Growth Effect by Storage Temperature, Soil Type and Treatment Chemical of Saururus chinensis (Lour.) Baill. (삼백초의 종근 저장온도, 용토 및 처리물질에 따른 생육특성)

  • Background: The objective of the present study was to investigate the effect of storage conditions on the growth of Saururus chinensis (Lour.) Baill. rootstock. Methods and Results: Rootstocks of S. chinensis were stored in either soil or vermiculite that had been treated with a control treatment, diluted wood vinegar (50 or 100-fold), DF-100 (50-fold), or 1-naphthylacetamide and at $5^{\circ}C$ or $15^{\circ}C$. After 8 weeks, the stored roots were planted in the field, and both plant height and leaf number were observed after transplantation. The greatest number of leaves ($5.60{\pm}0.80$) was produced by roots that had been stored in soil treated with 100 fold dilution of wood vinegar and at $5^{\circ}C$. Meanwhile the maximum plant height ($6.92{\pm}0.78cm$) at 30 d after transplanting was observed for rootstocks that had been stored in soil treated with the 100 fold dilution of wood vinegar and at $15^{\circ}C$, whereas the maximum plant height at 60 d after transplanting ($26.46{\pm}0.71cm$) was observed for rootstocks that had been stored in soil treated with the 100-fold dilution of wood vinegar and at $5^{\circ}C$. Therefore, the storage of rootstocks in soil treated with the 100-fold dilution of wood vinegar and at temperatures at or below $5^{\circ}C$ was most effective, and it can be used to prevent the decay of roots during the postharvest management of S. chinensis rootstocks. Conclusions: The results of the present study indicate that, among the parameters examined, the storage of roots in soil that had been treated with the 100-foil dilution of wood vinegar is the most effective method for improving the growth of S. chinensis.

Investigation for Streamflow Measurement of Small Stream using Dilution Discharge Method during Low Flow Season (갈수기 소하천에서 희석유량법을 이용한 유량측정의 적용성 검토)

  • In this study, dilution discharge method to measure streamflow using water quality data at small streams during low flow season was applied and compared with the conventional method using flowmeter combined with cross-sectional measurement. Streamflow were measured using both methods in Dal-cheon around Naesok Wastewater Treatment Plant located in Boeun, Chugbuk. This stream is reported to suffer from excessive algal growth in spring. An average flowrate difference of $0.004m^3/sec$ was obtained between the two methods. Once further tested for various conditions, the simple water quality method suggested in this study can be used effectively to estimate stream flowrates during low flow seasons where no measuring facilities such as weirs and flowmeters are not available or water depths are too small to measure flow velocities and exact cross-sections.

Direct Quantitation of Amino Acids in Human Serum Using a Stepwise-Dilution Strategy and a Mixed-Mode Liquid Chromatography-Tandem Mass Spectrometry Method

  • A quantitation method for free amino acids in human serum was developed using a stepwise-dilution method and a bimodal cation exchange (CEX)/hydrophilic interaction liquid chromatography (HILIC)-tandem mass spectrometry system equipped with an electrospray ionization source (ESI/MS/MS). This method, which was validated using quality control samples, was optimized for enhanced selectivity and sensitivity. Dithiothreitol (DTT) was used as a reducing agent to prevent the oxidation of a serum sample ($50{\mu}L$), which was then subjected to stepwise dilution using 3, 30, and 90 volumes of acetonitrile containing 0.1% formic acid. Chromatographic separation was performed on an Imtakt Intrada Amino Acid column ($50mm{\times}3mm$, $3{\mu}m$) in mixed mode packed with CEX and HILIC ligands embedded in the stationary phase. Underivatized free amino acids were eluted and separated within 10 min. As a result of the validation, the precision and accuracy for the inter- and intraday assays were determined as 2.11-11.51% and 92.82-109.40%, respectively. The lowest limit of quantification (LLOQ) was $0.5-4.0{\mu}g/mL$ and the matrix effect was 80.22-115.93%. The proposed method was successfully applied to the quantitative analysis of free amino acids in human serum.

