Changes in Dormant Phase and Bud Development of 'Fuji' Apple Trees in the Chungju Area of Korea (충주지역에서 '후지' 사과나무의 휴면단계 변화 및 눈 발달)

  • In this study, we investigated the onset and release of endo-dormancy under natural conditions by observing bud break characteristics in 'Fuji' apple trees using water cuttings. Through examinations of bud break rate and days to bud break, we found that the endo-dormancy of 'Fuji' apple tree continues for 70 d from 165 to 255 d after full bloom (DAFB), from late October to early January of the following year. In addition, within 20 d of first bud break, based on a final bud break rate of 60% or more, we able to identify the timing of the changeover from para-dormancy to endo-dormancy, and endo-dormancy to eco-dormancy. Analysis of the chilling requirement during the endo-dormancy period revealed that chilling accumulation up to 255 DAFB to release endo-dormancy amounted to 666 and 517 h based on the CH and Utah models, respectively. Observation of internal changes in the bud during endo-dormancy showed that flower bud differentiation begins from mid-July, and t ime of inflorescence o f the disk f lower is a vailable to f ind. The f lower buds subsequently developed slowly but steadily during endo-dormancy and in the following year in February, the developmental stage of each organ had progressed. Moreover, the flower buds of 'Fuji' apples were mostly healthy during the dormancy period, but some exhibited necrosis of flower primordium, due partial cell damage from the formation of ice crystals rather than a direct effect of the low temperature. Flower buds were formed in both the axillary buds of bourse shoots and terminal buds of spurs, but lower bud differentiation was observed for the terminal buds of spurs at rate of about 65% of total buds, which was directly related to the bud size and shoot diameter.

Effects of Removing Time of Flower Buds on Root Yield and Paeoniflorin Content in Paeonia lactiflora Pallas (작약(芍藥) 화뇌제거시기에 따른 근수량(根收量)과 Paeoniflorin함량(含量) 변화(變化))

  • This experiment was carried out to determine the proper time for flower bud removing to improve growth, yield and paeoniflorin content of root in peony(Paeonia lactiflora Pallas). The flower buds removement caused short stem length by $7{\sim}9cm$ compared with control, but number of stem and stem diameter were similar to that of control. The number of root, fresh root weight and paeoniflorin content in peony increased when their flower buds were removed at earlier stages. The root dry weight was highest of 1.647kg/10a, and increased by 27% compared with control when their flower buds were removed at early stage (bud diameter was less than 10mm). The root dry weight increased by 13% and 10%, respectively. when their flower buds were removed prior to flowering and at flowering stage. Paeoniflorin content in leaf was higher than that in stem, and was higher at early stage. Leaf and stem showed higher paeonif1orin content when their flower buds were removed.

Effects of Extract from Fermented Flower-buds of Panax ginseng C.A. Meyer on Mouse Cytokine IL-6, TNF-α Production (발효 인삼꽃 추출물의 경구 투여가 마우스 사이토카인 IL-6, TNF-α의 생성에 미치는 영향)

  • Panax ginseng C.A. Meyer has been used as a traditional medicinal ingredient and the ginseng flower-buds also proved to have good medicinal properties. In this study, in order to enhance immune activities of ginseng flower-bud, the ginseng flower-bud extract was being fermented by Bacillus subtilis KCTC 1022 (BS), Lactobacillus plantarum KCTC3 and Saccharomyces cerevisiae strain CHY1011 (SC). Mice were orally administered daily for two weeks at two different concentrations (100 and 200 mg/kg B.W.). Treatment samples were water extracts of ginseng flower-buds (FD), water extracts of fermented ginseng flower-buds (FM) and controls for saline solution. Cytokine production (IL-6, TNF-${\alpha}$) either stimulated with LPS or not stimulated with LPS was detected by the ELISA assay when using the cytokine kit. Cytokine was statistically increased at supplemented groups with LPS in both the 100 and the 200 mg/kg B.W. and treatment with FM significantly decreased the LPS-induced TNF-${\alpha}$ and IL-6 production more than the treatment with FD. The results of this study may suggest that supplementation with FM increases the immune function by regulating cytokine production capacity for activated macrophages.

  • Girdling and girdling+pinching treatments on 'Sekaiichi' apple remarkably reduced June drop and accelerated fruit growth in early stage of fruit development. Girdling+pinching and pinching reduced bourse shoot growth compared with non-treated control. There were no significant differences in flower bud formation in the following year among treatments. It was assumed that the optimum period for girdling is between 5 days before full bloom and full bloom period.

