Preparation of Functional Healthy Drinks by Ethanol Extracts from Defatted Safflower Seed Cake (탈지 홍화씨박 에탄올추출물 함유 기능성 건강음료의 제조)

  • Functional healthy drinks were processed with freeze dried powders of ethanol extract from of defatted safflower (Carthamus tinctorious L.) seed cake and some useful components of the drinks were investigated. Yield of freeze dried powder was the highest as 8.42% when it extracted with 60% ethanol (60% EFDP). Each drink contained 0.02% of freeze dried powder and ranged 10.6∼13.8% of soluble solid, 2.90∼3.68 of pH, 0.10∼0.83% of titratable acidity. ‘L’ value of drink-I (DSD-I) was the highest as 94.82$\pm$2.45, ‘b’ and ‘a’ value of drink-V (DSD-V) was highest as 27.15-2.65 and 28.67$\pm$2.69, respectively. Major free sugars of drink were 6015.3∼7918.2 mg% of glucose and 1511.4∼2091.0 mg% of sucrose. The content of citric acid was the highest as 179.2∼981.3 mg%. The content of total phenol in 60% EFDP was 99.17 mg% and that of drink-II(DSD-II) and DSD-V was 307.84 mg% and 224.06 mg%, respectively. Total flavonoid was contained as 50.29 mg% in 80% ethanol extract (80% EFDP) and 125.20 mg% in DSD-V. N-[2-(5-hydroxy-1H-indol-3-yl) ethyl] ferulamide (serotonin-I) was determined as high as 18.81 ppm in 80% EFDP and ranged 2.42∼2.89 ppm in drinks. N-[2-(5-hydroxy-lH-indol-3yl)ethyl]-p-coumaramide (serotonin-II) was determined as 30.17 ppm in 80% EFDP and ranged 3.79∼4.59 ppm in drinks. Acacetin, flavonoid compound were 9.83 ppm in amyloglucosidase hydrosis + 60% ethanol extract (A + 60% EFDP) and ranged 0.98∼1.26 ppm in drinks. Electron donating ability (EDA, %) was measured and compared with 100 ppm BHA as chemical antioxidant. EDA was 93.97$\pm$2.21% in A+60% EFDP, 94.79$\pm$2.26% in DSD-I, 94.69$\pm$1.37% in DSD-II, and 93.83$\pm$1.49% in BHA. DSD-II added with hot water extract solution from Korean ginseng and safflower yellow pigment recorded the highest sensory score.

Functional Components and Radical Scavenging Activity of Germinated Brown Rice according to Variety (품종별 발아현미의 기능성분 및 라디칼 소거 활성)

  • This study investigated the changes in chemical components, antioxidant compounds, and activity before (BG) and after germinated (AG) brown rice in order to promote the availability of these beneficial factors. The GABA content of BG and AG brown rice were 0.07~6.61 and 11.13~49.72 mg/100 g, respectively. The ${\alpha}$-amylase activity of AG brown rice was 1.77~70.25 unit/g fold higher than it was in BG brown rice. Total polyphenol and flavonoid contents, which are known to be related to antioxidation, were higher in BG brown rice than in AG brown rice. The total polyphenol contents of BG and AG brown rice were 10.52~36.38 and 11.38~26.33 mg/100 g, and the total flavonoid contents were 3.55~13.39 and 3.52~9.78 mg/100 g, respectively. Also, DPPH radical scavenging activity was 57.64~251.34 and 50.49~213.35 mg TE/100 g, respectively. ABTS radical scavenging activity and total tannin content showed a similar trend to DPPH radical scavenging activity. We expect that this data will be useful in the manufacturing of food products.

