A report of 21 unrecorded bacterial species of Korea belonging to the phylum Bacteroidota isolated in 2021

  • During screening for indigenous prokaryotic species in Republic of Korea in 2021, a total of 21 bacterial strains assigned to the phylum Bacteroidota were isolated from a variety of environmental habitats including pine cone, seaweed, soil, sea sediment, brackish water and moss. Based on the 16S rRNA gene sequence similarity value of more than 98.7% and formation of a robust phylogenetic clade with the type strain of the closest bacterial species, it was found that the 21 strains belong to independent and recognized bacterial species. There has been no official report that the identified 21 species have been isolated in Republic of Korea up to date. Therefore, 16 species in six genera of two families in the order Flavobacteriales, two species in two genera of two families in the order Cytophagales, one species in one genus of one family in the order Chitinophagales and two species in one genus of one family in the order Sphingobacteriales are proposed as unrecorded species of the phylum Bacteroidota isolated in Republic of Korea. Their Gram reaction, colony and cell morphology, basic phenotypic characteristics, isolation source, taxonomic status, strain ID and other information are described in the species descriptions.

  • 제주도 한라산의 숨은물벵뒤 습지는 여러 다른 생태계에 비해 접근이 용이하지 않아 생물다양성이 높다고 여겨져 왔으나, 세균의 다양성과 새로운 종에 대한 보고는 전혀 이루어지지 않았다. 본 연구에서는 제주도 한라산의 고산 습지인 숨은물벵뒤 습지에서 담수 및 토양시료를 채취하여 Bacteroidetes, Firmicutes, Actinobacteria문에 속하는 세균을 분리하였다. 분리된 세균 중 위의 3개의 문에 속하는 균주의 16S rRNA 유전자 염기서열을 구한 다음 표준균주와 비교하였으며, 16S rRNA 유전자 염기서열의 유사도가 표준균주와 98.7% 미만인 균주를 신종후보 균주로 간주하였다. 전체적으로 과 및 속 수준의 다양성은 높지 않았으며, 특정 계통에 집중되어 신종후보 균주가 발굴되었다. Bacteroidetes 문에는 Mucilaginibacter, Sphingobacterium, Pedobacter, Flavobacterium, Chryseobacterium 속에 속하는 13개의 후보신종이 확인되었다. Firmicutes 문에는 Paenibacillus, Lysinibacillus, Bacillus 속에 해당하는 13개의 후보신종이 배양되었다. Actinobacteria 문에는 Mycobacterium과 Nocardia에 속하는 후보신종 2개가 발굴되었다. 분리된 28개의 후보신종에 대하여 배양학적 특징, 생리학적 특징, 화학분류적 특징을 조사하였으며 본 논문에 기재하였다. 종합적으로 숨은물벵뒤 습지는 아직까지 배양되지 않은 많은 수의 신종 미생물을 포함하고 있는 생태계임이 확인되었다.

식재설계 및 시공시 조경수종 사용에 있어서의 정확성과 자생성 분석 (Analysis on Accuracy and Indigenity of Landscape Plants Species in Planting Design and Construction)

  • The purposes of this study was to investigate accuracy and indigenity of landscape plant species in planting design and construction at housing complex and offer basic data for improving precision of planting construction. The scientific name of landscape plant species was described in planting drawings at only two sites among twelve survey sites. According to analysis on indigenity of plant species in planting drawings, it was found that the component ratio of foreign species was the highest and the component ratio of native species at public housing complex was the lowest. The component ratio of landscape plant species in planting construction was severely differed to that of planting drawings. Also the component ratio of cultivar was higher than that of planting drawings due to planting of sevveral cultivars for one species in planting drawings. In the result of accuracy analysis on landscape plant species in planting construction, it was found that mean ratio of inacccurate species was 33.2% at public housing complex, 29.6% at local company housing complex and 26.4% at nationwide company housing complex. It was found that several cultivars were planted for one species in planting drawings. The representative species which were planted by several cultivars were Camellia japonica, Rododendron spp., Prunus spp. and Magnolia spp. and so on. In order to promote the precision and speciality of planting design and construction, scientific and cultivar name of plants should be described in planting drawings.

A report of 35 unreported bacterial species in Korea, belonging to the phylum Firmicutes

  • In an investigation of indigenous prokaryotic species in Korea, a total of 35 bacterial strains assigned to the phylum Firmicutes were isolated from diverse habitats including natural and artificial environments. Based on their high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with species of validly published names, the isolates were identified as 35 species belonging to the orders Bacillales (the family Bacillaceae, Paenibacillaceae, Planococcaceae, and Staphylococcaceae) and Lactobacillales (Aerococcaceae, Enterococcaceae, Lactobacillaceae, Leuconostocaceae, and Streptococcaceae). Since these 35 species in Korean environments has not been reported in any official report, we identified them as unrecorded bacterial species and investigated them taxonomically. The newly found unrecorded species belong to 20 species in the order Bacillales and 15 species in the order Lactobacillales. The morphological, cultural, physiological, and biochemical properties of the isolates were examined and the descriptive information of the 35 previously unrecorded species is provided here.

