Optimal Conditions for the Mycelial Growth of Coprinus comatus Strains

  • The principal objective of this study was to acquire basic data regarding the mycelial growth characteristics for the artificial cultivation of Coprinus comatus. 12 URP primers were employed to evaluate the genetic relationships of C. comatus, and the results were divided into three groups. Among six kinds of mushroom media, MYP medium was selected as the most favorable culture medium for C. comatus. The optimal temperature and pH ranges for the mycelial growth of C. comatus were $23{\sim}26^{\circ}C$ and pH 6${\sim}$8, respectively. The carbon and nitrogen sources for optimal mycelial growth were sucrose and tryptone, respectively.

  • 새로운 야생식용(野生食用)버섯의 인공재배(人工載培) 가능성(可能性)을 검토(檢討)하고자 조사(調査)한 결과(結果), 1. 배지(培地) 종류별(種類別) 균멱생장(菌生長)은CSA 배지(培地)에서 Coprinus comatus 양호(良好)하였으나 Pleurotus cornucopiae, Auricularia auricula-judae 및 collybia sp는 불량(不良)하였다. 2. Malt extract의 첨가량(添加量)은 5g까지 증가(增加)시켰을 때 Lepista nuda와 Auricularia auricula-judae는 균멱생장(菌生長) 양호(良好)하였으나 Pholiota squarrosa는 불량(不良)하였다. 3. 균주별(菌株別) 온도(溫度)에 따른 균멱생장(菌生長)에 있어서 Lepiota procera $20{\sim}30^{\circ}C$, Pholiota squarrosa는 $20{\sim}25^{\circ}C$에서 양호(良好)하였다. 그 외 다른 균주(菌株)는 25^{\circ}C$에서 균먹(菌)가 양호(良好)했다. 4. 종균조시(種菌造時) Pholiota squarrosa는 참나무톱밥, Pleurotus cornucopiae는 활엽수토밥, Coprinus comatus는 양송이퇴비(堆肥)에서 균멱생장(菌生長)이 양호(良好)하였다. 5. 배지재두별(培地材枓別) 버섯발생(發生)은 볏짚배지(培地)에서 Coprinus comatus, Lepiota, alborubescens, 활엽수톱밥에서는 Auricularia auricula-judae, Pleurotus cornucopiae, 그리고 참나무톱밥에서 Pholiota squarrosa의 자실체(子實體)가 발생(發生)되어 인공재배(人工栽培)가 가능(可能)하였다.

Physical, Mechanical, and Antimicrobial Properties of Edible Film Produced from Defatted Soybean Meal Fermented by Bacillus subtilis

  • In order to extend the shelf-life of packaged or coated foods, an antibacterial edible film was developed from soybean meal that had been fermented with Bacillus subtilis under the optimum condition of pH 7.0-7.5 and $33^{\circ}C$ for 33 h. The water vapor permeability of the fermented film ($86.0 mg/cm^2{\cdot}h$) was higher than those of normal soybean films ($66.9 mg/cm^2{\cdot}h$). Protein solubility of the fermented film was also higher than ordinary soy protein film at the pH range of 3 -10. The fermented soybean film had higher tensile strength and lower $\%$ elongation (elongation rate) than the ordinary soybean film, mainly because partial hydrolysis of proteins in the soybean film occurred during fermentation. Antimicrobial properties of the fermented film on foodstuffs were measured by placing the films on surime, jerked beef, and mashed sausage media; containing $10^2-10^3$ CFU/plate of foodborne pathogenic bacteria, and showed significantly higher inhibitory effects on the growths of all the indicating bacteria. The film could be used as a packaging material in the food industry. However, before direct application of the fermented film to the commercial food industry, its poor mechanical and antibacterial properties need to be improved.

꽃송이버섯의 액체배양조건 및 봉지재배 적합배지 재료선발 (Selection of suitable conditions of mycelial growth and materials of bag cultivation in Sparassis crispa)

