New Records of Two unknown Micro-filamentous Endophytic Green Algae in Korea: Phaeophila dendroides and Dilabifilum arthropyreniae (한국산 미기록 사상형 내생녹조 2종: 꼬인털속살이말 및 속살이마디말)

  • Morphological and molecular information about two microfilamentous endophytic algae of the Grateloupia lanceolata or Bryopsis sp. from Korea is given. Of two endophytes, Phaeophila dendroides is endophytic in Grateloupia lanceolata. It is green in color and composed of uniseriate branched filaments with long setae. Each cell had several pyrenoids. Undulate or twisted Phaeophila-type hair developed from vegetative cells. Dilabifilum arthropyreniae is endophytic in Bryopsis sp. The frond of this species consists of frequently irregular, branching uniseriate filaments. Each cell had a single pyrenoid without hairs. Sporangia were not observed. The ends of the filaments were curved. In the phylogenetic tree, based on tufA and ITS sequences, these two species are nested in the same clade as Phaeophila dendroides and Dilabifilum arthropyreniae, respectively. In this study, these two species are newly recorded in the Korean marine algal flora, based on the morphological and molecular data.

Distribution and Antifungal Activity of Endophytic Fungi in Different Growth Stages of Chili Pepper (Capsicum annuum L.) in Korea

  • This study investigated the distribution of endophytic fungi obtained from the surface sterilized tissues of leaves, stems and roots of chili pepper ($Capsicum$ $annuum$ L.) plants in seedling, flowering and fruiting stages in Korea and their antifungal activity. A total of 481 isolates were recovered and were identified using molecular techniques. Based on rDNA ITS gene sequence and phylogenetic analysis, 21 fungal genera were characterized, belonging to 16 Ascomycota and 5 Basidiomycota. $Penicillium$ in seedling stage, $Fusarium$ in flowering stage, $Colletotrichum$ followed by $Fusarium$, $Alternaria$ and $Xylaria$ in fruiting stage was predominant and $Alternaria$, $Cladosporium$ and $Fusarium$ were common in all growth stages. Among 481 endophytes 90 phenotypes were evaluated for the antimicrobial activity against three major pathogens ($Phytophthora$ $capsici$, $Colletotrichum$ $acutatum$ and $Fusarium$ $oxysporum$) of chili pepper. Among them 16 isolates inhibited the growth of at least one test microorganisms. Three strains showed a broad spectrum antifungal activity and displayed strong inhibition against chili pepper pathogenic fungi.

  • The microbiome makes a significant contribution to plant health and endophytes may generate positive effects for the host. However, there is a limited knowledge available concerning the kiwifruit endophyte. Therefore, we discuss endophyte microbiome community structures among the kiwifruit cultivars. Total reads numbered 17620 in cv. Hayward, 11515 in cv. Haegeum and 13613 in cv. Jecygold. The number of OTUs was follows: 112 in cv. Hageum; and 87 in cvs. Hayward and Jecygold. Most of the identified OTUs were phylum Proteobacteria and it emerged that Actinobacteria, Firmicute and Bacteroidetes were mainly present. While the largest difference in Proteobacteria content is between cv. Haegeum and cv. Jecygold, they shared more OTUs than cv. Hayward. As well, this study revealed the presence of remarkably dominated OTU of Pseudomonas spp. in kiwifruit sap regardless of cultivars. To the best of our knowledge, this study is the first tone to investigate the kiwifruit endophyte-microbiome community.

Fungal Endophytes in Roots of Aralia Species and Their Antifungal Activity

  • Endophytic fungi were isolated from surface sterilized root tissues of Aralia elata and Aralia continentalis, collected from farmer's field in Chungnam province, Republic of Korea, in 2005. Based on ITS sequence analysis, 24 fungal genera were characterized from 359 isolates, belonging to 22 Ascomycota, 1 Glomeromycota and 1 Oomycota. Strumella, Rhizopycnis and Entrophospora in A. elata and Rhizopycnis and Leptosphaeria in A. continentalis were the most abundant taxa. Out of 24 genera, Entrophospora, Leptodontidium, Neoplaconema, Paraconiothyrium, Rhizopycnis, Strumella and Tumularia were new to Korea. A total of 110 isolates were tested for antifungal activities against six plant pathogenic fungi. Out of these, 39 isolates showed antifungal activity against at least one plant pathogenic fungi. Four isolates of Pyrenochaeta, 1 isolate of Entrophospora and 1 unidentified fungus strongly inhibited the growth of six plant pathogenic fungi.