Anti-oxidant and α-Glucosidase Inhibition Activity of Extracts or Fractions from Diospyros lotus L. Leaves and Quantitative Analysis of Their Flavonoid Compounds (고욤잎 추출물과 분획물의 항산화 및 alpha-glucosdiase 저해 활성 및 플라보노이드 화합물의 정량)

  • Persimmon leaves were commonly consumed as beverages, but were also used as popular folk medicine in Asia. The purpose of this work was to assess the biological activities of Diospyros Lotus L. extracts (DLLE). Various solvent extracts, including n-Hexnae, $CHCl_3$, EtOAc, and n-BuOH fractions, were obtained from the methanol extract of Diospyros Lotus L. leaves. The increasing interest in the powerful biological activity of plant phenolics and flavonoids outlined the necessity for determining their content in medicinal herbs. In this study, the total polyphenol and flavonoid contents (TPC and TFC) in the EA fraction were higher than those of other fractions. The biological activities of DLLE were tested using the 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity assay, as well as superoxide dismutase (SOD) activity as an anti-oxidant effect and ${\alpha}$-glucosidase inhibitory activity as an anti-diabetic effect. The EA fraction with high TPC and TFC values showed the highest anti-oxidant effect and high ${\alpha}$-glucosidase inhibition. The EA fractions were further purified into eight fractions using open column chromatography. Higher anti-oxidant and anti-${\alpha}$-glucosidase activity were observed in polar fractions. The content of the flavonoids, including quercein-3-O-rutinoside, kaempferol-3-O-glucoside, myricetin, luteolin, and kaempferol, were analyzed in effective fractions using high-performance liquid chromatography (HPLC). The results suggest that DLLE have anti-oxidative and anti-diabetic effects and thus, have the potential as anti-diabetic materials and as a source for natural health products.

Quality and Antioxidant Characteristics of Cooked Rice with Various Mixed Grains in Korea (혼합잡곡 첨가 취반 밥의 품질 및 항산화특성)

  • This study evaluated the quality characteristics, polyphenolic compounds, and radical scavenging activity of cooked-rice added to commercially available mixed grains. L-value of cooked-rice with various mixed grains decreased compared to that of cooked-white rice; however, a- and b-values increased. Hardness and elasticity of cooked-rice added to various mixed grains were significantly lower in the pressure cooker compared to the electric cooker. There was no significant difference in adhesiveness and stickiness between rice from the electric cooker and pressure cooker. Total polyphenol and flavonoid contents of cooked-rice added to various mixed grains were significantly increased. The average total polyphenol content of cooked-rice added to various mixed grains cooked in an electric cooker and pressure cooker were $16.50{\pm}3.86$ and $15.88{\pm}3.52mg$ gallic acid equivalent /100 g, and flavonoid contents were $1.58{\pm}0.00$ and $1.55{\pm}0.02mg$ catechin equivalents/100 g, respectively. The average of 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was $9.27{\pm}2.62$ and $8.72{\pm}2.41mg$ trolox equivalent (TE)/100 g, and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was $22.89{\pm}4.60$ and $23.07{\pm}4.49mg$ TE/100 g for cooked-rice added to various mixed grains cooked in an electric cooker and pressure cooker, respectively. Phenol content and radical scavenging activity of cooked rice was in proportion to the amount of added grains, such as brown rice, colored rice, barley, soybean, and sorghum.

Changes of nutritional constituents and antioxidant activities by the growth periods of produced ginseng sprouts in plant factory (식물공장에서 생산된 새싹인삼의 생육 시기에 따른 영양성분 및 항산화 활성 변화)