  • 꽃송이버섯은 식용버섯으로 다른 식용버섯에 비해 1.3-${\beta}$-D-glucan이 높은 항암 및 면역증강 효과가 있은 버섯이다. 하지만 꽃송이버섯은 초근 한국과 일본에서 버섯을 재배하고 있지만 재배방법은 확립되어있지 않다. 본 연구는 꽃송이버섯에 대한 봉지재배기술 개발을 위해 균사생장 및 원기형성에 대한 조사를 수행하였다. 꽃송이버섯의 액체종균제조하기위해 보리를 당화하여 종균을 제조하였는데 당화는 시간이 경과 할수록 당도가는 증가하는 경향으로 8시간에 $16.3^{\circ}Brix$를 나타냈다. 당화된 액체배지에서의 균사생장조건은 온도 ${\beta}$, pH5.0~6.0 일때 균사생장이 양호하였다. 꽃송이버섯의 봉지재배시 낙엽송톱밥에서 균사배양 및 밀도가 양호하였다. 낙엽송톱밥, 밀가루, 옥수수가루를 7:1:2(부피비)의 혼합배지에서 균사생육이 48.5mm/45일로 비교적 빠른 생장속도를 보였고 자실체의 수량은 68.1g/500g를 생산하였다.

Antimicrobial Edible Film Developed from Defatted Corn Germ Meal Fermented by Bacillus subtilis

  • In order to extend the shelf-life of packaged or coated foods, an antibacterial edible film containing 1.8% of BLS was developed from the defatted corn germ meal, which had been fermented with Bacillus subtilis under the optimum condition of pH 7.0-7.5 and $33^{\circ}C$ for 33 h. Water vapor permeability of the fermented film $(88.3mg/cm^2\;h)$ was higher than those of the normal corn germ films $(75.8mg/cm^2\;h)$. Protein solubility of the fermented film was also higher than ordinary corn germ film at the pH range of 3-10. The fermented corn germ film had higher tensile strength and lower % elongation (elongation rate) than the ordinary corn germ film. The antimicrobial activity of the film was more than 50% of the maximum activity after film production with heat treatment at $90^{\circ}C$ and pH adjustment to 9. When the corn germ protein film with bacteriocin-like substance was applied on the mashed sausage media containing E. coli, the bacterial growth inhibition was higher than the ordinary corn protein film.

In Vitro Experiment to Evaluate 137Cs Dissolution in the Digestion Process of Mushrooms

  • Background: Several studies have reported that wild mushrooms contain high amounts of radioactive cesium (137Cs). After the Fukushima Daiichi Nuclear Power Plant Accident, a significantly high concentration of 137Cs has been detected in wild mushrooms, and their consumption may be the cause of the chronic internal exposure of local consumers to radioactivity. Therefore, an accurate evaluation of the internal radioactivity resulting from mushroom ingestion is needed. Materials and Methods: The 137Cs elution rate through the cooking and digestion stages was evaluated using in vitro experiments. The edible mushroom Pleurotus djamor was taken as a sample for the experiments. The mushrooms were cultivated onto solid media containing 137Cs. We evaluated the internal dose based on the actual conditions using the elution rate data. For various cooking methods, the results were compared with those of other wild edible mushrooms. Results and Discussion: From the elution experiment through cooking, we proved that 25%-55% of the 137Cs in the mushrooms was released during soaking, boiling, or frying. The results of a simulated digestion experiment after cooking revealed that almost all the 137Cs in the ingested mushrooms eluted in the digestive juice, regardless of the cooking method. The committed effective dose was reduced by 20%-75% when considering the dissolution through the cooking process. Conclusion: We found that cooking lowers 137Cs concentration in mushrooms, therefore reducing the amount of radioactivity intake. Besides, since there were differences between mushroom types, we demonstrated that the internal exposure dose should be evaluated in detail considering the release of 137Cs during the cooking stages.

Physicochemical Requirement for the Vegetative Growth of Schizophyllum commune Collected from Different Ecological Origins

  • Schizophyllum commune is an edible and medicinal mushroom widely distributed in the world. The optimal growth conditions for the mycelia of 10 strains of the fungus were investigated. The temperature suitable for the mycelial growth and density was obtained at $30{\sim}35^{\circ}C$. Among the tested conditions, the minimum mycelial growth was found at $15^{\circ}C$. In case of pH, the most favorable growth was found at pH 5. The results indicated that this mushroom well adapted to high temperature and low pH for its mycelial growth. Considering growth phenotype of mycelia, Hamada, Hennerberg, PDA and YM were the most suitable and Lilly, Glucose triptone, Glucose peptone and Hoppkins were the most unfavorable among tested media for the mycelial growth of S. commune. Out of tested carbon sources, dextrin and fructose were the most suitable and lactose, mannose and sorbitol were the unsuitable for the fungus. Compact mycelial density was obtained from most of the carbon sources. Among used nitrogen sources, calcium nitrate, potassium nitrate and alanine were the most appropriate and the most incompatible were ammonium phosphate, histidine, urea and arginine for mycelial growth of S. commune on the culture media. Calcium nitrate, histidine and potassium nitrate showed moderately thin or thin, and rest of nitrogen sources showed compact or moderately compact mycelial density.

