Antifungal Susceptibility Testing for the Dermatophytes Isolated from Dogs and Cats (개와 고양이에서 분리한 피부사상균에 대한 항진균제의 감수성시험)

  • The aim of this study was to determine optimal therapeutic dose of antifungal agents for dermatophytes. Forty nine dermatophytes were isolated form dogs and cats showing skin lesions and were tested for their in vitro susceptibility to nystatin, griseofulvin, terbinafine, ketoconazole and fluconazole by broth microdilution test. Terbinafine showed the lowest MIC value among the antifungal agents tested, and MIC values ranged from 0.002 to 0.016 $\mu\textrm{g}$/ml. Fluconazole showed the highest MIC values among the antifungal agents tested, and MIC values ranged from 32 to 512 $\mu\textrm{g}$/ml. MIC values of nystatin, griseofulvin and ketoconazole ranged from 1.12 to 4.48 $\mu\textrm{g}$/ml, 0.5 to 4.0 $\mu\textrm{g}$/ml, and 1.6 to 12.8 $\mu\textrm{g}$/ml, resistant to griseofulvin, and that antifungal susceptibility test was needed to determine optimal therapeutic dose of antifungal agents for each dermatophyte.

  • The effects of crude extract and bioactive fractions from Brachyglottis monroi on biological activity were investigated. The crude ethanol extract inhibited the growth of the Gram positive bacterium Bacillus subtilis (ATCC Strain number 19659, 1 mm zone at 150 μg/disk) and the dermatophyte Trichophyton mentagrophytes (ATCC 28185, 2 mm zone at 150 μg/disk), and toxic to P388 tumor cells (IC50 23.96 μg/ml at 75 μg/disk). Cytotoxic activity was strongly showed by Fr. 6 (P388 IC50 19.67 μg/ml at 75 μg/disk).

  • Dermatophytes such as Trichophyton mentagrophytes, Trichophyton rubrum and Epidermophyton floccosum are used in this study to confirm (a) The Dermatophytes could utilize the wool, cotton and nylon fiber as a nutrient source. (b) The degree of damage of fibers by the Dermatophytes growth. The results of the experiment are summarized as follows; 1. Dermatophytes could not utilize the wool, cotton and nylon fiber directly as a nutrient source without the exogenously applied nutrients. 2. It was presumed that Dermatophytes could utilize the knitted wool fabric as their nutrient source when nutrient was exogenously applied. since the knitted wool fabric was greatly damaged by T. mentagrophytes and T. rubrum growth. 3. The tensile strength of knitted wool fabric was significantly decreased by T. mentagrophytes and T. rubrum, but not by E. floccosum. However, the tensile strength of knitted nylon fabric was not particularly affected by the Dermatophytes. 4. The burst strength of knitted wool fabric was decreased by T. mentagrophytes ($77\%$). T. rubrum ($53\%$). and E. floccosum ($15\%$). Though the burst strength of knitted cotton fabric was decreased by Dermatophytes about $20\%$, that of knitted nylon fabric was not affected. 5. Observing the damaged wool fiber by scanning microscope, the inner part of wool fiber was permeated by T. mentagrophytes and T. rubrum.

  • A 4-year-old female American cocker spaniel was hospitalized with 6 months prolonged dermal problems in The Veterinary Medical Teaching Hospital of Seoul National University. Initial noticed signs by owner of this dog were rashes and papules on ear part and they were spreaded whole body. On initial physical examinations, papules, pustules, crusts, and erythema were presented on whole body. Diameters of focal scaly and erythematous alopetic lesions were varied ranging 1 to 4cm. Skin scraping for detecting ectoparasites and wood's light examinations for dermatophyte were negative. Any bacteria were not detected but Malassezia pachydermatis was detected on cytological examination and fungal culture. The dog was treated with Itraconazole (5mg/kg, q12h) and Cephalexin (20mg/kg, q12h) orally and then bathed with 2% chlorhexidine shampoo twice per week for 5 weeks. From 12 weeks after starting therapy, all dermal problems were disappeared although some mild pyoderma were once relapsed. Regular chlorhexidine bathing treatment so far has been continued for preventing recurrence.