  • During a comprehensive investigation of indigenous prokaryotic species in Korea, 25 and 10 bacterial strains assigned to the classes Alphaproteobacteria and Betaproteobacteria, respectively, were isolated from diverse environmental habitats, including soil, mud, tidal field, sea water, sand, rusted iron, and leaf. Based on their high 16S rRNA gene sequence similarities (>98.7%) and the formation of robust phylogenetic clades with type species, each strain was assigned to an independent and predefined bacterial species. Since there were no published or official reports regarding these 35 isolates in Korea, they - 25 species of 14 families in the 5 orders of Alphaproteobacteria and 10 species of 3 families in the two orders of Betaproteobacteria - have been reported as unrecorded species in Korea. In addition, Gram reaction, colony and cell morphology, basic biochemical characteristic, isolation source, and strain ID of each species are also described in the species description sections.

  • To study the biodiversity of bacterial species, here we report indigenous prokaryotic species of Korea. A total of 23 bacterial strains affiliated to the class Alphaproteobacteria were isolated from various environmental sources including seaweeds, seawater, fresh water, wetland/marsh, tidal sediment, plant roots, sewage and soil. Considering higher than 98.8% 16S rRNA gene sequence similarities and formation of a well-defined phylogenetic clade with named species, it was confirmed that each strain belonged to the predefined bacterial species of the class Alphaproteobacteria. There is no official report of these 23 species in Korea; 20 species of 16 genera (Mameliella, Yangia, Paracoccus, Ruegeria, Loktanella, Phaeobacter, Dinoroseobacter, Tropicimonas, Lutimaribacter, Litoreibacter, Sulfitobacter, Roseivivax, Labrenzia, Hyphomonas, Maricaulis, Thalassospira) in the order Rhodobacterales and 3 species of a single genus (Brevundimonas) in the order Caulobacterales. Gram-staining, cell morphology, basic biochemical characteristics, isolation sources, optimum temperature, growth media, and strain IDs are detailed in the species description as well as Table 1.

  • This study was carried out to differentiate the origins of Astragalus membranaceus Bunge and A. membranaceus Bunge var. mogholicus Nakai. To identify the variation of the RAPD patterns between domestic and foreign Astragalus species, 40 random primers were applied to ten accessions of A. membranaceus and six accessions of A. membranaceus var. mogholicus genomic DNA, respectively, Ten primers of 40 primers could be used to discriminate the origins and 33 polymorph isms among 44 scored DNA fragments (33 fragments are specific for A. membranaceus and A. membranaceus var. mogholicus) were generated using these primers, 75.0 % of which were polymorphic. Especially, three primers of ten primers, OPA17, OPA11 and OPB11, were useful to differentiate between domestic and foreign Astragalus species. RAPD data from the 10 primers were used for cluster analysis and cluster analysis of RAPD markers showed that the two groups are distinct genetically. Consequently, RAPD analysis was a useful method to discriminate between A. membranaceus and A. membranaceus var. mogholicus.

한국산 양귀비과 식물에 관한 본초학적 연구 (A Herbological Study on the Plants of Papaveraceae in Korea)

  • Objective : For the purpose of developing Korean herbology of the plants to Papaveraceae in Korea, the literatures of the successive generations have been thoroughly investigated to prepare this article.Methods : Books and research papers about Herbology which published at home and abroad were examined.Results : A list was made about Papaveraceae plant which were cultivated or grew naturally in Korea, after investigated the data on domestic and foreign Papaveraceae plants. Out of those lists, serviceable plants were selected and with those available plants, their distributions were analysed and parts which can be used as medicines were divided into 3 categories such as oriental medicine name, scientific name and non-official name. Papaveraceae's properties flavor, channels they use, effects, and toxicity were also noted. Papaveraceae (grew in Korea) were divided into 4 classes with 8 species. Out of those, 3 classes with 4 species were found serviceable which indicates 50% of all. Out of all 8 species of Papaveraceae family, Papaver genus were found 4 species, which were shown the most. Among 3 genus of 4 species, medicinal plants were all one species each. Out of all serviceable parts in Papaveraceae, Radix and Herba, Etc parts took first place as 2 species. There are toxic, the 3 species and 6 parts survey said.Conclusion : There were totaled to 4 genera and 8 species in Papaveraceae in Korea and among them medicinal plants are 3 genera, 4 species, some 50% in total.

Cupressaceae's properties flavor, channels they use, effects, and toxicity were also noted. Cupressaceae(grew in Korea) were divided into 4 classes with 20 species. Out of those, 2 classes with 4 species were found serviceable which indicates 20% of all. Out of all 20 species of Cupressaceae family, Juniperus genus were found 12 species, which were shown the most. Among 2 genus of Cupressaceae species, medicinal plants Juniperus genus and Thuja genus were found 2 species each. Out of all serviceable parts in Cupressaceae, Folium parts took first place as 4 species. The research also suggests that there are 1 species and 1 parts which are toxic. Conclusion : There were totaled to 4 genera and 20 species in Cupressaceae in Korea and among them medicinal plants are 2 genera, 4 species, some 20% in total.
