Glutathione peroxidase 3 (GPx3), an antioxidant enzyme, acts as a modulator of redox signaling, has immunomodulatory function, and catalyzes the detoxification of reactive oxygen species (ROS). GPx3 has been identified as a tumor suppressor in many cancers. Although hyper-methylation of the GPx3 promoter has been shown to down-regulate its expression, other mechanisms by which GPx3 expression is regulated have not been reported. The aim of this study was to further elucidate the mechanisms of GPx3 regulation. GPx3 gene analysis predicted the presence of ten glucocorticoid response elements (GREs) on the GPx3 gene. This result prompted us to investigate whether GPx3 expression is regulated by the glucocorticoid receptor (GR), which is implicated in tumor response to chemotherapy. The corticosteroid dexamethasone (Dex) was used to examine the possible relationship between GR and GPx3 expression. Dex significantly induced GPx3 expression in H1299, H1650, and H1975 cell lines, which exhibit low levels of GPx3 expression under normal conditions. The results of EMSA and ChIP-PCR suggest that GR binds directly to GRE 6 and 7, both of which are located near the GPx3 promoter. Assessment of GPx3 transcription efficiency using a luciferase reporter system showed that blocking formation of the GR-GRE complexes reduced luciferase activity by 7-8-fold. Suppression of GR expression by siRNA transfection also induced down-regulation of GPx3. These data indicate that GPx3 expression can be regulated independently via epigenetic or GR-mediated mechanisms in lung cancer cells, and suggest that GPx3 could potentiate glucocorticoid (GC)-mediated anti-infla-mmatory signaling in lung cancer cells.
Objective: Recently the demands for the effective and safe depigmentative and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The purpose of this study is to investigate the MKG(Methanol Extract of Kaempferia galanga) and their dermal bioactivity properties related to cosmeceuticals such as depigmentation. Methods: We assessed inhibitory effects of MKG on melanin production in B16/F10 melanoma cells, on mushroom tyrosinase activity, effects of MKG on the expression tyrosinase, TRP-1, TRP-2, GSK-$3{\beta}$, CREB, MITF in B16/F10 melanoma cells without cytotoxicity range. Cell viability was measured by MTT assay and tyrosinase activity was assessed using by DOPA staining, western-blot analysis. We measured inhibition of melanin synthesis and tyrosinase activity by down-regulation of melanogenic enzyme expressions in ${\alpha}$-MSH induced melanogenesis B16/F10 melanoma cells. Results: MKG inhibited tyrosinase-activity, total melanin contents and dendrite out-growth. MKG inhibited melanogenesis by down-regulation of tyorsinase, TRP-1, TRP-2, CREB, and MITF in B16/F10 cells. The treatment with MKG at the 12.5, $25{\mu}g/ml$ level significantly inhibited the melanin synthesis induced ${\alpha}$-MSH in B16/F10 melanoma cells compared with untreated control. Conclusion: These results suggest that MKG inhibit melanin biosynthesis which is involved in hyper-pigmentation. So MKG is considered to be used as a whitening components reducing cytotoxicity.
ERp29는 endoplasmic reticulum (ER) lumen에 존재하는 단백질로 protein disulfide isomerase (PDI) family에 속한다. 비록 관련 연구 결과는 조금 있지만 정확한 생물학적인 기능은 아직 분명하지 않지만, 분비단백질과정과 단백질 folding에 관여하는 것으로 알려 지고 있다. ERp29의 기능 연구를 위하여 yeast two-hybrid screening/GST pull-down assay방법을 사용하여 ERp29-결합단백질인 ADP-ribosylation factor 5 (ARF5)를 동정하였다. 이들의 결합은 정상적인 세포생리상태에서는 결합하지만 ER stress 상태에서는 떨어졌다. 이 결과는 ERp29의 기능 연구를 위하여 하나의 실마리를 제공할 것이다.
The aging of Korea is rapidly advancing in an unprecedented rate in the world. Indoor and outdoor safety accidents involving the elderly frequently occur, with elderly population increasing. Because the physical strength of elderly is decreased, it is necessary to develop devices that aid the everyday life of the elderly to prevent safety accidents. Among many activities, the motions of standing from and sitting on a chair require much physical strength from particular body parts, which causes an overload for particular body parts. To the elderly the repeating motion of standing from and sitting down on a chair is burdensome, because they do not have enough physical strength compare to young persons. As a result the elderly could be injured or falling during standing or sitting. Therefore, the development of a lift chair that considers the problems mentioned above is necessary to prevent the safety accidents of the elderly. The study designed a non-motorized lift chair that comfortably aids the standing and sitting down motion for the use of the elderly by reflecting the anthropometric data of the Korean elderly over 65 years old. Also the design was implemented to smoothly aid the standing motion by considering the knee angle of the elderly when standing. Because the regained strength to stand from a chair is depend on the knee angle. Unlike existing lift devices, this device utilizes a tilting mechanism that does not use electric power so that the product could be composed inexpensively than existing chairs released in the market.
