Removal and Inactivation of Viruses during Manufacture of a High Purity Antihemophilic Factor VII Concentration from Human Plasma

  • The purpose of this study was to examine the efficacy and mechanism of the cryo-precipitation, solvent/detergent (S/D) treatment, monoclonal anti-FVIIIc antibody (mAb) column chromatography, Q-Sepharose column chromatography, and lyophilization involved in the manufacture of antithemophilic factor VII(GreenMono) from human plasma, in the removal and/or inactivation of blood-borne viruses. A variety of experimental model viruses for human pathogenic viruses, including the bovine viral diarrhoea virus (BVDV), bovine herpes virus (BHV), murine encephalomyocarditis virus (EMCV), and porcine parvovirus (PPV), were all selected for this study. BHV and EMCV were effectively partitioned from a factor VII during the cryo-precipitation with a log reduction factor of 2.83 and 3.24, respectively. S/D treatment using the organic solvent, tri(n-butyl) phosphate (TNBP), and the detergent, Triton X-100, was a robust and effective step in inactivating enveloped viruses. The titers of BHV and BVDV were reduced from the initial titer of 8.85 and $7.89{log_10} {TCID_50}$, respectively, reaching undetectable levels within 1 min of the S/D treatment. The mAb chromatography was the most effective step for removing nonenveloped viruses, EMCV and PPV, with the log reduction factors of 4.86 and 3.72, respectively. Q-Sepharose chromatography showed a significant efficacy for partitioning BHV, BVDV, EMCV, and PPV with the log reduction the log reduction factors of 2.32, 2.49, 2.60, and 1.33 respectively. Lyophilization was an effective step in inactivating g nonenveloped viruses rather than enveloped viruses, where the log reduction factors of BHV, BVDV, DMCV, and PPV were 1.41, 1.79, 4.76, and 2.05, respectively. The cumulative log reduction factors of BHV, BVDV, EMCV, and PPV were ${\geqq}$11.12, ${\geqq}$7.88, 15.46, and 7.10, respectively. These results indicate that the production process for GreenMono has a sufficient virus-reducing capacity to achieve a high margin of the virus safety.

Comparative Evaluation of Washing Methods of Chinese Cabbages for Eliminating the Parasite Eggs in the Preparing Kimchi (김치 제조시 기생충란 제거를 위한 배추 세척방법의 비교평가)

  • Kimchi is one of the representative traditional foods in Korea, which is recognized as a kind of wellbeing foodstuffs. However, as increasing foreign-made Kimchi, its safety as food is at the forefront of public health concerns. We analyzed the washing methods of Chinese cabbages, which are the main materials in Kimchi, to prevent parasite contamination during preparing it. To decontaminate parasite eggs from Chinese cabbages, discard the discolored outer leaves from cabbages, spread the space of leaves with fingers and rinse more than 3 separate water tanks with streaming water (velocity more than 0.8m/sec). At each tank, Chinese cabbages were rinsed with more than 3 strokes upward and downward within the streaming water followed by moving back and forth more than 3 times, with 20cm in height and 30cm in width, respectively. Decontamination efficiency increased higher in parallel with streaming velocity of water, and with adding the vegetable detergent to the tank water.

Antimicrobial Activity of GC-l00X against Major Food-Borne Pathogens and Detaching Effects of It against Escherichia coli O157:H7 on the surface of Tomatoes (GC-100X의 주요 식품위해 미생물에 대한 항균효과와 토마토 표면에 부착된 Escherichia coli O157:H7에 대한 세척 효과)

  • GC-l00X is non-corrosive alkaline ionic water (pH 12). It is composed of hydroxyl radicals and supplemented with xylitol. Its antimicrobial activity was examined against 6 major food-borne pathogens; Staphylococcus aureus FRI 913, Salmonella enterica serova Enteritidis ATCC 13076, S. enterica serova Typhimurium Korean isolate, Vibrio parahaemolyticus ATCC 17803, Escherichia coli O157:H7 ATCC 43894 and Pseudomonas aeruginosa KCTC 1637 at three different temperatures (4$^{\circ}C$, $25^{\circ}C$ and 36$^{\circ}C$) with or without an organic material (2% yeast extract), respectively. The antimicrobial activities showed over 4 log-reductions (1.0$\times$10$^4$CFU/ml reduction) against all pathogens reacted at 37$^{\circ}C$ for 3 hours in the absence of the organic material. The activities showed same results when GC-l00X was diluted with same volume of distilled water or standard hard water (CaCO$_3$300 ppm). Its antimicrobial activity was more effective and quicker in Gram-negative bacteria than Gram-positive bacteria. Its washing efficacy against E. coli O157:H7 exposed to the surfaces of tomatoes (grapes) was compared with that of the other sanitizers such as other kitchen synthetic detergent and 100-ppm chlorine water. For the toxicological evaluation of the sanitizers, viable counts of E. coli O157:H7 penetrated into the core of tomatoes after washing products were also compared. The result revealed that GC-100X stock solution and its 5% diluted solution had similar washing effects to 100-ppm chlorine water and more effective than the other kitchen synthetic detergent. This result indicated that GC- l00X had antimicrobial activity and no toxicological side effects, therefore, could be useful for a new sanitizer to use in flood safety and kitchen hygiene.

