A Study on the Comparison of Chemical Characterization and Ellagic Acid Content Between Distribution Bokbunja and Korean Native Bokbunja (국내유통 복분자와 토종복분자의 이화학적 특성과 엘라그산 함량 비교연구)

  • The purpose of this study was to compare the chemical properties of the bokbunja distributed in Korea with the content of the bioactive substance ellagic acid. The bokbunja was Rubus coreanus group and Rubus occidentalis group were compared, domestic bokbunja and import bokbunja were compared. In bokbunja, free sugar was 30.89 ± 0.7 mg/g of Rubus coreanus and 29.05 ± 0.87 mg/g of Rubus occidentalis. and 27.28 ± 7.4 mg/g of domestic bokbunja and 21.58 ± 6.73 mg/g of import bokbunja. The free amino acids was 4.50 ± 0.08 mg/g of Rubus coreanus and 5.05 ± 0.08 mg/g of Rubus occidentalis. and 4.13 ± 1.09 mg/g of domestic bokbunja and 3.75 ± 0.31 mg/g of import bokbunja. Validation of the ellagic acid method was confirmed by comparing the retention time and spectrum of the standard and extract using HPLC. The calibration curve (R2) showed linearity of 0.9999. As a result of analyzing the ellagic acid content of each extraction solvent, DMSO and methanol mixture extracts were high, and Rubus coreanus was 2.56 mg/g and Rubus occidentalis was 3.16 mg/g, which was not significantly different (p < 0.05) In addition, the ellagic acid content of domestic bokbunja and import bokbunja was 2.83 mg/g and 2.99 mg/g, which was not significantly different (p < 0.05).

Quantitative Analysis of Ellagic Acid in Nymphaea tetragona Georgi Root by HPLC-UVD (HPLC-UVD를 이용한 수련(Nymphaea tetragona Georgi)의 Ellagic Acid 정량 분석)

  • A high performance liquid chromatography (HPLC) method for the quantitation of ellagic acid in Nymphaea tetragona was developed for the quality control of functional cosmetic ingredient, the extract of N. tetragona. Separation and quantitation were successfully achieved with a Kromasil C18 column ($5{\mu}m$, $250mm{\times}4.6mm$, i.d.) by isocratic elution of a mixture of acetonitrile containing 0.1% trifluoroacetic acid and water containing 0.03% phosphoric acid at a flow rate of 1.0 ml/min. The UV detector was used for the detection and the wavelength for quantitation was set at 254 nm. The presence of ellagic acid in the extract was determined by comparison of retention time and spiking with authentic standard. Analytical results showed good linearity ($R^2=0.99996$) in relatively wide concentration ranges. The R.S.D. for precision test was less than 3.0%. Recovery of the compound was 98.55~101.72% with R.S.D values less than 4.0%. In conclusion, this method has been successfully applied to the determination of ellagic acid in N. tetragona.

Comparison of whitening effect of Rubus coreanus fruit according to maturity (성숙도에 따른 복분자 열매의 미백 활성 비교)

  • Purpose: The Rubus coreanus fruit (RF) is an important traditional medicinal herb having antioxidant, anti-inflammatory, and immunoregulatory properties. These activities are known to change dramatically, depending on maturity of the RF. It is presumed that change of functional components, such as flavonoids, tannins, phenolic acids, triterpenoids and organic acids in RF, affect the various bioactivities. This study aimed to confirm changes in the anti-melanogenic effects of RF based on maturity, and to identify the bioactive compounds responsible. Methods: The cell viability of mature RF (MRF) and immature RF (IRF) extracts was investigated using B16F10 cells. To compare the anti-melanogenic effect of MRF and IRF extracts, we first assessed the melanin content. High-performance liquid chromatography analysis was performed to evaluate changes in the level of ellagic acid according to maturity of the RF. In addition, tyrosinase inhibitory activity of both extracts was examined. Results: MRF and IRF extracts (50-200 ㎍/mL) do not affect the cell viability of B16F10 melanoma cells. IRF extract more effectively inhibited melanin synthesis than MRF extract. The content of ellagic acid in IRF extract was higher than that obtained in MRF extract. Furthermore, greater inhibition of tyrosinase activity was observed after exposure to IRF extract than MRF extract. A positive correlation was determined between ellagic acid content and tyrosinase inhibitory activity, and a negative correlation was obtained between ellagic acid content and melanin content. Taken together, our results indicate that ellagic acid is one of the major bioactive compounds of RF that imparts a whitening effect. Conclusion: Our results indicate that ellagic acid in MRF and IRF extracts affect the anti-melanogenesis effect through inhibition of tyrosinase activity. Therefore, the ellagic acid rich IRF has greater potential for application as a natural and functional cosmetic material.

