참죽나무는 멀구슬과 나무로서 한국과 중국에 널리 분포하고 있다. 한방에서는 장염(腸炎), 이질(痢疾), 개선(疥癬) 등의 치료에 이용되고 있다. 본 연구는 참죽나무 잎에서 3종의 flavonol glycosides를 분리하여 NMR을 통해 구조를 확인한 결과, kaempferol-3-O-rhamnoside (1), quercetin-3-O-rhamnoside (2)와 quercetin-3-O-glucoside(3)로 구조 동정되었다. 분리된 화합물들의 생리활성은 matrix metalloproteinase-2 저해 활성과 인체암 세포주에 대한 세포독성과 apoptosis실험을 통하여 항암효과를 조사하였다. 그 결과 인체암 세포주에 대한 세포독성과 apoptosis 저해활성은 나타내지 않았으나, MMP-2활성저해 조사에서는 강한 저해 효과를 나타내었다. 특히 quercetin-3-O-glucoside는 암세포에 직접적인 독성을 보이는 것이 아니라 암의 침윤과 전이에 특이적으로 작용하는 물질로 보여진다.
Cudrania tricuspidata Bereau (Moraceae) have been used for anti-inflammatory, anti-hepatotoxic, anti-hypertensive and anti-diabetic activities. In order to investigate the efficacy of antioxidant activity, the bio-activity guided fraction and isolation of Biologically active substance were performed. $H_{2}O,\;30\%,\;60\%,\;100%$ MeOH and acetone fractions were examined on the antioxidant activity by DPPH method. It was shown that $30\%,\;60\%,\;100\%$ MeOH fractions have significantly antioxidant activity. From $30\%$ MeOH fraction, two dihydroflavonoid glycosides dihydroquercetin 7-O-$\beta$-D-glucopyranoside (I), dihydrokaempferol 7-O-$\beta$-D-glucopyranoside (V) were isolated and $60\%$ MeOH fraction, six flavonoids including quercetin 3-O-$\alpha$-L-rhamnopyranosyl($1\rightarrow6$)-$\beta$-D-glucopyranoside (II), quercetin 3-O-$\beta$-D-glucopyranoside (III), quercetin 7-O-$\beta$-D-glucopyranoside (IV), kaempferol 3-O-$\alpha$-L-rhamnopyranosyl($1\rightarrow6$)-$\beta$-D-glucopyranoside (VI), kaempferol 3-O-$\beta$-D-glucopyranoside (VII), kaempferol 7-O-$\beta$-D-glucopyranoside (VIII) were isolated. To investigate the antioxidant activities of each compounds, we measured radical scavening activity with DPPH method and anti-lipid peroxidative efficacy on low density lipoprotein (LDL) with TBARS assay. Four compounds of quercetin glycosides (I, II, III, IV) showed significant antioxidant activity.
굴피나무 수피를 채취하여 건조시킨 후 분말로 제조하여 95% EtOH로 추출하고 여기서 얻어진 EtOH 조추출물의 하나는 헥산(n-hexane), 디클로로메탄($CH_2Cl_2$)으로, 또 하나는 석유에테르(petroleumether), 디에틸에테르($Et_2O$) 및 에틸아세테이트(EtOAc)를 사용하여 순차적으로 분획하였다. 이중 디클로로메탄($CH_2Cl_2$) 가용부(6.0 g)는 EtOH-$CHCl_3$ (7:3, v/v)를 용출용매로 한 Sephadex LH-20 칼럼($72.0{\times}5.0cm$)을 사용한 칼럼크로마토그래피를 실시하였다. 또한 디에틸에테르($Et_2O$) 가용부는 $CHCl_3$-MeOH (9:3, v/v)을 용출용매로 한 silica gel 칼럼($42.0{\times}3.5cm$)을 사용하여 칼럼크로마토그래피를 실시하였다. 단리된 화합물들은 TLC로 확인한 후 $^1H$-, $^{13}C$-NMR, HMBC 등의 스펙트럼을 사용하여 화학구조를 규명하였고 EI-MS로써 분자량을 측정하였다. 추출물로부터 3, 3', 4', 5, 7-pentahydroxyflavone (quercetin), 3, 3', 4', 5, 5', 7-hexahydroxyflavone (myricetin), flavonoids 배당체인 kaempferol-3-O-${\alpha}$-L-rhamnopyranoside (afzelin), quercetin-3-O-${\alpha}$-L-rhamnopyranoside (quercitrin), myricetin-3-O-${\alpha}$-L-rhamnopyranoside (myricitrin)등을 단리할 수 있었다.
