Antimicrobial, Antioxidant and Cytotoxic Activities of Dendropanax morbifera Léveille extract for mouthwash and denture cleaning solution

  • PURPOSE. The purpose of this study was to analyze the antimicrobial, antioxidant activity and cytotoxicity of Dendropanax morbifera $L{\acute{e}}veille$ extract for assessing whether Dendropanax morbifera $L{\acute{e}}veille$ can be used for the development of natural mouthwash and denture cleaning solution. MATERIALS AND METHODS. The extract was obtained from branches of Dendropanax morbifera $L{\acute{e}}veille$. The solvent fractions were acquired by fractionating Dendropanax morbifera $L{\acute{e}}veille$ extract using n-hexane, ethyl acetate, chloroform and butanol solvent. Paper disc test was used to evaluate the antimicrobial and antifungal activity of Dendropanax morbifera $L{\acute{e}}veille$ extract and solvent fractions against Streptococcus mutans and Candida albicans. The analysis of antioxidant activity was carried out through DPPH radical scavenging assay. The cytotoxicity of Dendropanax morbifera $L{\acute{e}}veille$ extract was analyzed through MTT assay using normal human oral keratinocytes. RESULTS. Dendropanax morbifera $L{\acute{e}}veille$ extract showed antimicrobial activity against Streptococcus mutans and especially Candida albicans. The solvent fractions of Dendropanax morbifera $L{\acute{e}}veille$ showed strong antimicrobial activity against Streptococcus mutans and Candida albicans in n-hexane and butanol solvent fraction, respectively. Dendropanax morbifera $L{\acute{e}}veille$ extract also showed outstanding antioxidant activity. Butanol, ethyl acetate, and chloroform solvent fraction of Dendropanax morbifera $L{\acute{e}}veille$ tended to have increased antioxidant activity as the concentration increased. Dendropanax morbifera $L{\acute{e}}veille$ extract showed high cell survival rate in cytotoxicity test. CONCLUSION. Dendropanax morbifera $L{\acute{e}}veille$ extract turned out to have antimicrobial, antioxidant activity and cytophilicity. Based on these results, it is expected that Dendropanax morbifera $L{\acute{e}}veille$ is applicable as an ingredient for natural mouthwash and denture cleanser.

Phylogenetic Analysis of Dendropanax morbifera Using Nuclear Ribosomal DNA Internal Transcribed Spacer (ITS) Region Sequences (Internal transcribed spacer (ITS) region의 염기서열 분석에 의한 보길도산 황칠나무의 분자 계통학적 연구)

  • Dendropanax morbifera is an endemic tree species of Korea, it is restricted to the southern parts of Korea. The internal transcribed spacer (ITS) region of nuclear ribosomal DNA (nrDNA) for Dendropanax morbifera grown at Bogil-do, Korea was determined. We investigated the sequence-based phylogenetic relationships of plants related and clarified its taxonomical position. The determined sequences consisted of 689 residues. ITS1 was 222 bp long while ITS2 was 233 bp long. The 5.8S rDNA was 160 bp long. The ITS region sequences of the Dendropanax species included in this study were obtained from GenBank. Oreopanax polycephalus was used as the outgroup. A pairwise alignment was calculated using the Clustal X program. A phylogenetic tree was constructed by the neighbor-joining method using the Tree view program. Sequence similarities among species including D. morbifera Bogil-do isolate showed the range 92.6 to 99.7% in sequence-based phylogenetic analysis using total 615 base pairs of ITS1, 5.8S rDNA and ITS2. D. morbifera Bogil-do isolate exhibited the highest degree of relatedness to D. chevalieri, sharing 99.7% ITS region similarity. D. morbifera Bogil-do isolate also showed to D. trifidus, sharing 99.4% ITS region similarity.

Anti-bacterial properties and safety evaluation of disinfectant using Dendropanax morbifera (Hwangchil) extract for passenger cabin in the subway (지하철 객실 적용을 위한 황칠 추출물 소독제의 항균특성 및 안전성 평가)

  • Due to the syndrome coronavirus 2 (SARS-CoV-2) pandemic, the subway passenger cabin should be continuously sterilized. However, a disinfectant such as chlorine is toxic and can lead to different issues to human health. In this paper, we introduced a novel disinfectant based on natural product (Dendropanax morbifera extract). Via ultra-high performance liquid chromatography - mass spectrometer (UHPLC-MS), different compounds from Dendropanax morbifera extract showed antivirus potentials. Antimicrobial experiments confirmed that the air-disinfectant containing Dendropanax morbifera can eliminate harmful microorganisms including Gram (-), Gram (+), and yeast within 5 mins. The as-prepared air-disinfectant also showed high antivirus activity against H1N1, HRV, and EV71. Deodorization test also indicates that the as-prepared air-disinfectant can lower the harmful gas such as ammonia and trimethylamine in the atmosphere. To evaluate the potential of air-disinfectant containing Dendropanax morbifera in practical applications, different safety tests including acute oral toxicity, acute skin irritation, and eye irritation were conducted. Results showed that the as-prepared disinfectant did not negatively affect tested animals during these safety investigations.

