어류의 세균성 질병 예방을 위한 Probiotic균주의 선발 및 특성 (Screening and Characterization of Probiotic Strains for Prevention of Bacterial Fish Diseases)

  • 본 실험의 목적은 Gram 양성균 뿐만 아니라 어병세균인 Gram 음성세균에 대한 넓은 항균활성 및 내산성 그리고 항생제 내성을 갖고 있는 유용한 probiotic 후보균주를 선발하여 어류의 세균성 질병 예방 혹은 치료를 함으로써 양식산업의 효율성을 증대시키고자 한다. 20종의 유산균을 김치, 양식 넙치의 장 그리고 각종 젓갈류 등에서 분리하여 어류병원성 세균에 대한 항균능을 MRS agar상에서 agar spotted method에 의해 조사하여 Vibrio anguillarum, Edwardsiella tarda, 그리고 Streptococcus sp.에 대해 항균활성이 있는 7종의 균주를 1차 선발을 하였다. pH 및 항생제 내성능을 검토하여 probiotic 균주를 분리하였다. 분리된 후보 균중 창란젓에서 분리된 BK19에서 가장 큰 항균 spectrum과 pH 내성 그리고 항생제에 대한 내성이 확인되어 최종적으로 본 실험의 공시균주로 선발하여 생화학적, 형태학적 특성 및 16S rRNA 분석을 통하여 Lactobacillus sakei BK19로 동정하였다.

Metabolite profiling of fermented ginseng extracts by gas chromatography mass spectrometry

  • Background: Ginseng contains many small metabolites such as amino acids, fatty acids, carbohydrates, and ginsenosides. However, little is known about the relationships between microorganisms and metabolites during the entire ginseng fermentation process. We investigated metabolic changes during ginseng fermentation according to the inoculation of food-compatible microorganisms. Methods: Gas chromatography mass spectrometry (GC-MS) datasets coupled with the multivariate statistical method for the purpose of latent-information extraction and sample classification were used for the evaluation of ginseng fermentation. Four different starter cultures (Saccharomyces bayanus, Bacillus subtilis, Lactobacillus plantarum, and Leuconostoc mesenteroide) were used for the ginseng extract fermentation. Results: The principal component analysis score plot and heat map showed a clear separation between ginseng extracts fermented with S. bayanus and other strains. The highest levels of fructose, maltose, and galactose in the ginseng extracts were found in ginseng extracts fermented with B. subtilis. The levels of succinic acid and malic acid in the ginseng extract fermented with S. bayanus as well as the levels of lactic acid, malonic acid, and hydroxypruvic acid in the ginseng extract fermented with lactic acid bacteria (L. plantarum and L. mesenteroide) were the highest. In the results of taste features analysis using an electronic tongue, the ginseng extracts fermented with lactic acid bacteria were significantly distinguished from other groups by a high index of sour taste probably due to high lactic acid contents. Conclusion: These results suggest that a metabolomics approach based on GC-MS can be a useful tool to understand ginseng fermentation and evaluate the fermentative characteristics of starter cultures.

Bioconversion of Soybean Curd Residues into Functional Ingredients with Probiotics

  • Soybean curd residues (SCR) obtained from hot and cold manufacturing processes were fermented by indigenous microorganisms, Lactobacillus rhamnosus LS and Bacillus firmus NA-l for 15 h at 37$^{\circ}C$. The pH, acidity, viable cell counts, and tyrosine content were evaluated in samples with variations in sugar, starter and type of SCR. The raw Doowon SCR (D-SCR, cold-processed) fermented by indigenous microorganism had a 0.9% acidity and 6.7 ${\times}$ 10$^{7}$ CFU/g viable cell counts, compared with the 0.11 % acidity and 6.7 ${\times}$ 10$^{6}$ CFU/g viable cell counts of raw fermented Pulmuwon SCR (P-SCR, hot-processed). After fermentation of raw P-SCR with 1 % glucose and 1 % L. rhamnosus LS starter, the viable cell counts, tyrosine content and acidity were 4.7 ${\times}$ 10$^{8}$ CFU/g, 16.3 mg% and 0.9%, respectively. In addition, the raw P-SCR fermented with Bacillus firmus NA-l as co-starter had a 0.45% acidity, 2.4 ${\times}$ 10$^{8}$ CFU/g lactic acid bacteria, and 3.3 ${\times}$ 10$^{6}$ CFU/g Bacillus sp. In particular, the tyrosine content was increased 5 fold. The drying of fermented SCR was completed by hot-air drying (5$0^{\circ}C$) within 12 h; the dried P-SCR and D-SCR had 1.8 ${\times}$ 10$^{7}$ CFU/g and 5.3 ${\times}$ 10$^{6}$ CFU/g viable cell counts, respectively. The concentrate of methanol extract from fermented D-SCR inhibited the initial cell growth of E. coli, Staphylococcus aureus and Pseudomonas aeruginosa in liquid culture.