Review of the Current Research on Fetal Bovine Serum and the Development of Cultured Meat

  • The purpose of this review is to summarize studies that investigate blood and the main components of fetal bovine serum (FBS) in vertebrates, including major livestock, and review the current research on commercializing cultured meat. Detailed research on FBS is still lacking; however, some studies have shown that FBS consists of proteins, carbohydrates, growth factors, cytokines, fats, vitamins, minerals, hormones, non-protein nitrogen, and inorganic compounds. However, there are few studies on how the composition of FBS differs from blood or serum composition in adult animals, which is probably one of the main reasons for not successfully replacing FBS. Moreover, recent studies on the development of FBS replacers and serum-free media have shown that it is difficult to conclude whether FBS has been completely replaced or serum-free media have been developed successfully. Our review of the industrialization of cultured meat reveals that many basic studies on the development of cultured meat have been conducted, but it is assumed that the study to reduce or replace ingredients derived from fetuses such as FBS has not yet been actively developed. Therefore, developing inexpensive and edible media is necessary for the successful industrialization of cultured meat.

A. mellea의 균사생장(菌絲生長)에 알맞는 배지(培地)는 MSDS, 온도(溫度)는 $25^{\circ}C$, pH는 $6.2{\sim}7.0$, 배지(培地)의 수분함량(水分含量)은 65% (w/w), 톱밥배지(培地)의 가비중(假比重)은 0.12g/ml이고 종균제조(種菌製造)에 알맞는 배지(培地)는 포플러톱밥 이였다. 2. Lyophyllum sp.의 균사생장(菌絲生長)에 알맞는 배지(培適)는 MSDS, 온도(溫度)는 $25{\sim}30^{\circ}C$, pH는 6.2, 배지(培地)의 수분함량(水分含重)은 $75{\sim}80%$(w/w), 톱밥배지(培地)의 가비중(假比重)은 0.12g/ml이고 종균제조(種菌製造)에 알맞는 배지(培地)는 밀 배지(培地)이였다. 3. A. auricula-judae의 균사생장(菌絲生長)에 알맞는 배지(培地)는 MHS, pH는 8.0, 배지(培地)의 수분함량(水分含量)은 75%(w/w), 톱밥배지(培地) 의 가비중(假比重)은 0.12g/m이고 종균제조(種菌製造)에 알맞는 배지(培地)는 참나무톱방 이였다. 4. 공시균주중(供試菌株中) A. auricula와 Lyophyllum sp.는 포플러톱밥 배지(培地)에서 자실체(子實體)를 얻을 수 있어 식용(食用)버섯으로서 인공재배(人工栽培) 가능성(可能性)이 있었다.

RAPD분석을 통한 여름양송이(Agaricus bitorquis)의 교잡주 선발 (Selection of Agaricus bitorquis hybrid strains based on RAPD analysis)

  • 여름양송이(Agiricus bitorquis)는 Agaricus속에 속하는 버섯으로 일반적으로 양송이가 $16{\pm}2^{\circ}C$에서 발생되는 것과 달리 $25{\pm}1^{\circ}C$에서 자라는 고온성버섯이다. 우리나라에서 여름양송이는 대부분 오랜 기간 동안 재배를 하고 있다. 이에 따라 재배주기가 짧은 양송이 품종을 개발을 위한 교잡균주를 확보하였다. 수집된 균주 중 선발된 여름 양송이균주 ASI 1151와 ASI 1349에서 각각 100개 이상 단포자를 수집하였다. 배지별 단포자 발아시험의 결과에서 보면, CDA배지에서 73개의 단포자들의 발아되었고, 최소기간 20일, 최장기간 83일 후 포자발아가 진행되었다. 발아된 단포자들의 균사생장속도는 각 균주마다 달랐으며, RAPD마커 OPN-2를 가지고 ASI 1151에서 9개, ASI 1349에서 13개 동핵균주를 선발하였다. 또는 이 마커를 이용하여 99개의 교잡 중에 44균주의 친화성 유전자좌를 가진 동핵균주가 서로 교잡한 교잡주를 선발하였다.