  • The present experiments were conducted to investigate some of the factors affecting the number of haploids derived from anther culture of Nicotiana tabacum. Anther - donor plants grown under controlled environment room at 3$0^{\circ}C$ yielded more haploid than room at 18, 25 and 26-22-18$^{\circ}C$ in anther culture. Donor plants starved of fertilizer yielded more haploids as compared to those of the well fed with fertilizer in anther culture. Pretreatment of exercised flower bud at 5$^{\circ}C$ was shown to be more effective in anther culture than pretreatment at 7 and 1$0^{\circ}C$, and the optimum temperature and period of pretreatment were 4 or 6 days at 5$^{\circ}C$.

  • Sorting cut roses according to quality is very essential to increase the value of the product. Many factors are involved in determining the grade of cut roses: length, thickness, and straightness of stem, color and maturity of bud, and extra. Among these factors, the stem straightness and bud maturity are considered to be difficult to set proper classification criteria. In this study, a prototype machine and an analysis procedure were developed to grade cut roses according to stem straightness and bud maturity by utilizing color image processing and neural network. The test results indicated 15.8% classification error for stem straightness and 10.0% for bud maturity.

On-Line Sorting of Cut Roses by Color Image Processing (영상처리에 의한 장미 선별)

  • A prototype cut-flower sorter was developed and tested for its performance with five varieties of roses. Support plates driven by a chain mechanism transported the roses into an image inspection chamber. Color image processing algorithms were developed to evaluate the length, thickness, and straightness of stem and color, height, and maturity of bud. The average absolute errors of the system for the measurements of stem length, stem thickness, and height of bud were 19.7 mm, 0.5 mm, and 3.8 mm, respectively. The results of classification by the sorter were compared with those of a human inspector for straightness of stem and maturity of bud. The classification error for the straightness of stem was 8.6%, when both direct image and reflected image by a mirror were analyzed. The accuracy in classifying the maturity of bud varied among the varieties, the smallest for‘Nobless’(1.5%) and the largest for‘Rote Rose’(13.5%). The time required to process a rose averaged 2.06 seconds, equivalent to the capacity of 1,600 roses per hour.

  • Astragalus membranaceus has flowers that are similar to that of the legume family, but shows poor bearing when self-pollination is induced. Thus, this study was carried out observing the ripening procedure of pistils and stamens and development stages of pollen in the context of the birth and growth of the flower. As to the bearing of the flower of A. membranaceus, few pod setting and 13% pod setting were observed when self-pollination is induced by paper-bag covering or artificial pollination treated respectively. The result indicates that A. membranaceus is a cross-pollination plant. A pistil grew faster than a stamen until just before blooming. The flower size was about 17.0mm$\times$4.0mm. Pistils and stamens had the same length after flowering. Pollen mother cells passed through meiosis and mitosis when its length reached around 3.5mm, thus creating the tetrade when 4 mm long. Pollen attained full growth when the bud was about 10mm long. An anther was found to tend to dehisce when the length of a bud reached around 12.0mm. As to the shape of pollen, about 70 % were normal. 1% and 30 % were small or empty pollen respectively. The result indicates that pollen of A. membranaceus attains full growth just before anther dehiscence which occurs before blooming while pistils grow faster than stamens until before flowering.

  • The most important subject in cultivation of A. gigas NAKAI would be how to decrease the bolting rate. Belcause the decrease and poor of the yields and quality is imevitable in such field where the bolting rate is high. This study was carried out to investigate the time of flower-bud differentiation as a part of improvements of cultivation method. Shoot apex was fixed in Farmer's solution(ETOH 3:Acetic acid 1), thin paraffin ribbon was made, and the tissue was stained by Safranin solution. Differentiation time of flower-bud in A. gigas differed greatly from individuals by the seedling size, generally begun from middle of May to late of June, it was time that two leaves spread. In this results, Photoperiocid types of A. gigas NAKAI were regarded as LI type or LS type of dull-photosensitivity.

Effects of Night Temperature Treatment of Raising Seedlings before Transplanting on Growth and Development of Pepper (육묘(育苗) 야간온도(夜間溫度) 처리(處理)가 고추의 생육(生育)에 미치는 영향(影響))

  • This study was carried out to investigate effects of different night temperature treatments during nursery period on flower bud differentiation and growth of pepper cv. Cheongyang. Number of leaves, top fresh weight and top dry weight of pepper seedlings were increased with increasing the night temperature during nursery periods. And also flower bud differentiation and days to flowering were accelerated as increasing the night temperature. Plant height, stem diameter, branch length and intermode length of pepper after transplanting were height at the low night temperature ($28/11^{\circ}C$), but they were retarded at the high night temperature ($28/21^{\circ}C$) treatment. Number of lateral branches was significantly reduced at the high night temperature, but there was no a regular tendency in branching habit of the main stem by temperature treatments. Seedling growth before transplanting was retarded at the low night temperature but gradually recovered after transplanting into the plastic house. However, seedling growth at the high night temperature was shown in contrast to above response of the low night temperature.