Characteristics of Solid-state Fermented Feed and its Effects on Performance and Nutrient Digestibility in Growing-finishing Pigs

  • This study investigated the effects of solid-state fermentation of a compound pig feed on its microbial and nutritional characteristics as well as on pig performance and nutrient digestibility. A mixed culture containing Lactobacillus fermentum, Saccharomyces cerevisae and Bacillus subtilis was used for solid-state fermentation and solid-state fermented feed samples were collected on days 0, 1, 2, 3, 5, 7, 10, 15, 20 and 30 for microbial counts and chemical analysis. Lactic acid bacteria increased rapidly during the first three days of fermentation and then slowly declined until day 10 and, thereafter, the counts were maintained at about 6.7 log cfu/g for the duration of the fermentation period. Enterobacteria also increased during the first two days, and then fell below the detectable level of the analysis (3.0 log cfu/g). The pH of the fermentation substrate declined from 6.1 at the start of fermentation to 5.7 by day 30. The water-soluble protein content increased from 8.2 to 9.2% while the concentration of acetic acid increased from 16.6 to 51.3 mmol/kg over the 30-day fermentation. At the end of the 30-day fermentation, the solid-state fermented feed was used in a pig feeding trial to determine its effects on performance and nutrient digestibility in growing-finishing pigs. Twenty crossbred barrows ($14.11{\pm}0.77kg\;BW$) were allotted into two dietary treatments, which comprised a regular dry diet containing antibiotics and a solid-state fermented feed based diet, free of antibiotics. There was no difference due to diet on pig performance or nutrient digestibility. In conclusion, solid-state fermentation resulted in high counts of lactic acid bacteria and low counts of enterobacteria in the substrate. Moreover, feeding a diet containing solid-state fermented feed, free of antibiotics, can result in similar performance and nutrient digestibility in growing-finishing pigs to a regular diet with antibiotics.

발효소시지 제조에 적합한 스타터 선발 (Screening of Lactic Acid Bacteria as a Starter Culture in Fermented Sausage)

  • 본 연구에서는 전통발효식품 13종과 천연물 4종에서 27균주를 분리한 후 생육속도가 빠르고 pH 저하 능력이 우수한 3종의 균주를 1차 선발하였으며, 분자생물학적 방법을 이용하여 동정한 결과 Staphylococcus warneri, Sta. epidermidis, Lactobacillus plantarum과 99% 상동성을 보였다. 선발된 3균주의 발효소시지 스타터로서 이용가능성을 알아보기 위해 발효소시지의 환경과 유사한 model-system에서 배양하며 pH 저하 능력, 총산 생성 능력, 생육 능력, 아질산염 소거 능력을 발효소시지의 스타터로 많이 사용되고 있는 5균주 및 상업용 2균주와 비교하였다. Modelsystem에서 pH 저하능과 총산 생성능, 생육능은 관련성이 있었으며, 상업용 균주보다 분리한 Sta. epidermidis DO 10-1, Lac. plantarum MLK 14-2가 우수한 결과를 나타내었다. 아질산염 소거능의 경우에도 분리한 3균주가 상업용 균주보다 상대적으로 빠른 속도를 보였다. 분리한 3균주는 발효소지지 스타터로서의 발효 소거 능력은 우수할 것으로 보이지만 Staphylococcus는 잠재적인 위험성이 제기되는 균주이므로 Lac. plantarum이 발효소지지 제조에 가장 적합할 것으로 사료된다.

Effects of fermented feed on growth performance, nutrient metabolism and cecal microflora of broilers

  • Objective: To investigate the effects of enzyme-bacteria co-fermented feed on broilers, the basal diet (BF) was pretreated by microbial enzyme co-fermentation, and then different proportions of BF were replaced to study its effects on growth performance, nutrient metabolism and cecal microflora of broilers. Methods: Four hundred and eighty 1-day-old broilers were randomly divided into 6 groups. The control group was fed with BF, and groups 1 to 4 were treated with dried fermented feed (DFF) instead of 10%, 15%, 20%, and 25% the BF, and group 5 was treated with wet fermented feed (WFF) instead of 10% the BF, named BF, 10% DFF, 15% DFF, 20% DFF, 25% DFF, and 10% WFF, respectively. The trial period was 42 days. Results: The results showed that the average daily feed intake and average daily gain of 10% DFF, 15% DFF, and 10% WFF groups were significantly higher than those of the control group at 22 to 42 days and 1 to 42 days (p<0.05). Except for 10% DFF group, Firmicutes of all treatment were higher than that of control group. The Bacteroides of each treatment group were lower than that of the control group (p>0.05). At the same time, the nutrient apparent metabolic rate and cecal microbial abundance of each treatment group had an increasing trend (p>0.05). Conclusion: In conclusion, the feed fermented by enzyme and bacteria had a potential promoting effect on the growth performance and nutrient digestibility of broilers.