Dermatophyte and Cyclohexamide-Resistant Fungi Isolated from Patients with Tinea Capitis and from Air in Hospitals in Minia, Egypt

  • Out of 210 tinea capitis cases studies, 16 were negative when examined with KOH, among the remaining 194 cases, 123 were males (63.4%) and 71 were females (36.6%) and the age of incidence ranged between 7 and 15 years. Microsporum was the main causal agent being identified in 82 cases (42.3%) and was represented by 5 species among which M. canis was common in Egypt (55 cases, 28.4%). Trichophyton constituted 32% and was represented by 8 species among which T. violaceum was the most common (24.2% of total cases). Candida were isolated from 3.6% of total cases. The 47 species and twenty-five genera from nondermatophyte-cyclohexamide resistant fungi were recovered from the diseased skin tissue. Penicillium and Aspergillus were the most abundant followed by Scopulariopsis, Alternaria, Thermoascus, Chrysosporium and Cladosporium. Studies of the air-borne fungi in-door the hospital wards revealed the occurrence of 57 species belonging to 28 genera, among which Aspergillus flavus, Penicillium chrysogenum, p. corylophilum, A. niger, Tritirachium rosum and Alternaria alternata were the most common. Results of the out-door experiments were basically similar to those of the in-door experiments.

Antimicrobial Constituents from the Bacillus megaterium LC Isolated from Marine Sponge Haliclona oculata

  • Three compounds including 7,7-bis(3-indolyl)-p-cresol (1), cyclo-(S-Pro-R-Leu) (2) and cyclo-(S-Pro-R-Val) (3) were isolated from the strain of Bacillus megaterium LC derived from the marine sponge Haliclona oculata. All the isolated compounds showed antimicrobial activity at MIC values ranging from 0.005 to $5{\mu}g/mL$ against Gram-negative bacteria Vibrio vulnificus and V. parahaemolyticus, gram-positive bacteria Bacillus cereus and Micrococcus luteus, and the dermatophyte Trichophyton mentagrophytes. The results suggested that these compounds might have potential to be developed as agents treating dermatosis and controlling vibriosis in aquaculture.

  • The effects of crude extracts from medicinal plants on biological activity were investigated. The crude ethanol extract of H. paucistipula inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (2 mm inhibition zone at 150 ㎍/disc) and the dermatophyte Trichophyton mentagrophytes ATCC 28185, (7 mm inhibition zone at 150 ㎍/disc), and toxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC/sub 50/ 2.48 ㎍/㎖ at 75 ㎍/disc). This crude ethanol extract of H. paucistipula is the strongest antimicrobial and cytotoxic activities against P388 murine leukaemia cells ATCC CCL 46 P388D1.

  • DTM cultures were taken on 66 animals that presented with clinical signs of dermatophytosis or that had a positive Wood's lamp test. Of these cultures, 24 were Microsporum canis. Fifteen of the M canis cases were detectable by Wood's lamp. A dosage of 12.5mg/kg BID of ultramicrosize griseofulvin was effective in eliminating clinical signs in nearly all cases. This BID dosage was more effective than the same dose given once daily.

  • Pet dogs have been considered to be involved in the contamination of indoor air by serving as a source of providing molds at houses. Currently, information on the molds originated from pet dogs is rarely available in Korea. The present study was carried out to obtain basic information on the fungi present on pet dogs. For this, fungal isolation was performed to the skin and hairs of 70 pet dogs at different houses and veterinary hospitals. A total of 44 fungal isolates were obtained from skin (27 isolates) and hairs (17 isolates) of the dogs investigated. Based on the observation of microstructures and colony morphology, and the ITS rDNA sequence analysis, the fungal isolates were identified at the level of genus. The identified isolates belong to the genera of Alternaria, Aspergillus, Beauveria, Chrysosporium, Cladosporium, Penicillium, Scopulariopsis, and Trichoderma. Among these genera, Aspergillus (25%), Cladosporium (23%) and Penicillium (20.5%) were 3 major genera. 63% of the 44 isolates showed color changes on dermatophyte test medium (DTM). When we tested the growth ability of 44 isolates at $37^{\circ}C$, 45% of the isolates were able to grow. These results show that pet dogs could carry fungi having a potentiality of affecting on human health.

  • Human fungal infections are uncommon in normally healthy persons, being confined to conditions such as candidiasis (thrush) and dermatophyte skin infections such as athlete's foot. However, in the immunocompromised host, a variety of normally mild or nonpathogenic fungi can cause potentially fatal infections. Furthermore, the relativeease with which people can now visit 'exotic' countries provides the means for unusual fungal infections to be imported into this country. Mycoses appear in many different forms and areas. Fungal infections or mycoses are classified depending on the degree of tissue involvement and mode of entry into the host. These are Cutaneous, Subcutaneous, Systemic, and Opportunistic. Cutaneous mycoses specially appears symptoms on the skin. They are treated by amphotericinB, nystatin, grycelfulvin, micronazole and ketaconazole, etc, but these medicines are been careful about using, because most of them have serious side effects and toxicities. So, on the purpose of finding safe novel medicines, we have researched oriental medicines and search them to treat mycoses. In oriental medicines treating mycoses, we pay attention to orient applications that directly have an effect on disorder lesions. Oriental applications consists of various herbs and have a lot of forms, so we try to classify them as herbs and forms
