Comparative analysis of useful β-glucan and polyphenol in the fruiting bodies of Ganoderma spp. (영지버섯 균주별 자실체의 베타글루칸과 폴리페놀 함량 비교 분석)

  • This study was carried out to compare the medicinal effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. ${\beta}$-glucan and polyphenol are useful ingredient in mushrooms and they were known to have antioxidant activity. We analyzed ${\beta}$-glucan and polyphenol contents of fruiting body of Ganoderma spp., Cordyceps militaris, and Phellinus linteus. Most Ganoderma spp. exhibited ${\beta}$-glucan contents of 15 to 20%. Cordyceps militalis showed the highest ${\beta}$-glucan level of 25%. Interestingly, eight strains of Ganoderma spp. was analyzed to have higher contents of ${\beta}$-glucan than Phelinus linteus. Polyphenol contents was measured after extraction with different solvents. (D.W., 70% EtOH, 80% MeOH) The level of polyphenol in ASI 7020 strain was at maximum in the water extraction and ASI 7086 showed the highest level in the 70% EtOH extraction. The amounts of polyphenol in strain ASI 7113 was at maximum in the 80% MeOH extraction.

Screening of Bioactive Compounds from Mushroom Pholiota sp. (비늘버섯으로부터 생리기능성 물질의 탐색)

  • Extracts from 63 kinds of Pholiota sp. fruiting bodies were prepared using water and methanol, and then their physiological functionalities were investigated. The methanol extracts from Pholiota adiposa PAD030 showed high fibrinolytic activity and those of P. adiposa ASI PAD-022 showed potential inhibitory activity of 76.8% against ${\beta}-hydroxy-{\beta}-methylglutaryl(HMG)-CoA$ reductase. The highest antioxidant and tyrosinase inhibitory activities were found in the water extracts of Pholiota sp. PSP-015 (72.7%) and methanol extracts of P. nameko PNA-024 (69.5%), respectively. However, superoxide dismutase(SOD)-like activity and elastase inhibitory activity were low in almost of the extracts. The HMG-CoA reductase inhibitor from the fruiting body of P. adiposa PAD-022 which showed the highest functionality was extracted maximally when powder of the fruiting body was shaked at $30^{\circ}C$ for 12 h by methanol and its HMG-CoA reductase inhibitory activity was 80.2%.

Cultural Characteristics and Fruiting-body Productivity of New Variety 'Hwadam' (Lentinula edodes) According to Filter Treatment in Boxes (상자에 필터처리에 따른 표고 신품종 '화담'의 재배특성 및 자실체 생산성)

  • This study was carried out to develop a new method of cultivation of 'Hwadam' a new variety of Lentinula edodes, to improve fruiting body productivity and reduce labor. To determine the optimum filter number for box cultivation, L. edodes were assessed for their cultural characteristics and mushroom yield. During spawn running of L. edodes in the box, the temperature gradually increased to the highest value of 25.8℃, 23.7℃, and 23.0℃ on day 18 for the four filter, two filter, and no treatment groups, respectively. Oxygen concentration displayed an opposite trend to carbon dioxide concentration, reaching the lowest value on day 21. The oxygen concentration progressively increased as more filters were used (5.4%, 8.2%, and 8.9% treatment with no, two, and four filters, respectively. Growth of L, edodes in the box resulted in the highest yield (2,228 g/7kg substrate) and biological efficiency (70.7%) using four filters. The biological efficiency of the four filter treatment was 29.4% higher compared to bag cultivation (41.3%). Further studies are necessary to verify stable productivity of fruiting bodies through repeated cultivation.

Characteristics of newly bred spore-less cultivar Lentinula edodes 'Daedam' for sawdust cultivation (표고 톱밥재배용 무포자 신품종 '대담' 육성 및 특성)

  • A new spore-less cultivar Lentinula edodes 'Daedam' was bred from monokaryotic strains of 'LE15401-24' and 'LE192118-10'. The optimum temperature for mycelial growth of 'Daedam' on potato dextrose agar was 22~25℃. Total cultivation period of the new cultivar, from inoculation to its first harvest, was 134 days, similar to that of the control cultivar 'Hwadam'. Total yield of 'Daedam' was 222g per 3kg substrate, and was lower than that of control cultivar(266.0g). The fruiting body of 'Daedam' had a thick and small pileus and a longer stem compare to control cultivar. As a result of a analyzing the productivity of 'Daedam' on the different substrate types, the biological efficiency was 26.7% in the 1.2kg cylindrical substrate(CS), which was higher than that of the 3kg rod-type substrate(RS). 'Daedam' had a similar yield compared to 'Hanacham' in first fruiting body production, but the cultivation period was 40 days shorter. Therefore, 'Daedam' can only harvest fruiting bodies once, it is thought that it can be used as spore-less oak mushroom cultivar for short-term cultivation instead of 'Hanacham' in mushroom farms.

