수유기 동안 납의 투여는 급격한 체중의 변화를 유발하여 3주령 및 11주령 쥐의 경우 납을 투여한 군이 대조군에 비해 각각 50% 및 26% 체중 감소를 보였으나 뇌의 무게에는 유의적 변화를 관찰 할 수 없었다. 납 투여는 뇌 지방산 조성 변화에 큰 영향을 주지 않았지만 특히 AA와 DPAn-6와 같은 고도의 불포화 지방산 함량의 증가를 유발하였다. Motor ac-tivity 실험에서 납 투여군의 경우 대조군에 비하여 주어진 시간 내에서 움직이는 시간과 움직인 거리가 긴 것으로 보아 운동 활동성이 큰 것으로 여겨지나 두 군 간에 유의적 차이는 없었다. Elevated plus maze 실험에서 실험 1일째의 경우 open arm에 머무른 시간에는 유의적 차이가 없었으나 2일째에는 두 군에서 현저히 open arm에 머무른 시간이 감소하였으며 납 투여군의 경우 대조군에 비하여 머무른 시간이 감소하는 경향이었으나 통계적 유의성은 없었다. Open arm에 들어가는 횟수를 관찰한 결과, 실험 1일째의 경우 open arm에 들어가는 횟수가 납 투여군에서 유의적으로 적었고 2일째에는 두 군에서 모두 open arm에 들어가는 횟수가 감소하였으며 대조군에 비해 납을 투여한 쥐의 경우가 유의적으로 낮은 횟수로 open arm에 들어갔음을 관찰 할 수 있었다. 이상의 결과로부터 수유기 동안의 납 투여는 성숙 동물의 불안감을 증가시키는 효과를 나타내었다.
Objective : This study was designed to assess the protective effects of Chengwhabosimtang on the animal model of depression, chronic mild stress(CMS). Method : Male Sprague-Dawley rats were used for this experiment. The subjects were divided into 3 groups ( 1. CMS-drug: Chengwhabosimtang administered during CMS treatment, 2. CMS-vehicle: water administered, 3. normal ). After 4 weeks of CMS treatment, they were executed Forced swimming test(FST) and Elevated plus maze. Tyrosine hydroxylase(TH) in ventral tegmental area(VTA) and c-Fos in paraventricular nucleus(PVN) were measured. Result : 1. In FST, CMS-drug group showed significantly decreased immobility behavior. 2. CMS-drug group showed no significantly lower TH level in VTA than CMS-vehicle group. 3. CMS-drug group showed significantly less c-Fos expressed cell bodies in PVN than CMS-vehicle group. 4. In Elevated plus maze, CMS-drug group showed no significantly anxiety. Conclusion : These results suggest that Chengwhabosimtang may have protective antidepressant effects in CMS model rats. And these effects could be explained by the elevated stress-copying behaviors which are related with PVN of hypothalamus and dopaminergic neurons in VTA.
The bark of Magnolia officinalis has been used in traditional oriental medicine to treat a variety of mental disorders including anxiety and depression. The purpose of this study was to examine the effect of M. officinalis ethanol extract on stress-induced alterations in learning and cognitive function using a passive avoidance test (PAT) and also on anxiety-related behavior using the elevated plus-maze test (EPM) in female rats . The degree of Tyrosine hydroxylase (TH) in the region of the ventral tegmental area (VTA) and the locus coeruleus (LC) was measured using an immunohistochemical method. Corticosterone concentrations in serum were also measured. The ethanol extract from Magnolia officinalis was orally administered to female rats 30 minutes before evaluating their immobilization stress and anxiety-related behavior using an elevated plus-maze test and a passive avoidance test. Time spent in the open arms of the EPM increased in the M. officinalis-treated group compared with that of the saline-treated control group. In the passive avoidance test, the memory and cognitive function improved in the M. officinalis extract-treated group. M. officinalis extracts reduced elevated corticosterone concentrations in serum. Also, stress-induced TH increases were suppressed in the M. officinalis extract-treated group in the LC and the VTA region. These results suggest that M. officinalis might prove to be an effective anxiolytic anti-stress agent.
