The gene encoding an esterase enzyme was cloned from a metagenomic library of cow rumen bacteria. The esterase gene (est2R) was 2,120 bp in length, encoding a protein of 516 amino acid residues with a calculated molecular weight of 57,286 Da. The molecular weight of the enzyme was estimated to be 57,000 Da by SDS-PAGE. Est2R shared 35.6% amino acid identity with esterase (CAH19079) of uncultured prokaryote. The Est2R was most active at $20-40^{\circ}C$, and showed optimum at $30^{\circ}C$ and pH 8.0. The most activity of Est2R for the different chain length of p-nitrophenyl ester group as substrate was p-nitrophenyl acetate. Moreover, the enzyme was found to be most active without organic solvent, followed by 98% active with ethanol, and the enzyme activity was highly affected by the acetonitrile. The enzyme was significantly inhibited by $Zn^{2+}$ but stimulated by $Ca^{2+}$. So, novel esterase gene est2R is likely to obtain from cow rumen metagenome and supposed to use for industrial purpose.
멸종위기 천연기념물 어류 어름치(Hemibarbus mylodon)를 대상으로한 어름치 유전자 은행 구축 연구의 일환으로 뇌, 소화관, 근육, 간, 신장, 난소 및 정소 조직으로부터 expressed sequence tag (EST) library들을 구축하고 발현 유전자의 탐색을 실시하였다. EST 탐색을 통해 총 3,383개의 발현 유전자 염기서열 단편을 확보하였고 이들로부터 1,354개의 EST를 포함하는 총 333개의 contig들이 형성됨으로써 비교적 높은 빈도(69.8%)의 unigene 확보율을 나타내었다. EST의 조직 별 출현 양상은 orthologue들과의 상동성 정도 및 유추 기능의 대분류를 기준으로 분석할 때 각 조직들은 서로 다른 특징을 나타내었다. 어름치에서 발굴된 EST들은 zebrafish의 유전자들과 가장 높은 match 빈도를 나타내었다. 본 연구를 통해 확보된 EST library들과 염기서열 정보는 본 종의 장외 복원을 위한 효율적인 인공증식 기술 개발에 유용한 기초 정보로 활용될 수 있을 것이다.
Bacillus stearothermomophilus, a strong xylan degrader, was confirmed to express multiple esterase activities in addition to the major xylanolytic enzymes. One of the genes encoding the esterases was isolated from the genomic library of B. stearothermophilus constructed with EcoRl restriction endonuclease and pBR322 plasmid. Three recombinant plasmids showing the tributyrin degrading activity were selected from approximately 7, 000 E. coli HB101 transformants, and were found to have the same insert of a 3.2 kb DNA fragment. Restriction mapping and hybridization studies revealed that the gene(estII) on the hybrid plasmid (pKMG7) had originated from the B. stearothermophilus chromosome, and was distinct from the estl, another esterase gene of B. stearothermophilus isolated in the previous work. The E. coli cells harboring pKMG7 produced an acetylxylan esterase that exibited similar substrate specificity to the esterase encoded by the estI gene.
한우의 반추위에서 게놈 DNA를 분리하여 메타게놈 은행을 구축한 다음 carboxylesterase를 암호화하는 유전자를 클로닝 및 유전자를 선별하였다. 선별된 유전자의 DNA 염기서열 및 아미노산 서열을 분석하고 유전산물의 생화학적인 특성을 조사하였다. est1R 유전자는 2,465 bp로 366개의 아미노산 잔기를 가진 단백질을 암호화하였으며 이 단백질의 이론적인 분자량은 61,166 Da이었다. Est1R단백질은 PNB carboxylesterase (P37967), acetylcholinesterase (1EEAA) 및 chain A (1H23A)와 각각 5.9%, 6.1%, 6.1% 상동성을 가지고 있었다. 이러한 검색 결과 기존의 알려진 lipase 및esterase와의 상동성이 낮은 것으로 보아 새로운 그룹의 효소로 추정된다. Est1R효소의 최적 pH는 7.0 근방이었으며 최적 온도는 $40^{\circ}C$ 부근이었다. 한편 10% 유기용매를 함유한 기질의 효소활성측정에서 대조구에 비해 methanol은 95%의 상대적인 활성을 가진 반면에 hexane 용액에서는 그 활성이 반으로 감소하였다. 따라서 유기용매 농도의 작용성에 따라 이 효소의 산업적 이용성도 가능하리라 추정된다.