Bioassay-directed chromatographic fractionation of an ethyl acetate extract from leaves of sweet potato (Ipomoea batatas L.) afforded six quinic acid derivatives: 3,5-epi-dicaffeoylquinic acid (1), 3,5-dicaffeoylquinic acid (2), methyl 3,5-O-dicaffeoylquinate (3), methyl 3,4-dicaffeoylquinate (4), methyl 4,5-dicaffeoylquinic acid (5),4,5-dicaffeoylquinate (6), and two phenolic compounds: caffeic acid (7) and caffeic acid methyl ester (8) together with three flavonoids: quercetin 3-O-${\beta}$-D-glucopyranoside (9), quercetin 3-O-${\beta}$-D-glucopyranoside, isoquercitrin (10) and kaempferol 3-O-${\beta}$-D-glucopyranoside (11). The structures of these compounds were elucidated by the aid of spectroscopic methods. These compounds were assessed for antioxidant activities using three different cell-free bioassay systems. All isolates except 11 showed potent DPPH and superoxide anion radicals scavenging, and lipid peroxidation inhibitory activities. 3,5-epi-DCQA (1) and methyl quinates (3-5) along with flavonoide 9 were isolated for the first time from this plant.

The n-BuOH soluble fraction of the whole plant of Persicaria thungergii showed acetylcholinesterase inhibitory activity. Four flavonoid glycosides and a flavonoid were isolated from this fraction, and identified as quercitrin (1), luteolin-4'-O-${\beta}$-D-glucopyranoside (2), quercetin (3), quercetin-3-O-glucuronide (4), and isorahmnetin-3-O-glucuronid (5), by chromatographed and spectral data, respectively. All isolated compounds were showed acetylcholinesterase inhibitory activity, with $IC_{50}$ values of 243.1, 10.5, 39.1, 8.2 and $23.2{\mu}M$, respectively.

It has been well known that extended exposure to reactive oxygens causes severe damage to susceptible biomolecules. In this study, the effects of flavonoids including trifling and kaempferol from Ginseng leaves on single oxygen induced photohemolysis of erythrocytes and free radical scavenging activities were investigated . Each flavonoid aglycone (5-50UM) such as kaempferol, quercetin or baicalein exhibited a high protective effect against the photohemolysis. They protected the cells by scavenging 102 and free radicals. Although the free radical scavenging activities of the flavonoid glycosides were not much lower than those of their corresponding aglycones, their insolubility into lipid bilayers of membrane made them less effective in preventing the photohemolysis induced by 1O2. The 102 and free radical scavenging activities of flavonoids were estimated by the decomposition of the flavonoid by 1O2 and the bleaching of free radicals by the flavonoid, respectively. The solubilization of the flavonoid into micelle or erythrocytes was deduced from spectrophotometric and microscopic observations. The cooperation of L-ascorbic acid and a flavonoid, and a possible involvement of lipoxygenase or cyclooxygenase in the photohemolysis mechanism were discussed. 신선초의 건조분말을 사료에 5%수준으로 혼합하여 백서에 8주간 투여하였을 때, 총 cholesterol, LDL-cholesterol, VLDL-cholesterol의 농도를 낮추며, HDL-cholesterol의 농도는 높이는 효과가 있음이 관찰되었다. 신선초에 존재한다고 알려진 항산화성 flavonoid들 중 quercetin과 그 배당체의 하나인 isoquercitrin이 백서의 체내에서 측정된 반면, 다른 quercetin 배당체인 hyperoside는 측정되지 않았다. 이 결과는 장기간 신선초투여에 의해 신선초에 함유된 quercetin 또는 그 배당체가 실제로 체내로 흡수되어 약리작용을 발휘할 수 있을 가능성을 제시해 준다.
The major flavonoid component of Ginseng leaf is trifolin, a glycoside of kaempferol. To evaluate the antioxidative properties of trifolin and kaempferol on cellular membranes, we compared them with the other flavonoids through the 102-Induced photohemolysis of rabbit erythrocytes. All the flavonoid aglycones including kaempferol, quercetin and baicalein protected effectively the cells from the 102-caused damage in a dose- dependent manner, by scavenging 102 and free radicals in the cellular membranes. The solubilization of the flavonoid aglycones into micelles or erythrocyte membranes was deduced from spectro-photometric and microscopic observations. The flavonoid glycosides were not protective or less protective than their corresponding aglycones, and trifolin was the only glycoside that exhibited a solubilization into the membranes and a significant protection against the photohemolysis. We also tested some phenolic compounds contained in Ginseng, and found that they did not prevent the photohemolysis so effectively as kaempferol or trifolin.
All ten compounds were isolated from the methanolic extract of the whole plants of Diodia teres through repeated silica gel and Sephadex LH-20 column chromatography. Their chemical structures were elucidated as three iridoid glycosides, asperuloside, geniposidic acid and asperulosidic acid, a coumarin glycoside, scopolin, and six flavonoids, rutin, kaempferol-3-0-rutinoside, quercitrin, astragalin, isoquercitrin and quercetin by spectroscopic analysis.