  • For researching nitrogen-fixing bacteria associated with gramineous crops, we collected growing roots of rices, wheats, oats, barleys, ryes, and maizes at 19 sites of southern Korean peninsula. Endophytes and free living bacteria were isolated from those crop roots. Sixty-three isolates were classified on the basis of different morphology, size, color, host of colony, and the 16S rDNAs sequence. The analyses of PCR amplification for nifH gene and nitrogenase activity assay, revealed that all isolates contained nitrogen-fixing abilities. In addition, most of them have cellulase activity which is one of the common features of endophytic bacteria from plant.

  • Leaves of Lindera obtusiloba were collected from four sites in Gangwon Province, Korea. Endophytic fungi were isolated from the leaves and identified using ITS sequences of rDNA. Total twelve species belonging of endophytic fungi were identified; Alternaria alternata, Annulohypoxylon annulatum, Creosphaeria sassafras, Diaporthe eres, Discosia sp., Epicoccum nigrum, Glomerella acutata, Glomerella cingulata, Paraconiothyrium brasiliense, Pestalotiopsis neglecta, Phomopsis amygdali, Xylaria sp. The endophytic fungus, Phomopsis amygdali, was the most dominant species isolated from L. obtusiloba in this study and the fungal diversities varied in the different sites.

Diversity of Fungal Endophytes from Pinus koraiensis Leaves in Korea (한국의 잣나무에서 분리한 내생균의 다양성)

  • The composition of endophytic fungal species from Pinus koraiensis was studied in two areas (Yeongwol and Jincheon), Korea. To identify 113 isolates, rDNA ITS regions were sequenced. On the basis of the ITS sequence, Pestalotiopsis sp. was the most frequently isolated species in both areas. Pestalotiopsis sp. (68.5%) and Lambertella sp. (12.4%) were isolated frequently in Yeongwol, whereas Pestalotiopsis sp. (41.7%), Hypoxylon sp. (20.8%) and Phomopsis sp. (12.4%) were dominant in Jincheon. Hypoxylon sp. and Phomopsis sp. were not found in Yeongwol.

Four Species of Endophytic Fungi Isolated from Leaves of Woody Plants in Mt. Hambaek (강원도 함백산에 서식하는 목본식물의 잎에서 분리한 4종의 내생균)

  • In this study, endophytic fungi were isolated from leaves of four species of woody plants, Acer tagmentosum, Larix kaempferi, Abies holophylla, and Pinus koraiensis, on Mt. Hambaek, Gangwondo, Korea. The endophytic fungi were identified using morphological and sequences analysis of ITS regions. The fungal endophytes were identified as Talaromyces radicus, Myceliophthora verrucosa, Cryptosporiopsis diversispora, and Sphaerulina berberidis, which are the first record in Korea. The morphological and molecular phylogenetic characteristics of each strain were described.

  • Endophytic fungi were isolated from surface-sterilized roots of a species of orchid, Oreorchis patens, in Korea. The isolated strains were identified based on morphological characteristics and sequence analyses of the internal transcribed spacer (ITS) and large subunit (LSU) of rDNA regions. Two species, Thelonectria veuillotiana and Phialocephala humicola, are first reported in Korea with their descriptions.

  • We isolated endophytic fungal strains from the leaves of Abies koreana growing in Korea. The fungal strains were identified as Coleophoma parafusiformis and Coniochaeta ligniaria, based on morphological characteristics and sequence analysis of the internal transcribed spacer region and large subunit region of ribosomal DNA. These species have not been previously reported in Korea. In this study, we report the morphological characteristics and results of phylogenetic analysis of the two novel fungal strains.