The nucleotide sequence of the estI gene encoding acetylxylan esterase I of Bacillus stearothermophilus was determined and analyzed. The estI gene was found to consist of a 810 base pair open reading frame coding for a polypeptide of 270 amino acids with a deduced molecular weight of 30 kDa. This was in well agreement with the molecular weight (29 kDa) estimated by SDS-PAGE of the purified esterase. The coding sequence was preceded by a putative ribo some binding site 10 bp upsteam of the ATG codon. Further 53 bp upstream, the transcription initiation signals were identified. The putative $_{-}$10 sequence (TCCAAT) and $_{-}$35 seqence (TTGAAT) corresponded closely to the respective consensus sequences for the Bacillus subtiis major RNA polymerase. The G+C content of the coding region of the estI was 51% whereas that of the third position of codone was 60.2%. The N-terminal amino acid sequence of the EstI deduced from the nucleotide sequence perfectly matched the corresponding region of the purified esterase described previously. Comparison with the amino acid sequence of other esterases and lipases reported so far allowed us to identify a sequence, GLSMG at positions 123 to 127 of the EstI which was reported to be the highly conserved active site sequence for those enzymes. The nucleotide sequence of the estI revealed 55.7% homology to that of the xylC coding for the acetylxylan esterase of Caldocellum saccharolyticum.

A novel esterase gene, est7K, was isolated from a compost metagenomic library. The gene encoded a protein of 411 amino acids and the molecular mass of the Est7K was estimated to be 44,969 Da with no signal peptide. Est7K showed the highest identity of 57% to EstA3, which is an esterase from a drinking water metagenome, when compared with the enzymes with reported properties. Est7K had three motifs, SMTK, YSV, and WGG, which correspond to the typical motifs of family VIII esterases, SxxK, Yxx, and WGG, respectively. Est7K did not have the GxSxG motif in most lipolytic enzymes. Three additional motifs, LxxxPGxxW, PLGMxDTxF, and GGxG, were found to be conserved in family VIII enzymes. The results of the phylogenetic analysis and the alignment study suggest that family VIII enzymes could be classified into two subfamilies, VIII.1 and VIII.2. The purified Est7K was optimally active at 40ºC and pH 10.0. It was activated to exhibit a 2.1-fold higher activity by the presence of 30% methanol. It preferred short-length p-nitrophenyl esters, particularly p-nitrophenyl butyrate, and efficiently hydrolyzed glyceryl tributyrate. It did not hydrolyze β-lactamase substrates, tertiary alcohol esters, glyceryl trioleate, fish oil, and olive oil. Est7K preferred an S-enantiomer, such as (S)-methyl-3-hydroxy-2-methylpropionate, as the substrate. The tolerance to methanol and the substrate specificity may provide potential advantage in the use of the enzyme in pharmaceutical and other biotechnological processes.
The genus Acanthamoeba can cause severe infections such as granulomatous amebic encephalitis and amebic keratitis in humans. However, little genomic information of Acanthamoeba has been reported. Here, we constructed Acanthamoeba expressed sequence tags (EST) database (Acanthamoeba EST DB) derived from our 4 kinds of Acanthamoeba cDNA library. The Acanthamoeba EST DB contains 3,897 EST generated from amebae under various conditions of long term in vitro culture, mouse brain passage, or encystation, and downloaded data of Acanthamoeba from National Center for Biotechnology Information (NCBI) and Taxonomically Broad EST Database (TBestDB). The almost reported eDNA/genomic sequences of Acanthamoeba provide stand alone BLAST system with nucleotide (BLAST NT) and amino acid (BLAST AA) sequence database. In BLAST results, each gene links for the significant information including sequence data, gene orthology annotations, relevant references, and a BlastX result. This is the first attempt for construction of Acanthamoeba database with genes expressed in diverse conditions. These data were integrated into a database (http://www. amoeba.or.kr).