Cleaning Methods to Effectively Remove Peanut Allergens from Food Facilities or Utensil Surfaces (식품 시설 또는 조리도구 표면에서 땅콩 알레르겐을 효과적으로 제거하는 세척 방법)

  • Peanut is a well-known food allergen that causes adverse reactions ranging from mild urticaria to life-threatening anaphylaxis. Consumers suffering from peanut allergies should thus avoid consuming undeclared peanuts in processed foods. Therefore, effective cleaning methods are needed to remove food allergens from manufacturing facilities. To address this, wet cleaning methods with washing water at different temperatures, abstergents (peracetic acid, sodium bicarbonate, dilute sodium hypochlorite, detergent), and cleaning tools (brush, sponge, paper towel, and cotton) were investigated to remove peanuts from materials used in food manufacture, including plastics, wood, glass, and stainless steel. Peanut butter was coated on the surface of the glass, wood, stainless steel, and plastic for 30 min and cleaned using wet cleaning. The peanut residue on the cleaned surfaces was swabbed and determined using an optimized enzyme-linked immunosorbent assay (ELISA). Cleaning using a brush and hot water above 50℃ showed an effective reduction of peanut residue from the surface. However, removing peanuts from wooden surfaces was complicated. These results provide information for selecting appropriate materials in food manufacturing facilities and cleaning methods to remove food allergens. Additionally, the cleaning methods developed in this study can be applied to further research on removing other food allergens.

Cleaning Validation Studies for Multi-Purpose Facility : Vial Filling Machine (다품목 공용 제약설비인 바이알 충전기에 대한 세척공정 밸리데이션)

  • The purpose of this study is to evaluate the efficacy of stipulated cleaning process, and the prohibition of cross-contamination and microbiological contamination, which inadequate cleaning in multi-production could occur, through cleaning validation of multi-purpose facility used to produce five biopharmaceutical products as sterile injection. After production of five biopharmaceutical products such as hGH, rhGCSF, rhEPO, rhFSH and rhIFN using vial filling machine, the cleaning validation such as residual analysis of active ingredients or human serum albumin, measurement of total organic carbon (TOC), residual analysis of detergent and microbiological contamination were carried out. In the case of rhGH and rhGCSF clean validations, drug residues were not detected. Furthermore, in the case of rhEPO, rhFSH and rhIFN clean validations, human serum albumin residues were not detected. At TOC (total organic carbon) analysis, all clean validations gave the TOC of about average 137.93%, not more than 150% of acceptance criteria. At sodium analysis for the checking of residues of cleaning agent, sodium residues were not detected. In sterility test, they showed no microbiological contamination of bacteria and fungi. Thus, this cleaning validation was determined as successful in protection of cross-contamination and induction of safety in multi-purpose facility.

Feasibility Study on the Use of Liposomes for Detecting Food-borne Pathogenic Bacteria (식중독 세균 검출에 있어서 리포좀의 이용 가능성)

  • Feasibility tests on using liposomes for detecting food-borne pathogenic bacteria were studied with E. coli 0157:H7 as a model analyte. lmmunoliposomes, whose surface was conjugated with anti-E. coli 0157:H7 IgG and which encapsulated the marker dye, sulforhodamine B, were used for the detection label. Among the feasibility tests, the first test was to use a test-strip on which antibodies to anti-E. coli O157:H7 IgG were immobilized. In this format, immunoliposomes that did not bind to E. coli O157:H7 in sample were captured and then exhibited a visible signal which was inversely related with the number of E. coli O157:H7 in sample. The second test was a direct liposome assay followed by immunomagnetic separation. In this format, immunoliposomes which were bound to E. coli O157:H7 were lysed with detergent and produced a signal which was proportionally related with the number of E. coli O157:H7 in sample. The results from both formats indicate that liposomes can be utilized as a detection label.

Examination of Microbiological Contamination of Ready-to-eat Vegetable Salad (즉석 섭취 야채샐러드의 미생물 오염조사)

  • 120 samples of ready-to-eat salad product were purchased at department stores, marts and family restaurants in metro area. Coliform bacteria and food borne pathogenic bacteria were isolated from these samples. In 73 samples among the 120 salad product samples, coliform bacteria and food borne pathogenic bacteria were detected by 60.8% of isolated rate. Salad were classified into organic and non-organic salad. According to a salad type, salad were classified into vegetable salad and mixed vegetable salad with fried chicken and extra food. According to a packing type, packed salad product and salad-bar product were classified. After the classification, the results of each cases were compared. There is no statistical relation between cultivation or packing methods and contaminated bacteria. But the incidence number of microbial strains was significantly different between vegetable salad and mixed vegetable salad(p<0.005). In vegetable salad, more various strains were detected. E. coli was isolated in 10 cases among the 90 cases in non-organic vegetable and in 7 cases among the 30 cases in organic salad. Food borne pathogenic bacteria were isolated in non-organic vegetable salad product. Staphylococcus aureus was isolated in 4 cases of vegetable salad product and Salmonella spp. isolated in 1 case. After 5 times examination of each 4 market products, the total number of aerobic bacteria was average 4.8$\pm$0.19 log cfu/g. One sample from this product, saline and a detergent for vegetable were used for 3 minutes to notice the effect. As a result, when saline was used 5 times and detergent for vegetable was used 1 time, bacterial contamination was decreased up to 95.5%.