Comparison of Biological Activities of Dendropanax morbiferus by Different Cultivation Areas in Korea

  • Dendropanax morbifera H.Lev is an evergreen tree that lives in subtropical climates. About 75 species of D. morbifera live in East Asia, but only one species live in Korea(Korean dendropanax). D. morbifera belonging to Aralicacae family 98% of D. morbifera are distributed in Jeollanam-do in the Korea and the grow wild in Gyeongsangnam-do and Jeju Island. The harvest time and usage of D. morbifera were recorded in traditional medicinal books. The roots and stems of D. morbifera had been used for traditional medicine to treat migraine, menstrual irregularity and skin disease. And D. morbifera leaves are contain flavonoids and polyacetylene compounds. In this study, we were investigated the physiological activity of D. morbifera by different areas collected at the same time, and compared to characteristics of plants. D. morbifera collected from Jeollanam-do (Goheung-gun), Gyeongsangnam-do (Namhae-gun) and Jeju Island, and dried at $50^{\circ}C$ for three days. We used dried D. morbifera powder for antioxidant tests. Each sample was extracted with hot water under the same conditions. The contents of total polyphenols and total flavonoids from D. morbifera were identified. Also, we performed to DPPH radical scavenging activity, ABTS cation radical scavenging activity and Superoxide anion scavenging activity efficacy for antioxidant activity determination. The contents of total polyphenols and total flavonoids in hot water extract of D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do were higher than Jeju. However, D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do showed no significant difference those content of total polyphenols and total flavonoids. And the antioxidant capacity was showed the similar patterns in antioxidant activity.

Effect of Dendropanax morbifera extract Addition on Chungkukjang on Immune Response of Mice (황칠 추출물을 첨가한 청국장이 마우스의 면역기능에 미치는 효과)

  • This study is to investigate increase immunomodulatory activity of Chungkukjang added with Dendropanax morbifera extract. There are four groups divided that Control, Dendropanax morbifera extract group, Chungkookjang group and Chungkukjang added to Dendropanax morbifera extract group which of five mouses each group. Their immunological characteristics were compared with either general Chungkukjang or non treated group in the tested male ICR mice. There were no significant difference in body weight and organ weight. In the plaque forming cell assay, Dendropanax morbifera extract 250 and 500 mg/kg groups measured significant increase over the control group. The values of lymphocytes were significantly increased in the Dendropanax morbifera extract 500 mg/kg group campared with the control group. In the general Chungkukjang 400 mg/kg added with Dendropanax morbifera extract 500 mg/kg group, total serum immunoglobulin G concentration was significantly higher than the control group and their spleen tissues observed proliferation of white puls. These results demonstrated that Chungkukjang Added with Dendropanax morbifera extract was provided enhance of immunomodulatory activity and suggest that it can be used as various functional foods, based on foods promote immune system health.

Changes in Photosynthetic Performance and Water Relation Parameters in the Seedlings of Korean Dendropanax Subjected to Drought Stress (건조 스트레스에 따른 황칠나무 유묘의 광합성과 수분특성인자 변화)

  • Background: This study aimed to investigate out the influence of drought stress on the physiological responses of Dendropanax morbifera seedlings. Methods and Results: Drought stress was induced by discontinuing water supply for 30 days. Under drought stress, photosynthetic activity was significantly reduced with decreasing soil water content (SWC), as revealed by the parameters such as Fv/Fm, maximum photosynthetic rate ($P_{N\;max}$), stomatal conductance ($g_s$), stomatal transpiration rate (E), and intercellular $CO_2$ concentration (Ci). However, water use efficiency (WUE) was increased by 2.5 times because of the decrease in $g_s$ to reduce transpiration. Particularly, E and $g_s$ were remarkably decreased when water was withheld for 21 days at 6.2% of SWC. Dendropanax morbifera leaves showed osmotic adjustment of -0.30 MPa at full turgor and -0.13 MPa at zero turgor. In contrast, the maximum bulk modulus of elasticity ($E_{max}$) did not change significantly. Thus, Dendropanax morbifera seedlings could tolerate drought stress via osmotic adjustment. Conclusions: Drought avoidance mechanisms of D. morbifera involve reduction in water loss from plants, through the control of stomatal transpiration, and reduction in cellular osmotic potential. Notably photosynthetic activity was remarkably reduced, to approximately 6% of the SWC.

