Biological and Molecular Characterization of a Korean Isolate of Clover Yellow Vein Virus Infecting Canavalia ensiformis

  • Jack bean (Canavalia ensiformis) is one of healthy products for fermented or functional food in Korea and is widely distributed and cultivated worldwide. During August 2022, Jack bean plants showing symptoms of yellow flecks, chlorosis, necrotic spots and mosaic were observed in Jangheung-gun, South Korea. By transmission electron microscopy, flexuous filamentous virus particles of approximately 750×13 nm in size were observed in the symptomatic leaf samples. The infection of a Korean isolate of clover yellow vein virus (ClYVV-Ce-JH) was confirmed using double antibody sandwich enzyme-linked sorbent assay, reverse transcription polymerase chain reaction and high-throughput sequencing. The complete genome sequence of ClYVV-Ce-JH consists of 9,549 nucleotides (nt) excluding the poly (A) tail and encodes 3,072 amino acids (aa), with an AUG start and UAG stop codon, containing one open reading frame that is typical of a potyvirus polyprotein. The polyprotein of ClYVV-Ce-JH was divided into ten proteins and each protein's cleavage sites were determined. The coat protein (CP) and polyprotein of ClYVV-Ce-JH were compared at the nt and aa levels with those of the previously reported 14 ClYVV isolates. ClYVV-Ce-JH shared 92.62% to 99.63% and 93.39% to 98.05% at the CP and polyprotein homology. To our knowledge, this is the first report of a Korean isolate of ClYVV from Jack bean plants and the complete genome sequence of a ClYVV Jack bean isolate in the world.

Experimental and numerical investigation on the seismic behavior of the sector lead rubber damper

  • Beam-column joints in the frame structure are at high risk of brittle shear failure which would lead to significant residual deformation and even the collapse of the structure during an earthquake. In order to improve the damage issue and enhance the recoverability of the beam-column joints, a sector lead rubber damper (SLRD) has been developed. The SLRD can increase the bearing capacity and energy dissipation capacity, and also demonstrating recoverability of seismic performance following cyclic loading. In this paper, the hysteretic behavior of SLRD was experimentally investigated in terms of the regular hysteretic behavior, large deformation behavior and fatigue behavior. Furthermore, a parametric analysis was performed to study the influence of the primary design parameters on the hysteretic behavior of SLRD. The results show that SLRD resist the exerted loading through the shear capacity of both rubber parts coupled with the lead cores in the pre-yielding stage of lead cores. In the post-yielding phase, it is only the rubber parts of the SLRD that provide the shear capacity while the lead cores primarily dissipate the energy through shear deformation. The SLRD possesses a robust capacity for large deformation and can sustain hysteretic behavior when subjected to a loading rotation angle of 1/7 (equivalent to 200% shear strain of the rubber component). Furthermore, it demonstrates excellent fatigue resistance, with a degradation of critical behavior indices by no more than 15% in comparison to initial values even after 30 cycles. As for the designing practice of SLRD, it is recommended to adopt the double lead core scheme, along with a rubber material having the lowest possible shear modulus while meeting the desired bearing capacity and a thickness ratio of 0.4 to 0.5 for the thin steel plate.

복숭아순나방 성충 복부에서 발현하는 불포화효소의 RNA 간섭과 교미교란 (Mating Disruption of Grapholita molesta by RNA Interference of a Fatty Acid Desaturase Expressed in Adult Abdomen)

  • 복숭아순나방(Grapholita molesta)은 두 가지 주요 성페로몬 성분(Z-8-dodecenyl acetate and E-8-dodecenyl acetate)을 갖고 있다. 이 성페로몬 성분의 생합성 과정 분석은 포화지방산의 10번 탄소에 이중결합을 합성하는 불포화효소($10{\Delta}$ DES)가 종 특이적 광학이성체 형성에 필수적이라고 제시하였다. 그러나 이 효소의 분자적 특징에 대해서 분석되지 않았다. 본 연구는 복숭아순나방 성페로몬 샘의 전사체에서 $10{\Delta}$ DES로 추정된 불포화효소(Gm-comp1575)의 단백질 기능 영역을 분석하였다. Gm-comp1575 유전자는 370개의 아미노산 서열 정보를 암호하고 있으며 분자량은 약 43.2 kDa 그리고 등전위점(pI)은 8.77로 추정되었다. 이 불포화효소는 4개의 막투과영역을 지니고 있으며, 6개의 탄수화물 결합 위치가 아미노 말단과 세포내 영역에서 갖는 것으로 추정되었다. 분자계통분석은 Gm-comp1575가 다른 종에서 알려진 $10{\Delta}$ DES와 유사성이 높은 것으로 밝혀졌다. Gm-comp1575 전사체는 암컷 성페로몬 샘 및 다른 복부 조직에서 발현되었다. 이 유전자 발현에 대한 RNA 간섭 처리는 처녀 암컷으로 하여금 사과원에서 수컷을 유인하는 능력을 크게 감소시켰다. 이러한 결과는 Gm-comp1575가 복숭아순나방의 성페로몬 생합성과 관련이 있는 유전자라고 제시하고 있다.

