The study was carried out to establish an improved protocol for shoot organogenesis and plant regeneration from leaf explant cultures of Scrophularia buergeriana M. with the treatment of ethylene inhibitors [silver nitrate (AgNO$_3$), aminoethox-yvinylglycine (AVG), Cobalt chloride (CoCl$_2$)]. The regenerated shoots obtained from leaf explant cultures on MS medium containing 2 mg/l BAP, The additions of AgNO$_3$. AVG and CoC1$_2$ substantially improved the shoot regeneration frequency, at the optimal concentration of 7 mg/L, 7 mg/L, and 3 mg/L respectively, The regenerated shoots could be easily rooted with 0.1 mg/L IBA treatment. The noted plants were hardened and transferred to vermiculite with a 85% survival rate where they grew normally.
Adventitious buds appeared within 2 weeks on the base of the petiole explants and increased for two months. A maximum of regeneration (15.6%) was obtained on the medium containing $1.5\;{\mu}M$ TDZ in combination with $1\;{\mu}M$ IBA. To know which explants are the best for the induction of regeneration, three explants such as leaf, petiole and leaf-petiole were used. Among the explant types, the leaf-petiole explant was significantly more effective than leaf and petiole for promoting adventitious shoots, with leaf-petiole inducing at the highest regeneration frequency (33.7%). The regeneration frequency of adventitious shoots in the leaf-petiole explants was significantly affected by leaf size and the position of explants. The leaf-petiole smaller than 5 mm leaf in width was induced at the highest regeneration frequency (68.9%). The smaller leaf sizes, the greater regeneration frequency. Also when the leaves are nearer to the shoot tip, the regeneration frequency is higher. When the rooted micro-shoots were transferred to the soil after growing for 6 weeks in the media, the survival rate was 90%.
An efficient procedure was developed for adventitious shoot bud induction and plantlet regeneration from various explants of the ten genotypes of Pepper (Capsicum annuum L.) using Thidiazuron (TDZ). Among various treatments at 1.0-3.0 mg/L TDZ Induced maximum number of adventitious shoots depending upon the explant type and genotype compared to other treatments. Among the explants tested, leaf induced maximum number of adventitious shoots than the cotyledons. TDZ-mediated organo-genesis was possible in 10 pepper cultivars, the extent of the response being genotype-dependent. Of the ten genotypes tested, C. annuum cvs CA960, $G_4$ and X-235 were produced maximum number of adventitious shoots and Sell was the least, and all other genotypes gave moderate response. Elongation of multiple shoots was observed on medium supplemented with SA (0.05 mg/L) in combination of IAA (0.05 mg/L). Differences in ability for in vitro shoot regeneration and elongation depend upon the variety and explant type. The elongated shoots were success. Fully rooted on MS medium containing at 1.0 mG/L IAA. Plantlets regenerated from different explants of ten genotypes were found to be diploid (2n=24) and were devoid of any chromosomal aberrations. Regenerated plants were successfully established in soil where 85-90% of them developed into morphologically normal and fertile plants.

In order to develop an efficient micropropagation protocal for Eucalyptus pellita, on in vitro culture system has been was established by inducing axillary buds from greenhouse stock materials. Among 6different media tested, DKW medium was the best ot induce bast induce both shoot proliferation and growth. Average number of proliferated shoots of 403per explant was obtained at the concentration of 0.1mg/LBA. Most of the stem materials excreted phenolic compounds at the proximal part of the explant and caused darking of the media. Therefore, it was necessary to transfer frequently to a fresh medium and/or to add activated charcoal at the concentration of 0.02%(w/v). Generally on vitro roots were formed easily on 1/2DKW medium with NAA treatment. All the explants rooted at the medium containing 0.2mg/L NAA and displayed vigorous root growth in vitro culture conditions. After transferred to an artificial soil mixture (peatmoss: vermiculrite: perlite, 1:1:1, v/v/v) in the greenhouse, most rooted plantlets survived well without any morphological abnormalities. The results show that the species can be micropropagated effectively by the application of axillary bud culture system.
