Antioxidant activities of ethanol extracts from barley sprouts (새싹보리 에탄올 농도별 추출물의 산화방지 활성)

  • Phenolic compounds and antioxidant activities of ethanol extracts from barley sprouts were evaluated in this study. Barley sprouts were extracted using water and ethanol in various concentration (25, 50, and 75%) using reflux extraction methods. Ultra performance liquid chromatography (UPLC) analysis showed that barley sprouts are mainly composed of rutin, gallic acid, ferulic acid, and ${\rho}$-coumaric acid. The 75% ethanol extracts had higher total polyphenol contents ($44.01{\pm}1.32mg/g$) and total flavonoid contents ($102.96{\pm}2.49mg/g$). 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity ($EC_{50}$ value: $1.65{\pm}0.02mg/mL$) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activity ($EC_{50}$ value: $1.67{\pm}0.02mg/mL$) of the 75% ethanol extracts of barley sprouts were found to be the most effective. The 75% ethanol extracts of barley sprouts exhibited a strong reducing activity and ferric reducing antioxidant activity. As a result, the 75% ethanol extracts of barley sprouts showed stronger antioxidant activity than other extracts.

  • Objectives : This study is to effect of improving muscle atrophy through water extract on the solid-phase fermentation extraction with Phellinus linteus of discarded Schisandra chinensis in an atorvastatin-induced atrophy C2C12 cell. Methods : C2C12 myoblast were differentiated into myotube by 2% horse serum medium for 6 days, and then treated solid-phase fermentation(S-P) extract at different concentrations for 24h. To investigate the effect of S-P extract on the induction of muscle atrophy and expression of atrophy-related genes and apoptosis in differentiated C2C12 myotubes using a GSH, ROS, real-time PCR, western blots analysis. Results : As a result of treatment with atorvastatin at concentrations of 5, 10, and 20 uM on the 6th day of differentiation in C2C12 myotube cells, it was confirmed that the cell morphology was damaged in a concentration-dependent manner, and the length and thickness of the myotube also decreased in a concentration-dependent manner. Treatment with S-P extract (50, 100 and 200 ㎍/㎖) increased of GSH and inhibited ROS in the atorvastatin-induced muscle atrophy cell model at a concentration that did not induce toxicity. In addition, it was confirmed that it has an effect on muscle reduction by inhibiting apoptosis of muscle cells as well as being involved in protein production and degradation of muscle cells. Conclusions : Atorvastatin-induced atrophy C2C12 cell, S-P extract activates related to differentiation/generation and proteolysis, and inhibits cell death of atrophy in C2C12 cell. Based on this, it is necessary to prove its effectiveness through animal models and human application test, but it is considered to be discarded Schisandra chinensis can present the potential for development as a recycling industrial material.

Radiologic evaluation and survival rate of taper-straight designed implant with SLA surface: A 1-year follow-up retrospective study (SLA surface를 가진 taper straight designed implant의 방사선학적 평가와 생존율: 1년 추적 관찰을 통한 후향적 연구)

  • Purpose. This study was conducted to evaluate clinical usefulness by evaluating the marginal bone resorption and survival rate of an implant with a taper straight type SLA surface domestically available on the market recently. Materials and methods. 40 implants satisfying the including criteria were observed for one year of 125 implants of 83 adult men and women who had KISPLANT® implanted from August 2016 to December 2019 at the Department of Periodontology, Chonnam National University Dental Hospital. The marginal bone level was measured on periapical radiographs taken initially and 1 year later and we analyzed implant survival and success rates. A t-test was used for the analysis of the association between the marginal bone resorption and the severity of periodontitis, supportive periodontal therapy, the inserted site, the cause of extraction, immediate placement, and systemic disease. Results. After 1 year of loading, the mesial bone resorption was 0.74 ± 1.07 mm, and the distal bone resorption was 0.53 ± 1.04 mm. The marginal bone resorption of 2 mm or more occurred at 3 implants out of 40 implants after 1 year, so the success rate of implant was 92.5% and survival rate was 100%. There was a significant difference in mesial marginal bone resorption according to the inserted site and no significant differences were found between marginal bone resorption and the other factors. Conclusion. As a result of marginal bone resorption, success rate, and survival rate in this study, we found little marginal bone resorption and high survival and success rate. It can be concluded that they represent excellent clinical results.