소포체 스트레스에 대한 Protein Disulfide Isomerase의 세포보호효과 (Bombyx mori Protein Disulfide Isomerase (bPDI) Protects Sf9 Cells from Endoplasmic Reticulum (ER) Stress)

  • bPDI가 ER내 misfolding 단백질의 생성을 제한함으로써 곤충변역과 관계하는지를 해석하기 위하여 bPDI가 과발현(overexpression)되는 곤충세포주와 이와 반대로 bPDI가 억제발현(knock-down)되는 곤충세포주를 제작하여 bPDI가 곤충면역에 관련하는지를 해석하였다. bPDI가 과발현되는 세포주 (Sf9-bPDI)는 정상세포주(Sf9)나 pIZT/V5-His 벡터만 도입된 세포주(Sf9-pIZT)에 비하여 생존율이 30% 이상 높았지만, bPDI의 전사체 발현이 억제된 세포주(Sf9-bPDI-dsRNA)는 오히려 정상세포주나 pIZT/V5-His 벡터만 도입된 세포주에 비하여 생존율이 약 15%낮았다. 이와 같은 결과로써, bPDI는 ER내 misfolding 단백질의 생성을 제한함으로써 곤충의 ERSE과 밀접하게 관련할 것이라 추정할 수 있었다.

2 Cropping systems using field crops in unheated plastic house at paddy field

  • In Korea, the single span unheated plastic house cultivated crops from autumn to spring of the following year, removed the plastic film and frame, cultivated rice, set up a plastic house again and cultivated crops. The crops in the greenhouse are utilized mainly for the production of leaf vegetables such as lettuce, leek, and fruit vegetables such as strawberry, watermelon, oriental melon, etc. and raising high income. Because, the production of these crops has characteristics requiring a lot of labor and it is difficult to produce horticultural crops at unheated plastic houses as the rural population ages. Therefore, we conducted a test to develop a crop planting system to cultivate crops in single span unheated plastic houses, although the utilization of labor is less than that of horticultural crops. The prior cropping cultivated three cultivars of sweet potatoes early, the second produced cultivated sweet potatoes, corn and soybeans. In the cultivation of the previous cropping, the sweet potatoes were harvested on the 113th day after planting on March 30th, the yield was 822 kg/10a for Pungwonmi, 1,377 kg/10a for Jinhongmi, 1,483 kg/10a for the Dahomi. Because of differences, the yield of Pungwonmi cultivar was less than that other cultivars and the yield of open field cultivations, we will expect further research. In the cultivation of the succeeding crops sweet potatoes were planted on July 27 and harvested 110 days later and investigated. The product yield of Pungwonmi cultivar was 1,024 kg/10a, and the Jinhongmi, Dahomi cultivars were not at economic level for sale and were necessary to review. In succeeding-crops, corn tested the Ilmichal cultivar, seeded on 27th July, harvested on October 11th. The day of silking was 45 days after sowing, the yield was 1,156 kg/10a, the goods rate was 100% level. The beans in the succeeding cultivation crop were sowed on 27th July, the early maturing of the varieties coming to Hwangeumol and Saeol cultivar, on 17th October, the late maturing soybean Daewonkong cultivar were harvested on October 21st. The yield of early maturing two cultivars was 214 kg/10a, Daewonkong was 257 kg/10a, and 100 seeds weight which were more than the early maturing beans were also heavy. When calculating these incomes price-wise according to the harvest time, we were able to consider the income in the order of corn, sweet potato and soybean from the second term crop. Various studies such as varieties, mulching method, moisture management, control environment management, etc. are considered necessary to develop cropping systems with sweet potato and field crops in future unheated plastic house.

