  • Objective: The present study has been undertaken to investigate the effects of Korean Carthami Semen and Estradiol-Carthami Semen co-treatment on osteoporosis in ovariectomized rats. Method: In this experiment, the rats were ovariectomized. Rats were administered 3 kinds of medicine, Korean Carthami Semen, Estradiol, and Estradiol-Carthami Semen, which were taken orally. The levels of bone mineral density, osteocalcin, estradiol, ALP, phosphorus, calcium in serum, deoxypyridinoline in urine, calcium, phosphorus, ash weight of bone, body weight and uterus weight were measured. Results: These experimetal results, it appears that the only Carthami Semen have more less efficacy on osteoporosis but $E_2$-Carthami Semen co-treated group showed more effective on osteoporosis. Conclusions: Further west-east medicine combination study should be conducted to illustrate in depth the curing osteoporosis.

청년기 여성의 골밀도와 골대사지표와의 상관관계 (Relationship of Bone Mineral Density and Biochemical Bone Markers in Young Women)

  • Purpose: This study was measured to the bone mineral density(BMD) and biochemical bone markers in young women in order to identify the relationship between bone mineral density and biochemical bone markers. Methods: Forty two healthy young women were enrolled. BMD were checked Dual Energy X-ray Absorptiometry and biochemical bone markers were checked ELSA-OSTEO(CIS bio international, France)analyzed kit, Pyrilinks-D(Metra Biosystems Inc., U.S.A)analyzed kit. Data were analyzed with frequencies, percentages, means, and Pearson correlation coefficients. Results: 1) Young women forearm(radius & ulnar) BMD was $0.55g/cm^2$, lumbar($1{\sim}4$) BMD was $0.92g/cm^2$, neck of femur BMD was $0.75g/cm^2$, trochanter of femur BMD was $0.61g/cm^2$, ward's triangle of femur BMD was $0.68g/cm^2$. In biochemical bone marker, Osteocalcin was 21.94ng/ml, Deoxypyridinoline was 11.94nmol/nmolCr. 2) There was no significant correlation between BMD and biochemical bone markers. Conclusion: Results not indicated association between bone mineral density and biochemical markers. As seen in the small sample, future research on BMD and biochemical markers need to studies to the large sample.

Nano-Calcium Ameliorates Ovariectomy-Induced Bone Loss in Female Rats

  • In this study, we examined the effects of organic types of calcium derived from oyster shell (OS-Ca) and nano-calcium (Nano-Ca) on the bio-availability and physiological responses associated with bone health in ovariectomised rats. Increased body weight, which is one of the physiological effects of ovary removal, was significantly recovered by Nano-Ca treatment (p<0.05). The reduced calcium level in the liver in ovariectomised rat was increased significantly with OS-Ca and Nano-Ca treatment (p<0.05), suggesting improved calcium bio-availability. Alkaline phosphatase (ALP), osteocalcin, and deoxypyridinoline (DPD) were analysed as biochemical markers of bone metabolism and health in the presence or absence of OSCa and Nano-Ca. ALP, osteocalcin, and DPD levels increased following ovary removal and tended to decrease after treatment with Nano-Ca, indicating that Nano-Ca induces favourable bone metabolism. This result was reflected in the recovery of bone mineral density (BMD) and bone mineral content (BMC) of the femur after Nano-Ca treatment following ovary removal. Taken together, our data show that the tested calcium treatments, especially using Nano-Ca, enhanced the bioavailability or absorption of calcium and positively affected bone metabolism in ovariectomised rats.

  • Purpose: The purpose of this study was to investigate the effects of a 12-week Tai Chi on the bone mineral density and bone metabolic markers in postmenopausal women Methods: Data were collected from March to July, 2009. Fifty postmenopausal women were recruited for the study. Twenty two women were allocated to experimental group, and 28 to control group. The experimental group underwent Tai Chi exercise twice a week for twelve weeks. The control group was only notified with results of bone mineral density and bone metabolic markers. Bone mineral density was measured by using of DTX-200 (Osteometer MediTech, Hawthorne, CA, USA) at distal radius site and bone metabolic markers were measured by radioimmunoassay method. Collected data were analyzed by t-test, $X^2$-test, and Mann-Whitney test. Results: After 12 weeks of treatment, the Tai Chi group showed a significant difference in bone mineral density compared to control group but no significant effect on osteocalcin and deoxypyridinoline level. Conclusion: Our results suggest that 12 weeks of Tai chi may delay bone loss in postmenopausal women.

