Antibacterial efficacy and safety of copper sulfate pentahydrate to cultured fish (양식 어류에 있어서 황산동(copper sulfate pentahydrate)의 안전성과 살균효과에 대한 연구)

  • Antibacterial efficacy and safety of copper sulfate pentahydrate (5% solution) was evaluated in laboratory and field studies for disinfection of some fish bacterial pathogens in cultured fish, flounder(Paralichthys olivaceus), mirror carp(Cyprinus carpio), gold fish(Carrassius auratus) and eel(Anguilla japonica). Dipping of fishes for acute toxicity was performed for a period of 48h, and the TLm value(median tolerance limit) were 4.0ppm in mirror carp and flounder, 4.4ppm in rainbow trout, and 10.0ppm in gold fish. After exposure below than TLm value for 1 month, no side effects and histopathological changes were observed. The test drug were shown high antibacterial activities against most bacterial pathogens by exposure in the laboratory and field trials. However, Aeromonas salmonicida showed a resistance to the dosage. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1ppm for 6hr and the test drug is thought to be used effectively and widely as a disinfectant against most bacterial fish pathogens.

Toxicity of the Pufferfish Fugu stictonotus ('Ggachilbog') Collected at a Fish Market of Pusan (시판되고 있는 까칠복(Fugu stictonotus)의 독성)

  • Ten specimens (5 males and 5 females) of the pufferfish, fugu stictonotus ('gachilbog'), were collected at a fish market of Pusan, Korea in July 1993, and examined for anatomical distribution of toxicity by mouse assay method. The frequency of toxic specimens was $40\%\;for\;liver,\;60\%$ for ovary, $40\%\;for\;skin\;and\;60\%$ for bile in female puffers. The highest toxicities were 107, 107, 29 and 93MU/g for liver, ovary, skin and bile, respectively; and average toxicity $\pm S.E.\;values\;were\;14\pm11,\;48\pm22.4\pm3\;and\;12\pm9MU/g,$ respectively. The range of total toxicity was shown to be from 0 to 35,316MU. The characteristic pattern of toxin distribution observed on these specimens was exhibited; both muscle and testis were non-toxic, but others were weakly toxic. Also, there was significant difference for toxicity between male and female specimens.

  • Background: A quantitative structure-activity relationship (QSAR) model was adopted in the Registration, Evaluation, Authorization, and Restriction of Chemicals (REACH, EU) regulations as well as the Act on Registration, Evaluation, etc. of Chemicals (AREC, Republic of Korea). It has been previously used in the registration of chemicals. Objectives: In this study, we investigated the correlation between the predicted data provided by three prediction programs using a QSAR model and actual experimental results (acute fish, daphnia magna toxicity). Through this approach, we aimed to effectively conjecture on the performance and determine the most applicable programs when designating toxic substances through the AREC. Methods: Chemicals that had been registered and evaluated in the Toxic Chemicals Control Act (TCCA, Republic of Korea) were selected for this study. Two prediction programs developed and operated by the U.S. EPA - the Ecological Structure-Activity Relationship (ECOSAR) and Toxicity Estimation Software Tool (T.E.S.T.) models - were utilized along with the TOPKAT (Toxicity Prediction by Komputer Assisted Technology) commercial program. The applicability of these three programs was evaluated according to three parameters: accuracy, sensitivity, and specificity. Results: The prediction analysis on fish and daphnia magna in the three programs showed that the TOPKAT program had better sensitivity than the others. Conclusions: Although the predictive performance of the TOPKAT program when using a single predictive program was found to perform well in toxic substance designation, using a single program involves many restrictions. It is necessary to validate the reliability of predictions by utilizing multiple methods when applying the prediction program to the regulation of chemicals.