Comparison of Antioxidant, Wrinkle Improvement, and Whitening Efficacies of Extracts from Pinus koraiensis Cone Scale Using Extraction Methods (추출방법에 따른 잣나무(Pinus koraiensis) 구과피 추출물의 항산화, 주름개선 및 미백 효능 비교)

  • The present study evaluated the antioxidant activity, skin wrinkle reduction, and whitening activity of Pinus koraiensiscone scale extracts made with three different solvents (ethanol, supercritical fluid, and a mixture of both). Total polyphenol content was 11.03 mg/g GAE in the supercritical fluid extractand 33.79 mg/g GAE in the 70% ethanol extract. Electron donating ability of 1,000 ㎍/mL extract was 13.60% in the supercritical fluid extract, 91.37% in 70% ethanol extract, and 71.62% in mixed solvent extract. SOD-like activities in 100 ㎍/mL extract using supercritical fluid, 70% ethanol, and mixed solvents were 16.49%, 21.84%, and 10.7%, respectively. The ABTS+ radical scavenging activities of 1,000 ㎍/mL extract were 38.19%, 80.23%, and 78.72%for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Tyrosinase inhibitory activities in 1,000 ㎍/mL extract were 24.54%, 36.55%, and 15.23% for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Elastase inhibitory activities in 1,000 ㎍/mL extract were 15.62%, 22.56%, and 26.64% for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Skin astringent activity (81.23% with 5000 ㎍/mL extract) was only detected in the 70% ethanol extract. Supercritical fluid and mixed solvent extracts showed no such activity. Our analysis of Pinus koraiensiscone scale extracts show that the highest aggregate activity was found in the 70% ethanol extract, followed by mixed solvent and supercritical fluid extracts. Therefore, our results oppose the hypothesis stating supercritical extract has the highest total polyphenol content and antioxidant activity.

  • Analysis of research trends in specific subject areas is performed by examining related topics and subject changes by using topic modeling techniques through keyword extraction for most of the literature information (paper, patents, etc.). Unlike existing research methods, this paper extracts topics related to the research topic using the LDA topic modeling technique for the project information of national R&D projects provided by the National Science and Technology Knowledge Information Service (NTIS) in the field of artificial intelligence. By analyzing these topics, this study aims to analyze research topics and investment directions for national R&D projects. NTIS provides a vast amount of national R&D information, from information on tasks carried out through national R&D projects to research results (thesis, patents, etc.) generated through research. In this paper, the search results were confirmed by performing artificial intelligence keywords and related classification searches in NTIS integrated search, and basic data was constructed by downloading the latest three-year project information. Using the LDA topic modeling library provided by Python, related topics and keywords were extracted and analyzed for basic data (research goals, research content, expected effects, keywords, etc.) to derive insights on the direction of research investment.

A Study on the Estimation of Multi-Object Social Distancing Using Stereo Vision and AlphaPose (Stereo Vision과 AlphaPose를 이용한 다중 객체 거리 추정 방법에 관한 연구)

  • Recently, We are carrying out a policy of physical distancing of at least 1m from each other to prevent the spreading of COVID-19 disease in public places. In this paper, we propose a method for measuring distances between people in real time and an automation system that recognizes objects that are within 1 meter of each other from stereo images acquired by drones or CCTVs according to the estimated distance. A problem with existing methods used to estimate distances between multiple objects is that they do not obtain three-dimensional information of objects using only one CCTV. his is because three-dimensional information is necessary to measure distances between people when they are right next to each other or overlap in two dimensional image. Furthermore, they use only the Bounding Box information to obtain the exact coordinates of human existence. Therefore, in this paper, to obtain the exact two-dimensional coordinate value in which a person exists, we extract a person's key point to detect the location, convert it to a three-dimensional coordinate value using Stereo Vision and Camera Calibration, and estimate the Euclidean distance between people. As a result of performing an experiment for estimating the accuracy of 3D coordinates and the distance between objects (persons), the average error within 0.098m was shown in the estimation of the distance between multiple people within 1m.

