 'Miso'. Analysis of the genetic characteristics of the new commercial strain 'Goni' showed a major DNA profile as that of the parental Suhan when primer URP 1 was used, but different to 'Miso' that was used as a control. When screens were performed with primer URP 2, DNA patterns were similar both to that of the parents and 'Miso'. This new variety of the white Oyster mushroom has a clean and fresh image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.

Effect on Mycelial Growth and Fruit Body Development According to Supplementary Nutrition in Bag Cultivation of Neolentinus lepideus (잣버섯 봉지재배시 영양원이 균사배양 및 자실체 생육에 미치는 영향)

  • This study was carried out to elucidate suitable supplementary nutrition for Neolentinus lepideus cultivation. The main substrate for cultivation was douglas fir sawdust, and supplementary materials for experiment were corn meal, corn hull, corncob, beet pulp, and potato starch. The higher amount of total nitrogen source was showed in corn meal (0.45%) than any other nutritive materials. Incubation ratio and cultivation period showed the best 97% and the shortest 43 days on medium with corn meal substrate. The fruit body yield and biological efficiency on medium with 10% corn meal were highest 122 g/1 kg and 35%, respectively. Factor analyses concerning fruiting yield showed total nitrogen source (r = 0.95) and crude fat (r = 0.65). These results are recommended that N. lepideus cultivation is corn meal material as the source of the supplementary nutrition and total nitrogen source as the factor of fruiting body yield on the Douglas fir sawdust medium.

Fruiting body productivity of Lentinula edodes 'Hwadam' as determined by nutrient supplements and storage period of sawdust spawn (배지영양원 및 톱밥종균 저장기간에 따른 표고 '화담'의 자실체 생산성)

  • This study was conducted to establish an appropriate period of use of sawdust spawn at low temperatures and a nutrient supplement medium for cultivation of Lentinula edodes 'Hwadam'. Of the nutrient supplements, the total yield of rice bran (5%) + corn flour (5%) treatments were 673.3 g, which was higher than rice bran (551.6 g) and wheat bran (546.7 g) treatments, respectively. As shown by the growth of Lentinula edodes 'hwadam' during to the sawdust spawn storage period (at 4℃), the period of spawn running, browning, fruiting body formation, and development was 27 d, 81 d, 5 d, and 11-13 d, respectively, regardless of the length of the storage period at 4 ℃. After 3 months of storage of sawdust spawn, the number of fruiting bodies and yield decreased as the storage period increased. Therefore, the period of use of sawdust spawn (at 4 ℃) for the stable production of fruiting bodies of Lentinula edodes 'Hwadam' was a maximum of 3 months.

Fruitbody Development of Pleurotus ostreatus via Bottle Cultivation Using Recycled Substrate

  • This study was carried out to determine the possibility of bottle cultivation utilizing recycled oyster mushroom culture waste as a cultivating substrate for P. ostreatus. Total nitrogen percentage was 0.76%, 1.13%, 1.16%, 1.36%, and 1.38% in the 1-, 2-, 3-, 4-, and 5-time mixed substrate, respectively; 0.95%, 1.04%, 1.34%, 1.36%, and 1.25% in the 1-, 2-, 3-, 4-, and 5-time postharvest substrate, respectively; and 0.72% and 0.68% in the 2- and 3-time nonadditive substrate, respectively. Weight of the fresh fruiting body harvest was 115 g, 120 g, 117 g, 118 g, and 114 g on 1-, 2-, 3-, 4-, and 5-time mixed substrate, respectively; and 105 g and 45 g on 2- and 3-time nonadditive substrate, respectively. The first mixed substrate (fresh) and recycled substrates generated no significant difference in the weight of fresh fruiting bodies harvested.

Detection of RNA Mycoviruses in Wild Strains of Lentinula edodes in Korea

  • In general, mycoviruses remain latent and rarely cause visible symptoms in fungal hosts; however, some viral infections have demonstrated abnormal mycelial growth and fruiting body development in commercial macrofungi, including Lentinula edodes. Compared to other cultivated mushrooms, L. edodes is more vulnerable to viral infections as it is still widely cultivated under near-natural conditions. In this study, we investigated whether Korean wild strains of L. edodes were infected by RNA mycoviruses that have previously been reported in other parts of the world (LeSV, LePV1, LeV-HKB, LeNSRV1, and LeNSRV2). Using specific primer sets that target the RNA-dependent RNA polymerase genes of each of the RNA mycovirus, reverse transcription-polymerase chain reaction (RT-PCR) was used to detect viral infection. Viral infection was detected in about 90% of the 112 wild strains that were collected in Korea between 1983 and 2020. Moreover, multiple infections with RNA mycoviruses were detected in strains that had normal fruiting bodies. This work contributes to our understanding of the distribution of RNA mycoviruses in Korea and the impact of multiple viral infections in a single strain of L. edodes.

Studies on the development of mushroom media for bottle culture in new Pleurotus ostreatus 'Miso' (신품종 '미소' 느타리버섯 병재배 배지개발)

  • This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.