The acute toxicity and efficacy evaluation against Aeromonas salmonicida of aquatic drugs oxolinic acid, neomycin-oxytetracycline, and florfenicol in guppy (Poecilia reticulata) (구피(Poecilia reticulata)에서 수산용의약품 oxolinic acid, neomycin-oxytetracycline, florfenicol의 급성독성 및 Aeromonas salmonicida에 대한 약효 평가)

  • This study evaluated the acute toxicity induced by oxolinic acid (OA), neomycin-oxytetracycline combination (NEO-OTC) and florfenicol (FF) administered to guppy (Poecilia reticulata) by comparing standard formulations with commercial preparations (COOaqua curinpotion, COOaqua terafinpotion and COOaqua profenpotion, respectively) for ornamental fish at concentrations of 2-4%. NEO-OTC exhibited the highest acute toxicity in guppy, with no difference observed between the pstandard formulation and commercial preparation (LC50 = 126.08 mg/L and 112.44 mg/L, respectively). OA acute toxicity was assessed in the form of sodium salt, with an LC50 of 504.61 mg/L for the standard formulation and a slightly increased toxicity of 316.11 mg/L for the commercial preparation. In contrast, no mortality was observed during the 96-hour exposure period with the standard formulation in the form of oxolinic acid. The acute toxicity of FF was measured to be above 1,000 mg/L for the standard formulation; however, the commercial preparation significantly increased to 158.53 mg/L. These results indicate that toxicity can significantly increase in commercial formulations, especially those with low levels of active ingredients. This is presumed to be attributed to the organic solvents or solubilizing agents present in the commercial preparations, which may enhance toxicity. Additionally, guppy infected with Aeromonas salmonicida were effectively protected against mortality by administering OA, NEO-OTC and FF at concentrations of 50 mg/L, 100 mg/L and 15 mg/L, respectively, for 2 hours and at half the dose for 24 hours. This result indicates that liquid formulations containing low concentrations of antibiotics may partially increase toxicity, but there is no problem in effectively treating diseases in ornamental fish.

  • The pufferfish Takifugu vermicularis radiatus (Gukmeri-bog) was examined for toxicity. Thirty-one specimens, which had obtained from creeks of Inchon and Ansan in Spring and Autumn of 1992 and 1993, Korea, were assayed for anatomical distribution of toxicity by mouse bioassay method. Ovary, testis, liver and muscle showed very strong toxicity, and intestine and skin did moderate toxicity. Their average toxicities were moderately toxic in all. Moreover, the toxicities of liver and intestine were stronger in Spring than Autumn. Judging from the results, T. vermicularis radiatus is considered to be very harmful fish. Consequently, it is necessary to take a proper preventive step against food poisoning by pufferlish ingestion.

Toxicity of Puffer Fish, Takifugu poecilonotus (Heuinjeombok) and Takifugu vermicularis (Gukmaeribok) from Coastal Water of Korea (연안산 흰점복 (Takifugu poecilonotus)과 국매리복 (Takifugu vermicularis)의 독성)

  • The toxicity of two species of puffer fish, Takifugu poecilonotus (Heuinjeombok) and T. vermicularis (Gukmaeribok) collected from the coastal regions of Korea was determined using a mouse bioassay. In the T. poecilonotus collected in Jeju and Tongyeong, the proportion of toxic specimens containing ${\ge}10$ mouse units (MU) per gram exceeded 95% for the skin, liver, ovary, and fin, and approximately 30% for the testis and muscles. In each of the organs, the highest toxin levels were 79 MU/g in the muscle, hundreds (158-365) of MU per gram in the fin, intestine, testis, and gallbladder, but thousands (1,147-2,406) of MU per gram in the skin, liver, and ovary. In T. vermicularis collected from Incheon and Gunsan, the proportions of toxic specimens were 100% for the gallbladder, and 56-68% for the skin, fin, liver, and intestine however, no toxic muscle specimens were noted. The highest toxin scores were below 10 mouse units (MU) per gram in the muscle, 20-94 MU/g in the skin and fin, 319 MU/g in the intestine, and thousands (1,548-4,624) of MU per gram in the liver, gonad, and gallbladder. The toxicity in the muscle of T. vermicularis was deemed acceptable for human consumption, whereas the toxicities in the muscle of T. poecilonotus and the skin of both species of puffer fish were significantly high, such that special attention may be required when the fish is intended for human consumption.