Development of Kiln-Drying Schedules for Neolitsea sericea and Dendropanax morbifera Grown in Jeju Island (제주산 참식나무와 황칠나무의 건조스케줄 개발)

  • This research is to develop kiln-drying schedules for 2.7 mm thick boards of Neolitsea sericea and Dendropanax morbifera, which grow in the southern region of Korean Peninsula. These two species are lesser-known to woodcraft industry in this country. The proper USDA drying schedules for Neolitsea sericea and Dendropanax morbifera were found to be T10-C6 and T10-E6, respectively, by using the Quick Oven-drying Method. The boards dried in a kiln with the newly-developed schedules were revealed to be free from warping and checking.

First Report of Foliar Blight on Dendropanax morbifera Caused by Alternaria panax

  • Leaf spot and blight disease was observed on two-year-old seedlings of Dendropanax morbifera (Korean name: Hwangchil tree) during July of 2008 in Jindo Island, Korea. Symptoms included yellow-brown to dark brown irregularly enlarged spots frequently located along the veins of leaves. The lesions were often surrounded by chlorotic haloes. Severe leaf blight and subsequent defoliation occurred when conditions favored disease outbreak. The causal organism of the disease was identified as Alternaria panax based on morphological characteristics and sequence analysis of the internal transcribed spacer region of rDNA. A. panax isolates induced leaf spots and blight symptoms not only on D. morbifera but also on the other members of Araliaceae tested. This is the first report of foliar blight caused by A. panax on D. morbifera.

Antioxidant and Hepatoprotective Effects of the Ethanol Extract of Dendropanax morbifera Leveille on the t-Butyl Hydroperoxide-Induced HepG2 Cell Damages (황칠나무 추출물의 항산화 및 간세포보호효과)

  • Dendropanax morbifera Leveille, an endemic species in Korea, is best known as a tree that produces a resinous sap. Although D. morbifera is used in folk medicine for various diseases, its active ingredients are largely unknown. In this study, we investigated antioxidative activities of ethanolic extracts of three parts of this plant including leaves, debarked stems, and roots. The root extracts exhibited strong 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity compared with leaf or stem extracts. The root extracts showed hepatoprotective activity against t-butyl hydroperoxide-induced HepG2 cells, and reduced the ROS level in the cells. The root fractions lowered the mRNA level of COX-2 on lipopolysaccharide-stimulated Raw246.7 cells. These results suggest that ethanolic root extracts of D. morbifera are a source of antioxidant and hepatoprotective compounds, which indicate a potential for a botanical drug.

Effect of Dendropanax morbifera Pharmacopuncture on Sleep-related Hormones, Cognition in Psychosocial Stress-induced Rats (황칠(黃漆) 약침이 정신 스트레스를 받은 백서의 수면 관련 호르몬, 인지에 미치는 영향)

  • Objectives: The purpose of this study was to check the therapeutic effect of the Dendropanax morbifera Pharmacopuncture for insomnia and cognition caused by psychosocial stress. Methods: We separated Wistar rats (older than age 10 weeks, 300 g) into normal, control, positive control (Zolpidem administered, PC), and Dendropanax morbifera Pharmacopuncture experimental groups (DPA). Psychosocial stress such as circadian rhythm change, clammy straw, predator stress, restraint stress, noise bursts, and flashing lights were applied to the control, PC, DPA groups. The Dendropanax morbifera Pharmacopuncture group was given 0.5×, 1× and 2× amount concentration of Dendropanax morbifera Pharmacopuncture, respectively. The levels of Melatonin, Serotonin, Corticosterone, GABA, BDNF and CREB were measured, and the Y-maze test, weight and blood test were performed. Results: In all of the DPA groups, the Melatonin level showed no noticeable difference. In the DPA-2x group, Serotonin increased significantly. In all the experimental groups, Corticosterone decreased significantly and GABA showed increasing tendency. The DPA-1x and DPA-2x groups showed remarkable increase in BDNF and DPA-0.5x and the DPA-2x groups showed significant increase in CREB. The DPA-2x group showed remarkable increase in the Alternation behavior category of the Y-maze test. In all of the experimental groups the weight change showed increasing tendency, whereas no noticeable differences were found among all experimental groups regarding AST, ALT, BUN, Creatinine and CBC. Conclusions: Dendropanax morbifera Pharmacopuncture increases levels of serotonin and GABA, decreases corticosterone, increases levels of BDNF and CREB, and increases the ratio of alternation behavior in the Y-maze test. Thus, I suggest that Dendropanax morbifera Pharmacopuncture has the effect of treating insomnia caused by psychosocial stress, activating the brain, and improving cognition.