  • Background: Research on next-generation sequencing (NGS)-based HLA typing is active. To resolve the phase ambiguity and long turn-around-time of conventional high resolution HLA typing, this study developed a NGS-based high resolution HLA typing method that can handle large-scale samples within an efficient testing time. Methods: For HLA NGS, the condition of nucleic acid extraction, library construction, PCR mechanism, and HLA typing with bioinformatics were developed. To confirm the accuracy of the NGS-based HLA typing method, the results of 192 samples HLA typed by SSOP and 28 samples typed by SBT compared to NGS-based HLA-A, -B and -DR typing. Results: DNA library construction through two-step PCR, NGS sequencing with MiSeq (Illumina Inc., San Diego, USA), and the data analysis platform were established. NGS-based HLA typing results were compatible with known HLA types from 220 blood samples. Conclusion: The NSG-based HLA typing method could handle large volume samples with high-throughput. Therefore, it would be useful for HLA typing of bone marrow donation volunteers.

Physicochemical properties and biological activity of three-year-old and seven-year-old Platycodon grandiflorum extracts (3년근 도라지와 7년근 도라지의 이화학 특성 및 생리활성)

  • This study was designed to evaluate the physicochemical properties and biological activities (antioxidant activity and antimicrobial activities) of three and seven-year-old Platycodon grandiflorum extracts. Three and seven-year-old Platycodon grandiflorum contained crude saponins, free amino acids and minerals. Water extracts of the three and seven-year-old plants were prepared using reflux extraction methods. The total polyphenol contents (TPC), total flavonoid contents (TFC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and antimicrobial effects of the extracts were determined. The seven-year-old Platycodon grandiflorum extract had higher TPC ($5.08{\pm}0.07TAEmg/g$) and TFC ($3.80{\pm}0.07QUEmg/g$). DPPH radical scavenging activity ($IC_{50}$ value: $288{\pm}3.88{\mu}g/mL$) and ABTS radical scavenging activity ($IC_{50}$ value: $568{\pm}2.09{\mu}g/mL$). The three and seven-year-old Platycodon grandiflorum extracts exhibited a strong antimicrobial effect against three kinds of bronchus disease-inducing bacteria; the seven-year-old Platycodon grandiflorum extracts showed a stronger antimicrobial effect than the three-year-old Platycodon grandiflorum extracts.

Quantitative Determination of Marker Compounds in the Extracts of Camellia sinensis L. Sub-branches (Residual Products) by HPLC (HPLC에 의한 차나무 잔가지(부산물)의 추출물 내 지표 성분의 정량분석)

  • Background: Camellia sinensis L.(CS) is a perennial evergreen species of plant whose leaves are used to produce tea. In this plant species, the parts used are the leaves, sub-branch parts are thrown out. Methods and Results: Ethanol extract of sub-branch parts was used for isolation of major compounds by column chromatography. Structures were identified as caffeine (1), (-)-epicatechin (2) and (-)-epicatechin gallate (3) by interpretation of spectroscopic analysis, including $^1H$- and $^{13}C$-NMR. High-performance liquid chromatography (HPLC) method was used to compare the quantitative level of marker compounds in various extraction solvents of sub-branch parts of CS. The content of caffeine, (-)-epicatechin, and (-)-epicatechin gallate in 30% ethanol extract showed higher value with $3.28{\pm}0.57mg/g$, $5.53{\pm}0.88mg/g$, and $1.29{\pm}0.24mg/g$, respectively. Conclusions: These results indicated that not only leaves parts but also sub-branch, could be a good source for the functional material and pharmaceutical industry.

Automatic Text Summarization based on Selective Copy mechanism against for Addressing OOV (미등록 어휘에 대한 선택적 복사를 적용한 문서 자동요약)

  • Automatic text summarization is a process of shortening a text document by either extraction or abstraction. The abstraction approach inspired by deep learning methods scaling to a large amount of document is applied in recent work. Abstractive text summarization involves utilizing pre-generated word embedding information. Low-frequent but salient words such as terminologies are seldom included to dictionaries, that are so called, out-of-vocabulary(OOV) problems. OOV deteriorates the performance of Encoder-Decoder model in neural network. In order to address OOV words in abstractive text summarization, we propose a copy mechanism to facilitate copying new words in the target document and generating summary sentences. Different from the previous studies, the proposed approach combines accurate pointing information and selective copy mechanism based on bidirectional RNN and bidirectional LSTM. In addition, neural network gate model to estimate the generation probability and the loss function to optimize the entire abstraction model has been applied. The dataset has been constructed from the collection of abstractions and titles of journal articles. Experimental results demonstrate that both ROUGE-1 (based on word recall) and ROUGE-L (employed longest common subsequence) of the proposed Encoding-Decoding model have been improved to 47.01 and 29.55, respectively.