유산균들의 콜레스테롤 저하성, 내산성, 내담즙성, 항생제 내성 비교

  • For a probiotic yoghurt it is desirable to utilize lactic acid bacteria with a high survival rate and beneficial function to human beings. We have examined a variety of lactic acid bacteria to assess the acid and bile tolerance and antibiotic resistance. In addition, an in vitro culture experiment was performed to evaluate their ability to reduce cholesterol levels in the growth medium. Thirteen strains were selected from in vitro cholesterol assays and fed to Sprague-Dawley rats with a high-cholesterol diet. Among the 13 strains tested, 8 strains were shown to reduce serum cholesterol levels significantly after 24 days of administration in vivo. Rats were fed lyophilized yoghurt powder fermented with a combination of 3 selected strains: Bifidobacterium infantis AM-220, Lactobacillus AM-245, and Styeptococcus MA-1. The levels of total cholesterol and low density lipoprotein were significantly lower (p$\leq$0.05) in rats fed the yoghurt powder compared with control group. These studies suggest that yoghurt fermented with appropriately selected lactic acid bacteria may have a anticholesterolemic effect.

김치 젖산균과 효모의 혼합배양 방법에 의한 과채류즙의 발효양상 (Fermentation Aspects of Fruit-Vegetable Juice by Mixed Cultures of Lactic Acid Bacteria Isolated from Kimchi and Yeast)

  • Fermented beverage using lactic acid bacteria isolated from kimchi was investigated. Lactic acid bacteria KL 1, KD 6, KL 4 strains from kimchi, or obtained Lactobacillus acidophilus, Lactobacillus plantarum, Leuconostoc mesenteroides with and without yeast(Saccharomyces cerevisiae) were inoculated in fruit vegetable juice for single and mixed culture fermentation. During the fermentation by bacterial strain and yeast for 1~3 days at 30oC, various fermentation behaviors were observed. The growth rate of mixed culture of KL 1 and yeast was higher than that of single culture by KL 1 alone during the fermentation. The amount of organic acid produced by the mixed culture fermentation of KL 1 and yeast was 0.82%(3 day) or 0.58%(1 day) and with the final pH of 3.3(3 day) or 4.2(1 day). These mixed culture systems of isolated strains or other bacterial strains had almost similar results of growth rate and acid production. Among several bacterial strains, KL 1 was suitable for the mixed culture fermentation with yeast in terms of desirable fermentation behavior and organoleptical quality. The selected strain, KL 1 was identified as Leuconostoc spp. through the series of tests on carbohydrate fermentation and biochemical characteristics.

산소와 염농도가 한국전통 발효식품에서 생장하는 혐기성 세균과 Bifidus균의 생육에 미치는 영향 (Effects of Oxygen and Salt on the Growth of Bifidus and Anaerobic Bacteria Isolated from Korean Traditional Fermented Foods)

  • 한국 전통 발효식품의 숙성에 절대 혐기성 세균이 관여하는지를 보기 위해 김치류와 장류 14종의 시료를 복합 BL 배지 혹은 bifidus 세균 선택 배지인 BS agar에 접종하여 절대 혐기적으로 배양한 결과 복합 BL 배지에서 $10^7$ ~ $10^8$/g 시료의 세균이 생장하였으며, BS agar에서는 $10^3$ ~ $10^6$/g 시료의 colony가 형성되었다. 이들은 모두 호기성 조건에서도 생장 할 수 잇었으며, BS 배지에서 생장하는 세균은 fructose-6-phosphate phosphoketolase를 합성하지 않는 것으로 나타났다. 또한 표준 Bifidobacterium속 세균은 3% NaCl을 함유하는 배지에서는 생장할 수 없었다. 이상의 결과로부터 김치류와 장류 등 염 농도가 높은 발표식품에는 bifidus균은 생장할 수 없다고 판단된다.

Effects of Lactic Acid Bacteria Inoculant on Fermentation Quality and in vitro Rumen Fermentation of Total Mixed Ration

  • Fermented total mixed ration (TMR) is a novel feed for ruminants in South Korea. The purpose of this study was to evaluate the effects of lactic acid bacteria (LAB) on the quality of TMR and in vitro ruminal fermentation. Strains of three LAB spp. (Lactobacillus plantarum, L. brevis, L. mucosae) were used in fermentation of TMR. Inoculations with the three LAB spp. lowered pH and increased concentrations of lactic acid, acetic acid, and total organic acid compared to non-LAB inoculated control (only addition of an equivalent amount of water) (p<0.05). Bacterial composition indicated that aerobic bacteria and LAB were higher. However, E. coli were lower in the fermented TMR than those in the control treatment (p<0.05). Among the treatments, L. brevis treatment had the highest concentration of total organic acid without fungus detection. Gas production, pH, and ammonia-nitrogen during ruminal in vitro incubation did not differ throughout incubation. However, ruminal total VFA concentration was higher (p<0.05) in the LAB spp. treatments than the control treatment at 48 hours. Overall, the use of L. brevis as an inoculant for fermentation of high moisture. TMR could inhibit fungi growth and promote lactic fermentation, and enhance digestion in the rumen.