Development of an Isotope-Dilution Flow-Injection Electrospray/ Mass Spectrometric Method for the Accurate Determination of Glucosamine in Pharmaceutical Formulation

  • An isotope-dilution flow-injection electrospray/mass spectrometric method was developed for the accurate determination of glucosamine contents in pharmaceutical formulations. Samples were extracted by methanol. After spiking glucosamine-1-$^{13}C_1$ as an internal standard, the extracts were then analyzed by flow-injection ESI/MS in a selected ion monitoring (SIM) mode to detect [M+H]$^+$ ions of the analyte and its isotope analogue at m/z 180 and m/z 181, respectively. Confirmatory measurements were made by selectively monitoring the collisionally induced dissociation channels of m/z 180 $\rightarrow$ m/z 72 and m/z 181 $\rightarrow$73, respectively, to test the possibility of bias in the SIM method due to matrix interferences, but any significant bias in the SIM mode was not observed. Repeatability and reproducibility studies showed that the flow-injection ESI/MS method is a reliable and reproducible method which can provide a typical method precision of 1.0 %. Other results for the method validation are reported.

  • An isotope dilution-liquid chromatography/tandem mass spectrometric method was developed as a candidate reference method for the accurate determination of folic acid in infant milk formula. Sample was spiked with 13C5-folic acid and then extracted with phosphate buffer (pH 6) solution. The extract was further cleaned up by deproteinization followed by a C18 solid-phase extraction cartridge. The extract was analyzed by using LC/ ESI/MS/MS with selectively monitoring the collisionally induced dissociation channels of m/z 442 → m/z 295 and m/z 447 → m/z 295, which are the neutral glutamyl loss from the [M+H]+ ions of folic acid and 13C5-folic acid, respectively. LC/MS/MS chromatograms showed substantially reduced background from chemical noises compared to LC/MS chromatograms. Repeatability and reproducibility studies showed that the LC/MS/ MS method is a reliable and reproducible method which can provide less than 1.5 relative percentage of method precision.

  • Acetaminophen (N-acetyl-p-aminophenol) is one of the most popular analgesic and antipyretic drugs. An isotope dilution mass spectrometric method based on LC/MS was developed as a candidate reference method for the accurate determination of acetaminophen in pharmaceutical product. After spiking an isotope labeled acetaminophen (acetyl-$^{13}C_2$, $^{15}N$-acetaminophen) as an internal standard, tablet extracts were analyzed by LC/MS in a selected reaction monitoring (SRM) mode to detect ions at m/z $152{\rightarrow}110$ and m/z $155{\rightarrow}111$ for acetaminophen and acetyl-$^{13}C_2$, $^{15}N$-acetaminophen, respectively. The repeatability and reproducibility of the developed ID/LC-MS method were tested for the validation and assessment of metrological quality of the method.

Antimicrobial Activity of Houttuynia cordata Ethanol Extract against Major Clinical Resistant Microorganisms (주요 임상 내성균에 대한 어성초 에탄올 추출물의 항균효과)

  • The increase in resistance by pathogenic bacteria to multiple antimicrobial agents has become a significant treat, as the effective antimicrobial agents available for the patients infected by such resistant bacteria are reduced, or even eliminated. Several natural plant extracts have exhibited antibacterial and synergistic activity against various resistant microorganisms. Houttuynia cordata is frequently used by many traditional medicine practicioners for its antimicrobial, antiviral, and anti-inflammatory properties. This study investigated the antibacterial effects of H. cordata extract against clinical multi-resistant bacteria, and compared the two methods used for the antimicrobial susceptibility testing. Thirty isolates of Methicillin-resistant Staphylococcus aureus (MRSA, 10), Vancomycin-resistant Enterococcus faecium (VRE, 10), Carbapenem-resistant Acinetobacter baumannii (CRAB, 10) were included in this study. The antibacterial effect of H. cordata was tested by disk diffusion and microbroth dilution methods as per CLSI guidelines. In disk diffusion, all isolates (30) showed no inhibition to 30,000 ug/mL of H. cordata. But in the microbroth dilution method, $MIC_{90}$ of H. cordata was 4,096 ug/mL, 8,192 ug/mL and 4,096 ug/mL in MRSA, VRE and CRAB, respectively. These results demonstrate that H. cordata exhibits antibacterial activity against MRSA, VRE and CRAB. Moreover, the microbroth dilution method is a more effective method than disk diffusion to evaluate the antibacterial activity of natural products. The Disk diffusion method used to evaluate the antibacterial activity of natural products required new standard guidelines including inoculum concentration of bacteria.