Phenolic Acid Composition and Antioxidative Activity of Chestnut Endoderm (밤 삽피의 페놀산 조성과 항산화 활성)

  • Free, soluble esterified and insoluble bounded phenolic acids were separated from Eungi chestnut endoderm. The composition and contents of phenolic acid were analyzed by gas chromatography, and their antioxidant activity was examined by DPPH assay, 2-deoxyribose oxidation, and ferric thiocyanate method. Gallic, ellagic, salicylic, and gentisic acids in free phenolic acid fraction, gallic, ellagic, and protocatechuic acids in soluble esterified fraction, sianpic and gentisic acids were the major phenolic acids in insoluble bounded fraction. Marked differences were observed in the phenolic acid composition and contents among the fractions. Free phenolic acid fraction showed the strongest antioxidant activity. Results revealed chestnut endoderm could be a potential antioxidant source containing gallic and ellagic acids.

Method Development of Ellagic Acid as Marker Compound for Standardization of Gochang Bokbunja (Rubus coreanus Miquel) as Functional Ingredient (고창 복분자의 기능성원료 표준화를 위한 지표성분으로서 Ellagic Acid의 분석법 개발)

  • Method development and validation of ellagic acid for the standardization of Gochang Bokbunja as a functional ingredient and health food were accomplished. A Symmetry$^{(R)}$ (C18, $4.6{\times}250mm$, $5.0{\mu}m$) column was used with a gradient elution system of 1% formic acid in water and acetonitrile. This method was validated according to specificity, linearity, accuracy, precision test, and recovery test. Specificity was confirmed with identical retention time, and calibration curves of ellagic acid showed good linear regression ($R^2$ >0.9996). Relative standard deviations (RSD) of data from the intra- and inter-day experiments were less than 2.28% and 2.84%, except in the low limit of quality control (LLOQ, $1{\mu}g/mL$) sample. The results of the recovery test were from 89.17% to 97.92% with RSD values from 0.05 to 0.14%. Therefore, we performed analysis of ellagic acid as a marker compound in Gochang Bokbunja extracts. The amount of ellagic acid in Gochang Bukbunja was about $1.918{\mu}g/mg$ (0.192%) in the three times analysis, and RSD was less than 2.36% by the validated method. These results suggest that the developed HPLC method is simple, efficient, and could contribute to the quality control of Gochang Bokbunja extract as a functional ingredient.

Quantitative Analysis of Antioxidants in Korean Pomegranate Husk (Granati pericarpium) Cultivated in Different Site (HPLC에 의한 산지별 한국산 석류과피 중 항산화화합물의 함량분석)

  • The quantitative analytical method for major antioxidants, ellagic acid and punicalagin, in pomegranate husk (Granati pericarpium) were established by HPLC. The optimal HPLC conditions were as follows: Column; Agilent Zorbax Eclipse XDB-C18 ($4.6{\times}150mm,\;5{\mu}m$), mobile phase; 1% formic acid in water (A) and 1% formic acid in MeCN (B) (gradient elution of 5% to 100% B for 50 min), flow rate; 0.8 ml/min., detection; UV 254 nm. The optimal pre-treatment conditions for HPLC analysis were as follows: 5 g of pomegranate husk in 100 ml of 95% EtOH, refluxed for 3 h. Under these analytical conditions, punicalagin and ellagic acid contents in Korean pomegranates husks which were cultivated in five different sites were determined. As results, the ellagic acid and punicalagin (as a mixture of ${\alpha-\;and\;{\beta}-anomer$) contents were the highest in Haepyung pomegranate husk $(15.27{\mu}g/mg)$ and Jangsung pomegranate husk $(16.21{\mu}g/mg)$, respectively.

Activity Guided Isolation of Antioxidant Tannoid Principles from Anogeissus latifolia

  • Oxidative stress is an important causative factor in several human chronic diseases, such as atherosclerosis, cardiovascular disorders, mutagenesis, cancer, several neurodegenerative disorders, and the aging process. Phenolics and tannins are reported to be good antioxidants. Anogeissus latifolia (Combretaceae) bark has been used in the Indian traditional systems of medicine for curing a variety of ailments, but scientific validation is not available till date. Hence the present study was undertaken to isolate antioxidant compounds by activity-guided isolation. Inhibtion of diphenyl picryl hydrazyl (DPPH) and Xanthine oxidase along with photochemiluminescence assay were used as bioassay for antioxidant activity. Activity guided isolation was carried out using silica column and the compounds were quantified using HPLC. Ethyl acetate and butanol fraction exhibited potent antioxidant activity. Bioassay-guided isolation led to isolation of ellagic acid (1) and dimethyl ellagic acid (2) as the main active compounds, which along with gallic acid were quantified by HPLC. Thus we conclude that these three major tannoid principles present in A. latifolia, are responsible for the antioxidant potential and possibly their therapeutic potential.

  • The ethanolic extract of chestnut (Castanea crenata S. et Z., Fagaceae) inner shell (CISE) and one of its components, ellagic acid (EA), were evaluated for their protective effects against 1, 1-diphenyl-2-picryl hydrazine (DPPH) free radical generation and hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line. CISE and EA were shown to possess the free radical scavenging effect against DPPH radical generation, significantly. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from Chinese hamster lung (CHL) cell, assessed by single cell gel electrophoresis assay and 8-hydroxy -2'-deoxy guanosine (8-OH-2'dG) assay. Furthermore, topical application of CISE [$12.5\%$(w/w) cream] and ellagic acid [$1.0\%$(w/w) cream] for 14 days potently inhibited malondialdehyde (MDA) formation of mouse dorsal skin (a marker of lipid peroxidation) induced by ultraviolet B exposure. Therefore, CISE and its component, ellagic acid, may be the useful natural antioxidants by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage when topically applied.