Studies on Growth, Forage Yield, and Nutritive Value according to Different Seeding Dates of Barnyard Millet (파종기에 따른 사료용 피의 생육, 수량 및 사료가치에 관한 연구)

  • These experiments are being conducted to obtain data of plant height, panicle length, heading date, number of culm, and fresh and dry weight, crude protein, acid detergent fiber (ADF), neutral detergent fiber (NDF), total digestible nutrients (TDN) by six times seeding of 10 days interval ranging from May 1st to June 21st for two types of barnyard millet from the Suwon area in Korea. In the early-heading type, IT 170609 (Echinochloa crus-galli), the plant height and the number of culm per square meters are decreased by late seeding relatively, and heading date is 7 to 10 days earlier than the late seeding. And the dry weight of forage is rapidly decreased after the June 11th seeding. In the late-heading type, IT 195422 (Echinochloa frumentacea), the plant height and the number of culm per square meters are also decreased by late seeding relatively, and heading date is about 13 days earlier than late seeding. The dry weight of forage was rapidly decreased after the June 11th seeding. In both two types, the nutritive value of ADF, NDF, and TDN are not changed by late seeding, but crude protein is increased by late seeding. The TDN yields of late-heading type, IT 195400, show average amounts in the June 1st seeding, but the TDN yield of early-heading type, IT 170606, indicate safety amounts of dry weight to the March 21st seeding. And these data are available for cropping systems, cultivating barnyard millet in summer and cereals for forage in winter, to get high production of forage in Korea.

Biological Hazard Analysis of Paprikas, Strawberries and Tomatoes in the Markets (유통중인 파프리카, 딸기 및 토마토의 생물학적 위해요소 분포 조사)

  • The consumption of "ready-to-eat" agricultural products is recently increasing and the safety of these agricultural products is forefront of public concerns. The 120 samples of paprikas, strawberries and tomatoes, which are the representative exported agricultural products, were purchased at the department stores and discount stores in Daejeon. And we determined the microbiological and parasitological contamination level of these agricultural products using culture media, multiplex PCR, commercial bacterial detection kit and microscopy, and also evaluated the decontamination method. Mean counts of total aerobic bacteria from these agricultural products ranged from $1.3{\times}10^4$ CFU/g to $1.8{\times}10^5$ CFU/g, and mean counts of coliforms ranged from $1.4{\times}10^3$ CFU/g to $9.6{\times}10^3$ CFU/g. There was no significant difference in the level of bacterial contamination between the agricultural products from department stores and the ones from discount stores. Strawberry showed the highest contamination level for the bacteria and we also found the unidentified parasite eggs. Enterobacter cloacae was the most frequently isolated bacteria strain, but no food poisoning pathogenic bacteria except Staphylococcus aureus was isolated from the products by multiplex PCR. Compared to unwashed products, tab water-washed ones showed 80% decrease of the counts of total aerobic bacteria on the agricultural products, and the rates decreased more by incorporating detergent or ultrasonic wave treatment. We concluded that the biological contamination levels among paprikas, strawberries and tomatoes were the highest in strawberries, but there were not significant difference according to distribution systems.

Efficacy of Disinfectants and Sanitizers of Chlorine Oxide Bubbling Tablets (기포 발생형 타정제의 살균세정 효과)

  • By using effective sanitizers, early block for transmission of pathogens is the effective way to prevent epidermic outbreaks. Here we developed a chlorine oxide bubbling type of sanitizing tablets and evaluated the disinfectant and sanitization effects. The sanitizers showed 99.999% of sanitization effect for Escherichia coli ATCC 10536 and Staphylococcus aureus ATCC 6538 strains for 5 min±10 sec on 20±1℃ in clean condition by dilution-neutralization method. It reduced more than 5 log10 cfu/ml of a legal permission standard of colony reduction. When a few used socks and underwear soaked for one hour in the 0.1% of the sanitizing tablets, no microorganisms were grown on CHROMagar plates. However, on CHROMagar plates of the no sanitizing tablets treated control, about 6.5×104 cfu/ml of Staphylococcus sp., Klebsiella sp. and Enterococus sp. were grown. Furthermore, the sanitizing tablets killed approximately 1.5x108 cfu/ml of E. coli BL21 in 5 minutes. Therefore, we concluded that the chlorine based bubbling type of sanitizing tablets satisfied the legal standard for the regulation of food and drug safety for disinfectants and sanitizers to pathogens and daily supplies.
