Microalgae have been explored as potential host species for biofuel production. Environmental factors affect algal growth and cellular composition. The effects of several key environmental factors, such as temperature, light, and pH of the medium on the growth and biochemical composition of Scenedesmus obliquus were investigated in this study. The highest growth rate of microalgae was observed at an optimal temperature of 25℃, 150 μmol/(m2·s) light intensity, and pH 10.0. The biochemical composition analysis revealed that the carbohydrate content decreased at lower (20℃) or higher temperature (35℃), whereas the protein and lipid contents increase at these temperatures. The fluctuation of light intensity significantly affected the contents of protein, carbohydrate, and lipid. The protein levels varied greatly when the pH of the medium was below 7.0. The carbohydrate and lipid contents significantly increased at pH above 7.0.

This experiment was conducted to investigate the effect of light intensities on the growth responses, carbohydrate contents and the characteristics of leaf gas exchange in Miscanthus sinensis and Pennisetum purpurascens. The plant height and leaf length were increased to about 30% in the sun. However, those were reduced severely in the shade, and leaf necrosis was also observed. The representative growth index and the dry weight of 2 species were 50% higher than shade and the rate was reduced according to the decrease of light intensities. Total carbohydrate contents showed very similar changes to that of dry weight. However, any notable influences were observed at above the light intensities of 250~500${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ in the half shade. The size of spikes and the earliest spiking appeared in the sun and the spike color was decolorized as decreased in light intensities, irrespective of species. Photosynthetic rate of 2 species was 2 times higher in the sun than those in the shade, and it showed the typical photoresponses of sun plant. Stomatal conductances and intercelluar $CO_2$ concetration showed similar changes to that of photosynthetic rate. On the contrary, vapor pressure deficit was increased more in the shade than in the sun.
The effects of light and darkness on stomatal aperture and guard cell apoplastic pH in the intact leaf and in the isolated epidermal strips of Commelina communis have been investigated. Stomata in the intact leaf opened wide in the light. In contrast, stomata in the isolated epidermal strips did not respond clearly to light. To eucidate the relationship between the stomatal aperture and the guard cell apoplastic pH, apoplastic pH was measured. In the light the guard cell wall of intact leaf was acidified by pH 1.9 units, falling from pH 7.3 to pH 5.4 in the first 10 minutes. On the contrary, apoplastic pH of isolated epidermal strips changed slowly from pH 7.3 to pH 6.9 at 20 min. Stomata in the intact leaf closed rapidly in the dark. On the other hand, stomata in the isolated epidermal strips failed to close in dark. There was a slow increase in apoplastic pH on transfer to the dark after incubation for 1.5 h in the light and the level observed before the experiment was regained after around 40 min. When the isolated epidermal strips were transferred to the dark, apoplastic pH maintained a uniform level of around pH 7.2-7.4. These results indicate that the mechanism of stomatal opening and closing from isolated epidermal strips and intact leaves could be different.
BACKGROUND: This study was performed for the identification and quantification of glucosinolate (GSL) contents in seven varieties of rapeseed (Brassica napus L.) sprouts cultivated under dark and light conditions. METHODS AND RESULTS: Crude glucosinolates (GSLs) were desulfated by treating with aryl sulfatase and purified using diethylaminoethyl sepharose (DEAE) anion exchange column. Individual GSLs were quantified using high-performance liquid chromatography (HPLC) with electrospray ionization-tandem mass spectrometry (ESI-MS/MS). Eleven GSLs including six aliphatic (progoitrin, sinigrin, glucoalyssin, gluconapoleiferin, gluconapin, and glucobrassicanapin), four indolyl (4-hydroxyglucobrassicin, glucobrassicin, 4-methoxyglucobrassicin, and neoglucobrassicin) and one aromatic (gluconasturtiin) were identified based on the fragmentation patterns of MS spectrum. Aliphatic GSLs were noted as the predominant group with average 85.2% of the total contents. The most abundant GSLs were progoitrin which was ranged at $8.14-118.68{\mu}mol/g$ dry weight (DW). The highest total GSL amounts were documented in 'Hanra' ($146.02{\mu}mol/g$ DW) under light condition and 'Mokpo No. 68' ($86.67{\mu}mol/g$ DW) in dark condition, whereas the lowest was in 'Tamra' (30.13 and $14.50{\mu}mol/g$ DW) in both conditions. The sum of aliphatic GSLs attributed > 80% in all varieties, except 'Tamra' (67.7% and 64.9% in dark and light conditions, respectively) in the total GSL accumulation. Indolyl GSLs were ranged $2.41-15.73{\mu}mol/g$ DW, accounted 2.78-33.6% of the total GSLs in rapeseed varieties. CONCLUSION(S): These results provide valuable information regarding potential beneficial GSL contents individually. This study attempts to contribute to knowledge of the nutritional properties of the different varieties of rapeseed plants. These results may be useful for the evaluation of dietary information.

Objectives Rapid increase in engineered nanoparticles (ENPs) in many goods has raised significant concern about their environmental safety. Proper methodologies are therefore needed to conduct toxicity and exposure assessment of nanoparticles in the environment. This study reviews several analytical techniques for nanoparticles and summarizes their principles, advantages and disadvantages, reviews the state of the art, and offers the perspectives of nanometrology in relation to ENP studies. Methods Nanometrology is divided into five techniques with regard to the instrumental principle: microscopy, light scattering, spectroscopy, separation, and single particle inductively coupled plasma-mass spectrometry. Results Each analytical method has its own drawbacks, such as detection limit, ability to quantify or qualify ENPs, and matrix effects. More than two different analytical methods should be used to better characterize ENPs. Conclusions In characterizing ENPs, the researchers should understand the nanometrology and its demerits, as well as its merits, to properly interpret their experimental results. Challenges lie in the nanometrology and pretreatment of ENPs from various matrices; in the extraction without dissolution or aggregation, and concentration of ENPs to satisfy the instrumental detection limit.
