Vasodilatory Effect of the Fractions from Crataegus pinnatifida and Isolation of the Active Component (산사 분획의 혈관이완 활성과 성분분석에 관한 연구)

  • This study was performed for the investigation of vasodilatory effects of Crataegus pinnatifida Bunge and for isolation and structure determination of the constituent from the active fraction. The fruits of this herbal drug were extracted with 80% methanol, then fractioned successively with methylene chloride, ethylacetate and n-butanol. Among the fractions, ethyl acetate fraction exhibited the most effective vascular relaxation against phenylephrine-induced arterial contraction. In order to isolate the active constituent by activity-guided fractionation, this fraction was chromatographed on silica gel to yield seven subfractions. Among the subfractions, the active one showing the most potent vascular relaxation activity was further separated by prep. HPLC with reversed phase Microsorb C-18 column using 1 % acetic acid and methanol gradient solvent system to afford one pure compound, which revealed a potent vasodilatory effect. Instrumental analyses (NMR and mass spectrometry) of the isolated constituent indicated this compound to be (-)-epicatechin. The vasodilatory action mechanism of this compound should be further investigated.

Antimutagenic and Cytotoxic Effects of Ethanol Extract from the Inonotus obliquus (차가버섯 분획물의 항돌연변이 활성 및 암세포 성장억제효과)

  • This study was performed to determine the antimutagenic and cytotoxic effects of ethanol extract, ethylacetate fraction, water fraction and water soluble and insoluble polysaccharides I and II isolated from Inonotus obliquus using Ames test and SRB assay. Each sample itself did not show mutagenic effect. Among samples, the water unsoluble polysaccharides II in the presence of 50 $\mu\textrm{g}$/plate showed the strongest antimutagenic effect with over 90% against MNNG, 4NQO, B(a)P and Trp-P-l. However, ethyl acetate fraction (1 mg/mL) which had 90.8%, 94.3% and 83.5% inhibitory effect against on MCF-7, A549, AGS showed the strong cytotoxic effect compared to other samples.

Synergistic Antibacterial Activity of Ecklonia cava Extract against Anti-biotic Resistant Enterococcus faecalis (항생제 내성 Enterococcus faecalis에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과)

  • With continuing demand for the development of new, effective and safe therapies, an investigation was carried out to test the efficacy of an antibacterial agent derived from marine edible seaweed. The methanolic extract of Ecklonia cava from marine edible seaweed evinced potential antibacterial activity against Enterococcus faecalis. Among five solvent-soluble fractions of E. cava methanolic extract, the ethyl acetate soluble extract (EtOAc) exhibited the strongest antibacterial activity, with a MIC value of $128{\mu}g/mL $ against E. faecalis strains. Furthermore, a synergistic antibacterial effect between an antibiotic and the EtOAc fraction was assessed using fractional inhibitory concentration (FIC) indices. A combination of ciprofloxacin and the EtOAc fraction resulted in a ${\sum}FIC_{min}$ range of 0.188 and ${\sum}FIC_{max}$ of 0.508 to 563, suggesting that the ciprofloxacin-EtOAc fraction of E. cava combination resulted in an antibacterial synergy effect against E. faecalis.

Antioxidative and Antiobesity Activity of Nepalese Wild Herbs

  • A screening of Nepalese wild herbs for their antioxidant and antiobesity activity was carried out. The herbs including Allium hyposistum, Crateva unilocularis, Dryoathyrium boryanum and Cuscuta reflexa are widely used traditionally for various medicinal purposes in Nepal. The ethyl acetate fraction of D. boryanum showed polyphenol content of 266 ${\mu}g$GAE/mg with potent antioxidative activity assessed by DPPH free radical scavenging activity and hydrogen peroxide scavenging activity. The EtOAC fraction of D. boryanum also inhibited the lipid formation with 35% at 100${\mu}g/ml$ in 3T3-L1 cell model. Along with this, butanol fraction of C. reflexa also showed potent antioxidative activity and inhibition of 80% of lipid formation at the test concentration of 75 ${\mu}g/ml$ in 3T3-L1 cell line. This showed that these plant extracts have potential of antioxidant and antiobesity activity.