석조유구(石造遺構)를 통한 한국(韓國) 고대건축(古代建築)에 관한 연구(硏究) -삼국시대와 통일신라시대를 중심으로- (A Study on the Ancient Architecture in view of the Stone Remains (focused on the 3 Kingdom Period and Unificated Shilla Period))

  • The purpose of this study is to analogize the appearance of Korean Ancient Architecture in view of the Stone Remains from 3 Kingdom Period to Unificated Shilla Period. But in these period, there is no building remains but some stupas and fine arts. Especially, there are many architectural appearance and revealing signature in these Historical Stone remains. Architectural elements which are analogized by stone remains what has value as historical materials by preservation of original form from 3 Kingdom Preiod to Unificated Shilla Period are as follows : 1) Platform, the representative characteristic of Korean traditional architecture, was frame structure and accumulate structure. And circular or square footing stood a same shape column on it is put on the platform. 2) In the case of column, there used entasis column and inclined column and circular chamfer technique was applied on the top side of it. Upper side of column, capital and head pentrating tie that small bearing block was put on the center of it was joined. And longitu야nal rest(長舌) supported a cross beam. Capital and small bearing block had no bottom heel, and heel side was curved and straight. Centered bracket structure was often used, and multi bracket structure is not used yet. Inward incline technique was used. 3) Inward opening pair door which had lintel, threshold, doorjamb was usually used, Fixing stone was used for structural safety, and circular handle and lock was used for decoration. Handrail was used on the edge of wooden floor for decorative effect and safety. 4) Square rafter and circular rafter were used in the same period and so did flying rafter. Double eaves and single eave were used in the same period but, single eave was usually used. In this period, square rafter was usually used. This would be studied more by comparing with Japanese wooden architecture. 5) Hipped roof was used and half-hipped roof was not used yet. In front of th hip, there are small sculpture called Jap-Sang(雜像), and windbell was hang on the end of the hip rafter. Concave roof tile, convex roof tile, round eaver tile, decorative tile at end of roof ridge were used. Lotus style was well used on the face of roof tile for decoration. From the results of this study, wooden architecture of Unificated Shilla period was simple compare to Koryo dynasty and Chosun dynasty but, it had some brilliant character. It was hard work that analogized the form of non-existent wood architecture of Ancient Korean period by restricted stone remains. But, in addition to the results of this study and research of old documentations, more study should be go on.

애니메이션 창세기 첫 페이지의 오류, 그 기원과 수정 (Desperately Seeking an Icon (with 8 Legs):)

  • 알타미라 동굴의 다리 여덟 달린 멧돼지 벽화는 움직임을 표현하고자 하는 인류의 오랜 욕망을 보여주는 예로 제시되곤 한다. 그러나 여덟 개의 다리는 서로 다른 시기에 그려진, 이른바 더블 레이어의 결과물이다. 그럼에도 불구하고 알타미라 동굴벽화를 애니메이션의 시원과 결부시키는 설명은 별다른 의문이나 검증, 출처의 인용 등이 없이 꾸준히 재생산되고 있다. 여러 개의 다리를 움직임과 연결시키는 것은 시대를 초월한 시지각과는 무관하다. 그것은 특정한 시기에 발명된 움직임 표현 코드로 보아야 한다. 여기에는 19세기 후반에 시도된 연속촬영술, 그 중에서도 에티엔 쥘 마레의 시간사진술이 결정적인 역할을 한다. 일련의 연속적인 움직임들을 하나의 프레임 속에 겹쳐서 인화한 마레의 사진술은 20세기 초에 뒤샹과 발라를 비롯한 아티스트들의 회화 작업에 반영되면서 움직임 표현 코드로 형성되었다. 그 무렵부터 등장하기 시작한 애니메이션 매뉴얼들도 움직임을 분석하고 구현하기 위한 방법으로 마레의 시간사진술 이미지를 도입하였다. 결국 다리 여덟 개의 알타미라 동굴벽화를 움직임의 표현이라고 이해하는 것은 오늘날의 시각 코드를 통해 과거를 바라보고자 하는 오류이다.

Citrus reticulata 감과 과피 내 미세구조 변화 (Sequential Changes of Pericarp Ultrastructure in Citrus reticulata Hesperidium)

  • Citrus reticulata 감과 과피조직 내에서 탈리를 전후하여 신속하게 진행되는 특정 미세구조의 분화양상을 전자현미경적으로 연구하였다. 특히, 색소체 전환이 진행되어 뚜렷한 외과피층을 이루는 후각세포 내에서 일어나는 미세소기관 분해물질의 형성 및 특성에 초점을 두어 연구하였다. 색 변화가 수반된 발달 중의 외과피 유세포들은 세포벽이 비후되면서 1차공역이 잘 발달하는 후각세포로 분화하며, 대부분의 색소체는 유색체로 전환되어 티라코이드는 거의 소실되고 지질입자를 다량 함유하는 불규칙적인 형태로 변형되었다. 이때 세포막이나 액포막에서는 국부적으로 형성된 수많은 소낭들이 집적되어 세포질 내부로 이동하였고, 세포간 1차공역에 발달하는 원형질연락사를 통한 소낭의 집적현상 또한 빈번히 일어났다. 집적된 소낭들은 융합되어 전자밀도가 높은 구형 또는 일정한 형태가 없는 구조(EDB)로 되며, 이동하여 엽록체, 미토콘드리아 등의 주요 세포소기관을 점차 포위하였다. 소액포들은 융합하여 거대액포가 되며, 세포질 및 EDB에 포위된 소기관들은 점차 분해되기 시작하였다. EDB는 리파제 확인실험에 의해 지질성 물질로 이루어진 구조로 확인되었다. 과피발달 초기에 선행된 중과피 분화는 세포 내에서 급격히 진행된 용해현상에 의해 수많은 소액포들로 이루어진 거대한 세포 공강을 형성하며 밀착되어 불규칙적인 세포벽과 함께 해면성으로 되었다. 이와 같이 본 연구에서는 감과가 식물체에서 분리되기 직전에서부터 탈리 후까지 일어나는 과피의 발달 및 노화현상을 추적하였다.