A novel esterase gene (est7S) was cloned from an enriched metagenomic library derived from an oil-spill area. The gene encoded a protein of 505 amino acids, and the molecular mass of the Est7S was estimated to be 54,512 Da with no signal peptide. Est7S showed the highest identity of 40% to an esterase from a sludge metagenome compared to the characterized enzymes with their properties, although it showed 99% identity to a carboxylesterase in the genome sequence of Alcanivorax borkumensis SK2. Est7S had catalytic triad residues, Ser183, Glu312, and His420, and the GESAG motif in most family VII lipolytic enzymes. Est7S was purified from the crude extract of clone SM7 using Sephacryl S-200 HR and HiTrap Q column chromatographies. The purified Est7S was optimally active at $50^{\circ}C$ and pH 10.0. Est7S showed a high specific activity of 366.7 U/mg protein. It preferred short length esters, particularly p-nitrophenyl acetate, efficiently hydrolyzed R- and S-enantiomers of methyl-3-hydroxy-2-methylpropionate, and glyceryl tributyrate. These properties of Est7S may provide potential merits in biotechnological applications such as detergent and paper processing under alkaline conditions.
본 연구는 누에 수정란 초기에 발현하는 유전자를 대량 선발하고, 유용 유전자의 프로모터를 개발하기 위한 연구의 일환으로 추진하였다. 산란 후 2 ~ 16시간이 경과한 누에알로부터 cDNA 유전자은행을 제작하였다. 제작된 cDNA 유전자은행으로 전체 960개 클론을 무작위 추출하여 부분 염기서열 분석을 통해 EST를 제작하였다. 분석된 652개 ESTs 중 염기서열 상동성 분석을 통해 156개의 기존 알려진 유전자와 178개의 미지의 유전자로 구성된 334개 독립유전자를 최종 선발하여 'eegEST'로 명명하였다. eegEST 분석 결과, 기존 염기서열 정보가 알려진 156개 독립유전자 중 2회 이상 출현한 유전자 수는 143개로 전체의 34%를 차지하였으며, Hsp20.8 유전자(12회)와 ubiqutin-like 유전자(11회)가 가장 높은 출현 빈도를 나타내었다. 또한 eegEST 독립유전자의 추정 기능에 따른 분류에서 곤충 수정란 발생초기에 확인할 수 있는 기관 형성과 관련한 유전자가 전체 24%를 차지하고 있었다. 본 연구에서 작성된 누에 수정란 초기 발현유전자 데이터베이스(eegEST)는 곤충 발생학 연구를 위한 정보제공 뿐 아니라 형질전환누에 제작을 위한 프로모터 개발 연구에 활용될 수 있을 것으로 기대한다.
넙치 (Paralichthys olivaceus) 신장에서 추출한 mRNA로부터 cDNA library를 제작하고 이를 이용하여 EST (Expressed sequence tag) 분석을 하였다. 넙치의 신장 cDNA library에서 무작위로 선별한 390개의 EST를 조사한 결과, 250개의 EST는 이미 밝혀진 유전자와 유사성이 있는 것으로 나타났으며, 140개의 EST는 새로운 유전자로 밝혀졌다. 유전자의 기능이 밝혀진 250개의 EST 중 14 (5.6%)개의 EST는 이미 알려진 넙치 EST와 상동성이 있는 유전자로 확인되었고, 198 (79.2%)개의 EST는 다른 생물에서 알려진 유전자와 상동성이 있는 것으로 나타났다. 그러나 38 (15.2%)개의 EST는 전혀 기능이 알려지지 않은 새로운 유전자로 밝혀졌다. EST 분석은 새로운 유전자 뿐만 아니라 기능적으로 중요한 유전자를 탐색하는데도 아주 강력한 연구 방법이다. 이에 따라 본 연구에서는 넙치 신장 EST 분석을 통해 C-, L-type lectin과 MHC class II-invariant chain like proteins (li) 같은 면역기능과 관련이 있는 여러 개의 유전자를 확인하였고, 이들 유전자들은 질병감염 후 유전자 발현에서 나타나는 차이를 분석하는데 이용되는 cDNA microarray 연구에 유용하게 사용될 것으로 보인다.