Anti-oxidative effects of fractionated Pulsatilla koreana $N_{AKAI}$ extracts (백두옹 분획층의 항산화 효과)

  • Objectives : This study was designed to investigate the effects of anti-oxidation of fractionated Pulsatilla koreana $N_{AKAI}$ (PK) extracts. And we examined to determine that a certain fractionated extract has the best anti-oxidative effects between the fractionated PK extracts. Methods : Anti-oxidative effects of fractionated PK extracts was measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cell was measured. After these examination, we determined a fraction that has best anti-oxidative effects. Results : Fractionated PK extracts inhibited radicals effectively. Also in RAW 264.7 cell, intracellular oxidation has inhibited by PK extracts. In these tests, ethyl acetate (EA) fraction has the best anti-oxidative effects among PK extracts. Conclusions : This results demonstrate that PK extracts exhibit anti-oxidative effects. And EA fraction has the best inhibition effects among the six fractions of PK.

Antioxidant Activity of Fractions from 70% Methanolic Extract of Sonchus oleraceus L.

  • The potential antioxidant activities of different fractions from a 70% methanolic (MeOH) extract of Sonchus oleraceus were assayed in vitro. All of the fractions exception of n-hexane showed a strong antioxidant activity, especially the ethyl acetate (EtOAc) fraction, which showed the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity ($IC_{50}=19.25{\mu}g/mL$). The results of hydroxyl radical scavenging activity and a reducing power assay showed concentration dependence, the EtOAc fraction demonstrating a better result than the other fractions at the same concentration in the studies. Additionally, the fractions' total phenolic (TP) contents was measured, phenolic compounds such as tannic acid, p-coumatric acid, quercetin, epicathchin, and kaempferol being detected by high performance liquid chromatography (HPLC). Meanwhile, a regression analysis revealed a moderate-to-high correlation coefficient between the antiradical activity and the TP contents, suggesting that fractions obtained from the 70% MeOH extract of S. oleraceus are of potential use as sources of antioxidant material.

Anti-inflammatory, Anti-oxidative and Anti-bacterial Activities of the Constituents Extracted from Leaves of Talipariti hamabo

  • Talipariti hamabo is a plant growing around salt marshes in the Lava Coast region of Jeju Island, Korea. In this study, the extract of T. hamabo leaves was investigated for the anti-inflammatory, anti-oxidative and anti-bacterial activities and their active constituents were identified. In the anti-inflammatory tests using lipopolysaccharide-stimulated RAW264.7 cells, the ethyl acetate (EtOAc) fraction inhibited the nitric oxide production without causing cell toxicity. Moreover, the EtOAc fraction reduced pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-6) as well as prostaglandin E2. In the anti-oxidative studies with DPPH and ABTS+ radicals, potent scavenging activities were observed in the EtOAc and n-butanol fractions. Upon the anti-bacterial tests using Staphylococcus epidermidis, EtOAc and n-butanol fractions exhibited good activities. Through the phytochemical studies on EtOAc fraction, three components were isolated by repeated column chromatography; oleic acid (1), p-hydroxyphenethyl-trans-ferulate (2), nicotiflorine (3). Based on these results, the extract of T. hamabo leaves can be developed as natural resources for cosmetic applications.

Antioxidant Activity and Inhibition Activity against α-Glucosidase and PTP1B of Chrysanthemum indicum L. Extract and Fractions (감국(Chrysanthemum indicum L.) 추출물과 분획물의 항산화 및 α-Glucosidase와 PTP1B 저해 활성)