  • Biochemical markers of bone turnover has received increasing attention over the past few years, because of the need for sensitive and specific tool in the clinical investigation of osteoporosis. Bone markers should be unique to bone, reflect changes of bone loss, and should be correlated with radiocalcium kinetics, histomorphometry, or changes in bone mass. The markers also should be useful in monitoring treatment efficacy. Although no bone marker has been established to meet all these criteria, currently osteocalcin and pyridinium crosslinks are the most efficient markers to assess the level of bone turnover in the menopausal and senile osteoporosis. Recently, N-terminal telopeptide (NTX), C-terminal telopeptide (CTX) and bone specific alkaline phosphatase are considered as new valid markers of bone turnover. Recent data suggest that CTX and free deoxypyridinoline could predict the subsequent risk of hiP fracture of elderly women. Treatment of postmenopausal women with estrogen, calcitonin and bisphosphonates demonstrated rapid decrease of the levels of bone markers that correlated with the long-term increase of bone mass. Factors such as circadian rhythms, diet, age, sex, bone mass and renal function affect the results of biochemical markers and should be appropriately adjusted whenever possible. Each biochemical markers of bone turnover may have its own specific advantages and limitations. Recent advances in research will provide more sensitive and specific assays.

The Effect of Lactobacillus gasseri BNR17 on Postmenopausal Symptoms in Ovariectomized Rats

  • Clinical and preclinical studies have reported that Lactobacillus gasseri BNR17, a probiotic bacterial strain isolated from human breast milk, reduces body weight and white adipose tissue volume. In order to further explore the actions of L. gasseri BNR17, we investigated the anti-menopausal effects of L. gasseri BNR17 in an ovariectomized (OVX) rat model. The serum alanine aminotransferase levels of the rats in the OVX-BNR17 group were lower than those of the rats in the OVX-vehicle only (OVX-Veh) group. Upon administration of L. gasseri BNR17 after ovariectomy, calcitonin and Serotonin 2A levels increased significantly, whereas serum osteocalcin levels showed a decreasing tendency. Compared to the rats in the OVX-Veh group, those in the OVX-BNR17 group showed lower urine deoxypyridinoline levels, lower pain sensitivity, and improved vaginal cornification. Furthermore, L. gasseri BNR17 administration increased bone mineral density in the rats with OVX-induced femoral bone loss. These results suggest that L. gasseri BNR17 administration could alleviate menopausal symptoms, indicating that this bacterium could be a good functional probiotic for managing the health of older women.

녹차가루가 당뇨 쥐의 골 대사 지표 및 골밀도에 미치는 영향 (Effects of Green Tea Powder on Bone Markers and Bone Mineral Density in STZ-Induced Diabetic Rats)

  • The purpose of this study was to examine the effects of dietary green tea powder supplementation on bone metabolism in streptozotocin-induced diabetic rats. Thirty-two male Sprague-Dawley rats (body weight $210{\pm}3g$) were divided into two groups, diabetic and non-diabetic groups. Each group was randomly divided into two subgroups which were fed with the control and 1% green tea powder diets. The serum and urine concentrations of calcium and phosphorus were determined. Serum osteocalcin and ALP and urinary DPD crosslinks value were measured in order to monitor bone formation and resorption. Bone mineral density (BMD) and bone mineral content (BMC) were estimated using PIXImus in the spine and femur. Body weight gain and FER were lower in the diabetic group than in the non-diabetic group regardless of diets. The serum concentration of calcium and phosphorus were not changed among all groups. Urinary calcium and phosphorus excretion were higher in the diabetic group than in the non-diabetic group regardless of diets; however, they were not significantly different by green tea powder intake. Serum alkaline phosphatase (ALP) was increased in the diabetic group than in thenon-diabetic group. Further, there were no significant differences in serum osteocalcin and urinary deoxypyridinoline crosslinks value among all groups. The levels of spine and femur bone mineral density of the diabetic group were significantly lower than that of the non-diabetic group. Within the diabetic group, spine BMD was significantly higher in rats fed with the green tea powder diet than in rats fed the control diet. Therefore, this study suggests that green tea powder has a beneficial effect on bone health, although it is not directly applicable to humans.