This study was performed to evaluate the effects of Quibitang in the chronic mild stress(CMS) model of depression in rats. Chronic exposure to mild unpredictable stress was found to depress the consumption of sucrose solution in rats for 8 weeks. These CMS-treated rats were stratified into Quibitang gruop and saline group. And control rats were also stratified into Quibitang group and saline group. The change of the consumption of sucrose solution and the body weight were measured, and open field test, elevated startle response and plus maze test were performed, to investigate the anti-depression effect of Quibitang. The results were as follows: 1. The consumption of sucrose solution was significantly reversed in Quibitang-treated group at 9th, 11th, 12th week, but there was no significant change at 10th week 2. CMS schedule decreased body weight. CMS-treated groups showed decrease of body weight after 5 weeks. After 10 weeks, Quibitang group showed lower body weight than saline group in CMS-treated groups 3. In open field test, Quibitang group showed significant difference of locomotion, latency. 4. In elevated startIe test, Quibitang group showed no significant change of startle response. 5. In plus maze test, Quibitang group showed no significant change of plus maze-time and plus maze-error.
본 연구는 Sprague-Dawley 계통의 4주령 랫드에서 MSG의 반복경구투여 독성평가와 행동실험을 통해 어린랫드의 인지능력과 불안감에 미치는 영향을 평가하기 위해서 시행하였다. 시험군은 MSG 대신 distilled water를 투여한 대조군(Control, n = 4), MSG를 3 g/kg을 투여한 군(Low, n = 4), MSG를 5 g/kg을 투여한 군(High, n = 4)으로 나누어 4주 동안 주5회 경구투여를 하였다. MSG의 안정성을 확인하기 위해 다음과 같은 관찰 및 검사를 하였다. 검사항목으로는 체중의 변화, 임상증상, 행동실험인 T-maze, Elevated-plus-maze와 혈액학적 검사, 혈청생화학적 검사, 병리조직학적 검사를 관찰한 결과 모든 투여군 및 대조군에서 특이할 만한 임상증상과 체중변화는 관찰되지 않았으며, 폐사 및 빈사 동물은 시험 전 기간을 통하여 발견되지 않았다. 행동실험인 T-maze 결과 MSG 고용량 투여군에서 움직이는 횟수가 감소하다 증가하는 양상을 보였고, Elevated-plus-maze 실험에서는 MSG 고용량 투여군의 Open arm의 출입빈도가 증가하는 등 유의적인 변화가 나타났다. 혈액학적 검사 및 혈청생화학적 검사에서는 대조군과 비교 시 유의적인 변화가 관찰되었지만 그 수치가 정상범위 안에 포함되기 때문에 이상을 나타내지 않았다. 병리조직검사 또한 약간의 염증발생 소견이 나왔지만 정상범위 안에 포함되기 때문에 이상을 나타내지 않았다고 판단했다. 따라서, 어린랫드를 이용한 MSG의 반복투여 독성시험 결과 특이할만한 신체적인 변화는 나타나지 않았으나 고용량의 MSG 투여는 어린랫드에서 인지능력의 저하 및 행동의 불안을 유발할 가능성이 있다고 사료된다.
The alcoholic extract of dried roots of Rubia cordifolia dose dependently reduced blood sugar level in alloxan treated rats. The extract also reduced ulcer index, plasma corticosterone in cold restrain stressed rats in dose related manner. The mice treated with alcoholic extract of Rubia cordifolia spent more time in the open arm of the elevated plus maze indicating anxiolytic activity. The extract also antagonized the amnesic effect of scopolamine in mice as indicated by reduced transfer latency in the elevated plus maze. Thus the plant bears potential for use in diabetes, stress, anxiety, and dementia.