Peirce의 귀추법 양식을 이용한 교육 대학생들이 생성한 가설의 특징 분석 (The Features of the Hypotheses Generated by Pre-service Elementary Teachers Using the Form of Peirce's Abduction)

  • 본 연구는 '가설 생성 과정에서 귀추법 사용을 유도하는 틀', 즉 '귀추법 양식'을 고안하고, 그 틀을 사용하여 생성한 가설의 특징을 틀을 사용하지 않고 생성한 가설의 특징과 비교하여 분석하는 것을 목적으로 하였다. '귀추법 양식'은 Peirce가 제시한 귀추법의 의미와 형식에 관한 고찰을 통해 (i) 의심의 대상을 명기하는 단계, (ii) 의심의 대상을 믿음의 대상이 되게 하는 설명을 명기하는 단계, 그리고 (iii) 그 설명을 가설로 명기하는 단계로 구성되었다. 예비 교사 34명을 대상으로 하여 동일한 대상에게 두 가지 방법을 모두 사용하게 한 결과, 고안된 '귀추법 양식'을 사용하여 가설을 생성한 경우는, 첫째, 가설의 의미를 혼동하거나 충실히 반영하지 못한 경우로 볼 수 있는 유형들이 전혀 발견되지 않거나 상대적으로 적었고, 둘째, '부분적으로 유사성을 지닌 다른 현상이나 상황에 관한 기존 지식으로부터 유추하여 문제 상황의 원인에 관해 설명하는 유형'의 비율이 특정한 틀을 제공하지 않은 경우에 비해 두 배 이상 높았다. 결국, '귀추법 양식'을 사용하여 생성된 가설은 그렇지 않은 경우보다 상대적으로 '현상의 원인에 관한 설명과 지식의 확장 초래'라는 가설의 본래적 의미에 좀 더 충실한 특징을 가지고 있었다. 이는 '귀추법 양식'이, 그 형태가 비교적 간단함에도 불구하고, 과학 수업 현장에서 바람직한 형태의 가설이 생성되도록 돕는 하나의 방안이 될 수 있음을 보여주는 것이기도 한다.

NDP Kinases Suppressed Bax-Dependent Apoptosis in Yeast System

  • Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic reaction follows a ping-pong mechanism in which the enzyme is transiently phosphorylated on a histidine residue conserved in all nucleoside diphosphate kinases. Beside their role in nucleotide synthesis, these enzymes present additional functions, possibly independent of catalysis, in processes such as differentiation, cell growth, tumor progression, metastasis and development. To clone murine nm23-M5, several expressed sequence tags (ESTs) of the GenBank data base, selected according to their homology to nm23-H5 cDNA, reconstituted a complete open reading frame (GenBank AF222750). To test whether murine NDPKs (1, 2, 3, 4, 5, and 6) can inhibit Bax-mediated toxicity in yeast, co-transformation was performed respectively. The yeast S.cerevisiae was transformed with a copy expression plasmid containing the histidine selection marker and expressing murine Bax under the control of a galactose-inducible promoter. Several clones were selected and found to be growth inhibited when Bax expression was induced with galactose. A representative clone was transformed again with a copy expression plasmid containing the tryptophane selection marker and expressing either murine Bcl-xL or NDPK under the control of a galactose-inducible promoter. Several subclones of the double-transformants were selected and characterized. The ability of Bcl-xL and NDPKs to suppress Bax-mediated toxicity was determined by growing yeast cells overnight in galactose media and spot-testing on galactose plates starting with an equal number of yeast cells as determined by taking the OD$_{600}$. Ten-fold serial dilutions were used in the spot-test. Plates were grown at 3$0^{\circ}C$ for 2-3 days. All murine NDPKs suppressed Bax dependent apoptosis. Futher study will be peformed whether Bax-toxicity inhibition was caused by NDP kinase activity or additional function.n.