Characterization of a new commercial strain 'Guseol' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 품종 '구슬'의 육성 및 그 특성)

  • To develop new variety of oyster mushroom, 63 intra-specific hybrids between the strain Suhan and #Nongi201 were developed using hyphal anastomosis technique in 2004. The Po2008-275 hybrid between the dikaryon strain 04-154(Suhan x #Nongi201) and the monokaryon strain derived from ASI2487 were developed using hyphal anastomosis in 2008. The Po2008-275 was shown the best cultural characteristics, selected to be a new variety and named as 'Guseol'. The new commercial strain, 'Guseol' had dark grey pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Guseol' were of an excellent quality in that not only the stipe was thick and long but also the pileus was small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $10{\sim}16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body was about 3 to 5 days depending on the temperatures. The shape of fruiting body was thin funnel shape. Fruiting body production per box($43{\times}43{\times}12cm$) was about $1545{\pm}400.9g$ which was almost 137% quantity compared to that of parental strain 04-154. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Guseol' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the mycelial growth of this mushroom were much faster in mushroom complete medium. Similar results were observed with other variety '#Chunchu2'. Analysis of the genetic characteristics of the new commercial strain 'Guseol' showed a major DNA profile as that of the parental 04-154 when primer URP 1, primer URP 2 and primer URP 5 were used, but different to '#Chunchu2' that was used as a control. This new variety of the dark grey oyster mushroom had smart and high quality image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.

The Artificial Cultivation of Pleurotus eryngii (II) -Morphological Characteristics of Fruit Body and Cultural Conditions- (Pleurotus eryngii균의 인공재배 (II) -자실체의 형태적 특성 및 재배조건에 관하여-)

  • These studies were carried out to develop an artificial cultivation method. The diameter and thickness of pileus ranged $1.5{\sim}7.0\;cm$ and $0.8{\sim}3.0\;cm$, respectively. The diameters of stipe were $1.2{\sim}2.5\;cm$ and the lengthes were $4.5{\sim}9.0\;cm$. The spore fingerprint was white. The sizes were spore $10.8{\sim}12.2{\times}4.35{\sim}5.65\;{\mu}m$, basidia $50.0{\sim}59.2{\times}7.4{\sim}7.8\;{\mu}m$, nalsistidia $21.75{\sim}28.7{\times}4.8{\sim}6.1\;{\mu}m$, pileus hymenium cell $50.6{\sim}66.0{\times}4.4{\sim}6.7\;{\mu}m$, and stipe hymenium cell $28.6{\sim}33.0{\times}5.5{\sim}6.6\;{\mu}m$. The thirty percent mixture of rice and wheat bran into sawdust gives the high density of mycelia and the good development of fruiting structure. The optimum water contents of sawdust substrates were $60{\sim}65%$ in which condition the mycelium grows well and gives high density. In PP bottle cultivation, the first fruiting period was $6{\sim}8$ days earlier in nonscratching samples than scratching ones, but the quantity of fruiting body was higher in scratching samples than nonscratching ones. In the case of PP bag cultivation, the first fruiting was 10 days faster, and the quantity of fruiting bodies was 30% higher in samples with 30% wheat bran than those with rice bran. The fleshiness of stipe was $2{\sim}3$ times harder than that of pileus.