The Antioxidant Activities of Three Solvent(Ether, Butanol, Water) Extrats from Chestnut Inner Shell in Soybean Oil (대두유에 대한 율피의 용매분획별(Ether, Butanol, Water) 항산화효과)

  • This study was carried out to compare the antioxidant activities of the ether, butanol, water extracts from chestnut inner shell(Castanea crenata Sieb. et Zucc.) with those of tocopherol and BHA in soybean oil. All additives were added to soybean oil on the quantities of 0.02%. Comparing the antioxidant activities under autooxidation condition at $45.0{\pm}0.5$ for 42days of the extracts were recorded in the order of butanol>ether>control>BHA>tocopherol depending on the solvent. Under the condition at $60.0{\pm}0.5^{\circ}C$ for 32days, the butanol extracts represented the high antioxidation effect, however, there was no significant differences between the ether extracts and control. Under thermal oxidation condition, the ether extract showed stronger antioxidant activity than those of the butanol extract. In the results of polyphenol compound analysis, ellagic acid, quercetin, morin, naringenin, flavanol were included in the ether extracts and ellagic acid, naringenin, gallic acid, flavanol were included in the butanol extracts, respectively. Among them, ellagic acid in ether extract and gallic acid and naringenin in butanol extracts seed to increase the antioxidant activities in the substrate oil.

A Study on the Comparsion of Nutrients Content and Ellagic Acid Content Between Distribution Bokbunja and Korean Native Bokbunja (국내유통 복분자와 토종복분자의 영양성분학적 차이점과 Ellagic acid 함량 비교연구)

  • 국내 복분자는 서양에서 유래한 서양복분자(Rubus occidentalis)와 국내 자생종을 개량한 토종복분자(Rubus coreauns)가 혼용되고 있으며, 전통 한방약재로서 미숙과를 중심으로 국내에서 유통되고 있는 복분자의 영양성분적인 특성과 주요성분인 ellagic acid 함량을 비교하였다. 토종복분자는 광양에서 재배되고 있는 복분자를 수집하였으며, 외래종 서양복분자는 고창에서 재배되고 있는 복분자를 수집하여 사용하였다. 나머지는 국내에서 유통되고 있는 국내산과 중국산 복분자를 경동약령시장과 금산약령시장으로부터 구매하여 사용하였다. 영양성분으로는 조지방, 조단백질 그리고 탄수화물 함량을 측정하였으며, 유리당, 지방산, 유리 아미노산 17종의 함량을 측정하였다. 조지방의 경우 토종복분자는 1.90 %, 서양복분자는 3.03 % 이였으며, 중국산 유통품은 2.28 %, 기타 국내산 유통품의 경우 2.89 %으로 중국산복분자의 조지방 함량이 낮은 것을 확인하였다. 그러나 탄수화물 함량의 경우 70.28~71.85 %로 복분자간의 함량에 큰 차이를 발견할 수 없었다. 유리당의 경우에는 토종복분자의 경우 glucose가 19.03 mg/g, fructose 16.29 mg/g이 측정되었고 고창 서양복분자의 경우 glucose가 16.29 mg/g, fructose 12.76 mg/g이 측정되어 유리당의 총 함량은 차이가 없는 것으로 확인되었으나 토종복분자의 경우 glucose의 함량이 조금 높은 것을 확인하였다. 복분자의 지방산 조성을 비교한 결과 고창 서양복분자에서 불포화지방산의 함량이 19.49 mg/g 으로 광양 토종복분자의 7.69 mg/g에 비하여 월등히 높은 것을 확인되었으며, 불포화지방산 중 linoleic acid (12.19 mg/g), oleic acid (1.88 mg/g)와 linolenic acid (5.43 mg/g) 함량이 높았다. 복분자의 아미노산의 함량은 광양 토종복분자의 경우 4.50 mg/g, 고창 서양복분자의 경우 5.05 mg/g으로 유리아미노산의 함량은 유사한 것으로 나타났다. 특히 아미노산 17종 성분 중 asparagine(0.65~0.84 mg/g), arginine(0.51~1.00 mg/g)과 threonine(0.99~1.63 mg/g)의 함량이 높았다. 지표성분으로 ellagic acid의 함량은 광양 토종복분자의 경우 2.56 mg/g, 고창의 서양복분자의 경우 3.16 mg/g으로 측정되어 서양복분자가 조금 높은 것으로 나타났다. 국내 유통되고 있는 중국산 복분자의 ellagic acid의 경우 2.99 mg/g, 기타 국내산 유통 복분자의 경우 2.83 mg/g으로 광양 토종복분자와 유사한 것으로 나타났다. 위의 연구결과는 국내에서 유통되는 토종 및 서양 복분자를 원료로 하는 기능성식품에 대한 제품개발의 기초자료로서 활용 될 수 있을 것으로 기대한다.