BACKGROUND: Photoreversibility, a reversion of the inductive effect of a brief red light pulse by a subsequent far red light pulse, is a property of photo responses regulated by the plant photoreceptor phytochrome B. Plants use photoreceptors to sense photo signal and to adapt and modify their morphological and physiological properties. Phytochrome recognizes red light and far red light and plays an important role in regulating plant growth and development. METHODS AND RESULTS: The reversal responses of growth and fruiting characteristics were investigated to increase the yield of oriental melon (Cucumis Melo L. var. Kumsargakieuncheon) by means of controlling light quality in a plastic house. Red (R:660nm) and far red (FR:730nm) lights were subsequently irradiated on the whole stems and leaves of the oriental melon plant during growing periods, using red and far red LEDs as light sources, from 9:00 PM daily for 15 minutes. The intensities of R and FR light were 0.322-0.430 μmol m-2s-1 and 0.250-0.366 μmol m-2s-1, respectively. Compared to R light irradiation, combination of R and FR light irradiation increased the length of internode, number of axillary stems, number of female flowers, and fruit number of oriental melons. The results of treatment with R were similar to R-FR-R light irradiation in terms of length of internode, number of axillary stems, number of female flowers, and number of fruits. When FR treatment was considered, R-FR and R-FR-R-FR light irradiation had similarities in responses. These reversal responses revealed that oriental melon showed a photoreversibility of growth characteristics, flowering, and fruiting. CONCLUSION: These results suggested the possibility of phytochrome regulation of female flower formation and fruiting in oriental melon. The fruit weight of the oriental melon was the heaviest with the R light irradiation, while the number of fruits was the highest with the FR light. With the FR light irradiation, the fruit weight was not significantly higher compared to that of the control. Meanwhile, the yield of oriental melon fruits increased by 28-36% according to the intensities of the FR light due to the increases of the number of fruits.
One of the fundamental elements in developing mixed reality applications is to effectively analyze and apply the environmental lighting information to image synthesis. In particular, interactive applications require to process dynamically varying lighting sources in real-time, reflecting them properly in rendering results. Previous related works are not often appropriate for this because they are usually designed to synthesize photorealistic images, generating too many, often exponentially increasing, light sources or having too heavy a computational complexity. In this paper, we present a fast light source estimation technique that aims to search for primary light sources on the fly from a sequence of video images taken by a camera equipped with a fisheye lens. In contrast to previous methods, our technique can adust the number of found light sources approximately to the size that a user specifies. Thus, it can be effectively used in Phong-illumination-model-based direct illumination or soft shadow generation through light sampling over area lights.
The purpose of this study was to determine optimum condition for the cultivation of Chlorella sp. FC-21 using red light emitting diodes (LED). Specific growth rate and cell concentration were measured for the reactors at the illuminations of different light intensity of red LED. Under the illumination of red LED, specific growth rate increased as light intensity increased but cell concentrations decreased. To determine beneficial effect of aeration to cell cultivation, micro-air bubbles were aerated at 0.7 vvm in the reactor at the illumination of red LED. Two and ten times greater specific growth rate and cell concentration were obtained when aeration was applied. In case of blowing of carbon dioxide, pH of culture medium decreased below to pH 3, which resulted in decreases of cell concentration. From this study, we found that red LED with aeration were the most appropriate light source for the cultivation of Chlorella sp. FC-21.
This study was performed to determine optimum conditions of batch type cultivation of Isochrysis galbana cultivated under various wavelengths of light emitting dioes (LEDs). Among LEDs used in the cultivation, white LED was found to be the most effective light source, and light intensity of 3,000Lux resulted in the most effective for the cultivation of Isochyrysis galbana. Comparison with common light source, fluorescent light, showed less effective than that with white LED. Four different air flow rates were tested to overcome shading effects due to denser cell concentration in the solution. In results, cell growth rates and maximum cell concentrations were similar regardless of air flow rates. Three times greater cell concentrations, however, were observed when air was applied.
The effect of growth regulators (NAA, BA and $ extrm{GA}_3$) and light (blue, red and far-red) on the changes in total protein and thylakoid membrane protein pattern of callus from intergeneric protoplast fusion between Nicotiana tabacum and Solanum nigrw were investigated. When the callus were irradiated with different wavelengths of light, blue and red light accelerated the synthesis of total proteins and thylakoid membrane proteins. Particularly, red light led to an increase in the protein synthesis compared to blue light. When the callus were subjected to various combinations of growth regulators, NAA+$ extrm{GA}_3$ and NAA+BA treatments induced remarkable increase of total proteins and thylakoid membrane proteins accumulation, particularly in the combination of NAA+$ extrm{GA}_3$. NAA.$ extrm{GA}_3$ treatment with irradiation of red ligh showed highest value in the accumulation of total proteins and thylakoid membrane proteins. We conclude that simultaneous application of red light and NAA+$ extrm{GA}_3$ treatment may induce synergistic effect in the synthesis of total proteins and thylakoid membrane Proteins.