천궁 (Cnidium officinale)의 잎, 엽병 및 화기 외식편을 재료로 하여 2,4-D가 첨가된 배지에 배양하였을때 캘러스 형성은 어느 외식편에서나 비슷한 수준으로 유기되었지만 배형성능 캘러스는 화기유래 캘러스에서만 형성되었다. 체세포배발생의 유도는 2mg/L 2,4-D를 첨가한 MS배지에서 잘 일어났으며 2,4-D제거배지에서 계대배양하면 외식편당 78개의 체세포배가 형성되었다. 체세포배의 자엽수에 있어서 변이가 빈번하게 관찰되었으나 배지에 1% 활성탄을 첨가하면 비정상적인 자엽수를 갖는 체세포배의 발생률이 감소되는 효과를 얻을 수 있었다. 자엽기 체세포배는 MS 기본배지에서 저조한 발아율을 보이는 반면에 활성탄 첨가배지에서는 현저한 발아 향상을 보였다.
This study was conducted to establish the optimal condition for conservation of genetic resources and micropropagation of Lilium cernum. Induction of bulbet of L. cernum was highly effective (9.2 bulb/explant) on 1/2 SH (Schenk and Hildebrandt) medium supplemented with 1.0 mg/L TDZ (Thidiazuron) and 0.1 mg/L NAA (Naphthaleneacetic acid). The treatment of 0.1 mg/L NAA increased root development (6.4 root/explant) under the in vitro condition. In addition, treatments of AC (Activated Charcoal) and ventilation were enhanced to develop number of shoots and to elongate length of leaf, bulb and root. Futhermore, the process of short-term soil acclimatization was promoted to strengthen the plantlets induced under the in vitro condition.
This study was carried out to investigate the growth characteristics and propagation methods of L. obtusiloba as a woody landscape plant. The results are obtained as follows: L. obtusiloba was dioecious shrub and shaped with bush type. Leaves were ovate, obtuse, cordate, 7.05 cm long, 7.20 cm wide, and petiole 2.0 cm long. Flowers of L. obtusiloba were diclinous. Soil acidity ranged from pH 4.06 to 5.53 with the lowest at the Mt. Soo-Ri. Mt. Soo-Ri located near factory district, which was considered to damaged by environmental deteoration. While soil organic matter was highest at Mt. Soo-Ri, inorganic nutrients were low. L. obtusiloba grows in the area with low soil acidity and low content of inorganic. Therefore it seemed to be tolerant to air pollution. L. Obtusiloba was high seed germination rate in the plug box and its shoots were longer than seeding box and softwood cutting of L. obtusiloba showed the rooting rate of 50% at 5,000ppm on June 23. To develop a mass propagation method of Korean native L. obtusiloba through an axillary bud culture as a woody landscape plant, about 2∼3 cm shoots induced from explant were subcultured to new media contained different growth regulators. Shoots multiplied most effectively on a WPM containing 1.0 mg/l BA, producing 5.5 shoots with a shoot length of 2.5 cm per shoot explant.
Serratia marcescens (S. marcescens) emerged as an opportunist in the setting of immunodeficiency in the 1970s, when serious infections occurred in San Francisco hospitals after USA. Navy experiments had aerosolized the bacteria to study biologic warfare. We investigate the risks of S. marcescens in San Franciscans who undergo mastectomy with implant reconstruction. From 2007 to 2011, the senior author took breast capsule cultures for all patients at the time of tissue expander exchange/explant. Of the 142 women who had reconstruction, 23 had positive cultures. Only the two patients who were positive for S. marcescens developed clinical infections that required explantation. Both had postoperative chemotherapy with transient neutropenia, and both had close ties to San Francisco. Clinical signs of infection emerged for both patients months after initial surgery, despite having previously well healed incisions. Other patients were culture positive for Pseudomonas, Proteus, Enterococcus and MRSA and did not develop require explant. While the link between San Francisco and S. marcescens is controversial, a patient's geography is a simple screening tool when considering postoperative risks, especially in the immunocompromised. Closer monitoring for neutropenia during chemotherapy, and a lower threshold to administer S. marcescens targeted antibiotics may be warranted in these patients.
Protocorms were newly formed from the culture of axillary buds, obtained in the seedlings of Dendrobium moniliforme in vitro. Its formation ratio was calculated to 43.7% on MS medium containing 1.0 mg/L BA. To test their survival ratio, we gradually increased the inoculation of transplant populations from single to more than three, and then found that the ratio in three populations went up as high as 95.2% rather than those of one or two. In bioreactor, explant obtained from the axillary bud grew well in lower concentration as 1/4 MS medium, while clearly grew slow in a little bit high concentration as 1/2 MS medium. We found that the explant of axillary bud, obtained from the Dendrobium moniliforme seedlings, would grow five times after culturing in a bioreactor for six weeks in 1/4 MS medium.