Cilostazol은 phosphodiesterase III의 선택적 저해제로 알려져 있으며, 뇌졸중 치료에 일반적으로 사용되고 있다. Cilostazol을 처리한 경우, 국소 뇌허혈이 발생한 후에 혈관신생을 통해서 혈관형성이 향상된다는 것을 본 연구자들이 발표하였다. 혈관신생은 조직의 허혈상태를 극복하기 위해서 혈관재생을 촉진하는 중요한 과정으로써, 혈관내피세포의 증식, 이동, 모세관구조 형성의 다단계 과정으로 구성되어 있다. 이에 본 연구에서는 인간 뇌혈관내피세포를 이용하여 cilostazol이 혈관신생의 각 단계들에 어떤 영향을 미치는지 조사하였다. Cilostazol은 농도의존적으로 뇌혈관내피세포의 이동성을 촉진하였으나, 뇌혈관내피세포의 증식과 모세관구조 형성에는 영향을 미치지 않았다. Cilostazol이 세포이동을 조절하는 기전을 분석하기 위해서 cDNA microarray를 수행하였고, 세포이동에 관련성이 있는 5종의 후보 유전자들을 선택하여 real-time PCR을 통해 해당 유전자의 발현을 검증하였다. Cilostazol에 의해서 발현양이 조절되는 유전자들로써, phosphoserine aminotransferase 1 (PSAT1)와 CCAAT/enhancer binding protein ${\beta}$ ($C/EBP{\beta}$)은 발현이 증가하였고, tissue factor pathway inhibitor 2 (TFPI2), retinoic acid receptor responder 1 (RARRES1), RARRES3는 발현이 감소하였다. 이상의 결과를 통해서 cilostazol이 혈관내피세포의 이동을 촉진하여 혈관신생을 향상시킬 수 있음을 제안할 수 있으며, 뇌혈관내피세포에 대한 cilostazol의 조절기전에 대해서 더욱 상세히 규명을 한다면 혈관형성을 통하여 허혈성 질환을 치료할 수 있는 유용한 정보가 될 것으로 기대한다.
최근 국내외에서 도로포장설계법에 경험적/역학적 개념을 적용한 설계법의 개발 및 이의 활용을 위한 연구가 매우 활발하게 진행 중에 있다. 미국의 경우 AASHTO 2002 설계법, 우리나라의 경우 한국형 도로포장설계법의 개발이 진행 중에 있고, 개발되는 설계법에 도로포장재료의 역학적 물성치 평가가 상당히 중요한 역할을 하게 구성되어 있다. 따라서 설계법에 이용될 국내 아스팔트 혼합물의 재료물성의 평가가 매우 시급한 실정이다. 이 재료의 물성치를 평가하는 방법 중 최근에 많이 적용되는 방법이 동탄성계수시험이다. 본 연구에서는 아스팔트 포장체의 시간에 따른 노화특성을 고려한 아스팔트 포장체의 동탄성계수를 평가하였고, 이를 이용하여 동탄성계수 예측방정식을 제안하였다. 시험에 이용된 아스팔트 혼합물은 KS 표준규격에 사용되는 9종(SMA 및 SBS 포함)과 3종류의 아스팔트 바인더를 이용하였다. 보통 공장에서 배합, 생산되어 현장으로 이동하고 다짐이 완료되기까지 걸리는 시간을 고려하면 단기노화조건은 $135^{\circ}C$ 4시간이 적절할 것으로 판단되며, 공정이 빠르고 관리가 잘되는 조건을 반영 한다면 $135^{\circ}C$ 2시간이 적절할 것이다. 장기노화 시험은 공용단계의 아스팔트 바인더의 노화를 시험하였다. 전체적으로 노화가 진행되면서 동탄성계수가 높아짐을 알 수 있다. 최대입경이 클수록 그 정도는 작아지는데 그 이유는 아스팔트 바인더의 효과 보다 골재의 효과가 그 비중이 증가함에 따른 것이다.