위령선 약침이 MIA 유도 골관절염에 미치는 영향 (Effects of Clematidis Radix Pharmacopuncture on MIA induced Osteoarthritis Rat)

  • Objectives : This study was designed to investigate the anti-osteoarthritic effects of clematidis radix pharmacopuncture on the MIA-induced osteoarthritis rats. Methods : The subjects were divided into 4 groups ; Normal rat group(N, n=5), MIA-induced osteoarthritis control rat group(C, n=5), MIA-induced osteoarthritis rat group injected with normal saline at ST36(NS, n=5), and MIA-induced osteoarthritis rat group injected with clematidis radix pharmacopuncture at ST36(CR, n=5). The experiment was conducted over a period of 21 days after injecting MIA. We analyzed body weight, hind paw weight distribution, liver and renal function, immunocytes, cykokines, inflammatory mediators, inflammatory proteins and mRNA expressions, as well as histopathological changes. Results : The CR group showed a significant increase in the hind paw weight distribution, and significant decreases in IL-$1{\beta}$, IL-6, $PGE_2$, $LTB_4$, osteocalcin, deoxypyridinoline level, the protein expression of COX2, arachidonate 5 lipoxygenase, and the mRNA expression of COX2, TNF-${\alpha}$, IL-$1{\beta}$, and NOS-II. In terms of the joint damages induced by osteoarthritis, the CR group showed a greater protective effect than group C in histopathologic observation (H&E, Safranin-O staining). Conclusion : These results demonstrated that clematidis radix pharmacopuncture had anti-inflammatory and analgesic effects. In addition, these results showed that it inhibited the progression of osteoarthritis.

Method : In this experiment, the rats were ovariectomized. Rats were administered by KCF. The levels of bone mineral density, osteocalcin, ALP, calcium, phosphorus in serum, calcium, phosphorus, deoxypyridinoline in urine, calcium, phosphorus, ash weight of bone, body weight and uterus weight were measured. Results : The levels of femoral and fibula-tibial bone mineral density were significantly increased in comparison with OVX group at 4, 8 weeks in KCF group. The levels of serum osteoclacin showed significant decrease in comparison with OVX group at 4, 8 weeks in KCF group. The levels of serum ALP showed significant decrease in comparison with OVX group at 4 week in KCF group. The levels of serum calcium showed significant increase in comparison with OVX group at 8 week in KCF group. The levels of urine calcium and phosphoruls showed significant decrease in comparison with OVX group in KCF group. The levels of femoral and fibula-tibial calcium didn't show significant changes in comparison with OVX group in KCF group. The levels of femoral and fibula-tibial phosphorus didn't show significant changes in comparison with OVX group in KCF group. The levels of femoral and fibula-tibial ash weight didn't show significant changes in comparison with OVX group in KCF group. The levels of body weight were significantly decreased in comparison with OVX group at 4, 8 weeks in KCF group. The levels of uterus weight were significantly increased in comparison with OVX group in KCF group. Conclusion : Reviewing these experimetal results, it appeared that KCF had efficacy on treatment of osteoporosis.

청아원가감 추출물이 난소적출로 유발된 흰쥐의 골다공증 모델이 미치는 영향 (Effect of Cheongawongagam Extract on the Ovariectomized Rat Model of Osteoporosis)

  • This study was conducted to evaluate the efficacy of Cheongawongagam on osteoporosis rat. A total of 35 rats were divided into seven groups; Normal control(SD-Nr), experimental control group(OVX-CTL), positive control group(OVX-17β-E2) and herb extracts group[Eucommia ulmoides(OVX-EU-E), Juglandis semen(OVX-JR-SE), Acanthopanax senticosus(OVX-AS-E) extract and Cheongawongagam extracts(OVX-JAEG-E)]. All control group, and herb extracts group were ovariectomized. After the 3 weeks recovery period, herb extract group were orally administered 200 mg / kg of the EU-E, JR-SE, AS-E and JAEG-E for 12 weeks. In the OVX-CTL, 17β-estradiol(E2) was administered subcutaneously on the back of the rats at a dose of 0.03 ug/sc. Their body weight, serum total cholesterol, triglyceride, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), Leukotriene B4 (LTB4), calcium (Ca), estradiol, osteocalcin, and deoxypyridinoline (DPD) concentration were measured. Also, we investigated mRNA expression of inflammatory cytokine, MMP-2, MMP-9, and bone tissue. As a result, total cholesterol was significantly decreased in the OVX-AS-E and OVX-JAEG-E. ALP was significantly increased and osteocalcin, DPD was significantly decreased in OVX-JAEG-E. The expression of inflammatory mediators (TNF-α, IL-1β, LTB4, COX-2, NOS-2), inflammatory cytokines IL-1β and MMP-9 mRNA were significantly decreased in OVX-JAEG-E. Histologic examination of the femur showed that bone mineral density, and bone mass were increased and bone marrow were decreased in the OVX-JAEG-E group. The above experiment shows that cheongawongagam extract were effective in the prevention and treatment of osteoporosis.