  • Ginseng sprouts, which can be eaten from leaves to roots, has the advantage of not having to use pesticides without being affected by the season by using smart farms. The optimal cultivation timing of sprout ginseng was checked and the nutritional content and antioxidant activity were compared and analyzed. The values of total fatty acids and total minerals were no significant changes during the growth periods. The contents of total amino acids were slightly decreased to 45 days and after increased to 65 days. When the growth period was 65 days, arginine had the highest content of 3309.11 mg/100 g. The total phenolic contents were high at 3.73 GAE mg/g on the 45 days, and the total flavonoid contents were also the highest at 9.04 RE mg/g on the 45 days. The contents of total ginsenoside was not noticeable for the growth periods (29.83 on 25 days→32.77 on 45 days→26.02 mg/g on 65 days). The ginsenoside Rg2 (0.62 mg/g), Re (8.69 mg/g), Rb1 (4.75 mg/g) and Rd (3.47 mg/g) had highest contents on 45 days during growth. The values of phenolic acids and flavonols were gradually increased to 45 days (338.6 and 1277.14 ㎍/g) and then decreased to 65 days. The major compounds of phenolic acids and flavonols were confirmed to benzoic acid (99.03-142.33 ㎍/g) and epigallocatechin (416.03-554.64 ㎍/g), respectively. The values of 2,2-diphenyl-1-picrylhydrazyl (44.27%), 2,4,6-azino-bis (3-ethylbenzothiazoline-6-sulphnoic acid) diammonium salt (75.16%), and hydroxyl (63.29%) radical scavenging activities and ferric reducing/antioxidant power (1.573) showed the highest activity on the 45 days as well as results of total phenolic and total flavonoid contents.

Anti-oxidative Activity of the Extracts from Houttuynia cordata Thunb. Fermented by Lactic Acid Bacteria (어성초(Houttuynia cordata Thunb) 유산균 발효물의 항산화 활성)

  • This study was performed to evaluate the possibility of application of lactic acid bacteria fermentation to increase the anti-oxidative activity of extracts from Houttuynia cordata Thunb. Houttuynia cordata Thunb. was fermented by two species of lactic acid bacteria, Leuconostoc mesenteroides 4395 and Lactobacillus sakei 383. The anti-oxidative activities of Houttuynia cordata Thunb. extracts were analyzed both before and after fermentation. Anti-oxidative activity was determined by in vitro assays to measure 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging and super oxide dismutase (SOD)-like activities, and by determining total flavonoid and total phenolic compound contents. The extracts of fermented Houttuynia cordata Thunb. had higher anti-oxidative activity than the unfermented control. The DPPH scavenging activity of the extracts after fermentation by Leuconostoc mesenteroides 4395 at 30℃ for 5 days was 71.67±0.52%, and after Lactobacillus sakei 383 fermentation at 35℃ for 5 days was 70.11±0.67%; these activities were both about 20% higher than the control. Increases of about 10 mg GAE/g of total phenolic compounds were found in both fermented extracts and both contained about 6 mg quercetin equivalents/g of total flavonoids, compared with 35.90±0.61 mg/g and 21.69±1.52 mg/g in the control, respectively. These results also suggested that fermentation time and temperature were important factors in determining the anti-oxidative effect of extracts from fermented Houttuynia cordata Thunb. These findings should be valuable for the development of medicines or functional foods with antioxidative activity.

Comparison of NaCl and D-Pinitol Content of Freeze-Dried Ice Plant, Natural and Purified Commercial Salts and their Radical Scavenging Activity

  • This study investigated the suitability of freeze-dried ice plant (Mesembryanthemum crystallinum) as the source of an edible healthy salt alternative by examining the content of NaCl, D-pinitol, total phenols, total flavonoids, and DPPH radical scavenging activity compared with those of natural and purified salts. Our results showed that the NaCl content of freeze-dried ice plant, natural and purified salts was 19, 94 and 99%, respectively. The D-pinitol content of freeze-dried ice plant was 777 mg/100 g, whereas D-pinitol was not detected in either natural or purified salts. The total phenol and flavonoid contents of freeze-dried ice plant were 115 mg GAE/100 g and 985 mg RE/100 g, respectively. The DPPH radical scavenging activity of freeze-dried ice plant was markedly higher than that of natural and purified salts. Altogether, these results show that freeze-dried ice plant contains both NaCl and bioactive compounds and can be used as a source of edible salt with positive health effects.