일반적으로 노화, 영양부족 및 다양한 만성질환에 의하여 유발되는 근위축은 근육 단백질 합성 억제 및 분해증가를 통하여 근섬유 및 근육의 밀도를 감소시키는 것으로 알려져 있다. 본 연구에서는 근위축과 관련된 in vitro 실험을 위한 C2C12 근아세포에서 근관세포로의 분화과정을 확립하고, 분화가 유발된 C2C12 근관세포를 대상으로 dexamethasone 및 hydrogen peroxide에 의한 근위축 유발 및 관련 단백질들의 발현 변화를 조사하였다. 먼저 C2C12 근아세포에 분화배지를 처리하였을 경우 근관세포로 분화가 유발되었으며, 분화와 관련된 단백질인 myogenin 및 myoD의 발현이 증가하는 것으로 나타났다. 분화가 유발된 C2C12 근관세포에 세포독성이 없는 조건의 dexamethasone 및 hydrogen peroxide를 처리하였을 경우 근관의 지름이 감소하였으며, 이러한 현상은 musclespecific ubiquitin ligases인 MAFbx/atrogin-1 및 MuRF1의 발현 증가와 함께 muscle-specific transcription factor인 myogenin 및 MyoD의 발현 감소와 관련이 있다는 것을 확인하였다. 본 연구 결과는 근위축과 관련된 in vitro 실험 모델의 구축을 위한 최적의 분화조건 확립과 함께 dexamethasone 및 hydrogen peroxide를 근위축 유도제로 사용할 수 있는 가능성 을 제시하는 것이다.
Nucleoside adenosine 유도체의 하나인 cordycepin (3′-deoxyadenosine)은 Cordyceps 속에서 유래된 활성 물질 중의 하나로서 항염증, 항산화 및 항암활성을 포함한 다양한 약리학적 효능이 있는 것으로 잘 알려져 있다. 본 연구에서는 AGS 인체 위암세포의 증식에 미치는 cordycepin의 영향과 관련 기전 연구를 시도하였다. Cordycepin의 처리에 따라 AGS 세포의 생존율이 처리 농도 의존적으로 감소되었으며, DNA 단편화 및 flow cytometery 분석에 따른 apoptosis 유발 또한 유의적으로 증가하였음을 확인하였다. 이러한 cordycepin 처리에 따른 AGS 세포의 apoptosis 유도에는 TRAIL, DR5 및 FasL의 mRNA 및 단백질의 발현 증가가 연관되어 있었다. 아울러 cordycepin은 Bcl-2 family 중 pro-apoptotic 인자인 Bax의 발현은 증가시켰으며, anti-apoptotic 인자인 Bcl-2 및 Bcl-xL의 발현은 전사 및 번역 수준에서 억제시켰다. 이러한 현상들은 extrinsic 및 intrinsic apoptosis의 initiator caspase (caspase-8 및 -9) 뿐만 아니라 effector caspase인 caspase-3의 활성과 PARP 단백질의 절단 증가와 연관성이 있었다. 따라서 AGS 세포에서 cordycepin에 의한 apoptosis의 유발은 death receptor 활성과 mitochondria 기능 손상을 포함한 multiple apoptotic pathway가 관여할 것으로 생각된다. 비록 좀 더 세심한 기전 연구의 결과가 뒤따라야 되겠지만, 본 연구의 결과는 cordycepin의 항암작용을 이해하는데 중요한 자료가 될 것이며 향후 수행될 추가 실험을 위한 기초 자료로서 그 가치가 매우 높을 것으로 생각된다.
신장 질환은 초고령화 사회로 접어들고 있는 현대 사회에서 발병율이 매우 높은 질환이다. 이러한 신장 질환을 초기에 발견하면 약물이나 식이요법으로 막을 수 있지만 악화될 경우 인공투석이나 신장이식을 받아야 한다. 따라서 본 논문에서는 혈액투석에 따른 성대 및 강도의 변화율을 측정, 분석하여 신장과 음성과의 상관성을 분석하였다. 이를 위해 혈액투석 치료를 받는 피실험자를 대상으로 혈액투석 전과 후의 음성을 수집하고 음성 분석학적 요소의 적용을 통해 비교, 분석하였다. 즉, 신장에 이상이 있을 때 목소리가 가라앉는다는 동양의학적 진단 이론에 기반하여 혈액투석 치료 전의 음성과 치료 후의 음성을 각각 수집하고 수집된 음성 자료에 성대의 떨림과 강도 분석을 적용하여 혈액투석 치료 전과 후의 음성 변화를 측정, 분석하였다.
Purpose: The purpose of this study was to recognize the importance of balance in order to minimize daily living restrictions and to prevent falls due to aging, and develop an eight-week theraband exercise program to benefit the elderly. Method: A total of 20 elderly participants were divided into two groups. Group A used warm-up and finishing exercises for eight weeks including stretching and rhythm exercises conducted in a theraband exercise. Group B performed no exercises. Both A and B groups at weeks 1, weeks 4, weeks 8 were measured for deltoid and quadriceps femoris strength, balance and proprioception. Result: There was a significant difference in deltoid and quadriceps femoris muscle strength, balance and proprioception between Group A and Group B(p<0.05). Conclusion: The results of this study show that theraband exercises are thought to be able to contribute to the promotion of deltoid and quadriceps femoris muscle strength, balance and proprioception. It also contributes to minimize activity of daily living restrictions and prevent fall down in elderly. However in this study, generalization has been limited because of exercise duration, limited number of experiments and intensity modulated of theraband.