  • In this study, we evaluated the polyphenol content, flavonoid content, antioxidant and biological of 70% EtOH extract and its fractions (dichloromethane, ethyl acetate and water) of the plant Chrysanthemum indicum L. (CI) belonging to family chrysanthemum. antioxidant activity was determined 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS), and superoxide scavenging activity. The biological activity tests included measurement of ${\alpha}$-glucosidase and PTP1B. The results show that content of polyphenol and flavonoid in 70% EtOH of CI 30.20 mg and 31.58 mg, respectively. Among the different samples, ethyl acetate (EA) fraction showed the highest level of polyphenol (87.87 mg) and flavonoid (65.12 mg). The DPPH, ABTS and superoxide dismutase(SOD) antioxidant assay of EA fraction showed 93.84%, 93.38% and 84.68% inhibition respectively. The EA fraction also showed the highest antioxidant activity compared to 70% EtOH extract and other fractions. EA fraction ($20{\mu}g/mL$) showed the highest 30.29% and 99.24%, respectively compared to other samples. The results of this study show that the EA fraction contained more antioxidant and antidiabetic components than 70% EtOH extract and other fractions. In conclusion, we believe that CI may be useful natural antioxidant and functional material.

Cellular Protective Effects of Peanut Sprout Root Extracts (땅콩나물 뿌리 추출물의 세포 보호 효과)

  • In this study, the cellular protective effect and antioxidative property of peanut sprout root extracts were investigated. Cellular protective effects of peanut sprout root extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction of extracts exhibited a cellular protective effect in a concentration dependent manner. Particularly, the aglycone fraction of extracts showed prominent cellular protective effects in a concentration range (5~50 ${\mu}g/mL$). They are more effective than that of (+)-${\alpha}$-tocopherol, known as a lipid peroxidation chain blocker. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of peanut sprout root extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extracts ($OSC_{50}$; 1.59 ${\mu}g/mL$) showed a similar ROS scavenging activity compare with that of L-ascorbic acid (1.50 ${\mu}g/mL$), known as a strong antioxidant. On the other hand, the order of free radical (1,1-diphenyl-2-picrylhydraxyl, DPPH) scavenging activity ($FSC_{50}$) was (+)-${\alpha}$-tocopherol > 80% MeOH extract > aglycone fraction > ethyl acetate fraction. These results indicate that peanut sprout root extracts can function as an antioxidant in biological systems, particularly skin exposed to solar UV radiation by scavenging $^1O_2$ and other ROS, and to protect cellular membranes against ROS.

Antifungal Activity of Wood Extracts of Larix leptolepis and Pseudotsuga menziesii againstTrichoderma spp. (Trichoderma 속에 대한 낙엽송 및 미송 추출물의 항균활성)

  • This study was undertaken to determine inhibitory compounds from extracts of the softwood (larix leptolepis, Pseudotsuga menziesii) sawdust against Trichoderma spp. The sawdust of L. leptolepis and P. menziesii were hot water extracted, which were with fraction extracted organic solvents. The organic solvent extractions were carried out by n-hexane, methylene chloride, ethyl acetate. The antifungal activity of hot water extracts of L. leptolepis sawdust was determined to be 20.6% inhibition at a concentration of 1,000 ppm against Trichoderma spp. The antifungal activity of P. menziesii sawdust was outstanding about 60.3% against Trichoderma spp. The yields of the fractions of n-hexane soluble, methylene chloride soluble and ethyl acetate soluble from the hot water extract of L. leptolepis sawdust were 4.0%, 6.0% and 8.0%, repectively. However, the yields of the fractions of three solvents of P. menziesii sawdust were 8.0%, 13.0 and 14.0% correspondingly. The antifungal activity of n-hexane soluble fraction from hot water extracts of L. leptolepis sawdust was highest to about 68.5% to 79.9% against Trichoderma spp. compared to others. The antifungal activities of n-hexane soluble fraction from hot water extracts of P. menziesii sawdust showed 68.5%, 71.4%. 71.9%, 75.7% and 82.3% against T. aggressivum, T. atroviride, T. harzianum, T. koningii and T.viride, respectively. The n-hexane soluble fraction revealed much higher antifungal activity than the other fractions did. This study demonstrated that the n-hexane fraction of the hot water extracts of L. leptolepis and P. menziesii exhibited the greatest antifungal activity against Trichoderma spp.