Chemical Components of Propolis and Its Ethanolic Extracts (프로폴리스 및 알콜 추출물의 화학성분)

  • In order to use as a new functional food material, chemical components of propolis and its extracts were surveyed. The contents of crude fat, nitrogen free extract, crude protein, ash and crude fiber in propolis were 86.41%, 7.32%, 2.71%, 1.05% and 0.20%, respectively. The mineral contents were in the order of Na (120.40 mg%), Ca (115.40 mg%), K (105.87 mg%) and Ca were higher in water extract than alcohol extract. Free sugars were composed of sucrose 152 mg%, glucose 114 mg% and fructose 6 mg%. The major amino acids of propolis were lysine 395.29 mg%, cystine 267.66 mg% and glutamic acid 248.14 mg%, respectively. Eight fatty acids in propolis were identified and the major fatty acids were oleic acid (51.89%), myristic acid (20.86%) and palmitic acid (20.28%). Myricetin, quercetin, apigenin and kaempferol were shown as major flavonols and total flavonol contents were higher in 50% ethanol extract than any other extracts. Major Polyphenol compounds in four kinds of extracts were gallic acid, chlorogenic acid, catechin, epigallocatechin gallate, epicatechin and epicatechin gallate.

Effect of Extraction Methods on Antioxidant Activities of Mori ramulus (추출방법에 따른 상지 추출물의 항산화 활성)

  • The objective of this study was to examine the antioxidant activities of extracts from Mori ramulus using different extraction methods (HE, hot water extraction; EE, 50% ethanol extraction; UE, ultrasonic extraction; PE, pressured extraction). The extraction yield of PE (3.07%) was higher than that of UE (1.43%), EE (1.18%), and HE (1.07%). The total phenolic and flavonoid contents of EE were 334.66 mg/g, and 35.64 mg/g, respectively. The oxygen radical absorbance capacity (ORAC) of EE ($3,483.37{\mu}M/g$ FW) was higher than those of HE ($2,687.52{\mu}M/g$ FW), UE ($2,300.45{\mu}M/g$ FW), and PE ($2,117.62{\mu}M/g$ FW). The DPPH and ABTS radical scavenging activities of EE at $1,000{\mu}g/mL$ were 65.84%, and 97.52%, respectively. The superoxide radical scavenging activity of EE was 67.77~98.74% ($100{\sim}500{\mu}g/mL$) higher than those of other extracts. The ferric reducing antioxidant power and reducing power of EE were $189.00{\sim}974.80{\mu}M$, and 0.12~0.82, respectively. The tyrosinase inhibitory activity of EE (23.25~67.20%) improved with an increase treatment concentration. The antioxidant and tyrosinase inhibitory activities of EE were significantly higher than those of other extracts. In conclusion, we provided experimental evidence that extracts from Mori ramulus have potential as functional materials.

Effects of Quality Characteristics and Antioxidant Activity of Korean Cultivated Wild Ginseng Extract (산양삼의 품질특성 및 항산화 활성에 미치는 영향)

  • In this study, we investigated the nutritional and functional constituents as well as quality characteristics and antioxidant activity of Korean cultivated wild ginseng (KG). The chemical compositions and amino acid content of KG were 7.56% water, 73.01% carbohydrates, 12.58% protein, 1.99% lipids, and 5.54% ash as well as 16.17 mg/g of amino acids, respectively. The major ginsenoside and minor ginsenoside contents of KG were 15.94 mg/g and 0.04 mg/g, respectively. The total polyphenol and flavonoid contents of KGE (Korean cultivated wild ginseng with 70% ethanol extract) were 8.93 mg GAE/g and 3.96 mg RHE/g, respectively. KGE also showed higher antioxidant activity than the other extracts (KGW, Korean cultivated wild ginseng with water extract) with regard to DPPH and ABTS radical scavenging activities (57.75% and 70.73%, respectively), nitrite oxide scavenging activity (44.01%), SOD-like activity (78.05%), reducing power activity ($1.08OD_{700nm}$), and ferrous ion-chelating activity (65.33%). Additionally, KGE had higher elastase, collagenase, and tyrosinase inhibition activities than KGW. These results suggest that KGE can be used as a bioactive and functional material in the food industry.