Scutellaria baicalensis Georgi is one of the most important medicinal herbs in traditional chinese medicine. The object of this study was to determine the effects of water extracts of Scutellaria baicalensis (SB) and Scutellaria baicalensis metabolite (SBM) on the anxiolytic-like activities in the elevated plus-maze (EPM) test. The water extracts of SB (100, 200, or 400 mg/kg), and SBM (100 mg/kg) were orally administered to male SD rats for 3 day. This study was performed to investigate the anxiolytic-like effects Panax ginseng in mice using the elevated plus-maze model. Furthermore, the anxiolytic-like effects of Panax ginseng were compared to a known active anxiolytic drug, diazepam. Ginseng total saponin (GTS, 100 mg/kg) from red ginseng (RG), sun ginseng (SG) total extract (50 mg/kg), butanol fraction of SG(25 and 50 mg/kg) and ginsenosides ($Rb_1,\;Rg_1,\;and\;Rg_5$ and Rk mixture) significantly increased the number of open arm entries and the time spent on the open arm, compared with that of control. However, Red ginseng (RG) total extract (l00 mg/kg), GTS (25, 50 mg/kg), SG total extract (25 mg/kg) and ginsenosides ($Rg_{3}-R\;and\;Rg_{3}-S$) did not increase the number of open arm entries and the time spent on the open arm. On the other hand, butanol fraction of RG (l00 mg/kg), total extract of SG (50 mg/kg), butanol fraction of SG (50 mg/kg), ginsenosides ($Rb_{1},\;and\;Rg_{5}$ and Rk mixture) decreased the locomotor activity, in a similar fashion to diazepam. These data support that ginseng has the anxiolytic-like effects and the anxiolytic potential of SG was stronger than that of RG. Ginsenosides $Rb_{1},\;Rg_{1},\;and\;Rg_{5}$ and Rk mixture play important role on the anxiolytic-like effects of Panax ginseng. We provide evidence that ginseng and some ginsenosides may be useful for the treatment of anxiety.
The purpose of this study was to characterize the putative anxiolytic-like effects of the aqueous extract of the root of Polygala tenuifolia ( AEPT) using an elevated plus maze (EPM) and hole-board apparatus in mice. The AFPT was orally administered at 50, 100, 200 or 400 mg/kg to ICR mice, 1 h before the behavioral evaluation in the EPM respectively. Control mice were treated with an equal volume of saline, and positive control mice with buspirone (2 mg/kg). Single treatments of the AEPT significantly increased the percentage of time spent and arm entries into the open arms of the EPM vedrsus saline controls (P<0.05). Moreover, there were no changes in the locomotor activity and myorelaxant effects in any group compared with the saline controls. In the hole-board test,single treatments of the AEPT (200 and 400 mg/kg) significantly increased the number of headdips versus saline controls (P<0.05). In addition, the anxiolytic-like effects of the AEPT were blocked by WAY 100635(0.3mg/kg, I.p), a5-$HT_{1A}$ receptor antagonist not by flumazenil, a $GABA_{A}$ antagonist. These results indicate that P. tenuifolia is an effective anxiolytic agent, andsuggest that the anxiolytic-like effects of P. tenuifolia is mediated via the serotonergic nervous system.
Background : The aim of this study was to evaluate the effect of Punsimgieumgamibang on insomnia and anxiety. which is widely used in mental diseases. Method : We admitted Punsimgieumgamibang into rats which illumination of the sleeping periodhas been reversed, and measured its activity rate during the sleeping period. Also we studied the effect of Punsimgieumgamibang on anxiety using analyses of Punsimgieumgamibang on anxiety using analyses of behavior of rats submitted to the elevated plus-maze test.Result : 1) The sleeping ratio of the Pusimgieumgamibang group was higher than the control group on the whole, and the difference of the groups in the third time block was significant statistically. 2) We could not find any statistical information in all of the 4 anxiety related behavior of the elevated plus-maze experiment. and there was a tendency to a higher measurement in the Punsimgieumgamibang group animals in the total arms visitation in the elevated plus-maze, open arms visitation, and the amount of time of stay in arms. Conclusion : It is suggested that Pusimgieumgamibang has effects on insomnia.