  • Marine natural products isolated from the sponge Fascaplysinopsis cf reticulata, in French Polynesia, were investigated as an alternative to antibiotics to control pathogens in aquaculture. The overuse of antibiotics in aquaculture is largely considered to be an environmental pollution, because it supports the transfer of antibiotic resistance genes within the aquatic environment. One environmentally friendly alternative to antibiotics is the use of quorum sensing inhibitors (QSIs). Quorum sensing (QS) is a regulatory mechanism in bacteria which control virulence factors through the secretion of autoinducers (AIs), such as acyl-homoserine lactone (AHL) in gram-negative bacteria. Vibrio harveyi QS is controlled through three parallel pathways: HAI-1, AI-2, and CAI-1. Bioassay-guided purification of F. cf reticulata extract was conducted on two bacterial species, i.e., Tenacibaculum maritimum and V. harveyi for antibiotic and QS inhibition bioactivities. Toxicity bioassay of fractions was also evaluated on the freshwater fish Poecilia reticulata and the marine fish Acanthurus triostegus. Cyclohexanic and dichloromethane fractions of F. cf reticulata exhibited QS inhibition on V. harveyi and antibiotic bioactivities on V. harveyi and T. maritimum, respectively. Palauolide (1) and fascaplysin (2) were purified as major molecules from the cyclohexanic and dichloromethane fractions, respectively. Palauolide inhibited QS of V. harveyi through HAI-1 QS pathway at 50 ㎍ ml-1 (26 μM), while fascaplysin affected the bacterial growth of V. harveyi (50 ㎍ ml-1) and T. maritimum (0.25 ㎍). The toxicity of fascaplysin-enriched fraction (FEF) was evaluated and exhibited a toxic effect against fish at 50 ㎍ ml-1. This study demonstrated for the first time the QSI potential of palauolide (1). Future research may assess the toxicity of both the cyclohexanic fraction of the sponge and palauolide (1) on fish, to confirm their potential as alternative to antibiotics in fish farming.

Acute Toxicity of Chlorine Dioxide (ClO2) to Juvenile Black Seabream Acanthopagrus schlegelii and Red Seabream Pagrus major (감성돔(Acanthopagrus schlegelii) 및 참돔(Pagrus major) 치어에 대한 이산화염소(ClO2)의 급성 독성)

  • This study evaluated acute toxicity of chlorine dioxide (ClO2 ) to juveniles of black seabream Acanthopagrus schlegelii (19.4±2.3 g, 10.7±0.4 cm) and red seabream Pagrus major (74.9±8.2 g, 15.9±1.0 cm). Thirty juveniles for each species were exposed to target ClO2 concentrations of 0, 0.05, 0.1, 0.125, 0.15, 0.2, 0.3, 0.4, and 0.5 mg/L in triplicate for eight days. Half lethal concentrations for 96 hours were found at 0.14 and 0.24 mg ClO2/L for black seabream and red seabream, respectively. Red seabream larger than black seabream in body weight appears to be more resistance to chlorine dioxide. However, regardless of species or size, specific loading rates of chlorine dioxide to total fish weight (daily feeding amount of ClO2/total fish weight) were similar, showing 1.3 and 1.1 g ClO2/kg fish·day-1 for black seabream and red seabream.

  • This study was initiated to understand the mechanism of selective toxicity of diazinon and carbofuran to killifish and loach. Conventional LC50 was calculated from fish test. IC50 with acetylcholinesterase activity was estimated using whole body and wet brain homogenate of the two fish species. Acetylcholinesterase activity of killifish was approximately twice as high as that of loach. The selective toxicity of diazinon to killifish and loach was partly (14 : 4) explained by the IC50 of diazoxon, a toxic metabolite of diazinon. IC50 of carbofuran also partly (14 : 3.4) contributed to the selectivity. These result suggested that the enzymatic method might be utilized as a screening tool for the chemicals affecting fish species of environmental concern with certain limitations which should be overcome in future studies.

Acute and subacute toxicity of trichlorfon in guppies (Poecilia reticulata)

  • The aim of the present study was to determine the acute and subacute potential of trichlorfon in guppies (Poecilia reticulatus). We first defined the 24 h median tolerance limit ($TLm_{24h}$) of the fish to trichlorfon. Guppies were then treated with $TLm_{24h}$ and 1/10 $TLm_{24h}$ trichlorfon concentrations to evaluate if any histological alterations occurred. The $TLm_{24h}$ value of trichlorfon was 11 ppm. This concentration resulted in acute toxicity to the gills and kidneys with edema, hyperplasia of the gill lamellae, and vacuolated degeneration and necrosis of renal tubular cells. In the case of subacute toxicity using a 10-fold dilution of the $TLm_{24h}$ value (1.1 ppm), no behavioral changes, external lesions or histopathological changes were observed. Therefore, safe concentration of trichlorfon might be 1.1 ppm in guppy for controlling parasitic infections.