  • -ray fluorescence analysis of $Ta_2O_5$, $Nb_2O_5$ and $ZrO_2$ in tin-slag samples using the simple dilution method was studied. The method is to correct mathematically the calibration curve to the linear line by the dilution. One synthesized standard having similar composition to the sample and tin slag samples were diluted with anhydrous $Li_2B_4O_7$ at the level of 1%, 2% and 3% of the sample content respectively. The diluted samples were fused at $1150^{\circ}C$ for 30 minutes and these glass beads were finely ground and pelletized. Measuring the X-ray intensities with these pellets, analytical results were calculated by the equation derived from J. Scherman's equation for the characteristic X-ray intensity of an element. Analytical results agreed with the reference values obtained by the standard calibration method within allowable error range and were reproducible.

Antimicrobial Resistance of Seventy Lactic Acid Bacteria Isolated from Commercial Probiotics in Korea

  • In this study, lactic acid bacteria were isolated from 21 top-selling probiotic products on Korean market and their antimicrobial resistance were analyzed. A total 152 strains were claimed to be contained in these products and 70 isolates belonging to three genera (Bifidobacterium, Lactobacillus, and Lactococcus) were obtained from these products. RAPD-PCR showed diversity among isolates of the same species except for two isolates of Lacticaibacillus rhamnosus from two different products. The agar dilution method and the broth dilution method produced different MICs for several antimicrobials. With the agar dilution method, five isolates (three isolates of Bifidobacterium animalis subsp. lactis, one isolate of B. breve, one isolate of B. longum) were susceptible to all nine antimicrobials and 15 isolates were multi-drug resistant. With the broth microdilution method, only two isolates (one isolate of B. breve and one isolate of B. longum) were susceptible while 16 isolates were multi-drug resistant. In this study, only two AMR genes were detected: 1) lnu(A) in one isolate of clindamycin-susceptible and lincomycin-resistant Limosilactobacillus reuteri; and 2) tet(W) in one tetracycline-susceptible isolate of B. longum B1-1 and two tetracycline-susceptible isolates and three tetracycline resistant isolates of B. animalis subsp. lactis. Transfer of these two genes via conjugation with a filter mating technique was not observed. These results suggest a need to monitor antimicrobial resistance in newly registered probiotics as well as probiotics with a long history of use.

The Effects of Supplementation of BSA or Fatty Acid Free BAS on the Motility of Fresh or Cryopreserved Rooster Spermatozoa (BSA 또는 Fatty Acid Free BSA 첨가가 닭 희석 정자와 동결 정자의 생존성에 미치는 영향)

  • This study examined factors affecting the analysis of motility of chicken semen. The viability of spermatozoa was estimated using varying dilution ratios and supplementation with BSA or fatty acid free (FAF)-BSA as protein sources in semen diluent. Fresh semen was examined after preparing dilutions in beltsvile poultry semen extender (BPSE) of 1/8, 1/16 and 1/32 at $25^{\circ}C$. The motility of incubated semen at each dilution was observed at 3 min (89.9%, 69.9% and 53.2%), 30 min (86.7%, 71.4% and 51.7%), 1 h (89.5%, 74.0% and 53.5%) and 3 h (78.5%, 66.5% and 45.7%), respectively. The addition of BSA or FAF-BSA to BPSE diluent significantly increased the viability of semen in 1/32 dilution with results of 53.2% (control), 84.8% (BSA) and 92.9% (FAF-BSA) (p<0.05). This phenomenon was also observed in the dilution of frozen semen, where FAF-BSA treatment increased the viability of thawed semen from 17.6% to 34.0% in a 1/8 dilution (p<0.05). When the protein sources were used in the dilution, the survival rates of diluted chicken semen were also increased with time lapse. These results show that FAF-BSA may act to protect chicken semen and is suitable as a basic component of chicken semen diluent for the method of analyzing rooster semen after freezing.