Characterization of a new commercial strain 'Goni' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 백색느타리 품종 '고니'의 육성 및 그 특성)

  • To develop new white variety of Oyster mushroom, all white varieties which have been collected and kept in the lab were revived and screened their cultural characteristics. 84 intra-specific Oyster mushroom hybrids between the white-colored mutants Suhan and Wonhyeong were developed using hyphal anastomosis technique in 2007. The Po2007-63 ($2842-7{\times}0205-7$) was shown the best cultural characteristics, selected to be a new variety and named as 'Goni'. The new commercial strain, 'Goni' has white pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Goni' are of an excellent quality in that not only the stipe is thick and long but also the pileus is small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25-30^{\circ}C$ and $10-16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body is about 3 to 5 days depending on the temperatures. The shape of fruiting body is thin funnel shape. Fruiting body production per bottle was about $91{\pm}13$ g which is almost 97% quantity compared to that of other variety 'Miso'. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Goni' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the growth of mushroom were much faster in mushroom complete medium at $20-25^{\circ}C$, but not at $25^{\circ}C$. Similar results were observed with other variety 'Miso'. Analysis of the genetic characteristics of the new commercial strain 'Goni' showed a major DNA profile as that of the parental Suhan when primer URP 1 was used, but different to 'Miso' that was used as a control. When screens were performed with primer URP 2, DNA patterns were similar both to that of the parents and 'Miso'. This new variety of the white Oyster mushroom has a clean and fresh image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.

Characteristics and pathogenicity of Cladobotryum mycophilum isolated from cobweb disease of button mushroom (Agaricus bisporus) in Korea

  • Cobweb disease symptoms were observed in a mushroom farm in Buye, Korea during a disease survey in 2008-2011. Five isolates of Cladobotryum sp. were obtained from the infected caps and stipes. These isolates of Cladobotryum sp. were identified as C. mycophilum based on their morphological, cultural characteristics and analysis of the ITS sequences. Early symptoms were noticed as round, fleshy, yellowish brown lesions on mushroom caps. Late symptoms progressed when the parasitic fungus formed white cobweb circular colonies on dead or damaged pinheads, spread on the surface of the casing, and covered entirely fruiting bodies. Optimal temperature and pH for mycelial growth on MEA is $23^{\circ}C$ and 6.0. Microscopically the spores of the fungus are large and most 2~3 celled produced on vertically branched conidiophores. Mushroom caps turned dark brown and shrunk due to soft rot. Testing of sensitivity to selected fungicides showed that isolate was highly resistance to Mancozeb and Thiophanate-methyl, moderately sensitivity to Iprodione, and highly sensitivity to Benomyl, Prochloraz-Mn and Carbendazim.

Isolation and Identification of Fusicoccum Species from Quercus dentata

  • An imperfect fungus Fusiococcum species was isolated from Quercus dentata. A naturally infected Daimyo oak tree was collected and showed elongate wounds on the stem. The fungal cultures were initially white and cottony, and later turned dark gray. Numerous solitary pycnidia were developed on the medium surface, and typically spherical. Yellowish conidial masses were exuded from pycnidia on the culture plates. Conidial masses were swollen and measured as approximately 100 to $300{\mu}m$ in length. It appeared that conidia were usually held together in globose to oval drops. Conidia were hyaline, single-celled (nonseptate), ellipsoid to fusoid, and measured as approximately $8.0{\times}2.7{\mu}m$. Based on these cultural and morphological characteristics, the fungal isolate was identified as a species of Fusicoccum Corda. To preserve and examine fungal spores exuded from pycnidia on the medium surface, a vapor fixation procedure for scanning electron microscopy was employed in this study. The specimens were exposed to the vapor of 2% (v/v) glutaraldehyde and 2% (w/v) osmium tetroxide each for 2 h. With the vapor fixation we obtained excellent retention of conidial masses in this study. The simple and versatile procedure for demonstrating fungal spores and their exudation from fruiting bodies would facilitate characterization of diverse pathological and environmental isolates as they are in native environments.

  • Nine species of genus Ganoderma different in cultural characteristics each other were separated by histo-anatomical differences. Differences among the species and geographic distribution of G. lucidum were also analyzed to provide the criteria for the classification of Korean cultivation or wild type strains. Korean cultivation and wild type strains were quiet different from other species as well as Taiwan and North American G. lucidum strains in histo-anatomical traits of Ganoderma. Pore color of Korean G. lucidum strains was less brown than those of Taiwan and North American strains. Shapes of pores were round or circular in Korean strains but ellipsoidal or angular in Taiwan or North American strains. Pore numbers of Korean strains were more than those($4{\sim}6/mm$) of Taiwan or North American strains. Hardness of the pileus of Korean strains was much more than that of Taiwan or North American strains. Such characteristics of Korean strains were different from those of other species of Ganoderma. Korean G. lucidum strains could be classified into the other group because they had many different traits in growth characteristics of fruiting bodies and histo-anatomical characteristics from those of Taiwan or North American G. lucidum and other species of G. lucidum complex.
